RESUMO

The aim of this study was to evaluate the reasons of quality deviation of a concentrate from a predefined standard. Five treatments were established: T1 - Control, standard concentrate formulation (SCF); T2 - PXMore5, SCF with more 5% vitamin-mineral premix (VMP); T3 - PXLess5, SCF with less 5% VMP. All three treatments used a 400kg batches in an INTECNIAL mixer; T4 - FeedMixer, SCF using a 4,000kg batch in an IMOTO mixer; T5 - PremixMixer, SCF using a 1,200kg batch in an MUYANG mixer. For each treatment, bags of 20 kg were stored in three storage places for four months. Water activity of concentrate was affected by temperature and air relative humidity in different storage places. Regarding the kind of mixer, the greatest variation in concentration of crude protein, mineral residue, copper, zinc, and selenium was observed in the PremixMixer. Adjustments are imperative in the handling and use procedures of this kind of mixer to meet the quality requirements required in the concentrate production. Analyzing the effect of the mineral-vitamin premix level, no difference could be defined with the evaluated parameters.(AU)


O objetivo deste estudo foi avaliar as razões do desvio de qualidade de um concentrado de um padrão predefinido. Foram estabelecidos cinco tratamentos: T1 - controle, concentrado com formulação padrão (CFP); T2 - PXMais5, CFP com 5% a mais de vitaminas e minerais da pré-mistura (PVM); T3 - PXMenos5, CFP com 5% a menos de PVM. Todos esses três tratamentos utilizaram lotes de 400kg em um misturador INTECNIAL; T4 - FeedMixer, CFP usando um lote de 4.000kg em um misturador IMOTO; T5 - PremixMixer, CFP usando um lote de 1.200kg em um misturador MUYANG. Para cada tratamento, sacos de 20kg foram armazenados em três ambientes distintos por quatro meses. A atividade de água do concentrado foi afetada pela temperatura e umidade relativa do ar em diferentes locais de armazenamento. Em relação ao tipo de misturador, a maior variação na concentração de proteína bruta, resíduo mineral, cobre, zinco e selênio foi devido ao PremixMixer. Ajustes são imperativos nos procedimentos de manuseio e uso desse tipo de misturador para atender aos requisitos de qualidade exigidos na produção de concentrado. Ao se analisar o efeito do nível da pré-mistura de vitaminas e minerais, nenhuma diferença pôde ser definida com os parâmetros avaliados.(AU)

The objective of this study was to investigate the frequency of intestinal parasitic infections and infection with Toxocara spp. in children from a day care center in the city of Rio Grande, Rio Grande do Sul, Brazil. Methods: The study was conducted using fecal samples from 50 children, ages three to six years, using Ritchie, Faust, Rugai, and Kinyoun's techniques. A closed and structured epidemiological questionnaire was used to collect data from the parents/guardians of the children. Serological studies for detection of antibodies to Toxocara spp. by immunoenzymatic assays using Toxocara excretion and secretion antigen (TES) were conducted on sera collected from 41 children. Results: The frequency of enteroparasites was 18%, with 43.9% of the children testing seropositive for Toxocara spp. Low family income and low literacy levels of parents/guardians were common factors between the families of the parasite-positive children. For the children who tested positive for Toxocara spp., most of the parents/guardians reported using only water for sanitizing raw vegetables and fruits. Conclusions: The high seropositivity rates for Toxocara spp. indicate that children were exposed to this parasite, and it is important to reduce the risk of infection. In addition, the seropositivity for enteroparasites and the interviews with the parents/guardians indicate the need to educate the studied population regarding the modes of intestinal parasite transmission and the prophylactic measures needed to prevent their dissemination.

Métodos: Para o estudo, foram coletadas amostras fecais de 50 crianças de três a seis anos, analisadas pelas técnicas de Ritchie, Faust, Rugai e Kinyoun, sendo aplicado um questionário epidemiológico fechado e estruturado aos pais/responsáveis ​​das crianças. Também foi realizada uma pesquisa sorológica de anticorpos para Toxocara spp. em 41 crianças, pelo ensaio imunoenzimático, associado ao antígeno de excreção e secreção (ES), com soros pré-adsorvidos com antígeno somático de Ascaris lumbricoides. Resultados: A frequência de enteroparasitos foi de 18%, enquanto que 43.9% das crianças foram soropositivas para Toxocara spp.. Baixa renda familiar e baixo nível de alfabetização dos pais/responsáveis ​​foram fatores comuns entre as famílias das crianças parasitadas. A maioria dos pais ou responsáveis das crianças, com diagnóstico positivo para enteroparasitos ou para Toxocara spp., relatou utilizar apenas água para a higienização de verduras e frutas. Conclusões: As altas taxas de soropositividade para Toxocara spp. indicam que as crianças foram expostas a esse parasito, sendo importante minimizar o risco de infecção. Além disso, a positividade de 18% para enteroparasitos e os dados obtidos com as entrevistas realizadas com os pais ou responsáveis demonstram a necessidade da realização de trabalhos com a população estudada que visem a divulgação sobre as parasitoses intestinais, especialmente sobre medidas profiláticas especificas.

