RESUMO

To contribute knowledge of an important experimental model for studies on skin embryology, a study was carried out to describe the morphological events of the skin during the intrauterine life of Cavia porcellus from the 10th to the 60th day of gestation. Embryos and fetuses were dissected, and the skin of the nasal, cranial, lumbar and anal regions was processed byoptical microscopy. At 30 days the first hairs, called lanugos, were observed in the cranial region. The morphological description showed that a few days can make a great difference in development.


Con el objetivo de contribuir al conocimiento de un importante modelo experimental para estudios sobre embriología de la piel, se llevó a cabo un estudio para describir los eventos morfológicos de la piel durante la vida intrauterina de Cavia porcellus desde el día 10 hasta el día 60 de gestación. Los embriones y los fetos se disecaron y se procesó la piel de las regiones nasal, craneal, lumbar y anal, mediante microscopía óptica. A los 30 días se observaron los primeros vellos, llamados lanugos, en la región craneal. La descripción morfológica mostró que unos pocos días pueden marcar una gran diferencia en el desarrollo.

This study describes the development of the central nervous system in guinea pigs from 12th day post conception (dpc) until birth. Totally, 41 embryos and fetuses were analyzed macroscopically and by means of light and electron microscopy. The neural tube closure was observed at day 14 and the development of the spinal cord and differentiation of the primitive central nervous system vesicles was on 20th dpc. Histologically, undifferentiated brain tissue was observed as a mass of mesenchymal tissue between 18th and 20th dpc, and at 25th dpc the tissue within the medullary canal had higher density. On day 30 the brain tissue was differentiated on day 30 and the spinal cord filling throughout the spinal canal, period from which it was possible to observe cerebral and cerebellar stratums. At day 45 intumescences were visualized and cerebral hemispheres were divided, with a clear division between white and gray matter in brain and cerebellum. Median sulcus of the dorsal spinal cord and the cauda equina were only evident on day 50. There were no significant structural differences in fetuses of 50 and 60 dpc, and animals at term were all lissencephalic. In conclusion, morphological studies of the nervous system in guinea pig can provide important information for clinical studies in humans, due to its high degree of neurological maturity in relation to its short gestation period, what can provide a good tool for neurological studies.(AU)


Este estudo descreve o desenvolvimento do sistema nervoso central em guinea pig do 12º dia pós-concepção (dpc) até ao nascimento. No total, 41 embriões e fetos foram analisados macroscopicamente e por microscopia de luz e eletrônica. O fechamento do tubo neural foi observado no dia 14 e o desenvolvimento da medula espinhal e diferenciação das vesículas primitivas do sistema nervoso central foram observados no dia 20. Histologicamente, o tecido cerebral indiferenciado foi observado como uma massa de tecido mesenquimal entre os dias 18 e 20 e no 25º dia o tecido no interior do canal medular apresentou maior densidade. No dia 30 o tecido cerebral apresentou-se diferenciado, período no qual a medula espinhal preenchia todo o canal vertebral e foi possível observar os estratos cerebral e cerebelar. No dia 45 as intumescências cervical e lombar foram visualizadas e os hemisférios cerebrais estavam divididos, com uma clara distinção entre substância branca e cinzenta no cérebro e cerebelo. O sulco mediano dorsal da medula espinhal e a cauda equina foram evidentes apenas no dia 50. Não houve diferenças estruturais significativas em fetos de 50 e 60 dpc e animais a termo eram todos lisencefálicos. Estudos morfológicos do sistema nervoso em guinea pig podem fornecer informações importantes para estudos clínicos em seres humanos devido ao alto grau de maturidade neurológica em relação ao seu período de gestação curto, fato que servir como excelente ferramenta em estudos neurológicos.(AU)

This study aimed to characterize the patterns of arterial vascularization in swine hearts. Ninety swine hearts were submitted to the Spalteholz diaphanization technique in order to dissect the coronary arteries. Three types of arterial vascularization patterns were characterized through the behaviorof the rami circumflexus and interventricularis, namely: balanced, right and left types. The balanced pattern was the most frequently (42.2%); in this case, the rami circumflexus and interventricularis occupied their respective sulci. The right type (40%) was further categorized into three vascularization subtypes. In the first, ramus circumflexus dexter branched from the ramus interventricularis subsinuosus. In the second, the arteria coronaria dextra branched from ramus interventricularis subsinuosus and ramus circumflexus. In the third model, arteria coronaria sinister branched from ramus interventricularis paraconalis. The left type (17.7%) exhibited two subtypes. In the first, ramus interventricularis paraconalis ran through the entire corresponding sulcus and the ventral third of sulcus interventricularis subsinuosus, and ramus interventricularis subsinuosus occupied the dorsal and middle third of its respective sulcus. In the second, ramus interventricularis subsinuosus branched from arteria coronaria dextra and ran through the dorsal and medium thirds of its respective sulcus, and the ventral third was occupied by the collateral branch of ramus circumflexus sinister. Our results reinforce the thesis that the blood distribution system through the coronary artery in swine is similar to human, not only in qualitative but also by a quantitative comparison.


