Coturniculture has increased significantly in the last decades. There are several pathogens that can affect these birds. Among the diseases, fowl typhoid stands out as a disease with a potentially great impact to the poultry industry. The objective of this the study was to evaluate the effect of doses and administration routes of live 9R vaccine on protection of Japanese quails against experimental infection with Salmonella Gallinarum (SG). Two hundred and fifty birds were used, divided into five groups: G1, oral vaccination with one dose; G2, oral vaccination with 2 doses; G3, subcutaneous vaccination with one dose; G4, subcutaneous vaccination with two doses and G5 not vaccinated. All birds from all five groups were challenged with SG at an age of 45 days. SG was quantified in the periods of one, four, seven and twelve days after the challenge. The presence of clinical signs and macroscopic lesions of the disease were observed. The groups vaccinated by subcutaneous route had a higher egg production and lower mortality rate. Birds receiving a dose of the vaccine by subcutaneous route also showed lower amount of SG in the liver and spleen seven days after the challenge.(AU)


A coturnicultura tem aumentado significativamente nas últimas décadas. Existem vários patógenos que podem afetar essas aves. Entre as doenças, o tifo aviário se destaca como uma doença de grande impacto para a indústria avícola. O objetivo deste estudo foi avaliar o efeito de doses e vias de administração da vacina viva 9R na proteção de codornas japonesas contra infecção experimental por Salmonella Gallinarum (SG). Foram utilizadas duzentos e cinquenta aves, divididas em cinco grupos: G1, vacinação oral com uma dose; G2, vacinação oral com 2 doses; G3, vacinação subcutânea com uma dose; G4, vacinação subcutânea com duas doses e G5 não vacinado. Todas as aves dos cinco grupos foram desafiadas com SG aos 45 dias de idade. A SG foi quantificada nos períodos de um, quatro, sete e doze dias após o desafio. Foi observada a presença de sinais clínicos e lesões macroscópicas da doença. Os grupos vacinados por via subcutânea apresentaram maior produção de ovos e menor taxa de mortalidade. Aves recebendo uma dose da vacina por via subcutânea também apresentaram menor quantidade de SG no fígado e baço sete dias após o desafio.(AU)

In commercial layer poultry farming, molt induction is an important tool used by egg producers to prolong the production cycle of laying hens. Conventional molt induction programs involve total feed withdrawal, which raises questions about animal welfare and increased infection susceptibility. The high incidence of paratyphoid salmonellosis infections in commercial poultry farming is still an important health challenge because in addition to affecting the birds, such infections also cause public health problems. In this context, experiments were performed with laying hens at 79 wk of age to compare the conventional forced molting method (fasting) with an alternative method (free wheat bran supply) and determine their effect on the persistence of vaccine antibodies against Newcastle disease, the control and reduction of experimentally inoculated Salmonella Enteritidis, and the performance and egg quality of hens. A reduction (P < 0.05) of Salmonella Enteritidis in the crop and lower production of corticosterone were observed in the birds that received wheat bran compared with those subjected to total fasting. Moreover, a better performance (P < 0.05) with regard to egg production, egg mass, and feed conversion/kg and dozen eggs was observed in the hens that received the alternative treatment compared to the conventional forced molting method. Thus, the use of wheat bran for forced molting was found to be feasible and met the welfare needs of the hens.

This study evaluated the influence of the bird's age on the quality of the shell and percentage of bacterial penetration in commercial eggs. White-shelled commercial eggs were used, laid by light laying hens in their first laying cycle at 21, 39, 51, and 62 weeks of age. Shell quality evaluations comprised: egg weight, specific gravity, percentage and thickness of shell, number and size of pores. For evaluations regarding bacterial penetration, strains of several enterobacterias and one salmonella were used, all of which resistant to Nalidixic acid (100µg/ml). The method employed for evaluation of bacterial penetration was filling the eggs with growth medium. The data were subjected to variance analysis with 5% of probability using SAS (Education Analytical Software, 2013). Eveb though increase in the laying hen's age caused reduction of the quality of eggshells, it failed to affect the percentage of penetration of the bacterial samples evaluated.(AU)


