RESUMO

The data in this article are related to the research publication "Digestion of micellar casein in duodenum cannulated pigs. Correlation between in vitro simulated gastric digestion and in vivo data" (Miralles et al., Food Chemistry, 2021, 343, 128428). Pig duodenum effluents were collected with a T-shaped cannula 15 min before and during digestion over 150 min after casein intake. The casein degradation profile of individual pigs during digestion is presented. All identified peptide sequences at different digestion times for six subjects are provided. The peptide profile of digests in the form of heat maps is shown for αs1-, αs2-, ß- and κ-casein. The sum of amino acids belonging to peptides released from ß- and αs1-casein has been used to determine correlation coefficients and range the inter-individual variability. Finally, the global amino acid composition, isoelectric point and sequence length of all released peptides has been determined.

Correlation and validation of the results of simulated gastrointestinal digestion of food compounds towards in vivo data is essential. The objective of this work was to monitor the digestion of milk micellar casein in the porcine upper intestinal tract and to match the outcome with the gastric in vitro digestion following the Infogest harmonized protocol. In pig duodenum, small amounts of intact caseins were present in all samples, while caseins were observed up to 60 min of gastric in vitro digestion. The peptide profile generated after in vitro and in vivo digestion showed clear similarities with specific overrepresented regions rich in proline and other hydrophobic residues. The statistical comparison of the in vivo and in vitro peptidome resulted in satisfactory correlation coefficients, up to 0.8. Therefore, the in vitro protocol used was a robust and simple model that provides a similar peptide profile than that found in porcine duodenum.

The production of caseinophosphopeptides from a casein-derived by-product generated during the manufacture of a functional ingredient based on antihypertensive peptides was attempted. The casein by-product was submitted to tryptic hydrolysis for 30, 60 and 120min and further precipitated with calcium chloride and ethanol at pH 4.0, 6.0 and 8.0. Identification and semi quantification of the derived products by tandem mass spectrometry revealed some qualitative and quantitative changes in the released caseinophosphopeptides over time at the different precipitation pHs. The by-product was also subjected to simulated gastrointestinal digestion. Comparison of the resulting peptides showed large sequence homology in the phosphopeptides released by tryptic hydrolysis and simulated gastrointestinal digestion. Some regions, specifically αS1-CN 43-59, αS1-CN 60-74, ß-CN 1-25 and ß-CN 30-50 showed resistance to both tryptic hydrolysis and simulated digestion. The results of the present study suggest that this casein-derived by-product can be used as a source of CPPs.

RESUMO

Azacitidine is now considered one of the standard-of-care agents for patients with high-risk myelodysplastic syndromes who are not candidates for high-dose chemotherapy. Considering the mechanism of action of the agent, it is critical to maintain adequate dose intensities for prolonged periods of time in order for treatment to be effective. Therefore, aggressive prevention as well as treatment of side effects is critical. The drug mainly causes hematological toxicity that is managed with growth factor support, blood transfusions, and dose and schedule adjustment. Nonhematological side effects are mainly gastrointestinal and cutaneous in nature, and can be easily managed with symptomatic treatment and correct administration techniques.

Antimicrobial peptides derived from food proteins constitute a new field in the combined use of antimicrobial agents in food. The best examples of milk-derived peptides are those constituted by bovine lactoferricin [lactoferrin f(17-41)] (LFcin-B) and bovine alpha(s2)-casein f(183-207). The aim of this work was to study if the antimicrobial activity of a natural compound employed in food preservation, nisin, could be enhanced by combination with the aforementioned milk-derived peptides. Furthermore, the possibility of a synergistic effect between these peptides and bovine lactoferrin (LF) against Escherichia coli and Staphylococcus epidermidis was also studied. Finally, the most active combinations were assayed against the foodborne pathogens Listeria monocytogenes and Salmonella choleraesuis. Results showed a synergistic effect when LFcin-B was combined with bovine LF against E. coli. In the same way, the combination of LFcin-B with bovine LF was synergistic against Staph. epidermidis. Bovine LF and nisin increased their antimicrobial activity when they were assayed together with bovine alpha(s2)-casein f(183-207). It is important to note the synergistic effect among LFcin-B and bovine LF, because both compounds might be simultaneously in the suckling gastrointestinal tract and could, therefore, have a protective effect on it. The other synergistic effect high-lighted is that between alpha(s2)-casein f(183-207) and nisin against L. monocytogenes because of the ability of L. monocytogenes to develop resistance to nisin.

RESUMO

Beta-lactoglobulin (beta-Lg) is currently an important source of biologically active peptides. These peptides are inactive within the sequence of the precursor protein, but they can be released by in vivo or in vitro enzymatic proteolysis. Once released, these peptides play important roles in the human health, including antihypertensive, antioxidant and antimicrobial activities as well as opioid-like features and ability to decrease the body-cholesterol levels. Bioactive peptides derived from beta-Lg are currently a point of intensive research. Their structure, biological significance and mechanism of action are briefly presented and discussed in this review.

