Introduction: The use of treated municipal wastewater (TWW) represents a relevant opportunity for irrigation of agricultural crops in semi-arid regions to counter the increasing water scarcity. Pharmaceutically active compounds (PhACs) are often detected in treated wastewater, posing a risk to humans and the environment. PhACs can accumulate in soils and translocate into different plant tissues, reaching, in some cases, edible organs and entering the food chain. Methods: This study evaluated the uptake and translocation processes of 10 PhACs by olive trees irrigated with TWW, investigating their accumulation in different plant organs. The experiment was conducted in southern Italy, in 2-year-old plants irrigated with three different types of water: freshwater (FW), TWW spiked with 10 PhACs at a concentration of 200 µg L-1 (1× TWW), and at a triple dose (3× TWW), from July to October 2021. The concentration of PhACs in soil and plant organs was assessed, collecting samples of root, stem, shoot, leaf, fruit, and kernel at 0 (T0), 50 (T1), and 107 (T2) days of irrigation. PhACs extraction from soil and plant organs was carried out using the QuEChERS method, and their concentrations were determined by high-resolution mass spectrometry coupled with liquid chromatography. Results: Results of uptake factors (UF) showed a different behavior between compounds according to their physicochemical properties, highlighting PhACs accumulation and translocation in different plant organs (also edible part) in 1× TWW and 3× TWW compared to FW. Two PhACs, carbamazepine and fluconazole, showed interactions with the soil-plant system, translocating also in the aerial part of the plant, with a translocation factor (TF) greater than 1, which indicates high root-to-leaf translocation. Discussion: Findings highlight that only few PhACs among the selected compounds can be uptaken by woody plants and accumulated in edible parts at low concentration. No effects of PhACs exposure on plant growth have been detected. Despite the attention to be paid to the few compounds that translocate into edible organs, these results are promising for adapting wastewater irrigation in crops. Increasing knowledge about PhACs behavior in woody plants can be important for developing optimized wastewater irrigation and soil management strategies to reduce PhACs accumulation and translocation in plants.

RESUMO

Nematode samplings in various areas and crops of Greece were carried out and the recovered nematode species were characterized using morphological and molecular data. Seven species of plant-parasitic nematodes were recovered, three of which are reported for the first time in Greece, including Hemicycliophora poranga, Helicotylenchus dihystera and Tylenchorhynchus zeae. Four other recovered species had already been reported in Greece, including Bitylenchus hispaniensis, Helicotylenchus microlobus, Nanidorus minor and Scutellonema brachyurus. D2-D3 segments of 28S rRNA gene for all of these nematode species are provided.

BACKGROUND: Nematodes of the genus Heterorhabditis are important biocontrol agents as they form a lethal combination with their symbiotic Photorhabdus bacteria against agricultural insect pests. This study describes a new species of Heterorhabditis. METHODS: Six Heterorhabditis nematode populations were recovered from agricultural soils in Jammu and Kashmir, India. An initial examination using mitochondrial and nuclear genes showed that they belong to a new species. To describe this new species, a variety of analyses were conducted, including reconstructing phylogenetic relationships based on multiple genes, characterizing the nematodes at the morphological and morphometric levels, performing self-crossing and cross-hybridization experiments, and isolating and characterizing their symbiotic bacteria. RESULTS: The newly discovered species, Heterorhabditis casmirica n. sp., shares 94% mitochondrial cytochrome C oxidase subunit I gene (COI) sequence identity with Heterorhabditis bacteriophora and Heterorhabditis ruandica, and 93% with Heterorhabditis zacatecana. Morphologically, it differs from H. bacteriophora in its infective juvenile phasmids (present vs. inconspicuous) and bacterial pouch visibility in the ventricular portion of the intestine (invisible vs. visible); genital papilla 1 (GP1) position (at manubrium level vs. more anterior), and in its b ratio (body length/neck length), c ratio (tail length/bulb width), and D% [(excretory pore/neck length) × 100]. Other morphological differences include anterior end to the nerve ring distance (77-100 vs. 121-130 µm), V% [(anterior end of vulva/body length) × 100] (46-57 vs. 41-47) in hermaphroditic females; rectum size (slightly longer than the anal body diameter vs. about three times longer), phasmids (smaller vs. inconspicuous), body length (0.13-2.0 vs. 0.32-0.39 mm), body diameter (73-150 vs. 160-220 µm), anterior end to the excretory pore distance (135-157 vs. 174-214 µm), and demanian ratios in amphimictic females. Morphological differences with H. ruandica and H. zacatecana were also observed. Furthermore, H. casmirica n. sp. did not mate or produce fertile progeny with other Heterorhabditis nematodes reported from India. It was also discovered that H. casmirica n. sp. is associated with Photorhabdus luminescence subsp. clarkei symbiotic bacteria. CONCLUSIONS: The discovery of H. casmirica n. sp. provides novel insights into the diversity and evolution of Heterorhabditis nematodes and their symbiotic bacteria. This new species adds to the catalog of entomopathogenic nematodes in India.

