RESUMO

The objective of this study was to investigate the effects of chop length and inoculation of barley silage on feeding behavior, rumen fermentation, and growth performance of finishing feedlot steers. Barley forage (22-35% DM) was chopped to a theoretical chop length (TCL) of either 1.0 (short chop; SC) or 2.0 cm (long chop; LC) and ensiled without or with an esterase-producing bacterial inoculant in a 2 × 2 factorial arrangement of treatments with TCL and inoculation as the main factors. The resultant silages were then incorporated into 4 finishing diets and fed to 80 feedlot steers (n = 20) housed in feedlot pens equipped with a GrowSafe system. Each pen consisted of 8 intact and 2 cannulated steers. Feeding behavior and ruminal pH were measured continuously using the GrowSafe system and ruminal indwelling pH probes, respectively. On average, inoculated silages had higher acetic acid concentrations and lower NDF concentrations. Increasing the TCL from 1.0 to 2.0 cm increased DMI by 0.5 kg/d (P = 0.001), but intake was unaffected ( P= 0.56) by inoculation. Feed efficiency and ADG were not affected (P ≥ 0.46) by TCL, inoculation, or TCL × inoculation interaction. Steers fed LC silage exhibited a reduced eating rate compared to those fed SC silage (8.6 vs. 9.2 kg DM/h) and consequently spent more time (77.5 vs. 70.2 min/(visit∙d)) at the feed bunk. Inoculation also reduced the area under the curve and duration of ruminal pH below 5.8, 5.5, and 5.2 for steers fed the LC diet but increased (P ≤ 0.003) these parameters for those fed the SC diet. It was concluded that incorporation of longer chopped silage into a finishing diet increased DMI, with responses of ruminal pH to inoculation differing between SC and LC silage. Increasing the TCL of barley silage from 1.0 to 2.0 cm may have no additional benefits to finishing feedlot operators as it did not improve rumen function or the growth performance of feedlot steers.

The objective of this study was to develop emission factors (EF) for methane (CH4) emissions from enteric fermentation in cattle native to Benin. Information on livestock characteristics and diet practices specific to the Benin cattle population were gathered from a variety of sources and used to estimate EF according to Tier 2 methodology of the 2006 Intergovernmental Panel on Climate Change (IPCC) Guidelines for National Greenhouse Gas Inventories. Most cattle from Benin are Bos taurus represented by Borgou, Somba and Lagune breeds. They are mainly multi-purpose, being used for production of meat, milk, hides and draft power and grazed in open pastures and crop lands comprising tropical forages and crops. Estimated enteric CH4 EFs varied among cattle breeds and subcategory owing to differences in proportions of gross energy intake expended to meet maintenance, production and activity. EFs ranged from 15.0 to 43.6, 16.9 to 46.3 and 24.7 to 64.9 kg CH4/head per year for subcategories of Lagune, Somba and Borgou cattle, respectively. Average EFs for cattle breeds were 24.8, 29.5 and 40.2 kg CH4/head per year for Lagune, Somba and Borgou cattle, respectively. The national EF for cattle from Benin was 39.5 kg CH4/head per year. This estimated EF was 27.4% higher than the default EF suggested by IPCC for African cattle with the exception of dairy cattle. The outcome of the study underscores the importance of obtaining country-specific EF to estimate global enteric CH4 emissions.

