RESUMEN

In this work the impact of an innovative protein stabilization method (TiO2-based composite sorbent material coupled with a prototype device operating under continuous flow) has been tested in terms of protein and oxidative stability of white wines. Optimal process parameters (duration 60 min; flow rate 1.5 ± 0.1 L/h in 6 cycle rates/h) ensured an average 32.5 % reduction of total proteins; the nanoporous TiO2 film supported on inert glass beads acted as selective sorbent for pathogenesis-related proteins (PRPs, 10-60 kDa) responsible for wine instability, based on the protein stability studies (heat-test) performed in the experimental wines. The stabilization process has been tested for the release of contaminants (Ti), and the innovative treatment has been proven to preserve wine from oxidation also delaying the browning onset under extreme storage conditions.

RESUMEN

In the present work, a nondestructive device set up for a rapid and reliable freshness assessment of rainbow trout during 10 days of storage in ice was evaluated. The device was characterized by a vector network analyzer interfaced with an open coaxial probe to be placed in contact with the fish eye. The acquisition of the reflected scattering parameter (S11), which is the ratio between the amplitude of the reflected and the incident signal, was assessed in the 50 kHz-3 GHz spectral range. S11 is composed of a real part and an imaginary part, and both parts were used to predict quality index method for freshness evaluations. Partial least squares regression predictive models of the demerit scores related to fish eye attributes (eye pupil and eye shape) and the day of storage were set up. The main results showed that both the real and imaginary parts of the S11 decrease as a function of storage time. The combination with multivariate analysis allowed to set up predictive models of the storage time and the demerit scores with R2 values up to 0.946 (root mean square error [RMSE] = 0.88 days) and 0.942 (RMSE = 3.17 demerit scores related to the fish eyes attributes), respectively (external validation). According to our results, the proposed cheap solution appears a useful tool for the freshness assessment of rainbow trout. PRACTICAL APPLICATION: This work shows that dielectric properties have the potential to discriminate stored fish according to their freshness quality. A device based on this principle can play a significant role in the postharvest processes, contributing to higher product quality and safety and supporting producers and retailers during the qualitative inspections.

RESUMEN

The simultaneous use of UVA light irradiation coupled with low energy cold plasma generated by a dielectric barrier discharge prototype, results in significant enhancement of efficiency of the integrated process with respect to the sole plasma treatment. This effect has been demonstrated both on microbial inactivation of a food-borne pathogen, i.e. Listeria monocytogenes, and on the degradation of a compound of biological origin such as phenylalanine. In the latter case, the analysis of its reaction intermediates and the spectroscopic identification and quantification of peroxynitrites, allowed to propose mechanistic hypotheses on the nature of the observed synergistic effects. Moreover, it has been demonstrated that the process does not affect the quality of trout fillets, indicating its suitability as a chlorine-free, green, and sustainable tool for the decontamination of fish processing water.

A cheap technique based on an open-ended coaxial probe together with a vector network analyzer was set up. The vector network analyzer NanoVNA, a very tiny handheld device, is the affordable component that gives the instrumental chain a cost-effective perspective. The open-ended coaxial probe is a cable with an SMA gold-plated termination. User-friendly programs can be used to calibrate the instrument, carry out the measurements, and save data on PC. Simple liquid solutions (sodium chloride, citric acids, and saccharose) and more complex liquid food (milk, egg products, and fruit juice) were investigated. In addition, the temperature on the electric measurement of milk was measured to evaluate a possible influence for refrigerated storage products. The reflection parameters, such as the real and imaginary parts of S11, were used to build univariate and multivariate models. The best results in terms of coefficient of determination and related error were 0.997 (RMSE 0.05%) for sodium chloride and 0.965 (RMSE 0.71 °Brix) for fruit juice considering the univariate model, and 0.997 (RMSE 0.04%) for sodium chloride and 0.981 (RMSE 4.44%) for yolk using multivariate analysis. The proposed solution is non-destructive, cheap, rapid, and very attractive for potential lab and industrial applications.

The extraction of molecules with high added value plays an important role in the recovery of food waste. This work aimed to valorize tomato pomace, a by-product composed of skin and seeds, through extraction of carotenoids, especially lycopene and ß-carotene. The tomato pomace was dried using three different methods (freeze-drying, heat drying, and non-thermal air-drying) to reduce its weight, volume, and water activity and to concentrate the carotenoid fraction. These drying approaches were compared considering the extractive potential. Three solvent mixtures were compared, a traditional one (n-hexane:acetone) and two green deep eutectic solvent mixtures (ethyl acetate:ethyl lactate and menthol:lactic acid) in combination with different drying procedures. The extract obtained using ethyl acetate:ethyl lactate with non-thermal air-drying showed the highest contents of lycopene and ß-carotene (75.86 and 3950.08 µg/g of dried sample, respectively) compared with the other procedures.