Hydrodeoxygenation (HDO) kinetics of glycerol into 1,2-propanediol (1,2-PDO) in the liquid phase is studied on Cu-Pd/TiO2 catalysts. At a stirring speed higher than 480 rpm and an average diameter of the catalyst particles smaller than 89.5 µm, no mass transfer resistance artifacts are observed. The increasing temperature and H2 concentration promote the glycerol conversion and the selectivity to 1,2-PDO and disfavor the selectivity to acetol. Based on the experimental data, empirical kinetic pseudo-homogeneous expressions are proposed for glycerol disappearance, 1,2-PDO formation, and acetol formation in the catalytic system. Dependence of the disappearance rate of glycerol is closer to 1 with respect to glycerol and nonmeaningful with respect to H2. The formation rate of 1,2-PDO is not highly dependent on the initial concentration of glycerol or H2, and the formation rate of acetol is directly dependent on glycerol and inversely dependent on H2, since it accelerates acetol conversion to 1,2-PDO. The activation energies for glycerol disappearance (77.8 kJ/mol), 1,2-PDO formation (51.2 kJ/mol), and acetol formation (84.6 kJ/mol) evidenced the selective formation of 1,2-PDO in this catalytic system.

Antecedentes y objetivo La evolución digital supone una oportunidad para que la oftalmología se adapte a nuevos modelos asistenciales. Este estudio tuvo como objetivo conocer cómo la pandemia ha modificado la práctica clínica y actividades formativas del oftalmólogo especializado en superficie ocular, y analizar tendencias y necesidades emergentes. Materiales y métodos Estudio realizado mediante encuesta online. Un comité de 3 especialistas diseñó un cuestionario de 25 preguntas estructurado en: 1) Perfil del participante; 2) Impacto de la pandemia en el manejo del paciente y las actividades profesionales; 3) Tendencias y necesidades. Resultados Participaron 68 oftalmólogos clínicos. Hubo un alto grado de acuerdo (90%) en que la pandemia demoró las visitas de seguimiento oftalmológico y el diagnóstico. Los participantes coincidieron en que aumentó la frecuencia de pacientes con enfermedad de ojo seco (75%), orzuelo/chalazión (62%) y blefaritis (60%). Según un 28%, será habitual el teleseguimiento mediante teleconsulta de dolencias como ojo seco, glaucoma, diabetes, conjuntivitis, hiposfagmas, orzuelos, etc., especialmente en población joven. Esto será especialmente relevante en las enfermedades crónicas o leves de la superficie ocular, y en el seguimiento de pacientes tras operación de cataratas y retinopatía diabética. Conclusiones Durante la pandemia se ha percibido un aumento de la incidencia de ciertas enfermedades de superficie ocular. El seguimiento de enfermedades crónicas o leves de la superficie ocular se vincula a la necesidad de proporcionar formación específica tanto para el paciente como para el profesional que lo atiende, además de protocolos de cribado y derivación que optimicen el flujo asistencial (AU)

