RESUMO

Bed sediments and a dated sediment core were collected upstream and downstream from the city of Lyon (France) to assess the spatial and temporal trends of contamination by per- and polyfluoroalkyl substances (PFASs) in this section of the Rhône River. Upstream from Lyon, concentrations of total PFASs (ΣPFASs) in sediments are low (between 0.19 and 2.6 ng g-1 dry weight - dw), being characterized by a high proportion of perfluorooctane sulfonate (PFOS). Downstream from Lyon, and also from a fluoropolymer manufacturing plant, ΣPFASs concentrations reach 48.7 ng g-1 dw. A gradual decrease of concentrations is reported at the coring site further downstream (38 km). Based on a dated sediment core, the temporal evolution of PFASs is reconstructed from 1984 to 2013. Prior to 1987, ΣPFASs concentrations were low (≤2 ng g-1 dw), increasing to a maximum of 51 ng g-1 dw in the 1990s and then decreasing from 2002 to the present day (∼10 ng g-1 dw). In terms of the PFAS pattern, the proportion of perfluoroalkyl sulfonic acids (PFSAs) has remained stable since the 1980s (∼10%), whereas large variations are reported for carboxylic acids (PFCAs). Long chain- (C > 8) PFCAs characterized by an even number of perfluorinated carbons represent about 74% of the total PFAS load until 2005. However, from 2005 to 2013, the relative contribution of long chain- (C > 8) PFCAs with an odd number of perfluorinated carbons reaches 80%. Such changes in the PFAS pattern likely highlight a major shift in the industrial production process. This spatial and retrospective study provides valuable insights into the long-term contamination patterns of PFAS chemicals in river basins impacted by both urban and industrial activities.

The main objective of this study was to collect new data on the occurrence, levels of priority and emerging organic compounds in freshwater fish sampled in the Rhone River. The 34 studied contaminants included alkylphenols, bisphenol A, polybromodiphenylethers (PBDE), perfluorinated compounds, hexabromocyclododecanes (HBCD), hexachlorobenzene and hexachlorobutadiene (HCBD). About 50 fish samples (individual specimens or pooled fish) were collected from three sites located upstream and downstream of the Lyon metropolitan area in the Rhone River (France). Four species were caught at each site, namely: the barbel (Barbus barbus), the common bream (Abramis brama), the white bream (Blicca bjoerkna) and the chub (Squalius cephalus). Some contaminants were quantified in all the 32 fish samples analysed: 4-nonylphenol, α-HBCD, the six PBDE congeners (28, 47, 99, 100, 153, 154), perfluorooctanesulfonate (PFOS) and perfluorodecanoic acid. Twenty three of the 32 samples had a concentration of PFOS above the Environmental Quality Standards (EQS) (up to six times higher than the EQS), and all the 32 samples had concentrations of PBDE above the EQS (up to 4,000 times higher, with the sum of six PBDE varying from 4.5 to 182 ng/g dry weight). Clearly, the interest to consider PFOS and HBCD as new priority substances is confirmed. In contrast, the pertinence of a priority status for HCBD, which was never quantified in our study, might have to be reconsidered in the future.

Since 2005, restrictions have been because of fish consumption along the Rhone River because of high polychlorobiphenyl (PCB) concentrations, which have resulted inadverse economic consequences for professional fisheries in affected areas. French environmental authorities have expended considerable efforts to research sediment remediation strategies and development of sediment quality guidelines designed to protect the health of humans consuming Rhône River fish. Here we: (1) develop a bioaccumulation food-web model that describes PCB concentrations in three common freshwater fish species of the Rhône River, using Bayesian inference to estimate the input parameters; (2) test the predictive power of the model in terms of risk assessment for fish consumption; and (3) discuss the use of this approach to develop sediment quality guidelines that protect the health of humans consuming Rhône River fish. The bioaccumulation model predictions are protective for human consumer of fish and are efficient for use in risk assessment. For example, 85% of the predicted values were within a factor of 5 of measured CB153 concentrations in fish. Using sensitivity analyses, the major role played by sediment and diet behaviors on bioaccumulation process is illustrated: the parameters involved in the respiratory process (contamination from water) have little impact on model outputs, whereas the parameters related to diet and digestion processes are the most sensitive. The bioaccumulation model was applied to derive sediment concentrations compatible with safe fish consumption. The resulting PCB sediment thresholds (expressed as the sum of seven PCB indicator congeners) that are protective for the consumption of the fish species ranged from 0.7 to 3 ng/g (dw).