Este estudo teve como objetivo caracterizar os padrões de vascularização arterial em corações de suínos. Para tanto, 90 corações de suínos foram submetidos à técnica de diafanização de Spalteholz com a finalidade de dissecar as artérias coronárias. Através do comportamento dos rami circumflexus e interventricularis caracterizou-se três modalidades de vascularização arterial do coração de suínos, sendo eles os tipos equilibrado, direito e esquerdo. O equilibrado foi observado com maior frequência (42,2%), neste caso os rami circumflexus e interventricularis das artérias coronárias ocupavam seus respectivos sulcos. O tipo direito (40%) apresentou três subtipos de vascularização. No primeiro o ramus circumflexus dexter ramificava-se formando o ramus interventricularis subsinuosus. No segundo, a arteria coronaria dextra emitia o ramus interventricularis subsinuosus e ramus circunflexus. E no terceiro modelo, da arteria coronaria sinistra emergia o ramus interventricularis paraconalis. O tipo esquerdo (17,7%) apresentou dois subtipos. No primeiro, o ramus interventricularis paraconalis percorria todo o sulco correspondente e o terço ventral do sulcus interventricularis subsinuosus, o ramus interventricularis subsinuosus ocupava os terços dorsal e médio do seu respectivo sulco. No segundo, o ramus interventricularis subsinuosus oriundo da arteria coronaria dextra percorria apenas os terços dorsal e médio do seu respectivo sulco, ficando o terço ventral ocupado por colateral do ramus circumflexus sinister. Nossos resultados reforçam a tese de que a distribuição do suprimento sanguíneo pela artéria coronária em suínos é semelhante ao humano, não apenas de maneira qualitativa, mas também do ponto de vista quantitativo.

Yolk sac (YS) is the site of blood-cell production where primitive erythroid cells originate and complete their maturation. YS is a source of precursor cells, however its differentiation potential and suitability for cell therapies are not well described. YS can be a cell source when neovascularization is required. This study characterized YS canine cells, transduced with VEGF, to analyze then using Immunocytochemistry, flow cytometry and real time PCR. Immunocytochemistry: positive expression for CD105, PCNA, VEGF and vWF, flow cytometry for CD105, VEGF, PCNA, OCT-4 and RT-qPCR for VEGF, CD31, CD105, PCNA and FLT - 1, indicating that these cells have characteristics of endothelial progenitor and pluripotency. After transduction, the YS cells changed their morphology and showed endothelial-like cells. We suggest, because of their cell surface phenotype as well as their capacity to differentiate into endothelial-like cells, that canine YS represents a source of cells for neovascularization therapies.

Birds occupy a prominent place in the Brazilian economy not only in the poultry industry but also as an animal model in many areas of scientific research. Thus the aim of this study was to provide a description of macro and microscopic aspects of the ectoderm-derived structures in chicken embryos / fetuses poultry (Gallus gallus domesticus) from 1st to 19th day of incubation. 40 fertilized eggs, from a strain of domestic chickens, with an incubation period of 2-19 days were subjected to macroscopic description, biometrics, light, and scanning microscopy. All changes observed during the development were described. The nervous system, skin and appendages and organs related to vision and hearing began to be identified, both macro and microscopically, from the second day of incubation. The vesicles from the primitive central nervous system-forebrain, midbrain, and hindbrain-were identified on the third day of incubation. On the sixth day of incubation, there was a clear vascularization of the skin. The optic vesicle was first observed fourth day of development and on the fifth day there was the beginning of the lens formation. Although embryonic development is influenced by animal line as well as external factors such as incubation temperature, this paper provides a chronological description for chicken (Gallus gallus domesticus) during its embryonic development.

The poultry industry is a sector of agribusiness which represents an important role in the country's agricultural exports. Therefore, the study about embryogenesis of the domestic chicken (Gallus gallus domesticus) has a great economic importance. The aim of this study was to evaluate embryonic development of the endoderm in chicken (Gallus gallus domesticus). Forty fertilized eggs of domestic chickens, starting from the 1st day of gestation and so on until the 19 days of the incubation were collected from the Granja São José (Amparo, SP, Brazil). Embryos and fetus were fixed in 10% formaldehyde solution, identified, weighed, measured, and subjected to light and scanning electron microscopy. The endoderm originates the internal lining epithelium of the digestive, immune, respiratory systems, and the organs can be visualized from the second day (48 h) when the liver is formed. The formation of the digestive system was complete in the 12th day. Respiratory system organs begin at the fourth day as a disorganized tissue and undifferentiated. Their complete differentiation was observed at the 10 days of incubation, however, until the 19 days the syrinx was not observed. The formation of immune system at 10th day was observed with observation of the spleen, thymus, and cloacal bursa. The study of the organogenesis of the chicken based on germ layers is very complex and underexplored, and the study of chicken embryology is very important due the economic importance and growth of the use of this animal model studies such as genetic studies.