Non-thermal atmospheric plasma (NTAP) has gained attention as a decontamination and shelf-life extension technology. In this study its effect on psychrotrophic histamine-producing bacteria (HPB) and histamine formation in fish stored at 0-5°C was evaluated. Mackerel filets were artificially inoculated with Morganella psychrotolerans and Photobacterium phosphoreum and exposed to NTAP to evaluate its effect on their viability and the histidine decarboxylase (HDC) activity in broth cultures and the accumulation of histamine in fish samples, stored on melting ice or at fridge temperature (5°C). NTAP treatment was made under wet conditions for 30 min, using a dielectric barrier discharge (DBD) reactor. The voltage output was characterized by a peak-to-peak value of 13.8 kV (fundamental frequency around 12.7 KHz). This treatment resulted in a significant reduction of the number of M. psychrotolerans and P. phosphoreum (≈3 log cfu/cm2) on skin samples that have been prewashed with surfactant (SDS) or SDS and lactic acid. A marked reduction of their histamine-producing potential was also observed in HDC broth incubated at either 20 or 5°C. Lower accumulation of histamine was observed in NTAP-treated mackerel filets that have been inoculated with M. psychrotolerans or P. phosphoreum and pre-washed with either normal saline or SDS solution (0.05% w/v) and stored at 5°C for 10 days. Mean histamine level in treated and control groups for the samples inoculated with either M. psychrotolerans or P. phosphoreum (≈5 log cfu/g) varied from 7 to 32 and from 49 to 66 µg/g, respectively. No synergistic effect of SDS was observed in the challenge test on meat samples. Any detectable amount of histamine was produced in the meat samples held at melting ice temperature (0-2°C) for 7 days. The effects of NTAP on the quality properties of mackerel's filets were negligible, whereas its effect on the psychrotrophic HPB might be useful when time and environmental conditions are challenging for the cool-keeping capacity throughout the transport/storage period.

RESUMEN

Standard analytical methods for fish freshness assessment are based on the measurement of chemical and physical attributes related to fish appearance, color, meat elasticity or texture, odor, and taste. These methods have plenty of disadvantages, such as being destructive, expensive, and time consuming. All these techniques require highly skilled operators. In the last decade, rapid advances in the development of novel techniques for evaluating food quality attributes have led to the development of non-invasive and non-destructive instrumental techniques, such as biosensors, e-sensors, and spectroscopic methods. The available scientific reports demonstrate that all these new techniques provide a great deal of information with only one test, making them suitable for on-line and/or at-line process control. Moreover, these techniques often require little or no sample preparation and allow sample destruction to be avoided.

Dielectric barrier discharge plasma and photocatalysis have been proposed as tools for decontamination of process water, especially in food industry. The present investigation aims to redefine and identify the features of coupling the two technologies in terms of degradation efficiency of a model compound. Results show that, when the process is carried out in plasma activated water in the presence of irradiated TiO2, the efficiency of the integrated process is lower than the sum of the two processes acting separately. It is proposed that afterglow species, e.g. hydrogen peroxide and/or peroxynitrites could be activated by UVA light irradiation producing hydroxyl radicals in the liquid phase. Even if TiO2 limits this additional effect by acting as UVA screen barrier material, its decontamination efficiency under certain conditions results higher than that obtained with plasma systems. These results open the route to chlorine-free decontamination processes and redefine the application framework of this integrated approach.

This paper will show the electronic architecture of a portable and non-invasive soil moisture system based on an open rectangular waveguide. The spectral information, measured in the range of 1.5-2.7 GHz, is elaborated on by an embedded predictive model, based on a partial least squares (PLS) regression tool, for the estimation of the soil moisture (%) in a real environment. The proposed system is composed of a waveguide, containing Tx and Rx antennas, and an electronic circuit driven by a microcontroller (MCU). It will be shown how the system provides a useful and fast estimation of moisture on a silty clay loam soil characterized by a moisture range of about 9% to 32% and a soil temperature ranging from about 8 °C and 18 °C. Using the PLS approach, the moisture content can be predicted with an R2 value of 0.892, a root mean square error (RMSE) of 1.0%, and a residual prediction deviation (RPD) of 4.3. The results prove that it is possible to make accurate and rapid moisture assessments without the use of invasive electrodes, as currently employed by state-of-the-art approaches.

