RESUMO

This work highlights boosting the tumor targeting efficiency of epirubicin through loading on a new radionanosystem, based on the effective role of silver nanoparticles (AgNPs). Accordingly, PEGylated silver nanoparticles (PEG/AgNPs) were prepared in a size of 20.2 ± 0.1 nm. Additionally, epirubicin was loaded on PEG/AgNPs with a loading efficiency of 63 ± 3 %. Furthermore, both of PEG/AgNPs and EPI/PEG/AgNPs were radiolabeled with 131I isotope with radiolabeling yields of 85 ± 0.2 % and 90.3 ± 1 %, respectively. The in-vivo distribution of 131I-PEG/AgNPs and 131I-EPI/PEG/AgNPs were examined in healthy and tumor bearing mice models. Excitingly, 131I-EPI/PEG/AgNPs revealed a reticuloendothelial system (RES) avoidance and prolonged circulating time. In addition, 131I-EPI/PEG/AgNPs showed fast targeting of tumor site by 25.1 ± 0.1 %ID/g within 0.5 h after intravenous injection. Subsequently, the outcomes provided 131I-EPI/PEG/AgNPs as a new potential system for enhancement of tumor targeting and theranosis (therapy and/or imaging).

The rapid synthesis of two radiofluoronicotinamide derivatives, namely, [18F]MEL050 and [18F]MEL-2F has been simply performed starting from commercial materials. [18F]MEL-2F is a new, potential analogue PET-probe for melanoma imaging. [18F]MEL050 is already an excellent PET imaging probe for early specific diagnosis. The synthesis involves coupling step to obtain the precursor followed by radiofluorination. During the synthesis of the precursors different coupling reagents, such as HBTU, TFFH, HOBT, COMU and PyCIU have been applied. PyClU was found the best to reduce the coupling period to < 1h. The labeled compounds were isolated and purified by HPLC. In the in-vitro study three kinds of cells, namely, Melur (melanin free), KB-3 carcinoma cell line (non-melanoma) and B16-F10 melanoma cell line were used to evaluate the uptake of the radiotracers.

RESUMO

Radioiodinated MIP-1145, which specifically targets melanin, is an ideal candidate for targeted therapy of melanoma. An analogue of MIP-1145 lacking the iodo-substituent (desiodo-MIP-1145) was synthesized as a labeling precursor in three simple steps. The radioiodination of desiodo-MIP-1145 by iodine-125 was carried out via an electrophilic substitution reaction. An optimization study for the iodination reaction was carried out. The labeled compound was isolated and purified by means of electrophoresis and HPLC. The maximum radiochemical yield, 76%, was obtained with radiochemical purity greater than 99%. The log P value for [(125) I]MIP-1145 was measured as 4.5.

In this work, novel edible films were developed from peanut protein with sorbitol as a plasticizer. The peanut protein was extracted from peanut flour. Two groups of films were prepared. One group contained 10% in weight of natural oil/fat of the peanuts, while the other group contained no oil/fat. Six types of films with 37.5%, 44.5%, and 50% sorbitol content by weight, with and without fat, were chosen for mechanical properties characterization and microscopic observations. It was found that with the increase of sorbitol content, higher tensile strength and higher Young's modulus were achieved for the films without fat. However, films with fat exhibited maximum strength and modulus at 44.5% sorbitol content. Relationships between the mechanical properties and the microscopic features of these films were established. It was found that the higher the content of the sorbitol, the more evident the crystalline structure. Grant numbers: NCC9-51.