The aim of this study was to determine the effects of the use of a fibrolytic enzyme product, applied at ensiling either alone or in combination with a ferulic acid esterase-producing bacterial additive, on the chemical composition, conservation characteristics, and in vitro degradability of corn silage harvested at either conventional or high cutting height. Triplicate samples of corn were harvested to leave stubble of either a conventional (15cm; NC) or high (45cm; HC) height above ground. Sub-samples of chopped herbage were ensiled untreated or with a fibrolytic enzyme product containing xylanases and cellulases applied either alone (ENZ) or in combination with a ferulic acid esterase-producing silage inoculant (ENZ+FAEI). The fibrolytic enzyme treatment was applied at 2mL of enzyme product/kg of herbage dry matter (DM), and the inoculant was applied at 1.3×10(5) cfu/g of fresh herbage. Samples were packed into laboratory-scale silos, stored for 7, 28, or 70 d, and analyzed for fermentation characteristics, and samples ensiled for 70 d were also analyzed for DM losses, chemical composition, and in vitro ruminal degradability. After 70 d of ensiling, the fermentation characteristics of corn silages were generally unaffected by cutting height, whereas the neutral detergent fiber, acid detergent fiber, and ash concentrations were lower and the starch concentration greater for silages made with crops harvested at HC compared with NC. After 70 d of ensiling, the acetic acid, ethanol concentrations, and the number of yeasts were greater, and the pH and neutral detergent fiber concentrations were lower, in silages produced using ENZ or ENZ+FAEI than the untreated silages, whereas ENZ+FAEI silages also incurred higher DM losses. No effect of additive treatment was observed on in vitro degradability indices after 48h ruminal incubation. The use of a fibrolytic enzyme product, either alone or in combination with a ferulic acid esterase-producing inoculant, at ensiling did not improve corn silage fermentation or its nutritive value and resulted in some negative effects on these parameters. The effects of using a fibrolytic enzyme product at ensiling, either alone or in combination with a ferulic acid esterase-producing inoculant, did not differ between corn harvested at either NC or HC. Silage made from HC had a greater starch content and lower fiber content than NC silage, whereas cutting height did not affect the in vitro digestibility indices.

This study investigated the effects of a mixed bacterial inoculant possessing ferulic acid esterase (FAE) activity on silage fermentation characteristics, aerobic stability, and growth performance of growing feedlot steers. Whole-crop barley (Hordeum vulgare L.) forage (35% DM) was chopped and ensiled without a silage inoculant (UN) or with a mixed bacterial culture containing 1.0 × 10(11) cfu/g of Lactobacillus buchneri LN4017 that produces FAE, 2.0 × 10(10) cfu/g of Lactobacillus plantarum LP7109, and 1.0 × 10(10) cfu/g of Lactobacillus casei LC3200 at a combined rate of 1.3 × 10(5) cfu/g of fresh forage (IN) in mini and Ag-Bag (Ag-Bag Int. Ltd., Warrenton, OR) silos. Silages from the mini silos were assessed for the effect of inoculation on fermentation characteristics and aerobic stability, whereas silages from Ag-Bags were used to formulate 2 barley silage-based total mixed rations (UN and IN) that were fed to growing feedlot steers for 112 d. The IN silage exhibited a homolactic fermentation during the first 7 d of ensiling as reflected by an increased (P ≤ 0.02) lactic acid concentration and an accelerated rate (P < 0.01) of pH decline. Thereafter, fermentation of IN silage became more heterolactic, resulting in greater concentrations of acetic acid (P < 0.01) and pH (P < 0.01) but less (P < 0.01) lactic acid than UN silage. Inoculation did not affect DM losses (P = 0.52) from mini silos. The IN silage remained stable during 21 d, but temperature and yeasts counts in the UN silage increased after 5 d of aerobic exposure. Growing steers fed the IN silage diet had superior (P = 0.03) feed conversion efficiency compared with those fed UN silage. Inoculation of whole-crop barley silage with a mixed culture of homolactic lactic acid-producing bacteria and FAE-producing L. buchneri at ensiling changed fermentation from a homolactic to a heterolactic form during ensiling and improved aerobic stability of the silage and efficiency of BW gain of growing feedlot steers.

