RESUMO

An experience on the organization of therapeutic care for pregnant women, women-in-labour and puerperants is presented by the example of the city of Tolyatti. An organizational model related with rendering such care is defined; the model envisages the continuity of obstetrics, therapeutic and pediatric complexes, the general therapeutic practice, the practice of maternity welfare clinics, the practice of day-time inhospital policlinic facilities as well as the practice of somatic maternity hospitals. "Therapeutic consultations" and "Passport of extragenital pathology" came to be principally new technologies in Tolyatti.

A study was made of the functional role of the ArdA antirestriction motif (130-LLADVPETVALYFD-143) conserved among all known Ard (alleviation of restriction of DNA) proteins, which are encoded by self-transmissible plasmids and specifically inhibit type I restriction-modification systems. Conserved residues of the motif were individually changed, and the resulting mutants tested for in vivo activity. Hydrophobic L130, L131, and V138 were substituted with negatively charged E; negatively charged D133, E136, and D143 substituted with hydrophobic V; and D127, D150, and D154 neighboring the antirestriction motif substituted with V. Four substitutions (L130E, L131E, V138E, and D143V) substantially (25-1000 times) reduced the ArdA activity. The other substitutions within or beyond the motif had no appreciable effect. Substitutions L130A and L131A each reduced the ArdA activity 10- to 20-fold, indicating that high hydrophobicity of L130 and L131 is important for the ArdA function. Thus, the antirestriction role of ArdA is indeed due to its conserved motif.

The status of the taxa Elymus dahuricus Turcz. ex Griseb., E. excelsus Turcz. ex Griseb., E. woroschilowii Probat. and E. tangutorum (Nevski) Hand.-Mazz., all of which are sometimes treated as members of E. dahuricus s.l. or as constituting the E. dahuricus complex, has been investigated using morphological, biochemical (SDS-PAGE of endosperm proteins) and molecular (DNA-AFLP) variation characteristics. Populations include accessions from Siberia, Far East (Russia), Tien-Shan (Kirghizstan), and Tibet and Sichuan Provinces (China). Variation in the morphological characteristics and endosperm proteins patterns have been found in all populations of the complex. DNA variation was very low within populations but was different among populations; this was attributed to genetic drift. Seed fertility of artificial hybrids and F2-F3 progenies in 38 combinations was analyzed. All biotypes studied form a common recombination genepool (RGP). The reproductive compatibility of hybrid combinations decreases with increased geographical separation of the parents. Differences in protein patterns and in genetic variation drawn from DNA-AFLPs and phenotypical segregation in F2 on series of diagnostic characters does not support existing taxonomic treatments of the E. dahuricus complex in Southern Russia, and Central-Asian area. The E. dahuricus complex seems to be a polymorphic species having a wide geographical range and wide genetic variation. The different species need probably be relegated to infraspecific rank. We have refrained from making the necessary combinations pending examination of additional specimens, including specimens of taxa not included in this study and the type specimens of all taxa involved.

The IncW plasmid pSa contains the gene ard encoding an antirestriction function that is specific for type I restriction and modification systems. The nucleotide sequence of ard was determined and an appropriate polypeptide of about 33 kDa was identified in Escherichia coli T7 expression system. Analysis of deduced amino acid sequence of Ard encoded by pSa revealed that this protein has no significant similarities with the known Ard proteins (ArdA and ArdB types) except the "antirestriction" motif (14 amino acid residues in length) conserved for all known Ard proteins. This finding suggests that pSa Ard may be classified as a new type of Ard proteins which we designated ArdC. The remarkable feature of ArdC is that it has a high degree of similarity (about 38 % identity) to the N-terminal region of RP4 TraC1 primase which includes about 300 amino acid residues and seems to be essential for binding to the single-stranded DNA and TraC1 protein transport to the recipient cells during the conjugal transfer of plasmid DNA. ArdC also binds to single-stranded DNA. In addition, this protein is able in vitro to protect the single-stranded but not double-stranded plasmid DNA against the activity of type II restriction endonuclease HhaI that cleaves both single and double-stranded DNA. We suggest that like TraC1, ArdC would be transported as a result of their interaction with the single-stranded DNA of transferred plasmid strand during conjugative passage through the cell envelope to the recipient bacterium. Such properties of ArdC protein might be useful to protect immediately the incoming single-stranded DNA from the host endonucleases.

RESUMO

During the recent 10 years, 1987-1997, syphilis morbidity was found to increase 60-fold in Omsk Province. In 1997 adolescents constituted 6.0% in the structure of persons who contacted the disease. In this year more than 500 adolescent using drugs were registered in the region. The introduction of drugs by injection was practiced by 9.6% of the adolescents. The program aimed mainly at changing the behavior of young people with orientation on the choice of a healthy mode of life and theirs information about risk factors associated with sex transmitted diseases particularly HIV infection is discussed.

The biological activity of eight 1-oxa-4-aza-2-silacyclanes with the OSiCH(2)N fragment including 6-membered 2-sila-5-morpholinones (1-3) and 4-acyl-2-silamorpholines (4-6)and previously unknown 7-membered derivatives of salicylic acid (7, 8) was studied. Compounds 1 and 3-6 show the certain antihypoxic action. Compounds 2 (40 mg/kg), 4 (20 mg/kg), 6 (40 mg/kg), 7 (20 mg/kg) and 8 (40 mg/kg) reduce the physical serviceability of intact animals. Compound 1 (20 mg/kg) influences the physical serviceability in a moderate-positive way on the background of chlorophos-poisoning. Compounds 5-8 displayed protective properties against chlorophos-poisoning at the LD(50) dose and compounds 2, 4, 5, 7 at the LD(100) dose. Influence of compounds 1 and 2 on the emotional-research behavior of mice was studied.

RESUMO

In murine experiments, whether resistance to acute cooling (air and immersion cooling) can be predicted has been studied. Prognostic tests have demonstrated to be effective and feasible. The criteria examined for each type of cooling (which may be accounted for by the specific features of the air and immersion types of cold effects) have been found to be of different significance.