O presente estudo avaliou a influência da idade da ave sobre a qualidade da casca e na porcentagem de penetração bacteriana em ovos comerciais. Foram utilizados ovos brancos comerciais provenientes de poedeiras leves em primeiro ciclo de postura com 21, 39, 51 e 62 semanas de idade. As avaliações de qualidade da casca realizadas foram: peso do ovo, gravidade específica, porcentagem e espessura da casca, número e tamanho dos poros. Para as avaliações da penetração bacteriana, foram utilizadas cepas de diversas enterobactérias e uma salmonela, sendo todas resistentes ao ácido nalidíxico (100µg/mL). O método utilizado para a avaliação da penetração bacteriana foi por meio do preenchimento dos ovos com meio de cultura. Os dados foram submetidos à análise de variância com 5% de probabilidade utilizando-se o programa SAS - Statistical Analisys System (Education Analytical Software, 2013). O aumento da idade da poedeira promoveu a redução da qualidade da casca dos ovos, porém não foi capaz de influenciar a porcentagem de penetração das amostras bacterianas avaliadas.(AU)

This study investigates the effects of different doses of serotonin, its precursor 5-hydroxytry-ptophan (5HTP), and m-hydroxybenzylhydrazine inhibitor (NSD1015), administered via intraperitoneal for 5 consecutive days, on behavior and average body weight of broilers. We also measured the humoral immune response and quantification of Salmonella Enteritidis in broilers chickens that received the drugs evaluated and a Lactobacillus pool. The study was divided into 3 experiments: Experiment 1--administration of pharmaceuticals with choice of dosage; Experiment 2--administration of pharmaceuticals and a Lactobacillus pool in birds that were not challenged with S. Enteritidis, and Experiment 3--administration of pharmaceuticals and a Lactobacillus pool in birds challenged with S. Enteritidis. The ELISA was used to scan dosages of intestinal IgA and serum IgY. We used colony-forming units to quantify S. Enteritidis. The concentrations of IgA and IgY did not show significant differences (P>0.05) in Experiment 2. In Experiment 3, NSD1015 associated with Lactobacillus determined higher IgA concentrations, promoting greater stimulus to the immune system than 5HTP. Regarding quantification of S. Enteritidis in the cecal content of birds, 5HTP associated to Lactobacillus determined the smallest number of bacteria, showing possible interaction of 5-hydroxytryptophan and Lactobacillus spp. with the immune system of broiler chickens.

A study was conducted to investigate the effects of organic acids (OA) and competitive exclusion product (CE) on growth performance, intestinal morphology, and concentration of volatile fatty acids in the cecal content. The experiment lasted for 10 wk. Four hundred twenty 1-d-old female commercial cross turkey poults (British United Turkeys, BUT Big 9) were distributed into 4 treatments with 5 replicates/pen of 21 birds each. The birds were fed a basal diet without growth promoter (control), diet with lincomycin (44 mg/kg), diet with organic acids (2 g/kg), and diet with product of CE (10(9) cfu/kg). Dietary levels of other nutrients, housing, and general management practices were similar for all treatments. On the first week (d 0-7), the BW and BW gain of the birds that fed diets with OA were lower than in the control group. In the fattening phase (d 28-70), the feed intake of the OA-treated group was lower than compared with the control. The birds that received diet with OA and CE product presented higher concentrations of propionic acid, at 14 d, and butyric acid in cecal content at 28, 56, and 70 d, compared with the control. Dietary inclusion of additives had no significant effects on intestinal villus height, crypt depth, and villus:crypt ratio. Organic acids had negative effects either on early gain or feed intake throughout the study. Because the test was conducted under controlled experimental conditions, the additives that showed results similar to those found by using antibiotics should be studied further in commercial farms to obtain results that can be incorporated into practice.

AIM: To evaluate the use of organic acids (OAs) and competitive exclusion (CE) product administered continuously in the feed and transiently in drinking water on the control of Salmonella enterica subspecie enterica serotype Enteritidis (SE) prior to slaughter. METHODS AND RESULTS: The influence of treatments were evaluated on pH, population of the lactic acid bacteria (LAB) and bacteria of the family Enterobacteriaceae, concentration of volatile fatty acids and SE colonization in the crop and caecum. The birds were challenged with SE 24 h before being slaughtered, and then, the caeca and crop were removed and subjected to SE counts. Continuous administration of OAs reduced the population of bacteria from the Enterobacteriaceae family in both crop and caecum, positively influenced the butyric acid concentration and reduced SE colonization in the caecum. The diet supplemented with CE product positively influenced the quantity of LAB in the crop and caecum, elevated the butyric acid concentration and reduced both Enterobacteriaceae quantity and SE colonization in the caecum. There was no effect from administering the treatments via drinking water on the variables measured. CONCLUSIONS: Continuous supplementation in feed with OAs and CE product reduced SE colonization of the caeca. SIGNIFICANCE AND IMPACT OF THE STUDY: Supplementation of OAs and CE product in diet to turkeys can reduce the SE load, potentially leading to a lower contamination risk of meat during slaughter.