An in vitro rumen batch culture study was completed to compare effects of common grasses, leguminous shrubs and non-leguminous shrubs used for livestock grazing in Australia and Ghana on CH4 production and fermentation characteristics. Grass species included Andropodon gayanus, Brachiaria ruziziensis and Pennisetum purpureum. Leguminous shrub species included Cajanus cajan, Cratylia argentea, Gliricidia sepium, Leucaena leucocephala and Stylosanthes guianensis and non-leguminous shrub species included Annona senegalensis, Moringa oleifera, Securinega virosa and Vitellaria paradoxa. Leaves were harvested, dried at 55°C and ground through a 1 mm screen. Serum bottles containing 500 mg of forage, modified McDougall's buffer and rumen fluid were incubated under anaerobic conditions at 39°C for 24 h. Samples of each forage type were removed after 0, 2, 6, 12 and 24 h of incubation for determination of cumulative gas production. Methane production, ammonia concentration and proportions of VFA were measured at 24 h. Concentration of aNDF (g/kg DM) ranged from 671 to 713 (grasses), 377 to 590 (leguminous shrubs) and 288 to 517 (non-leguminous shrubs). After 24 h of in vitro incubation, cumulative gas, CH4 production, ammonia concentration, proportion of propionate in VFA and IVDMD differed (p<0.05) within each forage type. B. ruziziensis and G. sepium produced the highest cumulative gas, IVDMD, total VFA, proportion of propionate in VFA and the lowest A:P ratios within their forage types. Consequently, these two species produced moderate CH4 emissions without compromising digestion. Grazing of these two species may be a strategy to reduce CH4 emissions however further assessment in in vivo trials and at different stages of maturity is recommended.

In industrialized countries, the initiation of antiretroviral therapy (ART) is based on virological, immunological and clinical markers. The objective of this study was to identify treatment gaps when ART initiation is based on clinical staging alone. The method employed was a retrospective study of 5784 patients enrolled in an HIV treatment programme in two urban and two rural sites in Ghana. Of the patients, 29.5% were in clinical Stages I and II and had a CD4+ T-lymphocyte count less than 200 cells/mm(3). Significantly more patients in clinical Stage I from urban sites (37.0%) had a CD4+ T-lymphocyte count less than 200 cells/mm(3) as compared with patients from rural sites (23.8%) (P value <0.05). In addition, more men (39.9%) in clinical Stage I had a CD4+ T-lymphocyte count less than 200 cells/mm(3) when compared with women (27.4%) (P value <0.05). In conclusion, clinical staging cannot identify a relatively large number of patients who need ART. A wider availability of CD4+ T-lymphocyte count testing will optimize the identification of patients eligible for ART.

Mycobacterium ulcerans disease (Buruli ulcer) is a skin-ulcerating infection common in some parts of the tropics. We have investigated cytokine secretion after stimulation of whole blood from Buruli ulcer (BU) patients in a region of endemicity in Ghana with M. ulcerans sonicate or culture filtrate antigens to investigate the development of the response over time and its specificity by comparison with the response to Mycobacterium tuberculosis sonicate in human immunodeficiency virus-negative tuberculosis patients. Significant gamma interferon (IFN-gamma) production in response to whole-blood stimulation with M. ulcerans sonicate was detected in patients with ulcers, which was higher than that in patients with nodules but similar to subjects with healed BU. The mean IFN-gamma response in household contacts of BU patients was not significantly different from that in healthy control subjects from an area of nonendemicity. Results in patients with untreated, smear-positive pulmonary tuberculosis and tuberculosis patients on treatment for more than 2 weeks showed that BU patients responded better to M. ulcerans antigens than tuberculosis patients. In contrast, interleukin-10 results were higher in patients with active M. ulcerans disease than in those with healed lesions, but the pattern of response was similar to that seen in tuberculosis. A similar pattern of cytokine secretion was found using M. tuberculosis sonicate as an antigen. Neither of the two culture filtrate antigens of M. ulcerans appeared to be more specific than M. ulcerans sonicate. In the early stages of M. ulcerans disease there was a mixed Th1 and Th2 cytokine response, but the Th1 response emerged as the dominant type.