A Distrofia Muscular de Duchenne (DMD) é uma miopatia severa de caráter recessivo ligada ao cromossomo X e o modelo animal de estudo mais relevante é o Golden Retriever Muscular Dystrophy (GRMD). Além das severas alterações que ocorrem na musculatura estriada, muitos estudos mostram que outras estruturas, inclusive viscerais, podem se mostrar alteradas nesta patologia. Desta forma, este trabalho objetivou análisar e comparar possíveis alterações estruturais e funcionais do rim em cães GRMD. Neste modelo de estudo, foi possível observar a presença das faces convexa e côncava, do hilo renal e dos pólos craniais e caudais dos rins. O órgão mostrou-se envolto por uma cápsula fibrosa. Em um corte sagital do órgão, notou-se a presença das regiões cortical e medular e da pelve renal. Na análise microscópica foi possível identificar a zona medular e cortical com suas estruturas: os corpúsculos renais formados pelo glomérulo e pela cápsula de Bowman, os túbulos contorcidos proximais e distais, os ductos coletores, vasos sanguíneos e os segmentos das Alças de Henle. As dosagens séricas de creatinina e uréia encontram-se dentro dos limites de normalidade. Desta forma, de acordo com os nossos resultados, podemos concluir que os animais afetados estudados, não apresentaram alterações estruturais ou funcionais dos rins, o que nos permitir sugerir que apesar da ingestão hídrica comprometida, a estrutura renal, mantem- se preservada nos animais GRMD.


Duchenne muscular dystrophy (DMD) is a severe myopathy of recessive X-linked character and the most relevant animal study model is the Golden Retriever muscular dystrophy (GRMD). In addition to the severe changes occurring in the striated musculature, several studies show that other structures, including viscera, may prove to be altered in this pathology. Thus, this study aimed to analyze and compare possible structural and functional alterations of the kidney in GRMD dogs. In this study model, it was possible to observe the presence of convex and concave faces, the renal hilum, and the cranial and caudal poles of the kidneys. The organ was surrounded by a fibrous capsule. In a sagittal section of the organ, the presence of the cortical and medullary regions and the renal pelvis were noticed. On microscopic examination, it was possible to identify the medullary and cortical zones and their structures: the renal corpuscles formed by the glomerulus and Bowman's capsule, the proximal and distal convoluted tubules, the collecting ducts, the blood vessels, and the segments of the loops of Henle. The serum creatinine and urea were within normal limits. Thus, according to our results, we may conclude that the affected animals under study showed no structural or functional changes in the kidneys, something which allows us to suggest that, despite the impaired water intake, renal structure remains preserved in GRMD animals.

BACKGROUND: The bone morphogenetic proteins (BMPs) belong to a unique group of proteins that includes the growth factor TGF-ß. BMPs play important roles in cell differentiation, cell proliferation, and inhibition of cell growth. They also participate in the maturation of several cell types, depending on the microenvironment and interactions with other regulatory factors. Depending on their concentration gradient, the BMPs can attract various types of cells and act as chemotactic, mitogenic, or differentiation agents. BMPs can interfere with cell proliferation and the formation of cartilage and bone. In addition, BMPs can induce the differentiation of mesenchymal progenitor cells into various cell types, including chondroblasts and osteoblasts. The aim of this study was to analyze the effects of treatment with rhBMP-2 on the proliferation of canine mesenchymal stem cells (cMSCs) and the tumor suppression properties of rhBMP-2 in canine osteocarcoma (OST) cells. Osteosarcoma cell lines were isolated from biopsies and excisions of animals with osteosarcoma and were characterized by the Laboratory of Biochemistry and Biophysics, Butantan Institute. The mesenchymal stem cells were derived from the bone marrow of canine fetuses (cMSCs) and belong to the University of São Paulo, College of Veterinary Medicine (FMVZ-USP) stem cell bank. After expansion, the cells were cultured in a 12-well Transwell system; cells were treated with bone marrow mesenchymal stem cells associated with rhBMP2. Expression of the intracytoplasmic and nuclear markers such as Caspase-3, Bax, Bad, Bcl-2, Ki-67, p53, Oct3/4, Nanog, Stro-1 were performed by flow citometry. RESULTS: We evaluated the regenerative potential of in vitro treatment with rhBMP-2 and found that both osteogenic induction and tumor regression occur in stem cells from canine bone marrow. rhBMP-2 inhibits the proliferation capacity of OST cells by mechanisms of apoptosis and tumor suppression mediated by p53. CONCLUSION: We propose that rhBMP-2 has great therapeutic potential in bone marrow cells by serving as a tumor suppressor to increase p53 and the pro-apoptotic proteins Bad and Bax, as well as by increasing the activity of phosphorylated caspase 3. STUDY DESIGN: Canine bone marrow mesenchymal stem cells associated with rhBMP2 in canine osteosarcoma treatment: "in vitro" study.