This study investigated the immune response of broiler chickens with oral treatment of a Lactobacillus spp. pool (PL) associated with microencapsulated recombinant proteins flagellin (FliC) and the subunit B of cholera toxin (CTB). Immune responses were evaluated by measuring IgA from intestinal fluid, serum IgY, and immunostaining of CD8(+) T lymphocytes present in the cecum. The evaluations were performed on d 0, 7, 14, 21, and 28 posttreatment. A significant increase (P < 0.05) was observed in IgA levels in all immunized groups, especially 3 wk after immunization. Treatments 2 (recombinant CTB) and 3 (recombinant FliC+CTB) showed the highest concentrations. Similarly, serum concentrations IgY (µg/mL) increased along the experiment, and the means for treatments 2 and 3 showed significant differences (P < 0.05) compared with controls, reaching concentrations of 533 and 540 µg/mL, respectively. The number of CD8(+) T lymphocytes in all treatments greatly differed (P < 0.05) compared with the negative control at 21 d posttreatment. However, only treatment 2 (recombinant CTB), 4 (PL), and 5 (recombinant FliC+ recombinant CTB + PL) remained significantly (P < 0.05) different from the control at 28 d posttreatment. Thus, it is concluded that the microencapsulated recombinant proteins administered orally to broiler chickens are capable of stimulating humoral and cellular immune response, and the combinations of these antigens with Lactobacillus spp. can influence the population of CD8(+) T cells residing in the cecum.

The effect of a commercial organic acid (OA) product on BW loss (BWL) during feed withdrawal and transportation, carcass yield, and meat quality was evaluated in broiler chickens. Two experiments were conducted in Brazil. Commercial houses were paired as control groups receiving regular water and treated groups receiving OA in the water. Treated birds had a reduction in BWL of 37 g in experiment 1 and 32.2 g in experiment 2. In experiment 2, no differences were observed in carcass yield between groups. Estimation of the cost benefit suggested a 1:16 ratio by using the OA. In experiment 3, conducted in Mexico, significant differences on water consumption, BWL, and meat quality characteristics were observed in chickens that were treated with the OA (P < 0.05). These data suggest this OA product may improve animal welfare and economic concerns in the poultry industry by reducing BWL and improving meat quality attributes.

The protection level against Salmonella Enteritidis was evaluated in chickens after in ovo treatment with different species of Lactobacillus spp. inoculated into the air cell or by immersion in broth culture. Two hundred forty embryonated eggs were distributed into 8 groups, corresponding to treatments with Lactobacillus acidophilus, Lactobacillus fermentum, Lactobacillus salivarius, and control. On d 18 of incubation, 4 groups were inoculated with 0.1 mL of inoculum in the air cell and 4 groups were immersed for 3 min in culture of each treatment. Two days after hatching, 0.5 mL of Salmonella Enteritidis culture was inoculated by the intraesophageal route. On d 5 of life, the chicks were euthanized and the ceca were processed to obtain Salmonella Enteritidis counts. There was no decrease in Salmonella Enteritidis colonization of chick ceca, regardless of treatment or route of administration. Lactobacillus spp. samples used in the treatment showed no probiotic potential in chicks when inoculated in ovo, in relation to Salmonella Enteritidis inhibition in poultry ceca.

