RESUMO

Data on the established DNA Bank were summarized. The DNA Bank included workers of the Mayak nuclear facility in the Southern Ural, who were exposed to chronic radiation predominantly from external gamma-rays in different doses, and their families for the future study on radiation mutations in somatic cells of parents and possible transmission of genome instability through the germline. At present the DNA Bank contains genetic material from 1500 individuals, among whom there are 223 families.

RESUMO

A DNA bank was created to preserve the genetic material of staff of the first Russian atomic plant, Production Association (PO) Mayak, who were exposed to chronic external gamma-irradiation at various doses in the early years of plant operation. Some of the workers experienced combined irradiation from external and internal (incorporated plutonium-239) sources. The unique genetic material of the subjects and their children can be used to study the remote consequences of irradiation by means of modern molecular-genetic techniques. At present, the bank contains the genetic material of 500 people. A database was constructed to include medical, demographic, professional, and dose characteristics of each individual and quantitative and qualitative parameters of each DNA preparation. The bank is continuously augmented with new DNA samples of the irradiated people and members of their families.

The comparative evaluation of the diagnostic value of new cholera eltor bacteriophages ctx+ and ctx-, as well as monophages X[symbol: see text]-3, 4, 5, demonstrated their high activity and specificity. Using of these bacteriophages epidemic potential of 95% Vibrio cholerae eltor strains ctx+ and 84.5% of V. cholerae eltor stains ctx- was determined. Commercial monophages X[symbol: see text]-3, 4, 5 were inferior to bacteriophages ctx+ and ctx- in their diagnostic value: only 55% of strains having gene ctxAB were found to be epidemically dangerous, i.e. 45% of strains capable of causing the disease were not detected. On the basis of the results obtained in this investigation cholera eltor bacteriophages ctx+ and ctx- were recommended for introduction into practical use, while further production of cholera diagnostic monophages X[symbol: see text]-3, 4, 5 was recommended to be stopped.

RESUMO

The reactogenicity and immunological effectiveness of different doses of cholera vaccine for oral administration (in tablets), containing choleragen toxoid, Ogawa and Inaba O-antigens and some of Vibrio cholerae exoenzymes, have been tested on 143 children aged 2-17 years. In this investigation the optimum immunization doses of the preparation have been established: two tablets containing 100,000 binding units (BU) of the toxoid and 2,500 units of O-antigen for children 11-14 years; four tablets containing 200,000 BU of the toxoid and 5,000 units of O-antigen for adolescents aged 15-17 years. An essential advantage of the oral vaccine over vaccines intended for parenteral administration lies in its capacity for inducing the accumulation of secretory antibodies (coproantibodies) in practically all vaccinees.

RESUMO

The reactogenic properties and immunological potency of modified cholera chemical vaccine (choleragen-toxoid + O-antigens Inaba and Ogawa) were tested in 278 volunteers aged 18 years and over in comparison with those of a commercial batch of monovalent cholera vaccine (choleragen-toxoid + O-antigen Inaba). The cholera vaccine, enriched with O-antigen Ogawa, was found to be safe; vaccination with this vaccine was not accompanied by the development of systemic and local reactions whose frequency and intensity met the requirements for the reactogenic properties of commercial cholera vaccine. The immunological potency of the bivalent vaccine with respect to strain Inaba was not inferior to that of the commercial vaccine; at the same time in persons immunized with the new preparation the titers of vibriocidal antibodies to strain of serovar Inaba were five-fold higher. The conclusion on the expediency of using cholera chemical vaccine enriched with O-antigen Ogawa was made.
