RESUMO

Microtus thomasi (Rodentia: Arvicolinae), a fossorial vole endemic to the SW Balkans, uses a variety of substrates but its underground behavior remains poorly understood. This study examines the architecture and utilization of M. thomasi burrow systems in NW Peloponnese, Greece. In particular, eight burrow systems were meticulously excavated and studied, with comprehensive measurements taken of key characteristics, including length, depth, soil mounds, and surface openings. Key coordinates were recorded using a differential GPS device for detailed mapping and fractal dimension analysis using the box-counting method was employed to assess burrow system complexity. Soil samples were analyzed for particle size and chemical composition, and vegetation types at each site were identified. We did not find statistically significant correlations between size and complexity of the burrow systems and soil composition, altitude, or specific soil components. On the other hand, we did observe statistically significant differences in tunnel diameter between two burrow systems and in tunnel depth between more. Moreover, our study showed that more than one same-sex individual can occupy a single burrow system and not just an adult male-female pair, that was previously recorded, indicating the need for further study of the social behavior of this vole species. This study provides valuable insights into the underground behavior of M. thomasi by providing information on the features of its burrow systems, thus contributing to our understanding of its biology.

RESUMO

The development of polymeric materials as antifouling coatings for aquaculture nets is elaborated in the present work. In this context, cross-linked polymeric systems based on quaternary ammonium compounds (immobilized or releasable) prepared under mild aqueous conditions were introduced as a more environmentally friendly methodology for coating nets on a large scale. To optimize the duration of action of the coatings, a multilayer coating method was applied by combining the antimicrobial organo-soluble copolymer poly(cetyltrimethylammonium 4-styrenesulfonate-co-glycidyl methacrylate) [P(SSAmC16-co-GMA20)] as the first layer with either the water-soluble copolymer poly(vinylbenzyl trimethylammonium chloride-co-acrylic acid) [P(VBCTMAM-co-AA20)] or the water-soluble polymers poly(acrylic acid) (PAA) and poly(hexamethylene guanidine), PHMG, as the second layer. The above-mentioned approach, followed by thermal cross-linking of the polymeric coatings, resulted in stable materials with controlled release of the biocidal species. The coated nets were studied in terms of their antifouling efficiency under accelerated biofouling conditions as well as under real conditions in an aquaculture field. Resistance to biofouling after three water-nutrient replenishments was observed under laboratory accelerated biofouling conditions. In addition, at the end of the field test (day 23) the uncoated nets were completely covered by marine contaminants, while the coated nets remained intact over most of their extent.

RESUMO

Copper-based bactericides have appeared as a new tool in crop protection and offer an effective solution to combat bacterial resistance. In this work, two copper nanoparticle products that were previously synthesized and evaluated against major bacterial and fungal pathogens were tested on their ability to control the bacterial spot disease of tomato. Growth of Xanthomonas campestris pv. vesicatoria, the causal agent of the disease, was significantly suppressed by both nanoparticles, which had superior function compared to conventional commercial formulations of copper. X-ray fluorescence spectrometry measurements in tomato leaves revealed that bioavailability of copper is superior in the case of nanoparticles compared to conventional formulations and is dependent on synthesis rather than size. This is the first report correlating bioavailability of copper to nanoparticle efficacy.

RESUMO

Bacterial systems have gained wide attention for depolymerization of lignocellulosic biomass, due to their high functional diversity and adaptability. To achieve the full microbial exploitation of lignocellulosic residues and the cost-effective production of bioproducts within a biorefinery, multiple metabolic pathways and enzymes of various specificities are required. In this work, highly diverse aerobic, mesophilic bacteria enriched from Keri Lake, a pristine marsh of increased biomass degradation and natural underground oil leaks, were explored for their metabolic versatility and enzymatic potential towards lignocellulosic substrates. A high number of Pseudomonas species, obtained from enrichment cultures where organosolv lignin served as the sole carbon and energy source, were able to assimilate a range of lignin-associated aromatic compounds. Comparatively more complex bacterial consortia, including members of Actinobacteria, Proteobacteria, Bacilli, Sphingobacteria, and Flavobacteria, were also enriched from cultures with xylan or carboxymethyl cellulose as sole carbon sources. Numerous individual isolates could target diverse structural lignocellulose polysaccharides by expressing hydrolytic activities on crystalline or amorphous cellulose and xylan. Specific isolates showed increased potential for growth in lignin hydrolysates prepared from alkali pretreated agricultural wastes. The results suggest that Keri isolates represent a pool of effective lignocellulose degraders with significant potential for industrial applications in a lignocellulose biorefinery.

RESUMO

This study presents the application of a simultaneous method for the determination of total organic carbon (TOC) and total nitrogen (TN) in marine sediments and soils, using a data set of 206 samples collected from coastal lagoonal/marine sedimentary environments and certified reference materials (CRMs). TOC and TN were determined using the high temperature (720°C) catalytic (Pt/Al2O3) oxidation method and the detection of TOC and TN was performed using an infrared or a chemiluminescence detector, respectively. Results from the abovementioned TOC method were compared with the results from the widely used Wakley-Black titration method, while TN results with these from elemental analysis. Analytical quality control experiments were performed using CRM samples. Method characteristics such as range of measurement, calibration, method detection limit (MDL), limit of quantification (LOQ), repeatability and reproducibility, were calculated. The precision and the accuracy of the methods are also discussed. Comparison of the two TOC methods of 206 data set yields a regression line of correlation coefficient with R2 = 0.985. Additional different levels of TOC concentrations of low <1%, moderate 1-5% and high 5-40% level were examined indicating very good correlations. The lowest correlation coefficient was observed in low concentrations TOC<1% (R2 0.825), mainly as a result of the limitation of titration method. The evaluation of TN results indicated that the catalytic oxidation method and the elemental analysishave a significant good correlation with R2 = 0.977. The results of precision and accuracy, as well as the calculated MDL and LOQ show that this is a reliable method. Moreover, it requires a small amount of the analyzed sample and the total analysis time is 10 min. Therefore, it can be easily applied for the fast and precise simultaneous determination of TOC and TN in sediment and soil samples.