This study aimed to characterize the in vitro and in vivo adhesion and immunomodulatory effect of Lactobacillus strains isolated from chickens. Lactobacillus samples isolated from 65-wk-old birds were identified by PCR; their adhesion was evaluated in vitro via basement membrane-type cell matrix and in vivo through carboxyfluorescein succinimidyl amino ester staining inoculation in 1-d-old birds and duodenum, jejunum, ileum, and cecum collections at 1, 4, 12, and 24 h after inoculation. The 5 best adhesive samples at the in vitro test formed a pool for total IgA and IgG measurement in sera and intestinal fluid. The birds were divided into groups by inoculation scheme: group 1 was treated with a pool of Lactobacillus spp. at 2-d-old and challenged 1 d later with Salmonella Enteritidis and then treated again with a pool of Lactobacillus spp. at 4 d of age; group 2 was treated with a pool of Lactobacillus spp. at 2 and 4 d of age; group 3 was challenged with Salmonella Enteritidis at 3 d of age; and group 4 was a negative control. Collections were taken at 7, 14, 21, 28, and 35 d after the first inoculation. The results suggest that basement membrane matrix use represents an important technique for triage of samples for subsequent in vivo evaluation and that carboxyfluorescein succinimidyl amino ester staining is efficient for identifying this bacterial characteristic. The Lactobacillus-treated groups (1 and 2) presented the highest IgA concentrations at the end of the experiment (12,054.6 and 10,568.4 ng/mL, respectively). The group 2 IgG values in intestinal fluid exceeded those of the other 3 groups (P < 0.05), peaking at 6.419 ng/mL. In most serum collections, the Lactobacillus-treated groups (1 and 2) did not differ significantly in IgG concentrations (P > 0.05), whereas group 3 presented the highest concentration of this antibody. It is concluded that there was greater adhesion of strains in the cecum and an important correlation between in vitro and in vivo results. These results also suggest the immunomodulatory action of Lactobacillus spp. in the chicken.

The objective of this work was to verify the possibility of transference of resistance to the antimicrobials between bacteria that are in the present normal microbiota of chickens and Salmonella Enteritidis. Samples of Lactobacillus spp. (L. spp.), Salmonella Enteritidis (SE) and Escherichia coli (E. coli) previously isolated from chickens, selected after the test of sensitivity antimicrobial in vitro according the standard method (National Committee for Clinical Laboratory Standards) utilizing those with resistance and sensibility to the antimicrobials inductors, named donor and receptor bacteria, respectively were used. Antimicrobials inductors were utilized to stimulate the transference of resistance to the antimicrobials between the bacteria. The possibility of transference was verified from the E. coli resistant to the SE and L. spp. Transference of a sample of L. spp resistant to the antimicrobials inductors to the SE was also verified. It was only possible to verify the transference of the resistance to the antimicrobials inductor when the donor bacteria was the E. coli and the bacteria receptor was SE. In the present study we conclude that the transference of resistance to the antimicrobials between bacteria is possible, however, not all bacteria participate in that trial, not transmitting and neither acquiring this resistance.


O objetivo deste trabalho foi verificar a possibilidade de transferência de resistência aos antimicrobianos entre bactérias normais da microbiota de frangos e Salmonella Enteritidis. Utilizamos amostras de Lactobacillus spp. (L. spp.), Salmonella Enteritidis (SE) e Escherichia coli (E. coli) previamente isolados de frangos, selecionados após prova de sensibilidade antimicrobiana in vitro conforme metodologia padrão (Comitê Nacional para Padrões Clínicos de Laboratório). Utilizamos aqueles com resistência e sensibilidade aos antimicrobianos indutores, chamados de bactérias doadoras e receptoras, respectivamente. Os antimicrobianos indutores foram utilizados para estimular a transferência de resistência aos antimicrobianos entre as bactérias. A possibilidade de transferência foi verificada da E. coli resistente para a SE e L. spp. Também foi verificada a transferência de uma amostra de L. spp resistente aos antimicrobianos indutores para a SE. Só foi possível verificar a transferência da resistência aos antimicrobianos indutores quando a bactéria doadora foi a E. coli e a bactéria receptora foi a SE. No presente estudo concluímos que a transferência de resistência aos antimicrobianos entre bactérias é possível, mas nem todas as bactérias participam desse evento, não transmitindo e nem adquirindo esta resistência.

O presente estudo avaliou a susceptibilidade de amostras de Escherichia coli isoladas de quadros de colissepticemia aviária da região centro-oeste paulista pelo Laboratório de Ornitopatologia da FMVZ-UNESP/Botucatu, SP. Constatou-se um grande número de amostras multirresistentes aos antibióticos testados, onde as drogas menos efetivas foram sulfonamida e tetraciclina. Todas as amostras mostraram-se sensíveis a norfloxacina e gentamicina.