Many chemical, physiological, and trophic factors are known to affect bioaccumulation of polychlorinated biphenyls (PCBs) in biota. Understanding the primary factors affecting fish contamination is critical for predicting and assessing risks to upper-trophic level consumers, including humans. Here we identify PCB contamination pathways that could explain within- and between-species variability in fish concentration levels. Three freshwater river fish species (barbel, chub and bream) were sampled at three sites along the Rhone River (France) where fish consumption is partially prohibited because of PCB levels exceeding the European health-based benchmark. The trophic position was assessed using an innovative approach based on stable isotope analyses and Bayesian inference, which takes into account both isotope data variability and parameter uncertainty. The effect of foraging habitat on fish contamination was addressed using stable isotope mixing models. The fish trophic position and PCB concentrations were found to be unrelated while the exploitation of sediment detrital carbon as a food source appeared to be a critical factor affecting fish contamination. Fish length, PCB concentration of the sediment, and individual fish foraging habitat (exploitation of detrital versus planktonic carbon sources) explained 80% of within- and between-species variability observed in PCB concentrations. These results, obtained for species that have overlapping TPs and exploit different carbon sources, reveal that the important factor in fish PCB contamination is not only what fish consume, but also and essentially the feeding location.

Among the numerous PCB congeners, most of the dioxin-like PCBs (DL-PCBs) need to be characterized by hyphenated techniques. It has been shown in several instances that these congeners are well related to the total PCB content in fish. We examined datasets collected mainly in France, on freshwater and marine fish and sediments. A statistical model linking DL- and indicator PCBs was developed for a dataset composed of freshwater fishes, and proved to predict well DL-PCBs from indicator PCBs in all other fish sets, including marine ones. Type II error rates remained low in almost all fish sets. A similar correlation was observed in sediments. Non-dioxin-like PCBs elicit various adverse effects and represent 95% of the total PCBs. A European guideline for them is needed; the correlation between DL- and indicator PCBs could help develop this standard in the future.

Atrazine (6-chloro-N2-ethyl-N4-isopropyl-1,3,5-triazine-2,4-diamine) is frequently detected at high concentrations in ground water. Bentazone [3-isopropyl-1H-2,1,3-benzothiadiazin-4(3H)-one 2,2-dioxide] plus alachlor (2-chloro-2',6'-diethyl-N-methoxymethylacetanilide) is a potential herbicide combination used as a substitute for atrazine. Thus, the objective of this study was to assess the environmental risk of this blend. Drainage water contamination by bentazone and alachlor was assessed in silty clay (Vertic Eutrochrept) and silt loam (Aquic Hapludalf) soils under the same management and climatic conditions. Drainage volumes and concentrations of alachlor and bentazone were monitored after application. Herbicides first arrived at the drains after less than 1 cm of net drainage. This is consistent with preferential flow and suggests that about 3% of the pore volume was active in rapid transport. During the monitoring periods, bentazone losses were higher (0.11-2.40% of the applied amount) than alachlor losses (0.00-0.28%) in the drains of the silty clay and silt loam. The rank order of herbicide mass losses corresponded with the rank order of herbicide adsorption coefficients. More herbicide residues were detected in drainage from the silty clay, probably due to preferential flow and more intensive drainage in this soil than the silt loam. Surprisingly, herbicide losses were higher in the drains of both soils in the drier of the two study years. This could be explained by the time intervals between the treatments and first drainage events, which were longer in the wetter year. Results suggest that the drainage phases occurred by preferential flow in the spring-summer period, with correspondingly fast leaching of herbicides, and by matrix flow during the fall-winter period, with slower herbicide migration.