Solid and liquid components coexist into glassy and amorphous structures of food complex matrixes. Both states admit movements, promoting physical modifications to a more thermodynamically stable system. Green and roasted coffee beans are principally characterized by a glassy structure that slowly evolves during storage. The aim of this study was to assess calorimetric and dielectric properties in combination, as a useful multi-analytical technique to improve the understanding of the motion mechanism of localized molecules. After equilibration at different water activities (aw) for the determination of sorption isotherms of green and roasted coffee, the glass transition temperature (Tg) of the samples has been measured by using differential scanning calorimetry (DSC). Increasing the aw from 0.155 to 0.512, the Tg shifted from 48.76 (±0.04) to 34.89 (±0.02) °C for green coffee and from 45.73 (±0.05) to 40.15 (±0.10) °C for the roasted one. The spectroscopic fingerprint of the matrix has been determined by dielectric measurements in terms of "gain" spectra (related to the imaginary part of permittivity). The maximum values of the determination coefficient (R2), obtained by linear correlation between spectral data and water activity or glass transition values for a specific frequency of the whole range (1.6 GHz-2.7 GHz), were 0.999 and 0.943 for green, and 0.997 R2 and 0.925 R2 for roasted coffee respectively.

Pulsed electric field (PEF) treatment is a non-thermal technology that has shown good potential for microbial inactivation. However, in many cases, it cannot be sufficient to avoid microbial proliferation, and the combination with other stabilizing technologies is needed. In the framework of the hurdle concept, several researches have been focused on the use of PEF in combination with heat and/or antimicrobials to increase its efficacy. This study investigated the inactivation effect of PEF on a strain of Saccharomyces cerevisiae (isolated from spoiled beverages) in a model system (growth medium). The efficacy of PEF treatment was evaluated in relation to different variables, such as electric field strength (25 and 50 kV/cm), treatment time (from 1 to 5 s), initial inoculum level (4 or 6 log cfu/ml), preheating at 50°C, medium pH (4 or 6), and addition of citral at sublethal concentration (i.e., half of minimum inhibiting concentration). The data from plate counting, modeled with the Weibull equation, showed that one of the main factors affecting yeast inactivation was the preheating of the suspension at 50°C. Indeed, higher cell load reductions were obtained with heat-assisted PEF, especially in the presence of citral. The effect of initial cell load was negligible, while pH affected yeast inactivation only without preheating, with higher death kinetics at pH 6. Flow cytometry (FCM) analysis confirmed higher mortality under these conditions. However, the occurrence of injured cells, especially in samples treated at pH 4, was observed. The ability of these cells to recover from the damages induced by treatments was affected by both citral and preheating. The synergic effects of PEF, preheating, and citral were likely due to the increase of membrane permeability (especially at pH 6), as the primary target of electroporation, which favored the solubilization of citral in the cell membrane, enhancing the efficacy of the whole process. The multi-analytical approach (traditional plate counting and FCM) allowed defining parameters to increase PEF efficacy against S. cerevisiae. Moreover, FCM, able to discriminate different physiological states of the yeast population, was helpful to better clarify the action mechanism and the potential recovery of cells after treatment.

This work studied water state of honey during crystallization, obtained statically and dynamically, by differential scanning calorimetry (DSC), water activity (aw) assessment and time domain nuclear magnetic resonance (TD-NMR). Crystallization was induced by adding 5% of crystallized honey to three honey samples with different fructose/glucose ratio, the key characteristic for honey crystallization. Samples were stored at 14 °C. Dynamic crystallization was obtained by using an impeller. DSC showed that the dynamic crystallization was faster than the static one, the latter characterized by two phases, showing different rates. The crystallization rate did not affect aw, that remained below 0.600. TD-NMR allowed to separately observe two kinds of protons, both pertaining to liquid sugars, one chemically exchanging with water and one not exchanging with it. The combination of techniques allowed speculating that the two crystallization methods led to crystals of different size and shape.

The increasing demand for high quality and safe food led to important technological innovations in food processing. Cold plasma appears as an emerging technology that has demonstrated efficiency in the removal of microbial contamination from fresh and minimally processed food. In this study, the proteomic profile of Salmonella Enteritidis SE86 subjected to cold plasma treatment was investigated. The number of viable S. Enteritidis SE86 cells was analyzed at different time intervals upon exposure to cold plasma and approximately 100 µg of S. Enteritidis SE86 protein extracts were analyzed by Multidimensional Protein Identification Technology (MudPIT). The results demonstrated that no significant changes in cell counts were detected for up to 20 min exposure to cold plasma, and 2 log reduction was achieved after 60 min. Overall, 1096 proteins were identified, with 249 detected only in plasma-treated samples, and 9 exclusive in non-treated control samples. The proteins uniquely detected in cold plasma-treated cells that showed higher abundance were glyoxalase I, ABC transporter substrate-binding protein and transcriptional activator OsmE, followed by some oxidoreductases. Proteins related with carbohydrate and nucleotide metabolism were mostly overexpressed in cold plasma treated cells, suggesting energy metabolism was increased.

