ABSTRACT

BACKGROUND: To explore the value of cell morphology, immunophenotype, and gene alterations of serosal effusion in the diagnosis of lymphoma. METHODS: Serosal effusion of 69 cases of lymphoma patients were collected, including 36 cases with malignant effusion and 33 cases with nonmalignant effusion. Ordinary cytology, liquid-based cytology, cellblock, and immunocytochemical staining were performed in each case, some cases were detected by fluorescence in situ hybridization for C-MYC, BCL2, and BCL6 gene translocations. T/B cell ratio in malignant and nonmalignant serosal effusions was analyzed and compared by flow cytometry (FCM) and immunohistochemical (IHC), respectively. The prognostic value of serous effusion in diffuse large B-cell lymphoma (DLBCL) was investigated and another 20 DLBCL cases without effusion were successively selected as control. RESULTS: The number of naive lymphocytes, apoptotic bodies, and mitotic figures were more common in malignant effusions compared with nonmalignant effusions (p < .01). The top three lymphomas in malignant effusion were DLBCL (19/36, 52.8%), mantle cell lymphoma (MCL) (4/36, 11.1%, 3 blastoid variant) and high-grade B-cell lymphoma (HGBL) (4/36, 11.1%). T/B cell ratio by FCM analysis ranged from 0.00 to 0.55 (mean 0.084) in malignant effusion, and 2.58 to 984.00 (mean 249.9) in nonmalignant effusion. The difference was significant (p = .017). The T/B cell ratio by IHC analysis ranged from 0.02 to 3.00 (mean 0.200) in malignant effusion, and 2.00-100.00 (mean 34.10) in nonmalignant effusion. The difference was significant (p = .017). In the effusions involving DLBCL, most effusions were present at the time of diagnosis (57.9%); single pleural effusions were more common (36.8%). The median overall survival times of patients with malignant effusion, nonmalignant effusion and DLBCL without serous effusion were 11, 17, and 23 months respectively (p = .04). Three patients of HGBL died, and the overall survival times were 5, 8, and 9 months, respectively. CONCLUSIONS: The cytomorphological characteristics combined with immunophenotype, FCM, gene rearrangement, and other tests can diagnose and classify patients with effusion as the first symptom. The T/B cell ratio is less than 1 by FCM or IHC suggesting a malignant serosal effusion. The presence of malignant effusion in DLBCL patients is an important clue for poor prognosis.

ABSTRACT

ObjectiveTo investigate the mRNA expression levels of various aquaporins (AQPs) in luteinized granulosa cells from follicles of different diameters. MethodsFrom March 25, 2022 to September 23, 2022 in our reproductive medicine center, 48 women undergoing in-vitro fertilization (IVF) were enrolled and divided into the antagonist group and the agonist group according to the ovarian stimulation protocol. Follicular fluid samples were collected on the day of oocyte pick-up and granulosa cells were extracted from follicles of different diameters: small (<13 mm), medium (13~18 mm) and large (≥18 mm). After RNA quantification, 22 cases (66 samples) were included for analysis and mRNA expression levels of AQPs were compared among the three follicle groups. ResultsThe mRNA expression of aquaporin 2 (AQP2) in luteinized granulosa cells increased with the increase of follicle diameter (linear trend P = 0.004) and the difference was statistically significant between two groups of large and small follicles (P = 0.017). Statistical difference was found in the antagonist group (P = 0.049 6), but not in the agonist group (P = 0.108). ConclusionThe mRNA level of AQP2 in luteinized granulosa cells increases with the increase of follicle diameter and its expression is related to the ovarian stimulation protocol, suggesting that AQP2 may play a role in follicle growth and follicular fluid formation, and its mRNA expression level may be regulated by follicle stimulating hormone (FSH) and luteinizing hormone (LH).