ANOVA-type data analysis, i.e.. determination of lowest-observed-effect concentrations (LOECs), and no-observed-effect concentrations (NOECs), has been widely used for statistical analysis of chronic ecotoxicity data. However, it is more and more criticised for several reasons, among which the most important is probably the fact that the NOEC depends on the choice of test concentrations and number of replications and rewards poor experiments, i.e., high variability, with high NOEC values. Thus, a recent OECD workshop concluded that the use of the NOEC should be phased out and that a regression-based estimation procedure should be used. Following this workshop, a working group was established at the French level between government, academia and industry representatives. Twenty-seven sets of chronic data (algae, daphnia, fish) were collected and analysed by ANOVA and regression procedures. Several regression models were compared and relations between NOECs and ECx, for different values of x, were established in order to find an alternative summary parameter to the NOEC. Biological arguments are scarce to help in defining a negligible level of effect x for the ECx. With regard to their use in the risk assessment procedures, a convenient methodology would be to choose x so that ECx are on average similar to the present NOEC. This would lead to no major change in the risk assessment procedure. However, experimental data show that the ECx depend on the regression models and that their accuracy decreases in the low effect zone. This disadvantage could probably be reduced by adapting existing experimental protocols but it could mean more experimental effort and higher cost. ECx (derived with existing test guidelines, e.g., regarding the number of replicates) whose lowest bounds of the confidence interval are on average similar to present NOEC would improve this approach by a priori encouraging more precise experiments. However, narrow confidence intervals are not only linked to good experimental practices, but also depend on the distance between the best model fit and experimental data. At least, these approaches still use the NOEC as a reference although this reference is statistically not correct. On the contrary, EC50 are the most precise values to estimate on a concentration response curve, but they are clearly different from the NOEC and their use would require a modification of existing assessment factors.

Fluvial suspended particulates collected from the Rhine River were investigated in terms of composition, structure and size distribution. Elemental analysis and Diffuse Reflectance Infrared Spectroscopy reveal that most particulate organic matter is formed from material derived from microorganisms. Transmission Electron Microscopy observations on resin-embedded samples and structural characterization from break-up experiments, show that fluvial particulate matter should be viewed as fractal aggregates organized by bacterial exopolymeric substances. The shape of particulate size distribution suggests that the formation and dynamics of suspended particulate matter are controlled mainly by physical processes. Finally, particulate growth and structure are consistent with a cluster-cluster aggregation scheme.

Antioxidant systems were studied in the freshwater bivalve Unio tumidus transplanted from a control site to four different contaminated areas, in order to study the biological response according to the contamination characteristics. Reduced and oxidized glutathione (GSH, GSSG), the activities of antioxidant enzymes such as selenium-dependent and non-selenium-dependent glutathione peroxidases (SeGPx and non-SeGPx), and glutathione reductase (GRd) were measured in the gills and the digestive gland of the mussels after 15 days of exposure at different sites. Lipid peroxidation (LPO) was evaluated by means of malondialdehyde measurements (MDA). The four sites investigated were located in the valleys of Fensch (F), Moselle (M), Lot et Garonne (LG), and Sarthe (S). At each site, the bivalves were placed upstream (Up) from an identified pollution source (a cokery, a laundry, or a foundry) and downstream (Do), close to the effluent outfall (Do(1)) or farther (Do(2)). The goal was to study the antioxidant response in relation to the pollution gradient. Metals and congeners of PAHs, PCBs, and organochlorinated pesticides were analyzed in the river sediments of each station. The exposure of the bivalves at the most highly polluted sites or close to the pollution source led to a sharp depletion in some antioxidant parameters, namely GRd, SeGPx, and GSH. The decrease in enzyme activities could reach 80% for GRd and 70% for SeGPx, while GSH depletion could yield 70%, leading then in an induction of lipid peroxidation, either in the digestive gland or in the gills. The higher the MDA concentrations, the lower the activity of these three antioxidant parameters, suggesting that they could be biomarkers for toxicity. Yet, a depletion in these parameters was sometimes insufficient for cytotoxicity to be induced, since lipid peroxidation failed to appear in some cases where antioxidant depletion was clear, although not so severe. The response of the gills and the digestive gland was not always paralleled, which can be explained by differences in the bioavailability of pollutants. In some cases, a relationship was not found between the antioxidant response and the degree and the type of contamination in sediments, suggesting that the effects could result from nonidentified pollutants or/and be indicators of bio-availability.