CLINICAL RELEVANCE: The Monobond Etch & Prime seems to be an efficient option for adhesive cementation of ultrathin veneers and full crown ceramic with good properties after two years of clinical follow-up. SUMMARY: The use of the self-etching ceramic primer combines the stages of acid conditioning and silanization in cementation procedures of ceramic restorations. The protocol is a simpler and safer alternative to the conventional protocol for surface treatment of silica-based ceramics. This case report describes the steps of an esthetic rehabilitation with ultrathin veneers and full crown based on lithium disilicate treated with a ceramic primer (Monobond Etch & Prime, Ivoclar Vivadent, Schaan, Liechtenstein). After two years of clinical follow-up, the restorations presented satisfactory esthetic and functional performance, color stability, surface and marginal integrity, and absence of cracks and debonding. More research is needed to investigate the clinical performance and longevity of the ceramic restorations treated with self-etching ceramic primers.

Avaliou-se a composição química e a resistência óssea do tibiotarso de frangos de corte aos 21 dias de idade. Foram determinados os percentuais ósseos de proteínas colagenosas (PC) e proteínas não colagenosas (PNC) e de cálcio, fósforo, potássio e sódio. Foram utilizados 650 pintinhos machos de marca comercial, alimentados com dietas à base de milho e farelo de soja. Foi utilizado delineamento em blocos ao acaso com cinco repetições e 26 aves por unidade experimental. Os tratamentos consistiram na suplementação da dieta basal com NH4Cl a fim de se obter cinco níveis -50; 0; 50; 100 e 150mEq/kg de balanço eletrolítico (BE). O nível de BE influenciou os teores de fósforo, potássio, sódio, PC e PNC, relação Ca:P e a resistência à quebra. A redução do balanço eletrolítico da dieta em nível inferior a 150mEq/kg influenciará negativamente a mineralização e a resistência óssea. A resistência à quebra do tibiotarso não está correlacionada com as concentrações dos minerais de forma individual, mas correlaciona-se negativamente com as concentrações de proteínas colagenosas e não colagenosas.


This study was carried out in order to evaluate the bone chemical composition and breaking force resistance of tibiotarsus birds at 21 days of age. The bone percentage of colagenous proteins (CP), non colagenous proteins (NCP) and minerals (calcium, phosphorus, potassium and sodium) was analyzed. A total of 650 commercial male broiler chicks were fed corn and soybean diets. A completely randomized block design with five replications of 26 birds per experimental unit was used. The treatments consisted of the basal ration supplemented with NH4Cl in order to obtain five levels (-50; 0; 50; 100 and 150mEq/kg) of electrolyte balance. The EB level affected the percentages of phosphorus, potassium and sodium, PC and PNC, Ca: P relation and breaking force resistance. The reduction of EB diets at levels below 150mEq/kg will affect negatively the mineralization and bone resistance. The breaking force of tibiotarsus is not correlated with the mineral concentration individually, but correlates negatively with the concentration of collagenous and non-collagenous proteins.

Determinaram-se os melhores valores de balanço eletrolítico (BE) para o desempenho de frangos de corte de um a sete, um a 14, um a 21 e um a 42 dias de idade. Utilizaram-se 720 pintos comerciais, machos, criados em galpão de alvenaria, em boxes cobertos com maravalha, alimentados com dieta basal composta por milho e farelo de soja combinada com valores de BE de 200; 250; 300; 350 e 400mEq/kg. Adotou-se o delineamento inteiramente ao acaso, com seis repetições de 24 aves para cada uma das 30 unidades experimentais. Avaliaram-se o ganho de peso, o consumo da dieta e a conversão alimentar aos sete, 14, 21 e 42 dias de idade. O melhor BE estimado para o ganho de peso aos 21 e 42 dias foi de 270mEq/kg, estimado pelo modelo quadrático de regressão. Para as demais variáveis, não houve diferenças entre os BE. Recomenda-se um valor de BE entre 270 e 300mEq/kG para frangos de corte para as fases inicial e de crescimento em regiões de clima quente.


