RESUMO

Cassava (Manihot esculenta Crantz.) has been a vital staple and food security crop in Tanzania for several centuries, and it is likely that its resilience will play a key role in mitigating livelihood insecurities arising from climate change. The sector is dominated by smallholder farmers growing traditional landrace varieties. A recent surge in virus diseases and awareness in the commercial potential of cassava has prompted a drive to disseminate improved varieties in the country. These factors however also threaten the existence of landraces and associated farmer knowledge. It is important that the landraces are conserved and utilized as the adaptive gene complexes they harbor can drive breeding for improved varieties that meet agro-ecological adaptation as well as farmer and consumer needs, thereby improving adoption rates. Here we report on cassava germplasm collection missions and documentation of farmer knowledge in seven zones of Tanzania. A total of 277 unique landraces are identified through high-density genotyping. The large number of landraces is attributable to a mixed clonal/sexual reproductive system in which the soil seed bank and incorporation of seedlings plays an important role. A striking divergence in genetic relationships between the coastal regions and western regions is evident and explained by (i) independent introductions of cassava into the country, (ii) adaptation to prevailing agro-ecological conditions and (iii) farmer selections according to the intended use or market demands. The main uses of cassava with different product profiles are evident, including fresh consumption, flour production, dual purpose incorporating both these uses and longer-term food security. Each of these products have different trait requirements. Individual landraces were not widely distributed across the country with limited farmer-to-farmer diffusion with implications for seed systems.

White clover (Trifolium repens L.) is a key component legume of temperate pasture agriculture and an important target for molecular marker-assisted plant breeding. A genetic map of white clover has been used to assess genetic control of agronomically important traits that vary in the F2(I.4RxI.5J) mapping family. Phenotypic analysis was performed for a range of vegetative morphogenesis traits (such as leaf area, internode length, plant height and plant spread) and reproductive morphogenesis and development traits (such as flowering date, floral intensity and seed yield), with both spatial and temporal replication. A multi-environment combined analysis (combined analysis) has been performed for traits assessed across multiple experimental datasets in order to identify consistent genetic effects. Quantitative trait locus (QTLs) were detected for the majority of traits, and the locations and magnitudes of QTL effects were compared between individual and combined analyses. This molecular genetic dissection of agronomic traits in white clover provides the basis for equivalent studies in more complex populations, design of marker-assisted selection strategies and comparative genetics with model legume species. Selection for QTLs derived from the combined analysis will permit robust improvement of phenotypic traits over different environments.

Date of first flowering was recorded for 40 years in the same agricultural varieties of white clover (Trifolium repens L.). Standard procedures, experimental conditions and germplasm allowed the unmediated effects of climatic trends to be observed. Flowering dates of the same varieties at the same site have become significantly earlier over this period, with an advance of first flowering of approximately 7.5 days per decade since 1978. Annual maximum, minimum and soil temperatures at the site increased during the 40-year period. First flowering dates (FFDs) were significantly negatively correlated with minimum and maximum temperatures during February and March and soil temperatures between January and April. Maximum, minimum and soil temperatures increased between 1962 and 2002 for these months. No significant correlation was seen between FFD and soil temperatures at other times of the year including the period of vernalisation during winter. The occurrence of ground frost in February was significantly correlated with later flowering. Total annual rainfall and the number of wet days in a year increased between 1962 and 2002. However, only rainfall in February had a significant impact on FFD.

Additive and multiplicative effects of colchicine treatment at the seedling stage and of chromosome doubling on chromosome-specific chiasma frequency at metaphase I have been measured in comparisons between C0 and untreated diploids and between C0 autotetraploids and C0 diploids. Early colchicine treatment increases the frequency of chromosome C univalents to 1.8% but has no similar effect on chromosomes A and D. Colchicine treatment has little net effect on mean chiasma frequency, deducting an average of 0.204 chiasmata per set but otherwise multiplying the mean by a factor of 1.182. These additive and multiplicative effects represent averages of six phenotypes. Chromosome doubling in a tetraploid-diploid chimaera subtracts an average of 0.265 chiasmata per set but otherwise doubles the numbers of chiasmata at the diploid level (x 2.134). Comparison of six diploids and tetraploids reveals modest average additive (+ 1.103) and multiplicative effects (x 1.190). The implications of these findings are discussed in the light of new analyses of previously published data. Key words : chiasmata, Crepis, colchicine, meiosis, polyploidy.

pBNiR1, a cDNA clone encoding part of the barley nitrite reductase apoprotein, was isolated from a barley (cv. Maris Mink) leaf cDNA library using the 1.85 kb insert of the maize nitrite reductase cDNA clone pCIB808 as a heterologous probe. The cDNA insert of pBNiR1 is 503 bp in length. The nucleotide coding sequence could be aligned with the 3' end of other higher plant nitrite reductase apoprotein cDNA sequences but diverges in the 3' untranslated region. The whole-plant barley mutant STA3999, previously isolated from the cultivar Tweed, accumulates nitrite after nitrate treatment in the light, has very much lowered levels of nitrite reductase activity and lacks detectable nitrite reductase cross-reacting material due to a recessive mutation in a single nuclear gene which we have designated Nir1. STA3999 has the characteristics expected of a nitrite reductase apoprotein gene mutant. Here we have used pB-NiR1 in RFLP analysis to determine whether the mutation carried by STA3999 is linked to the nitrite reductase apoprotein gene locus Nii. An RFLP was identified between the wild-type barley cultivars Tweed (major hybridising band of 11.5 kb) and Golden Promise (major hybridising band of 7.5 kb) when DraI-digested DNA was probed with the insert from the partial barley nitrite reductase cDNA clone, pBNiR1. DraI-digested DNA from the mutant STA3999 also exhibited a major hybridising band of 11.5 kb after hybridisation with the insert from pBNiR1.(ABSTRACT TRUNCATED AT 250 WORDS)

Forty-three patients with 69 feet affected by isolated metatarsus adductus et supinatus were reviewed. Of these, 20 patients (with 31 involved feet) had been treated expectantly and spontaneous resolution had occurred with time. The remaining 23 patients (with 38 feet) had required anteromedial release; the operative technique is described. Excellent results were uniformly achieved in both groups, with neither recurrence nor complications in the operatively treated feet. There was a consistent correlation between good clinical results and a naviculo -metatarsal angle of less than 100 degrees. The timing of soft-tissue release did not influence the final outcome.
