RESUMO

Moringa oleifera is a medicinal plant that has anti-inflammatory, antihypertensive, antidiabetic, tissue-protective, and antioxidant activities. Here, we evaluated the protective effect of M. oleifera leaf powder (MoLP) and 70% ethanol M. oleifera leaf extract (MoLE) on mitigating polycystic ovary syndrome (PCOS)-induced liver and kidney dysfunction via regulating oxidative stress in female albino mice (Mus musculus). The efficacy of M. oleifera was compared with metformin (standard medicine used to treat infertility in women). PCOS was induced by intramuscular injection of testosterone enanthate at 1.0 mg/100 g BW for 35 days. PCOS-induced mice were treated with MoLP (250 and 500 mg/Kg), MoLE (250 and 500 mg/kg), and metformin (250 mg/kg) orally for 14 days. Renal function test (RFT), liver function test (LFT), and oxidative stress biomarker malondialdehyde (MDA) were quantified in serum at 0, 7, and 14 days of intervention. Mice treated with M. oleifera and metformin showed a significant decrease (p < .001) in alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphate (ALP), total bilirubin, urea, creatinine, and a significant increase (p < .001) in total protein, albumin, globulin, and albumin/globulin (A/G) ratio. Oxidative stress decreased significantly (p = .00) with respect to treatments, exposure days, and their interaction in metformin and all M. oleifera-treated groups. M. oleifera leaf powder and extract reduce oxidative stress and enhance nephron-hepatic activity in PCOS-induced female albino mice.

Phenolic compounds in prickly pear [Opuntia ficus indica (L.) Mill.] are known to contribute to the antioxidant and antimicrobial activities of the prickly pear. The present study aimed to evaluate the antioxidants and in vitro antimicrobial potential in the hydroethanolic extracts of different parts (fruit, cladode, and pulp) of prickly pear. Different polyphenolic compounds were analyzed by using high-performance liquid chromatography. The results indicated that cladode possessed a higher quantity of phenolics compared with that observed in fruit and pulp. The most important phenolic compound in high quantity was gallic acid (66.19 µg/g) in cladode. The 100% aqueous extract of cladode exhibited the highest antioxidant (92%) and antimicrobial activities against Salmonella typhi (3.40 mg/ml), Helicobacter pylori (1.37 mg/ml), Escherichia coli (1.41 mg/ml), and Staphylococcus aureus (1.41 mg/ml). Principal component analysis (PCA) indicated that antioxidant activity and minimum inhibitory concentration (MIC) responses had a significant negative correlation with each other. Overall, the current results provided basic data for choosing prickly pear cladode with high antioxidant capacity for the development and consumption of antioxidant-based alternative medicines and value addition of formulated foods.

Organisms employ various enzymatic and non-enzymatic detoxification mechanisms to minimize the harmful effects of metal pollution in the terrestrial environment. We examined the effects of copper (Cu), lead (Pb) and their mixture (Cu + Pb) on glutathione (GSH), metallothionein (MTs), cytochrome P450 (CYP 450), carboxylesterase (CarbE), acetylcholinesterase (AchE) and glutathione S-transferase (GST) in Lycosa terrestris and Pardosa birmanica via two exposure routes, i.e., soil and food for 10, 20 and 40 days. The present results revealed that the accumulation of Cu and Pb in both spiders' species increase with exposure duration and depend on the route of exposure and type of metal. The activities of CarbE, GST, and MTs significantly increased with increasing metal body burden for all experimental treatments. The CYP 450 activity exhibited a significant time-dependent decrease with increasing Cu concentration in both species. The AchE activity was significantly inhibited on Pb exposure via soil and Cu + Pb exposure via both routes. The decrease in the level of GSH was measured on Cu + Pb exposure via both routes. Thus, all these enzymatic and non-enzymatic responses are sensitive to the metals tested and could serve as early warning indicators for assessing the effects of metal pollution in these species.

Heavy metal exposure induces oxidative stress in terrestrial organisms, which they counteract via activation of antioxidant biomarkers. The present study investigated the effects of copper (Cu) and lead (Pb) on the total antioxidant capacity (TAC) and antioxidant enzymes such as Catalase (CAT), Glutathione reductase (GR), Superoxide dismutase (SOD) and Glutathione peroxidase (GPX) in two spider species, namely Lycosa terrestris and Pardosa birmanica. The spiders were exposed to Cu and Pb separately (10 ppm) or in combination (10 ppm each) via two different exposure routes (i.e. food and soil) for 10, 20 and 40 days. The results showed that metal accumulation and antioxidant biomarker responses in spiders were metal- and species-dependent. Also, the levels of all antioxidant biomarkers increased significantly with increasing exposure time and metal load in the bodies of spiders via both exposure routes. The significant inhibition of TAC and antioxidant enzyme activities was only observed in single Pb treatment through soil exposure. In L. terrestris, the activities of detoxification enzymes and TAC were significantly enhanced on single Cu exposure than Pb via both exposure routes. However, in P. birmanica consistent variation among antioxidant parameters were observed depending on the metal load and exposure routes. The combined metal exposure caused more pronounced increase in the level of antioxidants compared to single metal exposure in both species, mainly via food exposure. These results suggest that the antioxidant enzymes and TAC are sensitive to single and combined metal exposure via both uptake routes. These data show that antioxidant parameters can be used potential biomarkers of oxidative stress associated with metal exposure and for monitoring environmental health using spiders as bioindicators.