ABSTRACT

OBJECTIVE@#To observe the effects of electroacupuncture on threshold of pain, gait, proliferation and differentiation of muscle satellite cell in rats with acute blunt trauma of gastrocnemius muscle, and to explore the possible mechanism of electroacupuncture in promoting the repair of acute injury of skeletal muscle.@*METHODS@#A total of 48 SD rats were randomly divided into a blank group (6 rats), a model group (24 rats) and an electroacupuncture group (18 rats). In the model group and the electroacupuncture group, the model of acute blunt trauma of gastrocnemius muscle was established by self-made impactor. In the electroacupuncture group, electroacupuncture was applied at "Chengshan" (BL 57) and "Yanglingquan" (GB 34) on the right side, with disperse-dense wave, in frequency of 2 Hz/100 Hz, once a day, 30 min each time. Electroacupuncture intervention was performed for 3, 7 and 14 days according to the sampling time. On the 1st, 3rd, 7th and 14th days after modeling, the mechanical withdrawal pain threshold of hindfoot was detected by Von Frey method; the standing time and the maximum contact area of the right hindfoot were recorded by Cat Walk XTTM animal gait analysis instrument; the morphology of the right gastrocnemius muscle and the number of inflammatory cells were observed by HE staining; the positive expression of paired box gene 7 (Pax7) and myogenic differentiation (MyoD) of the right gastrocnemius muscle was detected by immunofluorescence.@*RESULTS@#After modeling, the muscle fiber rupture and massive infiltration of red blood cells and inflammatory cells were observed in the right gastrocnemius muscle; after electroacupuncture intervention, the morphology of muscle fiber was intact and the infiltration of inflammatory cells was improved. Compared with the blank group, in the model group, the differences of mechanical withdrawal pain threshold between the left and right foot were increased (P<0.05), the standing time was shortened and the maximum contact area of the right hindfoot was decreased (P<0.05), the number of inflammatory cells and the positive expression of Pax7 and MyoD of the right gastrocnemius muscle were increased (P<0.05) on the 1st, 3rd, 7th and 14th days after modeling. Compared with the model group, in the electroacupuncture group, the differences of mechanical withdrawal pain threshold were decreased (P<0.05), the standing time was prolonged (P<0.05), the number of inflammatory cells of right gastrocnemius muscle was decreased (P<0.05) on the 7th and 14th days after modeling; the maximum contact area of the right hindfoot was increased (P<0.05), the positive expression of MyoD of the right gastrocnemius muscle was increased (P<0.05) on the 3rd, 7th and 14th days after modeling; the positive expression of Pax7 of the right gastrocnemius muscle was increased (P<0.05) on the 3rd day after modeling.@*CONCLUSION@#Electroacupuncture can effectively improve the pain threshold and gait in rats with acute blunt trauma of gastrocnemius muscle, and promote the repair of skeletal muscle injury, the mechanism may be related to the up-regulation of Pax7 and MyoD, so as to promoting the proliferation and differentiation of muscle satellite cell.

ABSTRACT

Objective To investigate the protein expression of Atg7 and Atg12-Atg5 in myocardium of streptozotocin (STZ)diabetic rats.Methods The diabetic model was made in rats by injection of STZ.The rats were divided into control group(NC)and diabetes model group(DM).The left ventricular weight index of rats was detected at the time of 10,20,30,40,60,80,100 d after model initiated.The protein expression of Atg7 and Atg12-Atg5 in myocardium of rats was assayed by western blot.The myocardial ultrastructure and autophagosome were observed by electron microscope. Results Compared with rats in NC group at the same period,the left ventricular weight index in DM group increased since the day of 40th.The protein expression of Atg7 and Atg12-Atg5 in myocardium of diabetic rats increased a little at 20 d(It had not a statistical significance),but reduced significantly at 80 d(P<0.01).The changes in the myocardial ultrastructure and decrease in autophagosome were both obvious in diabetic rats at 80 d.Conclusions With the develop-ment of diabetes mellitus,the autophagy level in myocardium of STZ diabetic rats finally is reduced.

ABSTRACT

Objective To investigate the effect of moxifloxacin(MXF)on inflammatory reaction of mice abdominal peritoneal macrophage induced by LPS.Methods The level of TLR4、SPHK1 and NF-κB mRNA was determined by realtime-PCR;The relative levels of TLR4、SPHK1 and NF-κB were determined by Western blot.TNF-αand IL-1 se-creted by macrophages were detected by ELISA.Results At low and medium concentration(8,16 mg/L)of the MXF have inhibitory effect on the increasing of TLR 4,SPHK1,NF-κB expression level and cell supernatant of TNF-αand IL-1 in LPS stimulation of mice peritoneal macrophages.High concentration(64 mg/L)of the MXF promoted the role of inflammation.Conclusions The MXF of low and medium concentrations can inhibit the inflammatory response in LPS stimulation of mice peritoneal macrophages,and high concentration of the MXF showed contrary effects.