This will be especially relevant in chronic or mild pathologies of the ocular surface, and in the follow-up of patients after cataract and diabetic retinopathy interventions. Conclusions During the pandemic, an increase in the incidence of certain ocular surface diseases has been perceived. The telematic follow-up of chronic or mild pathologies of the ocular surface entails the need to provide specific training for both the patient and the healthcare professional, in addition to screening and referral protocols that would optimize the flow of care (AU)

In C3 plants, carbon isotope composition (δ13C) is influenced by isotopic effects during diffusion from the atmosphere to the chloroplasts and carboxylation reactions. This work aimed to demonstrate if δ13C of leaf soluble carbohydrates (δ13Cleaves) and of dry matter from new-growth shoots (δ13Cshoots) of Prunus plants subjected to a period of water deficit was related to water use efficiency (WUE). For this purpose, three interspecific Prunus hybrids rootstocks (6-5, 7-7 and G × N) were gradually subjected to drought and then rewatered. Soil water content (SWC) decreased from 26.1 to 9.4% after 70 days of water shortage, when plants reached values of predawn leaf water potential (LWP) ranging from -3.12 to -4.00 MPa. Gas exchange, particularly net photosynthetic and transpiration rates, differed among the three hybrids, leading to different values of WUE. After 70 days of drought, a significant δ13C increase of 5.86, 4.28 and 4.99‰ was observed in 6-5, 7-7 and G × N, respectively. Significant correlations between δ13C and other parameters (substomatal CO2/atmospheric CO2 ratio, stomatal conductance and stem water potential) were found in all hybrids. The rewatering phase caused a recovery of the physiological status of the plants. The isotope composition of δ13Cshoots was correlated with the average WUE measured during the whole experiment. δ13Cleaves and δ13Cshoots were positively related (r = 0.87; p < 0.001). The isotopic signature was a reliable screening tool to identify Prunus genotypes tolerant to drought stress. The results suggest the possibility of using δ13C as an integrated indicator of level of drought stress in plants subjected to prolonged stress conditions.

The heat shock protein family 70 (Hsp70) comprises chaperone proteins that play major multiple roles in Plasmodium asexual and sexual development. In this study, we analyzed the expression of Hsp70-1 in gametocytes, gametes, zygotes, and its participation in ookinete formation and their transition into oocysts. A monoclonal antibody against recombinant Hsp70-1 revealed its presence in zygotes and micronemes of ookinetes. Compared to wild type parasites, Hsp70-1 knockout ookinetes produced fewer oocysts in Plasmodium-susceptible Anopheles albimanus mosquitoes. This may indicate a defective transformation of ookinetes into oocysts in the absence of Hsp70-1. The presence of this protein in micronemes suggests its participation in mosquito infection, probably aiding to the adequate structural conformation of proteins in charge of motility, recognition and invasion of the insect midgut epithelium.

We demonstrated recently that CuPd/TiO2-Na bimetallic catalysts synthesized by sequential wet impregnation are active, selective, and stable for the hydrodeoxygenation (HDO) of glycerol into propylene glycol at low H2 pressure. The present study reports on the nature and distribution of Cu and Pd surface species in CuPd/TiO2-Na bimetallic catalysts using different scanning transmission electron microscopy techniques that supply cluster-specific alloying details. In particular, we used atomic-resolution Z-contrast imaging, X-ray energy-dispersive spectroscopy, and electron energy-loss spectroscopy. We also include X-ray photoelectron spectroscopy results. Our analysis shows that the metallic nanoparticles adopt mainly five different structures according to how the Cu and Pd atoms coordinate among themselves: a homogeneous CuPd alloy structure (45-61%), a Cu shell/CuPd core (15-23%), a smaller number of particles formed by Cu on the surface and Pd in the nucleus (10-17%), and there are also nanoparticles formed only by Pd (4-7%) or by Cu (8-13%). We determined that there is a inhomogeneous distribution of Cu and Pd in the bimetallic nanoparticles, with Cu being predominant on the surface (between 76 and 90% of the total area analyzed for each particle). Most bimetallic nanoparticles have sizes below 6 nm, the Pd monometallic nanoparticles are in the 2-4 nm range, whereas the monometallic Cu nanoparticles are larger than 8 nm. Bimetallic nanoparticles with sizes smaller than 6-7 nm are fundamentally made up of Cu0-Pd0 and Cu1+-Pd0. The nanoparticles with sizes greater than 7 nm consist of Cu2+ and Cu2+-Pd2+. Our obtained results also help describe reports about the activation of HDO by Pd-Cu in the absence of H2, an effect apparently not observed with other bimetallic systems.

