RESUMEN

Pyroelectric nanostructures could effectively generate temperature-mediated reactive oxygen species (ROS) through the pyroelectric effect, providing promise for treating hypoxic tumors, and therefore, the synergistic application of photothermal therapy (PTT) and pyroelectric dynamic therapy (PEDT) presents an intriguing approach for cancer therapy. However, this method still faces challenges in improving pyroelectric catalysis and achieving precise tumor localization. In this study, a nano-heterojunction based on CeO2-BaTiO3 nanorods (IR1061@PCBNR) is reported, which exhibits highly effective pyroelectric catalysis for simultaneous tumor-targeted dynamic therapy and gentle photothermal therapy through the utilization of the rich oxygen vacancies. The oxygen vacancies create active sites that facilitate the migration of pyroelectrically-induced charge carriers, improving charge separation and ROS generation. IR1061@PCBNR also demonstrates high tumor penetration while minimizing damage to normal cells. This precise nanomedicine strategy holds great potential for advancing dynamic cancer therapies by overcoming the limitations of conventional approaches. This article is protected by copyright. This article addresses the finite-time neural predefined performance control (PPC) issue for state-constrained nonlinear systems (NSs) with exogenous disturbances. By integrating the predefined-time performance function (PTPF) and the conventional barrier Lyapunov function (BLF), a new set of time-varying BLFs is designed to constrain the error variables. This establishes conditions for satisfying full-state constraints while ensuring that the tracking error meets the predefined performance indicators (PPIs) within a predefined time. Additionally, the incorporation of the nonlinear disturbance observer technique (NDOT) in the control design significantly enhances the ability of the system to reject disturbances and improves overall robustness. Leveraging recursive design based on dynamic surface control (DSC), a finite-time neural adaptive PPC strategy is devised to ensure that the closed-loop system is semi-globally practically finite-time stable (SPFS) and achieves the desired PPIs. Finally, the simulation results of two practical examples validate the efficacy and viability of the proposed approach.

Solar-driven water purification has been deemed a promising technology to address the issue of clean water scarcity. However, traditional solar distillers often suffer from low evaporation rates under natural sunlight irradiation, while the high costs of the fabrication of photothermal materials further hinders their practical applications. Here, through the harnessing of the complexation process of oppositely charged polyelectrolyte solutions, a polyion complex hydrogel/coal powder composite (HCC)-based highly efficient solar distiller is reported. In particular, the influence of the charge ratio of polyanion-to-polycation on the solar vapor generation performance of HCC has been systematically investigated. Together with a scanning electron microscope (SEM) and the Raman spectrum method, it is found that a deviation from the charge balance point not only alters the microporous structure of HCC and weakens its water transporting capabilities, but also leads to a decreased content of activated water molecules and enlarges the energy barrier of water evaporation. As a result, HCC prepared at the charge balance point exhibits the highest evaporation rate of 3.12 kg m-2 h-1 under one sun irradiation, with a solar-vapor conversion efficiency as high as 88.83%. HCC also exhibits remarkable solar vapor generation (SVG) performance for the purification of various water bodies. In simulated seawater (3.5 wt% NaCl solutions), the evaporation rate can be as high as 3.22 kg m-2 h-1. In acid and alkaline solutions, HCCs are capable of maintaining high evaporation rates of 2.98 and 2.85 kg m-2 h-1, respectively. It is anticipated that this study may provide insights for the design of low-cost next-generation solar evaporators, and broaden the practical applications of SVG for seawater desalination and industrial wastewater purification.

Ovarian cancer is a relatively common tumor in women with the highest mortality among female reproductive system tumors. The lack of apparent early symptoms and effective screening strategies often leads to ovarian cancer being diagnosed at an advanced stage. Immunotherapy relying on tumor-associated antigens might improve the treatment of ovarian cancer. Cancer-testis antigens (CTAs) are ideal tumor-associated antigens, and MAGE-A, NY-ESO-1, CT45, and Sp17 are classic CTAs highly expressed in ovarian cancer. Here, we review the research on CTAs in ovarian cancer, including prognostic value and advances in immunotherapy, all of which are essential for developing a theoretical basis for targeted therapy strategies.

Glioblastoma (GBM) is the most common malignant intracranial tumour with intrinsic infiltrative characteristics, which could lead to most patients eventually relapse. The prognosis of recurrent GBM patients remains unsatisfactory. Cancer cell infiltration and their interaction with the tumour microenvironment (TME) could promote tumour recurrence and treatment resistance. In our study, we aimed to identify potential tumour target correlated with rGBM microenvironment based on the gene expression profiles and clinical information of rGBM patients from The Cancer Genome Atlas (TCGA) database. LRRC15 gene with prognostic value was screened by univariate and multivariate analysis, and the correlation between macrophages and LRRC15 was identified as well. Furthermore, the prognosis correlation and immune characteristics of LRRC15 were validated using the Chinese Glioma Genome Atlas (CGGA) database and our clinical tissues by immunochemistry assay. Additionally, we utilized the transwell assay and carboxy fluorescein succinimidyl ester (CFSE) tracking to further confirm the effects of LRRC15 on attracting microglia/macrophages and tumour cell proliferation in the TME. Gene profiles-based rGBM microenvironment identified that LRRC15 could act in collusion with microglia/macrophages in the rGBM microenvironment to promote the poor prognosis, especially in mesenchymal subtype, indicating the strategies of targeting LRRC15 to improve macrophages-based immunosuppressive effects could be promising for rGBM treatments.

BACKGROUND: Sarcomas are rare heterogeneous tumours, derived from primitive mesenchymal stem cells, with more than 100 distinct subtypes. Radioresistance remains a major clinical challenge for sarcomas, demanding urgent for effective biomarkers of radiosensitivity. METHODS: The radiosensitive gene Kinesin family member 18B (KIF18B) was mined through bioinformatics with integrating of 15 Gene Expression Omnibus (GEO) datasets and The Cancer Genome Atlas (TCGA) database. We used radiotherapy-sh-KIF18B combination to observe the anti-tumour effect in sarcoma cells and subcutaneous or orthotopic xenograft models. The KIF18B-sensitive drug T0901317 (T09) was further mined to act as radiosensitizer using the Genomics of Drug Sensitivity in Cancer (GDSC) database. FINDINGS: KIF18B mRNA was significantly up-regulated in most of the subtypes of bone and soft tissue sarcoma. Multivariate Cox regression analysis showed that KIF18B high expression was an independent risk factor for prognosis in sarcoma patients with radiotherapy. Silencing KIF18B or using T09 significantly improved the radiosensitivity of sarcoma cells, delayed tumour growth in subcutaneous and orthotopic xenograft model, and elongated mice survival time. Furthermore, we predicted that T09 might bind to the structural region of KIF18B to exert radiosensitization. INTERPRETATION: These results indicated that sarcomas with low expression of KIF18B may benefit from radiotherapy. Moreover, the radiosensitivity of sarcomas with overexpressed KIF18B could be effectively improved by silencing KIF18B or using T09, which may provide promising strategies for radiotherapy treatment of sarcoma.