The present study evaluated the susceptibility of Escherichia coli samples isolated from poultry with colisepticemia in midwestern São Paulo State at the Ornitopathology Laboratory of FMVZ-UNESP/Botucatu, SP. A large number of samples were found that were multi-resistant to the antibiotics tested, the less effective drugs being sulfonamide and tetracycline. All samples were susceptible to norfloxacin and gentamicin.

Inglúvios e cecos de reprodutoras comerciais de frangos de corte foram utilizados para o isolamento de Lactobacillus spp. As estirpes isoladas foram caracterizadas como Gram-positivo, catalase negativo, produtoras de gás em glicose, não produtoras de H2S em triple sugar iron e identificadas pela reação em cadeia da polimerase como Lactobacillus reuteri e Lactobacillus salivarius. A utilização da técnica spot-on-the-lawn para avaliação da inibição in vitro permitiu a análise de vários microrganismos simultaneamente. Todas as estirpes isoladas inibiram in vitro S. Enteritidis fagotipo 4, S. Enteritidis fagotipo 28, S. Typhimurium, S. Pullorum, S. Agona, S. Anatum, S. Dublin e S. Senftenberg.


Crops and ceca of commercial broiler breeders were used for the isolation of Lactobacillus spp. The isolated samples were characterized by Gram positive staining, negative catalase test, production of gas from glucose, negative for H2S production from triple sugar iron, and were identified by the polymerase chain reaction as Lactobacillus reuteri and Lactobacillus salivarius. The use of the spot-on-the-lawn technique, with modifications, for the evaluation of the in vitro inhibition made it possible the simultaneous analysis of several microrganisms. All the crop and ceca isolated microorganisms presented an in vitro inhibitory effect against strains of Salmonella Enteritidis fagotype 4, S. Enteritidis fagotype 28, S. Typhimurium, S. Pullorum, S. Agona, S. Anatum, S. Dublin, and S. Senftenberg.

Lactobacilos foram isolados do inglúvio e cecos de reprodutoras pesadas e caracterizados como Gram-positivo, catalase negativo, produtores de gás em glicose e não produtores de H2S em triple sugar iron e pela fermentação de carboidratos. Utilizaram-se os iniciadores: Lac 1/23-10C para detecção de Lactobacillus acidophilus, L. crispatus, L. amylovorus, L. gasseri, L. helveticus e L. jensenii; Lac 2/LU-1' para L. acidophilus; Fer 3/Fer 4 para L. fermentum; Reu 1/Reu 2 para L. reuteri e Sal 1 e Sal 2 para L. salivarius. L. reuteri e L. salivarius foram identificados pela reação em cadeia de polimerase (PCR) e pelo teste bioquímico, enquanto L. acidophilus, L. fermentum e Lactobacillus sp. somente pelo teste bioquímico. Os resultados obtidos na PCR foram mais precisos quando comparados aos obtidos com o método bioquímico, que demonstrou ser subjetivo devido às variações na fermentação de carboidratos, principalmente na diferenciação entre L. fermentum e L. reuteri.


Lactobacilli were isolated from crops and ceca of broiler breeders and characterized by positive Gram staining, negative catalase test, production of gas from glucose, and negative for H2S production from triple sugar iron, and carbohydrates fermentation. Primers: Lac1/23-10C for detecting Lactobacillus acidophilus, L. crispatus, L. amylovorus, L. gasseri, L. helveticus, and L. jensenii; Lac2/LU-1' for L. acidophilus; Fer3/Fer4 for L. fermentum; Reu1/Reu2 for L. reuteri, and Sal1/Sal2 for L. salivarius were used. L. reuteri and L. salivarius were identified by both polymerase chain reaction (PCR) and biochemical tests. However, L. acidophilus, L. fermentum, and Lactobacillus sp. were only identified by biochemical tests. PCR results were more precise, considering the variability of carbohydrate fermentation among the strains, especially for identifying L. fermentum and L. reuteri.

RESUMO

The aim of this study was to evaluate the occurrence of potentially zoonotic intestinal protozoan infections in exotic and wildlife Brazilian birds. Fecal samples from 207 birds of 45 species were examined. Infections by Balantidium sp., Entamoeba sp., and Blastocystis sp. were observed in 17 individuals (8.2%) of Gnorimopsar chopi, Oryzoborus angolensis, Sporophila caerulescens, Ramphastos toco, Aratinga leucophtalmus, and Pavo cristatus.