In conventional olive growing, frequent soil tillage strongly reduces the complexity and diversity of the agro-ecosystem. Here, a metabolomic analysis was carried out on the xylem sap (XS) of olive plants (Olea europaea L.) from a grove located in Southern Italy (Basilicata region). The orchard has been divided in two plots that have been managed for 18 years with two different systems: a) 'sustainable management' (Smng), with no-tillage, fertigation and internal C-inputs (spontaneous weeds and pruning residues), and b) an adjacent rainfed 'conventional management' (Cmng), that included soil tillage and mineral fertilization. The XS was extracted from olive shoots in two sampling times (ST1: May; ST2: October) using a Sholander pressure chamber, and its metabolome analyzed by ultra-high performance liquid chromatography (UHPLC) coupled to a hybrid quadrupole-time-of-flight mass spectrometer (QTOF-MS). The discriminating compounds were 94 at ST1 and 119 at ST2, and 35 of them were in common between the two sampling times. The majority of the discriminating metabolites (73 on 94 at ST1, and 109 on 119 at ST2) were found at higher concentration in the XS of Smng plants, compared to that of Cmng ones. Most of the discriminating metabolites found in XS (about 80%, both at ST1 and ST2) were involved in plant secondary metabolism, mainly for plant chemical defense, growth regulation and signal transduction. The most prevailing class of compounds included terpenoids, phytohormones, alkaloids, sterols/steroids, retinols/retinoids, tocopherols and carotenoids. For the first time, we have demonstrated that the XS of a tree crop significantly responds to a shift of soil management. Generally, the plants of the Smng plot showed an up-regulated secondary metabolism. The results of our study encourage the use of a set of sustainable agricultural practices in a productive orchard, in order to enhance plant physiological status, increase yield quantity/quality, safeguard the environment and ameliorate human health.

Under suitable conditions, low-quality, treated urban wastewater (TWW) is an additional water resource for irrigation in water-scarce environments but its use in agriculture requires a careful monitoring of a range of hygiene parameters, including human pathogenic bacteria (HPB). DNA-based microbiological analyses on soil, xylem sap, and leaves surface (phyllosphere) were carried out in an olive (Olea europaea L.) grove located in Southern Italy (Basilicata region). The experimental grove has been managed in two plots for 18 years. The experimental plot (WWtr) was drip irrigated daily with TWW (2800 m3 ha-1 year-1), while the control plot (RFtr) was rainfed. The results of the 16S-rRNA-based metagenomic analysis demonstrated that the phyllosphere had the lowest number of potential HPB (6), compared to soil (22) and xylem (26) compartments. Gammaproteobacteria, including potential HPB, like Pseudomonas and Acinetobacter spp., were significantly higher in WWtr soil and xylem sap, compared to RFtr. A similar trend was observed for Burkholderia spp. (Betaproteobacteria) and Mycobacterium spp. (Actinobacteria). The Firmicutes Enterococcus, Staphylococcus and Streptococcus spp. were more abundant in WWtr xylem sap. The pathogenic Clostridium perfringens was found higher on WWtr leaves (relative abundance 7.17 in WWtr and 1.33 in RFtr) and Enterococcus faecalis in WWtr xylem sap (93.22 in WWtr and 7.08 in RFtr). On the basis of the results obtained, the irrigation with TWW can be considered a realistic and safe agronomic practice in Mediterranean orchards, and an opportunity for farmers and consumers.