Major concerns exist regarding the environmental and human health risks caused by exposure to heavy metals. Spiders are often used as a model in ecotoxicological studies to assess soil pollution. Here, we measured the bioaccumulation of copper (Cu) and lead (Pb) in spiders, Lycosa terrestris and Pardosa birmanica, by inductively coupled plasma mass spectrometry (ICP-MS). We investigated whether Cu and Pb accumulation differs according to different spider species, single versus combined metal exposure, and routes of exposure. Spiders were exposed to 10 mM CuSO4 and 10 mM PbCl2 solutions separately or in combination (10 mM + 10 mM) through different exposure routes (spiked soil and food) for 6 weeks. The effect of metals on the survival and body mass of exposed and unexposed (control) spiders was determined. We found that in both spider species, accumulation of metals increased with exposure time. In single metal exposure, Cu accumulation from food was higher than soil exposure in both spider species, whereas the opposite was observed for Pb. The simultaneous uptake of Cu and Pb significantly decreased from food and soil, respectively. Soil exposure caused more accumulation of metals in L. terrestris than P. birmanica. Metal exposure via contaminated food caused higher mortality compared to soil exposure. Body mass of both spider species was significantly decreased and negatively correlated with metal's concentration. Overall, our results show that bioaccumulation efficiency of Cu and Pb differs significantly in spiders exposed to metal's mixture compared to single metal exposure and is dependent on the exposure route, the type of metal, and spider species. More understanding of the effects of exposure to metal mixture and exposure routes is essential for designing and supporting risk assessment and ecological monitoring programs.

RESUMO

During the last few decades, substantial scientific and technological efforts have been focused on the development of neuroprostheses. The major emphasis has been on techniques for connecting the human nervous system with a robotic prosthesis via natural-feeling interfaces. The peripheral nerves provide access to highly processed and segregated neural command signals from the brain that can in principle be used to determine user intent and control muscles. If these signals could be used, they might allow near-natural and intuitive control of prosthetic limbs with multiple degrees of freedom. This review summarizes the history of neuroprosthetic interfaces and their ability to record from and stimulate peripheral nerves. We also discuss the types of interfaces available and their applications, the kinds of peripheral nerve signals that are used, and the algorithms used to decode them. Finally, we explore the prospects for future development in this area.

OBJECTIVES: Intrathecal (IT) trial is a prognostic interventional pain management procedure employed to determine the potential success of treating intractable pain with an implantable infusion device system. There is a dearth of data regarding trials with continuous infusion of combination therapy (e.g. opioid combined with local anesthetic). The objective of the this study was to determine the overall outcomes of continuous infusion IT trials and factors influencing long-term success of IT therapy in patients with chronic intractable pain post-laminectomy. MATERIALS AND METHODS: This is a retrospective analysis of all patients with lumbar failed back surgery syndrome (FBSS) who were trialed with a combination of hydromorphone and bupivacaine with a temporary externalized IT catheter from March 2007 to June 2014. RESULTS: From a cohort of 62 patients fulfilling the inclusion criteria, 54 (87.10%) patients had successful IT trials. No significant differences were found between successful and failed trial patients with regards to age, sex, pre-trial pain numeric rating scale scores, pre-trial morphine equivalent daily dose, or trial dosages. Significant positive correlations were found between pretrial oral opioid intake and end of trial hydromorphone dose and hydromorphone dose escalation at 12 months and 24 months. CONCLUSIONS: Patients with refractory low back pain due to FBSS who underwent successful combination IT trial with hydromorphone and bupivacaine infused through a temporary IT catheter had significantly improved pain intensity scores following permanent implant. Higher pre-trial MEDD was correlated with higher trial and post-implant opioid doses and higher rates of opioid dose escalation post-implant.

The presence of pesticides in the environment is highly toxic to environment and human health. Aim of the study was determination, quantification and assessment of associated health risk due to presence of pesticide residues in chicken eggs using high pressure liquid chromatography. HPLC method was successfully employed and validated. From collected samples pesticides were extracted in presence of petroleum ether and acetonitrile. Bifenthrin and Difenoconazole residues were found in all samples with different concentration exceeding maximum residue limits (MRL) of Codex Alimentarius Commission. However imidacloprid was not detected in any sample. Concentration of bifenthrin in house egg samples ranged from 0.256206 to 4.112387 mg/kg while in poultry farm samples it varied from 1.5862 to 5.80796 mg/kg. Difenoconazole was found in concentration of 0.02835 mg/kg, 1.7668 mg/kg, 3.7205 mg/kg, 21.8937 mg/kg 21.9835 mg/kg, 19.26407 mg/kg in samples collected from houses while and in poultry farm samples its detected concentration was 10.939 mg/kg, 12.3296 mg/kg, 29.3617 mg/kg, 18.6116 mg/kg, 40.0523 mg/kg and 19.2335 mg/kg. Concentrations of both pesticides Bifenthrin and Difenoconazole exceeded the MRLs (0.05 mg/kg). Health risk index surpassed 1 (the cut off value) for Difenoconazole in seven samples while for Bifenthrin values were less than 1, indicating the possibility of potential medium to long term health risk associated with ingestion of contaminated eggs.(AU)
