RESUMO

Systematic investigations into the impact of haemorheological parameters through ionizing rays are not available in literature. The main factors influencing the flow capacity of the blood were determined in vitro by using a set of special methods. Even by using high dosages of irradiation (600 R) only those changes in the flow capacity of the blood could be identified which lay in or near the dispersion area of the methods. These investigations carried out in vitro only take into account the immediate effect of the blood and not rheological changes caused by the intact organism after irradiation.

RESUMO

There are no systematic and complex investigations regarding the hemorheologic action of the betablockers propranolol and talinolol. The results on the other ones are contradictory. On the basis of a critical survey of the literature, in-vitro-experiments and studies in animals (rats) and humans may conclude that there are no remarkable deteriorations of rheological parameters in the therapeutic dose range. This is valid for diseases only where the haemodynamic regulation doesn't prevail.

RESUMO

Benzotriazines are a new group of substances with surprisingly hemorheological effects. Especially red-cell deformability is influenced. In the case of decreased deformability some benzotriazines are comparable to pentoxifylline. The increasing importance of hemorheologically active agents in treatment concepts makes it seem promising to carry out further research into benzotriazines in animal experiments.

RESUMO

Some benzotriazines markedly reduce human whole-blood viscosity. Viscosity was measured electronically with a novel capillary viscosimeter. Because of the increasing interest in hemorheologically active drugs, the compounds merit further investigation.

By applying in vitro and additional in vivo investigations of analysing coagulation in human beings with the routine methods TPZW, PTT and TZ it was possible to exclude influences of hemostasis described in literature and often uncritically generalised in papers and articles as well as assumed influences for such medicaments as acetylsalicylic acid, ascorbic acid, chlorpromazine, fluphenazine, gentamycin, haloperidol, lincomycin, methyldopa, sodium fluoride and penicillin G in therapeutic doses or concentrations in vitro respectively which correspond to therapeutic or toxic doses. For such solutions of infusion as infucoll 6% and gelafusal there exist these influences in the sense of an acceleration of coagulation in vitro. They can be regarded as shiftings within the normal range of methods which have no clinical relevance. Infucoll M 40 will to a concentration of 0.25 mmol/l of blood which corresponds to therapeutic conditions, to a shortening of Tz falling below the normal range of the method. These findings correlate with data in literature and could be identified even in lower concentrations by excluding the effect of dilution. According to literature these relations are identical with those in vivo some minutes after infusion.

The Farnsworth-Munsell-100 Hue-Test (FMT) shows an increasing importance for judgement of colour vision disturbances and drug side effects on the ocular system, respectively. Because of the widening of caffeine use, the high incidence of intake of oral contraceptives and the absence of prospective studies, it is necessary to do it to investigate the possibility of influences on colour vision. 206 test persons of both sexes took part in our investigation with the FMT to clear influences of caffeine, oral contraceptives and the combination of both of them. A remarkable result is that caffeine with intake of oral contraceptives diminished the ability of colour discrimination in a wide range of visible coloured light. These investigations are in addition to views known from relevant literature.

With a newly developed photoplethysmographical measuring device it is possible to record the conjunctival pulse of the human eye. The device consists of the self-constructed photoplethysmographical amplifier unit, the sucking electrode from silicone hard-rubber with photo-detector and luminescence diode, and a 3-channel recorder (3-NEK). Electrocardiogram and the conjunctival pulse are recorded simultaneously. Parameters that are derived from the curves allow to judge on the circulation in the tissue region investigated. Investigations were carried out in 70 test persons without clinical or other signs of pathological conditions regarding eye or vessels (classified into sex and age groups up to 40 years and over). 133 recordings with six consecutive curve waves were examined. Additionally, the influence of vasoactive drugs on the conjunctival pulse curves was investigated. The topical application of Oculoguttae Tolazolini 2% and Oculoguttae Epinephrini 2% showed no influence compared with the reference curves. Contrary to this, differences were proved after application of Nitrangin liquidum systemically in all parameters tested. The photoplethysmography of the conjunctiva bulbi is a noninvasive running-on method with good reproducibility without troubles for the patient, with an easy performance and only a small time demand. The results qualified the method to test the influence of systemically applied vasoactive drugs.

Anticholinergic and antiserotonergic activities of the psychotropic drugs chlorpromazine, fluphenazine, imipramine and promethazine have been investigated quantitatively on the guinea-pig isolated ileum. Additional experiments were made to assure the comparability of receptor activity with human intestinal (ileum) preparations. Each of the four compounds exhibited a marked blocking potency both on muscarinic and serotonergic receptors. The antagonism was found to be slowly reversible and non-competitive in higher drug concentrations. Sodium sulfate often recommended in cases of poisoning with these drugs was nearly ineffective for evoking contractions in model investigations with drug concentrations which can be found in practical clinical conditions.

In the accessible literature the influence of beta-blockers on laboratory methods is estimated only little systematically and nearly exclusively on parameters of the glucose and lipometabolism. The influence on several methods in vitro is without relevance to practice. Own examinations showed that in 32 patients with essential hypertension of stage I (after Baumann and Nitschkoff) under therapeutic doses of 50--80 mg propranolol a day in none of the 18 chemical parameters tested after a four weeks' treatment significant deviations relevant to practice appeared in comparison to the parameters before treatment.

Only some few systematic studies have appeared in the literature relative to the interference of drugs with clinico-chemical screening and rapid tests. This question is dealt with in the light of in vitro and in vivo investigations of the interference of selected drugs with screening test reagents produced in the G.D.R. In contrast to speculations in the secondary literature, a practice-relevant interference of the drugs under investigation with the test procedures was an exception. In this sense, our following findings are of importance: --inhibitory action of ascorbic acid and sodium fluoride on the demonstration of glucose by Biophan G, --false positive indication of Biophan PAS in case of high concentrations of sulphamerazine, --interference of ascorbic acid with the Biophan P test.

RESUMO

The paper presents further investigations for a critical survey on the influences of drugs on laboratory methods. In controversion to the meanings you can find in the literature that ascorbic acid is most one of the important drugs to interfere with laboratory results we couldn't see in our systematical experimental investigation such results. Only in very extremly cases it seems to be right. Selected parameters of clinical chemistry (glucose, lactic dehydrogenase, aspartal-aminotransferase, alanine aminotransferase, alkaline phosphatase, protein, albumine, creatinine, butanol extractable iodine, ferrum) show under therapeutic conditions no influences of ascorbic acid, which can lead to diagnostic or therapeutic false interpretations. Above all the often mentioned example that glucose estimations in blood (reduction methods) can disturb if ascorbic acid is present, is abstracted in an uncritical manner how our experimental results may show.