Osmotic dehydration (OD) is a widely used preservation technique that consists in the reduction in food water activity by the immersion of the biological tissue in hypertonic solutions. The aim of this work was to analyze the effect of pulsed electric fields (PEF) in mass transfer as a pre-treatment of the OD using NMR. In this sense, PEF pre-treatments were done using three different voltages (100, 250 and 400V/cm) and 60 number of pulse. The OD of kiwifruit was carried out in 61.5% of sucrose solution at 25°C, for a contact period from 0 to 120min. The water distribution into the cellular tissue was studied by NMR relaxometry. In conclusion, NMR is an excellent technique for quantifying water molecules according to their interactions in the fruit tissue, obtaining the adsorbed water and opening the possibility to apply the BET model to fit the adsorbed isotherm over the whole range of water activity.

The main aim of the present research is to assess the possibility of quickly screening fatty acid alkyl esters (FAAE) in olive oils using time domain reflectometry (TDR) and partial least-squares (PLS) multivariate statistical analysis. Eighteen virgin olive oil samples with fatty acid alkyl ester contents and fatty acid ethyl ester/methyl ester ratios (FAEE/FAME) ranging from 3 to 100 mg kg(-1) and from 0.3 to 2.6, respectively, were submitted to tests with time domain resolution of 1 ps. The results obtained in test set validation demonstrated that this new and fast analytical approach is able to predict FAME, FAEE, and FAME + FAEE contents with R(2) values of 0.905, 0.923, and 0.927, respectively. Further measurements on mixtures between olive oil and FAAE standards confirmed that the prediction is based on a direct influence of fatty acid alkyl esters on the TDR signal. The suggested technique appeared potentially suitable for monitoring one of the most important quality attribute of the olive oil in the extraction process.

UNLABELLED: The nondestructive assessment of apricot fruit quality (Bora cultivar) was carried out by means of FT-NIR reflectance spectroscopy in the wavenumber range 12000 to 4000 cm⁻¹. Samples were harvested at four different ripening stages and scanned by a fiber optical probe immediately after harvesting and after a storage of 3 d (2 d at 4 °C and 1 d at 18 °C); the flesh firmness (FF), the soluble solids content (SSC), the acidity (A), and the titratable acidity (malic and citric acids) were then measured by destructive methods. Soft independent modeling of class analogy (SIMCA) analysis was used to classify spectra according to the ripening stage and the storage: partial least squares regression (PLS) models to predict FF, SSC, A, and the titratable acidity were also set-up for both just harvested and stored apricots. Spectral pretreatments and wavenumber selections were conducted on the basis of explorative principal component analysis (PCA). Apricot spectra were correctly classified in the right class with a mean classification rate of 87% (range: 80% to 100%). Test set validations of PLS models showed R2 values up to 0.620, 0.863, 0.842, and 0.369 for FF, SSC, A, and the titratable acidity, respectively. The best models were obtained for the SSC and A and are suitable for rough screening; a lower power prediction emerged for the other maturity indices and the relative predictive models are not recommended. PRACTICAL APPLICATION: The results of the study could be used as a tool for the assessment of the ripening stage during the harvest and the quality during the postharvest storage of apricot fruits.

The time history of subsequent tunneled wave packets can represent a more meaningful way to describe signal evolutions and to determine the group delay than the detection of a demodulated monopulse envelope. In the present experimental research, the delay of transmitted tunneled packets was found to be shorter than that measurable in vacuum, in good agreement with the phase time and Esposito's equation and independent of the barrier length. The wave packets did not show narrowing and "reshaping," so these phenomena did not appear to be the reason of the short delay experienced. The latter was not found to be proportional to the amplitude of the incident signal.

A granular cell tumor (GCT) in a 39-year-old white man is reported. It was localized in the intrapancreatic part of the common bile duct and caused obstruction of the bile downflow. The patient underwent radical surgical procedures because a malignant tumor was clinically suspected. Macroscopically, the tumor appeared as a duct stricture caused by diffuse infiltration of neoplastic cells in the walls. In the cytoplasm smaller and larger PAS-positive granules were present and constantly reactive to S-100 and NSE antibodies. Ultrastructurally, cytoplasmic granules appeared as membrane-bound vacuoles of variable size and shape containing debris, disrupted mitochondria, and myelin figures. No basal lamina around cell cytoplasm was observed. GCTs are relatively uncommon soft tissue tumors usually presenting in the skin and subcutaneous tissues or tongue. The prognosis in any location is quite good, but very rare malignant GCTs (1-2%) are documented. Complete excision reduces the risk of recurrence. Accurate operative diagnosis seems to be critical when the tumors are located in the intrapancreatic common bile duct as in this reported case. Gastro-pancreatico-duodenectomy is too radical a procedure for such a benign lesion and additional assessments and investigations are recommanded before such an extensive radical surgery.
