Plants are good sources of various bioactive substances and have significant importance for the discovery of new drugs. In this study, Croton macrostachyus plant roots from six different sites in the Central Gondar Zone, Ethiopia, were collected; and their phenolic and flavonoid contents, as well as antioxidant and antibacterial activities were investigated. Total phenolics and flavonoid contents of the root extracts were determined by the Folin-Ciocalteu and aluminum chloride methods, respectively. The antioxidant activity was determined by the DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging method. Moreover, the antibacterial activities were evaluated by disk diffusion method. Results revealed that total polyphenols and flavonoid contents were in the range of 802 ± 53-1557 ± 75 mg GAE/100 g and 342 ± 26-745 ± 32 mg CE/100 g, respectively. Root extracts of C. macrostachyus were found to have higher antioxidant activities ranging from 3.53 ± 0.38 to 6.38 ± 0.62 mg AAE/g sample. They also showed inhibition zones of 5.8, 6.2, 5.9 and 6.0 mm for Staphylococcus aurous, Staphylococcus pneumonia, Escherichia coli and Klebsiella pneumonia, and had equivalent potency with the reference Gentamicin antibiotic. Pearson correlation result indicated a strong relationship between total polyphenol contents and their respective antioxidant activities. This study articulates that the root extracts accumulated a substantial quantity of polyphenols and bears a considerable antioxidant activity.

Concentrations of essential (Cu, Mn, and Zn) and toxic (Cr, Cd, and Pb) trace metals in 30 raw cow's milk samples were quantified using flame atomic absorption spectrometry. The samples were collected from the Nara-Awudarda, Tana-Abo, and Kosoye Amba-Rass sites in North Gondar, Ethiopia, preserved in a deep freezer (-20 °C), and then digested by Kjeldahl apparatus with HNO3/H2O2 (5:2; v/v) at 300 °C for 2.5 h. The data were subject to principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA). Overall hazard quotient (HQ) and carcinogenic risk (CR) values were also estimated to assess metal-related health risks. The mean concentrations of Cr, Mn, Cu, Zn, Cd, and Pb in the milk samples ranged 0.468-0.828, 1.614-2.806, 0.840-1.532, 1.208-5.267, ND-0.330, and ND-0.186 mg/kg, respectively. The lowest values were obtained for Kosoye Amba-Rass milk samples, while the highest were found for those collected from Nara-Awudarda milk samples, probably due to high mineral enrichment and metal leaching (especially Cd and Pb) from coal deposits. PCA revealed clustering of samples with respect to their geographic origin. Validation of PLS-DA model showed 100% classification efficiency using external validation samples and detected Cd and Cu as trace metal markers. The HQ and CR values were within the safe level; however, the former is close to the alert threshold level for Nara-Awudarda milk samples. Thus, further studies on common foodstuffs, constituting a higher proportion in the local diet, are required in this area to provide a complete risk assessment.

The levels and distribution of trace metals (Cr, Mn, Co, Ni, Cu, Zn, Cd and Pb) at eleven water and sediment stations on the Little Akaki River (LAR) in Addis Ababa, Ethiopia, were determined. The binding forms of the metals in various geochemical fractions of the sediments were also quantified. The molar ratio of the sum of the simultaneously extractable metals (∑SEM) and acid-volatile sulphide (AVS)-as a measure for predicting metal-induced toxicity-was estimated. LAR trace levels in water for Cu, Zn, and, particularly Mn were, in most instances, higher than the recommended guidelines for healthy aquatic ecosystems. Total trace metal (TTM) contents in the LAR sediments at certain stations exceeded "threshold effect concentrations" and even "probable effect concentrations", especially in the cases of Zn, Cu, Ni, Pb, and at all stations for Mn. This became more apparent after applying "normalizations" to the relatively lower TTM adsorption capacities of coarse-grained, organic-poor sediments. Sequential extraction of the sediments showed that trace metals generally have a higher affinity for Fe-Mn oxide and organic matter/sulphidic fractions, followed by the residual fraction. Mn was relatively strongly bound to the exchangeable, carbonate bound fractions, whereas a large proportion of Cr was found in the residual fraction. The Σ[SEM]/[AVS] ratio pointed to potential metal-induced toxicity of sediments collected from seven out of the eleven stations. The results indicate that trace metal pollution pose risks to the health of ecosystems, and to human communities that use the river for a range of different purposes.