OBJECTIVE: This analysis estimates type 1 and type 2 diabetes direct costs in 2012, in terms of hospital care, outpatient visits, diagnostics and medications, in a local healthcare trust in Northern Italy (ULSS n.20 Verona). METHODS: The Johns Hopkins Adjusted Clinical Group (ACG®) System was used to analyze data, including hospital discharges, emergency room admissions, medical encounter records, disease registries, copayment exemptions, home care services, psychiatric services, rehabilitation services, and medications. Data from general practitioners and nursing homes were not directly available. Patients obtained from the first analysis were subsequently divided in two groups (type 1 and type 2 diabetes) according to ATC drug classification system and age. Costs were estimated from inpatient and outpatients fees and drugs costs. RESULTS: ULSS n. 20 takes care of about 480.000 people. We identified 974 people affected by type 1 diabetes (prevalence 0,2%) and 24.087 people affected by type 2 diabetes (prevalence 5,0%) among the residents in 2012. Hospitalization mean annual cost was 4.753,50€ (SD 9.330,19€) for type 1 diabetes and 1.718,08€ (SD 5.087,34€) for type 2 diabetes. Outpatient care mean annual cost was 1.401,76€ (SD 4.394,88€) for type 1 diabetes and 669,15€ (SD 2.121,24€) for type 2 diabetes. Medications mean annual cost was 1,369,35€ (SD1.781,18€) for type 1 diabetes and 874,07€ (SD 2.832,2€) for type 2 diabetes. CONCLUSIONS: ACG® diabetes data analysis agrees with data obtained by more expensive methods and seems to be a comprehensive and applicable tool to analyze chronic diseases dynamics in the Italian setting in order to prioritize future research and analyze the effects of interventions aimed to ensure the sustainability of public health services. Because of the combination between prevalence data and epidemiological trends, we could be at the eve of a dramatic increase of diabetes costs with major concerns for the Italian NHS ability to withstand.

BACKGROUND: Malaria parasites are transmitted by Anopheles mosquitoes. Although several studies have identified mosquito midgut surface proteins that are putatively important for Plasmodium ookinete invasion, only a few have characterized these protein targets and demonstrated transmission-blocking activity. Molecular information about these proteins is essential for the development of transmission-blocking vaccines (TBV). The aim of the present study was to test three monoclonal antibodies (mAbs), A-140, A-78 and A-10, for their ability to recognize antigens and block oocyst infection of the midgut of Anopheles albimanus, a major malaria vector in Latin America. METHOD: Western-blot of mAbs on antigens from midgut brush border membrane vesicles was used to select antibodies. Three mAbs were tested by membrane feeding assays to evaluate their potential transmission-blocking activity against Plasmodium berghei. The cognate antigens recognized by mAbs with oocyst-reducing activity were determined by immunoprecipitation followed by liquid chromatography tandem mass spectrometry. RESULTS: Only one mAb, A-140, significantly reduced oocyst infection intensity. Hence, its probable protein target in the Anopheles albimanus midgut was identified and characterized. It recognized three high-molecular mass proteins from a midgut brush border microvilli vesicle preparation. Chemical deglycosylation assays confirmed the peptide nature of the epitope recognized by mAb A-140. Immunoprecipitation followed by proteomic identification with tandem mass spectrometry revealed five proteins, presumably extracted together as a complex. Of these, AALB007909 had the highest mascot score and corresponds to a protein with a myosin head motor domain, indicating that the target of mAb A-140 is probably myosin located on the microvilli of the mosquito midgut. CONCLUSION: These results provide support for the participation of myosin in mosquito midgut invasion by Plasmodium ookinetes. The potential inclusion of this protein in the design of new multivalent vaccine strategies for blocking Plasmodium transmission is discussed.