A trial was carried out to determine the best electrolyte balance value (EB) for broilers performance from 1 to 7, 1 to 14, 1 to 21 and 1 to 42 days of age. A total of 720 one-day-old chicks, males, were reared in the floor covered with shaving woods and fed corn-soybean base diets combined with 200; 250; 300; 350 e 400mEq/kg of EB. A completely randomized experimental design with six replicates of 24 chicks each and 36 experimental units was used. Weigh gain (WG), feed intake (FC) and feed efficiency (FE) were evaluated at 7, 14, 21 and 42 days of age. The best values of EB estimated for the WG at 21 and 42 days was 70mEq/kg, estimated by a quadratic regression model. No differences among treatments were observed for the other studied traits. The results suggested EB should be around 270 up to 300mEq/kg in high temperature regions, for broiler chicks in inicial and growth phases.

We have previously demonstrated that Na+, K(+)-ATPase activity is present in both differentiated plasma membranes from Electrophorus electricus (L.) electrocyte. Considering that the alpha subunit is responsible for the catalytic properties of the enzyme, the aim of this work was to study the presence and localization of alpha isoforms (alpha1 and alpha2) in the electrocyte. Dose-response curves showed that non-innervated membranes present a Na+, K(+)-ATPase activity 2.6-fold more sensitive to ouabain (I50=1.0+/-0.1 microM) than the activity of innervated membranes (I50=2.6+/-0.2 microM). As depicted in [3H]ouabain binding experiments, when the [3H]ouabain-enzyme complex was incubated in a medium containing unlabeled ouabain, reversal of binding occurred differently: the bound inhibitor dissociated 32% from Na+, K(+)-ATPase in non-innervated membrane fractions within 1 h, while about 50% of the ouabain bound to the enzyme in innervated membrane fractions was released in the same time. These data are consistent with the distribution of alpha1 and alpha2 isoforms, restricted to the innervated and non-innervated membrane faces, respectively, as demonstrated by Western blotting from membrane fractions and immunohistochemical analysis of the main electric organ. The results provide direct evidence for a distinct distribution of Na+, K(+)-ATPase alpha-subunit isoforms in the differentiated membrane faces of the electrocyte, a characteristic not yet described for any polarized cell.

The effects of tricyclic antidepressants drugs (TCA) amitriptyline, imipramine and nortriptyline, on purified Electrophorus electricus (L.) acetylcholinesterase (AChE; acetylcholine hydrolase, EC 3.1.1.7) were studied using kinetic methods and specific fluorescent probe propidium. The antidepressants inhibited AChE activity by a non-competitive mechanism. Inhibition constants range from 200 to 400 microM. Dimethylated amitriptyline and imipramine were more potent inhibitors than the monomethylated nortriptyline. Fluorescence measurements using bis-quaternary ligand propidium were used to monitor ligand-binding properties of these cationic antidepressants to the AChE peripheral anionic site (PAS). This ligand exhibited an eight-fold fluorescence enhancement upon binding to the peripheral anionic site of AChE from E. electricus (L.) with K(D)=7 x 10(-7)M. It was observed that TCA drugs displaced propidium from the enzyme. On the basis of the displacement experiments antidepressant dissociation constants were determined. Similar values for the inhibition constants suggest that these drugs have similar affinity to the peripheral anionic site. The results also indicate that the catalytic active center of AChE does not participate in the interaction of enzyme with tricyclic antidepressants. These studies suggest that the binding site for tricyclic antidepressants is located at the peripheral anionic site of E. electricus (L.) acetylcholinesterase.

We have determined the effects of mercury and cadmium on the creatine kinase activity of the electric organ of Electrophorus electricus (L.) which catalyses the transphosphorylation reaction between phosphocreatine and magnesium adenosine-5'-di-phosphate and has essential sulfhydryl groups. The kinetic effects of these heavy metals, which have high affinity for sulfhydryl groups, on the creatine kinase activity were analysed with the three reaction components: phosphocreatine, adenosine-5'-di-phosphate and magnesium. The kinetic data were analysed with a non-linear regression program (Sigmaplot for Windows). Both metals inhibit creatine kinase activity in the micromolar range, mercury being a more potent inhibitor than cadmium. With phosphocreatine as substrate, mercury behaved as a mixed partial hyperbolic inhibitor, non-competitive inhibitor with adenosine-5'-di-phosphate, and with magnesium mercury behaved as a competitive inhibitor. Cadmium inhibition was shown to be of a classical competitive nature with respect to both substrates, phosphocreatine or adenosine-5'-di-phosphate, and non-competitive when magnesium was the variable in the reaction mixture. The results suggest that the binding site of mercury is at or near the phosphocreatine site, but it is not the same as adenosine-5'-di-phosphate, whereas cadmium competes with these substrates to bind at the same sulphydryl site.