A total of 107 different peptides, all derived from alphaS1-, alphaS2-, and beta-casein, were identified in different fractions of artisan or industrial Manchego cheese at 4 and 8 mo of ripening, and their sequences were examined. Most of these peptides are described for the first time in Manchego cheese. Taste characteristics (umami and bitter) were assigned based on their AA sequence and the position of these AA within the sequence. The umami taste was predominant in all fractions analyzed by the panelists, and the peptides EQEEL, QEEL, and EINEL, containing a high number of glutamic residues, were found within the fractions. However, in several fractions described as having umami characteristics, no peptides responsible for this taste were detected. Therefore, compounds other than peptides seem to be involved in the umami properties of water-soluble extracts lower than 1,000 Da of Manchego cheese.

RESUMO

Water-soluble fractions from Protected Denomination of Origin Manchego cheese, with molecular weight <1,000 Da, were fractionated using gel permeation chromatography and studied using both instrumental and sensorial analysis. In 2 of the fractions, panelists detected a floral, rose-like flavor. Analysis of these fractions by gas chromatography-mass spectrometry after simultaneous distillation extraction with dichloromethane identified 2-phenylethanol and phenylacetaldehyde as the compounds responsible for this flavor.

BACKGROUND: Legionella is a gram negative aerobic bacillus. A total of 42 species with 64 serogroups have been described, Legionella pneumophila serogroup 1 being the one that causes the disease most frequently. OBJECTIVE: Describe an outbreak of legionellosis with some characteristics different from those communicated previously. MATERIAL AND METHODS: Descriptive study of the epidemiological and clinical characteristics of the patients affected by an outbreak of pneumonia due to Legionella pneumophila serogroup 1, that occurred in Vigo in September and October, 2000, seen in the Hospital Xeral-Cies. RESULTS: A total of 33 patients, with a mean age of 61 years, were diagnosed. Fifty-nine percent had underlying diseases. The most frequent symptoms were fever, cough, dyspnea, diarrhea and awareness level alterations. Elevation of creatinphosphokinase in 79% and hyponatremia in 12%. The X-ray pattern observed most frequently was unilateral alveolar with predominant involvement of inferior lobes. A total of 77% received treatment with beta lactamics and macrolides, 13% beta lactamics and quinolones and 7% quinolones. Clinical and gasometric improvement were observed at a mean of 5 and 7 days, respectively. X-ray abnormalities improved in 62% in the first week. Eighteen percent had multiorgan failure and four patients died. In the analysis of the variables considered to have poor prognosis, only multiorgan failure demonstrated a significant association with mortality. DISCUSSION: The characteristics of the patients affected by an episode of legionellosis in our health care area were similar to those described in other outbreaks, except for the rapid resolution of the pulmonary infiltrates and low presence of hyponatremia. Multiorgan failure was the only factor associated with poor prognosis.

Introducción. La Legionella es un bacilo aerobio gramnegativo. Se han descrito 42 especies con 64 serogrupos, siendo Legionella pneumophila serogrupo 1 la que causa enfermedad con mayor frecuencia. Objetivo. Describir un brote de legionelosis con algunas peculiaridades distintas a las comunicadas con anterioridad. Material y método. Estudio descriptivo de las características epidemiológicas y clínicas de los pacientes afectados por un brote de neumonía por Legionella pneumophila serogrupo 1 ocurrido en Vigo durante septiembre y octubre del año 2000 atendidos en el Hospital Xeral-Cíes. Resultados. Se diagnosticaron 33 pacientes, con una edad media de 61 años. El 59% tenía enfermedades subyacentes. Los síntomas más frecuentes fueron fiebre, tos, disnea, diarrea y alteraciones del nivel de conciencia. Se detectó elevación de la creatinfosfoquinasa en el 79% e hiponatremia en el 12%. El patrón radiológico observado con mayor frecuencia fue alveolar unilateral con afectación predominante de lóbulos inferiores. El 77% recibió tratamiento con betalactámicos y macrólidos, el 13% betalactámicos y quinolonas y el 7% quinolonas. Se observó mejoría clínica y gasométrica a los 5 y 7 días de media, respectivamente. En el 62% las alteraciones radiológicas mejoraron en la primera semana. El 18% presentaron fallo multiorgánico y cuatro pacientes fallecieron. En el análisis de las variables consideradas de mal pronóstico, sólo el fracaso multiorgánico demostró una asociación significativa con la mortalidad. Discusión. Las características de los pacientes afectados por un brote de legionelosis en nuestra área sanitaria fueron similares a las descritas en otros brotes, a excepción de la rápida resolución de los infiltrados pulmonares y la baja presencia de hiponatremia. El fracaso multiorgánico fue el único factor que se asoció con mal pronóstico

In this study, a potent angiotensin-converting enzyme (ACE)-inhibitory activity was found in a commercial kefir made from caprine milk. The low molecular mass peptides released from caseins during fermentation were mainly responsible for this activity. Sixteen peptides were identified by HPLC-tandem mass spectrometry. Two of these peptides, with sequences PYVRYL and LVYPFTGPIPN, showed potent ACE-inhibitory properties. The impact of gastrointestinal digestion on ACE-inhibitory activity of kefir peptides was also evaluated. Some of these peptides were resistant to the incubation with pepsin followed by hydrolysis with Corolase PP. The ACE-inhibitory activity after simulated digestion was similar to or slightly lower than unhydrolyzed peptides, except for peptide beta-casein f(47-52) (DKIHPF), which exhibited an activity 8 times greater after hydrolysis.