The marine nematodes usually comprise free-living species, although a few are parasitic. However, several cases of free-living nematodes found accidentally in the digestive tract of certain vertebrates, especially fishes, have sometimes been recorded and categorized as pseudoparasites. In the present work, two species of marine fishes, the rhomboid crappie, Diapterus rhombeus, and the silvered crappie, Eucinostomus argenteus (Perciformes: Gerreidae), from Angra dos Reis on the coast of Rio de Janeiro (Brazil) were examined. Seven species of free-living marine nematodes were found in the digestive tract of these fish. Several of these species remain unknown as free-living forms in Brazil. The combination of the fish feeding strategies and the poor preservation of the body of the nematode specimens found could indicate that these nematodes are pseudoparasites, appearing in the fishes' digestive tracts through accidental ingestion and thereafter surviving for brief periods of time. Descriptions, illustrations and tables of measurements are provided for all species. Six of these species (Croconema torquens, Dorylaimopsis pellucida, Oncholaimellus labiatus, Parodontophora breviamphida, Prooncholaimus ornatus, Trissonchulus latus) have been reported for the first time from the Brazilian coast.

Accumulation of Ca in several fleshy fruit is often supposed to depend, among others, by climatic variables driving fruit transpiration. This study tests the whole causal chain hypothesis: VPD → fruit transpiration → Ca accumulation. Also there are evidences that relationship between fruit transpiration and Ca content is not always clear, hence the hypothesis that low VPD reduces the fraction of xylemic water destined to transpiration was tested by examining the water budget of fruit. Attached fruits of Actinidia deliciosa were subjected to Low (L) and High (H) VPD. Their transpiration was measured from early after fruit-set to day 157 after full bloom (DAFB). Fruits were picked at 70, 130 and 157 DAFB for Ca and K determinations and for water budget analysis. Cumulative transpired water was ∼ 70 g and ∼ 16 g H2O f(-1) in HVPD and LVPD, respectively. Calcium accumulated linearly (R(2) = 0.71) with cumulative transpiration when VPD was high, while correlation was weaker (R(2) = 0.24) under LVPD. Under low VPD the fraction of xylem stream destined to transpiration declined to 40-50%. Results suggest that Ca accumulation is coupled to cumulative transpiration under high VPD because under that condition cumulative transpiration equals xylem stream (which carry the nutrient). At LVPD, Ca gain by fruit is uncoupled from transpiration because ∼ 60% of the xylemic water is needed to sustain fruit growth. Results will apply to most fruits (apples, tomatoes, capsicum, grapes etc.) since most suffer Ca deficiency disorders and grow in changing environments with variable VPD, also they could be supportive for the implementation of fruit quality models accounting also for mineral compositions and for a reinterpretation of certain field practices aimed at naturally improve fruit Ca content.

BACKGROUND: Water temperature greatly influences the physiology and behaviour of teleost fish as other aquatic organisms. While fish are able to cope with seasonal temperature variations, thermal excursions outside their normal thermal range might exceed their ability to respond leading to severe diseases and death.Profound differences exist in thermal tolerance across fish species living in the same geographical areas, promoting for investigating the molecular mechanisms involved in susceptibility and resistance to low and high temperatures toward a better understanding of adaptation to environmental challenges. The gilthead sea bream, Sparus aurata, is particularly sensitive to cold and the prolonged exposure to low temperatures may lead to the "winter disease", a metabolic disorder that significantly affects the aquaculture productions along the Northern Mediterranean coasts during winter-spring season. While sea bream susceptibility to low temperatures has been extensively investigated, the cascade of molecular events under such stressful condition is not fully elucidated. RESULTS: In the present study two groups of wild sea bream were exposed for 21 days to two temperature regimes: 16 ± 0.3°C (control group) and 6.8 ± 0.3°C (cold-exposed group) and DNA microarray analysis of liver transcriptome was carried out at different time points during cold exposure.A large set of genes was found to be differentially expressed upon cold-exposure with increasingly relevant effects being observed after three weeks at low temperature. All major known responses to cold (i.e. anti-oxidant response, increased mitochondrial function, membrane compositional changes) were found to be conserved in the gilthead sea bream, while, evidence for a key role of unfolded protein response (UPR) to endoplasmic reticulum (ER) stress, during short- and long-term exposure to cold is reported here for the first time. CONCLUSIONS: Transcriptome data suggest a scenario where oxidative stress, altered lipid metabolism, ATP depletion and protein denaturation converge to induce ER stress. The resulting UPR activation further promotes conditions for cell damage, and the inability to resolve ER stress leads to severe liver dysfunction and potentially to death.