OBJECTIVE: To investigate the impact of HIV infection on the prevalence, incidence and short-term prognosis of squamous intraepithelial lesions (SIL), in a prospective study with 1-year follow-up. METHODS: Between 1993 and 1995, 271 HIV-positive and 171 HIV-negative women at high risk of HIV infection were recruited, 365 (82.6%) of whom completed the 1-year follow-up. The women underwent a Papanicolaou smear test at inclusion and at 6 and 12 months. Human papillomavirus (HPV) was detected at inclusion by Southern blot and PCR. RESULTS: The SIL prevalence ranged from 7.5% for HIV-negative to 31.3% for HIV-positive women with CD4 cell counts < 500 x 10(6)/l (P < 0.001). Other factors associated independently and significantly with SIL prevalence were HPV-16, 18, 33 and related types, HPV-31, -35, -39 and related types, lifetime number of partners, younger age, past history of SIL and lack of past cervical screening. The SIL incidence ranged from 4.9% in HIV-negative women to 27% in HIV-positive women with CD4 cells < 500 x 10(6)/l (P < 0.001). Progression from low- to high-grade SIL during follow-up was detected in 38.1% of HIV-positive women with CD4 cells < or = 500 x 10(6)/l but in no HIV-negative nor HIV-positive women with CD4 cells > 500 x 10(6)/l. HPV-16, 18, 33 and related types were also associated with higher incidence of SIL and progression from low- to high-grade SIL. CONCLUSION: HIV-induced immunodeficiency is associated with high prevalence, incidence and persistence/progression of SIL. A pejorative influence of HIV infection without marked immunodeficiency is less clear. HIV-positive women with SIL may thus benefit from early treatment when a useful immune response is still present.

The aim of this study was to evaluate the usefulness of antioxidant parameters in the freshwater bivalve, Unio tumidus, as biomarkers of exposure to pollutants and to study their potential interest in predicting toxicity. Selenium-dependent glutathione peroxidase (Se-GPx), non-selenium-dependent glutathione peroxidase (non-Se-GPx), glutathione reductase (GRd), catalase, and superoxide dismutase (SOD) activities; reduced (GSH) and oxidized (GSSG) glutathione levels; and lipid peroxidation were measured in the gills and digestive glands of Unio. Control mussels were encaged and transplanted for 15 and 30 days to sites where the contamination of sediments was analyzed, along a river receiving domestic and industrial sources of pollution. After 15 days of exposure, all antioxidant parameters of the bivalves transferred to the most polluted sites had strongly decreased compared with control values. This was particularly true for Se-GPx and GRd activities, which were inhibited by 60 and 80% in the two tissues, and for GSH levels (80% reduction in the gills and 60% in digestive glands). These decreases were associated in the gills with lipid peroxidation (measured by malondialdehyde content) and with a high level of contamination of sediments by polycyclic aromatic hydrocarbons and polychlorinated biphenyls. In the mussels exposed at the least polluted sites, the same parameters decreased in the gills, but to a lesser extent: 50% for Se-GPx and 32% for GRd activities, and 45% for GSH levels. The gills appeared more sensitive than the digestive glands. After 30 days of exposure, while Se-GPx, GRd, and GSH remained reduced, a significant induction of non-Se-GPx and catalase activities was recorded in the gills, which reflected an adaptation of the transplanted species to their unsafe environment. All the results indicated that antioxidant defense components, namely, Se-GPx, GRd, and GSH, are sensitive parameters that could be useful biomarkers for the evaluation of contaminated aquatic ecosystems. The relationship between the degree of deficiency of antioxidant defenses and lipid peroxidation suggests that these parameters could also be biomarkers for toxicity.

Electroplating effluents were tested for their genotoxicity with the micronucleus test on newt larvae. The metallic content of the tested samples was responsible for the induction of micronuclei in red blood cells (RBC). Then, iron (Fe3+), chromium (Cr3+, Cr6+) and zinc (Zn2+) which were identified in these samples, were tested either separately or combined, at their concentrations in the electroplating effluents. Fe3+ induced a high level of micronuclei at 12.5 and 25 mg/l (nominal concentrations). Both soluble and non-soluble forms of iron were responsible for these genotoxic effects. At lower concentrations (0.6 and 4.5 mg/l) Fe3+ was not systematically genotoxic. Zinc could not be considered genotoxic on newt. Cr3+ gave negative responses, but exposure to Cr6+ (1 mg/l) could result in a significant number of micronucleated RBC in some cases. The most dramatic genotoxic effects were registered when Fe3+ and Cr6+ were combined. This study demonstrates that interactions between pollutants and the effects of non-soluble chemicals on aquatic vertebrates and invertebrates can no longer be neglected.