AIMS: The effect of a lactic acid producing bacterial (LAB) inoculant on the elimination of Escherichia coli O157:H7 from barley forage was assessed. METHODS AND RESULTS: Triplicate mini-silos were prepared for four treatments and six sampling times (1, 3, 7, 15, 30 and 42 d post-ensiling). The treatments were (i) 10(5) cfu g(-1) Pediococcus pentosaceus and Propionibacterium jenzenii (P2); (ii) 10(5) cfu g(-1) E. coli O157:H7 strain 3081 and 10(5) cfu g(-1) E. coli Biotype 1 strains 719IE10, 719IE14 and 614ME49 (EC); (iii) P2 + EC; and (iv) the control (sterile distilled water). Triplicate mini-silos were opened at each sampling time for pH, volatile fatty acid (VFA) and lactate determinations and E. coli, E. coli O157:H7 and LAB were enumerated. On d 3 and 7, numbers of E. coli O157:H7 in P2 + EC were significantly lower than in EC (P < 0;05). Escherichia coli O157:H7 was not detected in P2 + EC and EC at 7 and 15 d post-ensiling, respectively. On d 15 through 42, E. coli Biotype 1 was not detected in P2 + EC or EC. Populations of LAB were higher in P2 and P2 + EC than in the control and EC on d 3 and 7 (P < 0.05). After 3 d of ensiling, lactate levels were higher (P < 0.05) and pH was lower (P < 0.05) in P2 and P2 + EC as compared to the control and EC. Bacteriocins of P2 were not found to be inhibitory to E. coli O157:H7 using the agar-spot procedure. Escherichia coli O157:H7 inoculated into the control silage at a level of 10(3) cfu g(-1) and exposed to aerobic conditions at 22 degrees C was not detected after 1 d and remained undetectable for the 28 d exposure period. CONCLUSIONS: Silage inoculant P2 increased lactate levels and decreased pH more rapidly during ensiling, which appeared to hasten the elimination of E. coli O157:H7 from the silage. SIGNIFICANCE AND IMPACT OF THE STUDY: Results emphasize the importance of adequate ensiling since E. coli O157:H7 may be maintained and spread among cattle through feed.

Microbial enzymes extracted from mixed ruminal microorganisms were incubated for 2 h with casein and Tween 60 or Tween 80 at 10 concentrations ranging from 0 to 2.0% (vol/vol) to determine the effects of these nonionic surfactants on protease activation and thiol reactivity (unmasking of thiol groups). Rate and extent of protein adsorption to cellulosic substrate (barley straw) was measured in the presence of 0, 0.05, 0.10, 0.25, and 0.50% (vol/vol) Tween 80. Degradation of cellulose by a rumen bacterial fraction was measured over 48 h of incubation with and without Tween 60 or Tween 80 at 0.25% (vol/vol). Maximum accelerations of protease activity achievable with Tween 60 and Tween 80 (calculated from a Michaelis-Menten kinetics model) were 99.2 and 166.8%, respectively. Concentrations of Tween 60 and Tween 80 at which half the maximal velocities were attained were 0.28 and 0.20% (vol/vol), respectively. Tween 80 increased (P < 0.05) the rate and extent of adsorption of microbial protein to barley straw, and the effect was related to concentration of Tween 80 up to 0.10% (vol/vol). Initial rates of cellulose degradation with no surfactant, 0.25% Tween 60, or 0.25% Tween 80 were 0.60, 0.87, and 1.04 micrograms/ml per h, respectively. These nonionic surfactants were effective for enhancing rumen microbial protease and cellulase activities. Thus, further study is warranted to determine their potential for improving ruminant feeding.

Production records for a flock of Djallonké sheep kept in the forest belt of Ghana were analysed to determine the effects of some environmental factors on the performance of the breed. The period under study covered April 1969 to November 1979. The mean ages at first conception and first lambing and the mean lambing interval were 498, 638 and 264 days respectively. The overall twinning rate and prolificacy were 30.7 and 130.6% respectively. The season of birth and age of dam had significant (P less than 0.05) effects on prolificacy and twinning rate. The birth weights of the lambs were affected significantly (P less than 0.05) by age of dam, type of birth, sex and season of birth with the mean birth weight being 1.77 kg. Weaning weight (mean of 8.36 kg) and pre-weaning growth rate (mean of 0.076 kg/day) were affected significantly (P less than 0.05) by types of birth. Type of birth significantly (P less than 0.05) affected pre-weaning mortality of lambs (overall mean of 20.95%).