RESUMO

The synthesis of environmentally friendly antimicrobial polymeric coatings, especially in the case of aquaculture, that inhibit the growth of bio-deposits is a very important issue that will contribute to the cost reduction of nets' cleaning process as well as the protection of the submarine wealth from the biostatic substances used so far. In the present work, the antimicrobial polymers P(SSAmC16-co-VBCHAMx) and the terpolymer P(SSAmC16w-co-VBCHAMx-co-GMAy) were synthesized, bearing quaternary ammonium compounds, electrostatically bound and covalently attached at the same polymer chain. The combination of the two types is of particular importance, as it can provide effective antimicrobial polymeric materials with self-polishing capabilities as a result of the released nature of the antimicrobial, in combination with the permanent local action of the immobilized species. The cross-linking reaction of the terpolymer P(SSAmC16w-co-VBCHAMx-co-GMAy) with the homopolymer polyacrylic acid (PAA) was tested at 120 °C in terms of the equivalent ratio between epoxy and carboxyl groups. The synthesized polymers were further used for the coating of aquaculture nets and tested in terms of antifouling efficiency in lab and scale-up conditions. Uncoated nets were also used in all applications for comparison reasons. The coated nets performed efficiently for 35 days in lab-scale and 66 days in scale-up conditions, showing a high antifouling activity in both fields compared to the uncoated nets.

RESUMO

Crosslinked polymeric materials based on a quaternary trimethylammonium compound were developed and evaluated as potential antifouling coatings. For this purpose, two water-soluble random copolymers, poly(4-vinylbenzyltrimethylammonium chloride-co-acrylic acid) P(VBCTMAM-co-AAx) and poly(N,N-dimethylacrylamide-co-glycidylmethacrylate) P(DMAm-co-GMAx), were synthesized via free radical polymerization. A water based approach for the synthesis of P(VBCTMAM-co-AAx) copolymer was used. Coatings of the complementary reactive copolymers in different compositions were obtained by curing at 120 °C for one day and were used to coat aquaculture nets. These nets were evaluated in respect to their release rate using Total Organic Carbon (TOC) and Total Nitrogen (TN) measurements. Finally, the antifouling efficacy of these newly-composed durable coatings was investigated for 14 days in accelerated conditions. The results showed that this novel polymeric material provides contact-killing antifouling activity for a short time period, whereas it functions efficiently in biofouling removal after high-pressure cleaning.

RESUMO

Many hirundine species construct their nests by carrying mud particles from adjacent areas. This study aimed to investigate for the first time the materials that mud-nesting hirundines choose for nest construction from a mineralogical and sedimentological perspective. For this purpose, we sampled nests of three sympatric species, namely the Barn Swallow (Hirundo rustica), the Red-rumped Swallow (Cecropis daurica) and the House Martin (Delichon urbicum), from southeastern Europe. Our results showed that all species tend to use clay minerals as a cement and especially smectite and illite and if these minerals are not present in the adjacent area, they use halloysite, kaolinite or chlorite. The amounts of clay minerals in the nests are generally low indicating that the studied species can accurately identify the properties of the nesting materials. Most of the non clay minerals that they use are the common, easily accessible colourless or white minerals with low specific gravity values such as quartz, feldspars and calcite. Grain size distribution analysis revealed that the amount of clay sized grains in the mud nests of all three species is relatively low, while the amount of larger grain particles decreases when the size of the non clay minerals is small. The Red-rumped Swallow showed an increasing preference for larger grain size particles and quartz, the Barn Swallow for finer grain size particles and calcite, and the preferences of the House Martin are in between the other two species. The three hirundine species present different nest building strategies and depending on the nest architecture, each of them seems to show preference for specific minerals and specific grain sizes.

RESUMO

BACKGROUND: Natural oil seeps offer the opportunity to study the adaptation of ecosystems and the associated microbiota to long-term oil exposure. In the current study, we investigated a land-to-sea transition ecosystem called "Keri Lake" in Zakynthos Island, Greece. This ecosystem is unique due to asphalt oil springs found at several sites, a phenomenon already reported 2500 years ago. Sediment microbiomes at Keri Lake were studied, and their structure and functional potential were compared to other ecosystems with oil exposure histories of various time periods. RESULTS: Replicate sediment cores (up to 3-m depth) were retrieved from one site exposed to oil as well as a non-exposed control site. Samples from three different depths were subjected to chemical analysis and metagenomic shotgun sequencing. At the oil-exposed site, we observed high amounts of asphalt oil compounds and a depletion of sulfate compared to the non-exposed control site. The numbers of reads assigned to genes involved in the anaerobic degradation of hydrocarbons were similar between the two sites. The numbers of denitrifiers and sulfate reducers were clearly lower in the samples from the oil-exposed site, while a higher abundance of methanogens was detected compared to the non-exposed site. Higher abundances of the genes of methanogenesis were also observed in the metagenomes from other ecosystems with a long history of oil exposure, compared to short-term exposed environments. CONCLUSIONS: The analysis of Keri Lake metagenomes revealed that microbiomes in the oil-exposed sediment have a higher potential for methanogenesis over denitrification/sulfate reduction, compared to those in the non-exposed site. Comparison with metagenomes from various oil-impacted environments suggests that syntrophic interactions of hydrocarbon degraders with methanogens are favored in the ecosystems with a long-term presence of oil.