El resultado más frecuente en una biopsia de próstata es el informe negativo del patólogo, debiendo decidir respecto a insistir en la búsqueda del cáncer. La rebiopsia a pacientes con APE menor a 10 ng/ml produce 10 por ciento de casos positivos para cáncer. Presentamos nuestra experiencia en rebiopsia. El hallazgo de cáncer se concentró en el grupo de pacientes con próstatas de menos de 40 grs. Ningún nuevo caso se encontró luego de una segunda biopsia entre pacientes con próstatas mayores a 90 grs. Pensamos que en un primer procedimiento de biopsia los falsos negativos se concentran entre los pacientes con próstatas de menor tamaño. En consecuencia nosotros biopsiamos todos los casos con 12 muestras desde el primer procedimiento.

A capillary electrophoresis method has been applied to the detection of illegal addition of milk from goat and/ or cow in Halloumi cheese, traditionally made with sheep milk. The electrophoretic profiles of the casein from Halloumi cheeses have revealed that caprine para-kappa-casein and bovine alphas1-casein peaks point to the presence of low percentages of goat's and/or cow's milk added to Halloumi cheese. Stepwise multiple linear regression has been used to predict these percentages with a standard error of the estimation of 2.14%. The analytical method combined with the statistical application is valid for the prediction of percentages higher than 2% of goat's and percentages of 5% of cow's milk added to the cheese either in fresh or ripened cheese. The standard error of estimation was higher for the prediction of cow's milk than for goat's milk.

Capillary electrophoresis (CE) was used to determine the whey protein to total protein ratio in raw and UHT milk samples with different degrees of proteolysis caused by storage. In raw milks, the analysis of samples taken at regular times demonstrated the influence of proteolysis in the whey protein to total protein determination, which was overestimated after 4 d of storage. In UHT milks, the overestimation of the whey protein to total protein ratio took place after 30 or 60 d of storage. However, the ratios alphaS1-CN/beta-CN and alphas1-CN/kappa-CN permitted detection of the samples of raw or UHT milk with degraded proteins. The distorted capillary electrophoretic pattern obtained for UHT milks made necessary an integration of the electropherograms in a "valley-to-valley" way. Results for raw milk samples were identical when "valley-to-valley" was compared to standard integration techniques. This CE method could be considered an alternative method to derivative spectroscopy for the determination of the whey protein to total protein of milk and could be used to detect samples with proteolysis.

Matrix-assisted laser desorption ionisation time-of-flight mass spectrometric analysis of the tryptic digest of beta-casein A2 and beta-casein B was performed before and after the separation of the peptides by LC. The overlapping of the chromatograms showed that all peaks were present in both samples, except for one only found in the tryptic digest of the A2 variant and two in the B variant. Experimental masses could be assigned to those peptides produced by tryptic digest of beta-casein variant. This peptide mapping strategy and current methodological improvements represent a promising tool for the identification of milk genetic variants with the difference of an amino acid substitution.

BACKGROUND AND AIMS: Biliary lipid absorption by the gall bladder mucosa and the cholesterol content of the gall bladder wall appear to play a role in cholesterol gall stone formation. As the scavenger receptor class B type I (SR- BI) regulates cellular cholesterol uptake, we studied its expression in human and murine gall bladders, its regulation by increased biliary lipid content, and its role in gall stone formation. METHODS AND RESULTS: Using immunohistochemistry, SR-BI was found in the apical domain of human gall bladder epithelial cells. Immunoblotting of isolated membranes from gall bladder epithelial cells showed a specific signal for the 82 kDa SR-BI protein. In C57BL/6 mice, SR-BI was also found in the gall bladder epithelium. Using western blot analysis, an inverse relationship was observed between biliary cholesterol concentration and SR-BI expression in murine gall bladder mucosa. By comparing lithogenic diet fed wild-type and SR-BI deficient mice, gall bladder wall cholesterol content and gall stone formation were not found to be dependent on SR-BI expression. CONCLUSIONS: (i) SR-BI is expressed in both human and murine gall bladder epithelium; (ii) biliary cholesterol hypersecretion is associated with decreased gall bladder SR-BI expression in mice; and (iii) murine SR-BI is not essential in controlling gall bladder wall cholesterol content and gall stone formation during diet induced cholelithiasis.