Because of recent interest in bacteriophage therapy in poultry, information regarding the interaction of bacteriophages and potential host bacteria in the environment should be collected. The present studies were initiated with a rather typical commercial broiler integrator within the south-central United States to examine environmental Salmonella levels in two broiler complexes, attempt to isolate Salmonella-lytic bacteriophages, and elucidate a possible reason for differing apparent Salmonella prevalence. Significantly (P < 0.05) less Salmonella was isolated from houses in complex 1 (15/44 [34%] Salmonella-positive drag swabs) as compared to houses in complex 2 (22/24 [92%]). A total of seven Salmonella-lytic bacteriophages were isolated from Salmonella-positive environments, and two bacteriophages were isolated from a single Salmonella-negative house. During the initial bacteriophage isolation, individual bacteriophages did not replicate in the Salmonella host isolated from the same environment, and lysis of additional Salmonella hosts relied on high numbers of bacteriophage to be present. This suggests that the presence of these bacteriophages in the environment of a commercial broiler house had little to no effect on the presence of Salmonella. This study highlights the need to find additional bacteriophage sources, more effective isolation methods, and more innovative approaches to using bacteriophages to treat enteric disease.

The expression of immune response in the form of leukocytic infiltrate by CD3+, CD4+, and CD8+ cells in the epithelium and in the intestinal lamina propria of chicks was studied in the present work by means of immunohistochemical reaction. The chicks were treated with Lactobacillus spp. or cecal microflora (CM) and experimentally challenged or not with Salmonella enterica serovar Enteritidis. The 320 birds utilized were divided into 4 groups containing 80 chicks each and submitted to treatments with Lactobacillus reuteri, Lactobacillus salivarius, Lactobacillus acidophilus, and CM. Each group was subdivided into 4 subgroups of 20 birds each and classified into a subgroup that did not receive treatment (negative control), subgroup treated, subgroup treated and challenged with Salmonella Enteritidis, and subgroup only challenged with Salmonella Enteritidis (positive control). The results obtained show that the treatment with L. reuteri, L. salivarius, L. acidophilus, or CM and challenged or not with Salmonella Enteritidis determine immune response in the form of leukocytic infiltrate by CD3+ and CD8+ lymphocytes followed by CD4+ in the epithelium and in the lamina propria of the duodenum, jejunum, and cecum of chicks up to 12 d of age. The quantity of CD3+ lymphocytes was significantly higher (P < 0.05) in the intestine of chicks treated with L. acidophilus or CM and challenged or not with Salmonella Enteritidis; however, the higher quantity of CD8+ lymphocytes was in the intestine of chicks treated with CM and challenged with Salmonella Enteritidis. The duodenum was the segment in which the immune response by T cells (CD3+, CD4+, and CD8+) was stimulated with the greatest intensity, followed by, respectively, the jejunum and cecum. The quantity of CD3+ lymphocytes present in the duodenum, jejunum, and cecum increases with the age of chicks, independent of the stimulus determined by treatments or challenge.

Salmonella enterica serovar Enteritidis-lysing bacteriophages isolated from poultry or human sewage sources were used to reduce Salmonella Enteritidis in vitro and in experimentally infected chicks. Cocktails of 4 different bacteriophages obtained from commercial broiler houses (CB4Ø) and 45 bacteriophages from a municipal wastewater treatment plant (WT45Ø) were evaluated. In experiment 1, an in vitro crop assay was conducted with selected bacteriophage concentrations (10(5) to 10(9) pfu/mL) to determine ability to reduce Salmonella Enteritidis in the simulated crop environment. Following 2 h at 37 degrees C, CB4Ø or WT45Ø reduced Salmonella Enteritidis recovery by 1.5 or 5 log, respectively, as compared with control. However, CB4Ø did not affect total SE recovery after 6 h, whereas WT45Ø resulted in up to a 6-log reduction of Salmonella Enteritidis. In experiment 2, day-of-hatch chicks were challenged orally with 3 x 10(3) cfu/chick Salmonella Enteritidis and treated cloacally with 1 x 10(9) WT45Ø pfu/chick 1 h postchallenge. One hour later, chicks were treated or not with a commercially available probiotic (Floramax-B11). Both treatments significantly reduced Salmonella Enteritidis recovery from cecal tonsils at 24 h following vent lip application as compared with controls, but no additive effect was observed with the combination of bacteriophages and probiotic. In experiment 3, day-of-hatch chicks were challenged orally with 9 x 10(3) cfu/chick Salmonella Enteritidis and treated via oral gavage with 1 x 10(8) CB4Ø pfu/chick, 1.2 x 10(8) WT45Ø pfu/chick, or a combination of both, 1 h postchallenge. All treatments significantly reduced Salmonella Enteritidis recovered from cecal tonsils at 24 h as compared with untreated controls, but no significant differences were observed at 48 h following treatment. These data suggest that some bacteriophages can be efficacious in reducing SE colonization in poultry during a short period, but with the bacteriophages and methods presently tested, persistent reductions were not observed.