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical efficacy of amphotericin B and voriconazole in the treatment of invasive fungal infection in children with acute lymphoblastic leukemia(ALL) during chemotherapy.</p><p><b>METHODS</b>The clinical data of 214 patients with acute lymphoblastic leukemia admitted in our hospital from March 2014 to February 2017 was retrospectively analyzed. Among them 65 patients had invasive fungal infection (IFI) during the chemotherapy period, out of them 35 patients treated with voriconazole were enrolled in group A; anather 30 patients treated with amphotericin B were enrolled in group B. The clinical factors that affected the incidence of IFI was statistically analyzed. And the efficacy and adverse reactions in children with ALL were compared after treatment for 8 weeks.</p><p><b>RESULTS</b>The incidence of IFI in children with ALL in this study was 30.37%, and was related with the duration of hospitalization and the level of neutrophil deficiency (P<0.05). The total effective rates of group A and group B were 72.28% and 43.33%, respectively, their difference was statistically significant between 2 group (P<0.05). The incidence of renal function impairment, digestive dysfunction and neurotoxicity were 8.57%, 5.71% and 5.71% in children with ALL treated with voriconazole respectively, which were lower than those in children treated with amphotericin B (P<0.05). The score index of Physical health, energy state, emotion control and overall health in group A were significantly higher than those in B group, and the difference was statistically significant (P<0.05).</p><p><b>CONCLUSION</b>The occurrence of IFI in children with ALL relats with the time of hospitalization and the level of neutrophils. The clinical effect of voriconazole is better, and the incidence of adverse reactions can be reduced, suggesting important clinical significance.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the therapeutic effects of percutaneous vertebroplasty versus percutaneous kyphoplasty for treatment of osteoporotic vertebral compression fractures.</p><p><b>METHODS</b>Fifteen patients (24 vertebrae) underwent percutaneous vertebroplasty and 15 (18 vertebrae) received percutaneous kyphoplasty for osteoporotic vertebral compression fractures. The postoperative recovery of the anterior vertebral body height, visual analogue scale (VAS) and locomotor activity before and after the operations were compared between the two groups.</p><p><b>RESULTS</b>All the patients were available for the follow-up lasting for 3-15 months (average 7 months). In the vertebroplasty group, the VAS score decreased from 8.68 preoperatively to 1.74 postoperatively and to 1.13 at the final follow-up. The VAS score in the kyphoplasty group decreased from 8.62 preoperatively to 1.72 postoperatively and to 1.21 at the final follow-up. Both vertebroplasty and kyphoplasty were effective in improving the anterior vertebral body height, but kyphoplasty showed a better effect. None of the patients developed serious complications associated with the operations.</p><p><b>CONCLUSION</b>Percutaneous vertebroplasty and percutaneous kyphoplasty both achieve rapid and significant improvement of back pain in patients with osteoporotic vertebral compression fractures, but kyphoplasty shows a better effect in terms of vertebral body height restoration and cement leakage reduction.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the clinical outcome of intracytoplasmic sperm injection (ICSI) in patients with previous fertilization failure after conventional IVF.</p><p><b>METHODS</b>Data from 20 ICSI cases (22 ICSI cycles) with previous complete failure of fertilization or with fertilization rate < or = 20% between January 2002 and December 2004 were retrospectively analyzed. The control group consisted of 100 consecutive ICSI cycles for male factor infertility in the same period.</p><p><b>RESULTS</b>The fertilization rate dramatically increased from 5.4% after conventional IVF to 76.9% after ICSI treatment (chi-squared = 264.66, P < 0.001). However, the fertilization rate in the subgroup with previous low fertilization was significantly lower than those in the control and in the subgroup without previous fertilization (67.9% vs 77.5%, 67.9% vs 84.2%). Compared with the control group, the subgroup without previous fertilization had a higher pregnancy rate and implantation rate, but only the difference in the implantation rate was statistically significant (40.5% vs 18.9%).</p><p><b>CONCLUSION</b>ICSI can overcome previous fertilization failure with conventional in vitro fertilization and thus improve the clinical outcome.</p>