Pintos de corte com um dia de idade foram tratados com microbiota cecal cultivada em condição de aerobiose, nos tempos de congelamento de 90, 200, 290 e 360 dias, e associada aos crioprotetores sacarose, trealose, dimetilsulfóxido (DMSO) e glicerol. Posteriormente as aves foram desafiadas com Salmonella Enteritidis, visando determinar a eficácia dos tratamentos em relação à quantidade de bactérias viáveis da microbiota que foi maior aos 90 dias (10,58 Log10 UFC/ml), quando as aves foram tratadas com sacarose, e menor aos 290 dias, quando tratadas com glicerol (7,73 Log10 UFC/ml). No tempo zero, todas as aves apresentaram Salmonella (100 por cento) quando tratadas com DMSO e glicerol, com colonização cecal de 4,9 e 5,2 Log10 UFC/g do conteúdo cecal, respectivamente; aos 360 dias nenhuma ave foi infectada, independente do tratamento. A microbiota cecal, independente de tratamento, sempre determinou menor quantidade de S. Enteritidis em qualquer um dos parâmetros pesquisados, quando comparada com a das aves não tratadas. O congelamento em nitrogênio líquido foi eficaz na manutenção da viabilidade da microbiota cecal até 360 dias.


One-day-old broiler chicks were treated with cecal microbiota cultivated under aerobiose conditions, frozen during 90, 200, 290 and 360 days and associated with different cryoprotectors such as sucrose, trehalose, DMSO and glycerol. Subsequently, the birds were challenged with Salmonella Enteritidis in order to determine the efficacy of the different treatments in relation to the quantity of viable bacteria, which was higher at 90 days when treated with sucrose (10.58 log10 CFU/ml) and lower at 290 days when treated with glycerol (7.73 log10 CFU/ml). The quantity of infected birds was 100 percent in 0 time, when the cecal colonization by S. Enteritidis was 4.9 and 5.2 log10 CFU/g of cecal content, respectively treated with DMSO and glycerol. No bird was infected at 360 days, irrespectively of the treatment. In all treatments, the cecal microbiota always determined a lesser quantity of S. Enteritidis for all the studied parameters compared to non-treated birds. Frozen in liquid nitrogen was effective in maintaining the viability of cecal microbiota during the experimental period of 360 days.

Ovos embrionados provenientes de matrizes pesadas foram inoculados na câmara de ar com microbiota cecal total, microbiota cecal diluída e cultura de Lactobacillus salivarius, no 18° dia de incubação. Dois dias após o nascimento, as aves foram desafiadas com Salmonella enterica sorovar Enteritidis (SE) e, cinco dias após o desafio, avaliou-se a presença da bactéria no fígado e ceco. O efeito de exclusão competitiva, após o desafio com SE, somente foi observado pela ausência da bactéria no fígado das aves tratadas in ovo com L. salivarius. A inoculação in ovo de microbiota cecal indefinida ou diluída não reduziu a colonização de SE no fígado e no ceco das aves, incluindo, neste último, também o tratamento com L. salivarius. Nenhum dos tratamentos in ovo determinou índice de eclodibilidade superior a 65 por cento.


Commercial 18-day-old incubating chicken embryos were inoculated with total or diluted cecal microbiota and Lactobacillus salivarius cultures directly into the inner air sac. Two days after hatching, the chicks were challenged with Salmonella enterica serovar Enteritidis (SE), and five days later the presence of bacteria in cecum and liver was evaluated. The competitive exclusion effect was determined by the search for SE in the liver of chicks treated in ovo with L. salivarius and challenged with SE. The in ovo inoculation of total or diluted cecal microbiota, in addition to the L. salivarius treatment did not significantly decrease the colonization of SE in liver and cecum. All treatments resulted in hatchability of 65 percent or less.
