RESUMO

In this paper, we innovatively proposed a highly uniform vesicle preparation scheme based on the intervesicle mechanical self-constraint effect of vesicle crowding. By adjusting the spacing of discrete microwell structures, we observed that during the self-assembly of phospholipid molecules in microwells to form giant unilamellar vesicles (GUVs), the scale swelling of the vesicles during the continuous growth process would lead to the crowding of vesicles in adjacent microwells, thus inducing the formation of intervesicle mechanical self-constraint effect. The results of the experiment showed that this paper obtained the optimized discretized microwell structure (micropillar side: 30 µm; pitch: 0 µm), and the corresponding lipid mass was measured and determined, yielding homogeneous giant GUVs of 37.9 ± 2.0 µm. In this paper, homogenized GUVs (∼40 µm) with different cholesterol concentrations (10, 20, and 30%) were obtained by this method, and the above vesicles were subjected to controlled electroporation experiment under external electric fields of 23, 31, and 41 kV/cm, respectively. It showed that the mechanical self-constraint effect of vesicle crowding induced by patterned microstructures during the self-assembly of phospholipid molecules significantly enhances the size homogeneity of GUVs, which would be helpful for the wide applications of GUVs in other areas such as cell-like models and controlled release of drugs.

Glass nanopipettes, as important sensing tools, have attracted great interest due to their wide range of applications in detecting single molecules, nanoparticles, and cells. In this study, we investigated the translocation behavior of nanorod particles through dual-nanopipettes using a transient continuum-based model based on an arbitrary Lagrangian-Eulerian approach. Our findings indicate that the translocation of nanorods is slowed down in the dual-nanopipette system, especially in the dual-nanopipette system with a nanobridge. These results are in qualitative agreement with previous experimental findings reported in the literature. Additionally, the translocation of nanorods is influenced by factors such as bulk concentration, initial location of the nanorod, and surface charge of the nanopipette. Notably, when the surface charge density of the nanopipette is relatively high and the initial location of the nanorod is in the reservoir, the nanorod can hardly enter the nanopipette, resulting in a relatively low translocation efficiency. However, the translocation efficiency can be improved by initially positioning the nanorod in one of the barrels. The resulting dual-blockade current signal can be used to correlate the characteristics of the nanorod.

Cell electrofusion is a key process in many research fields, such as genetics, immunology, and cross-breeding. The electrofusion efficiency is highly dependent on the buffer osmotic pressure properties. However, the mechanism by which the buffer osmotic pressure affects cell electrofusion has not been theoretically or numerically understood. In order to explore the mechanism, the microfluidic structure with paired arc micro-cavities was first evaluated based on the numerical analysis of the transmembrane potential and the electroporation induced on biological cells when the electrofusion was performed on this structure. Then, the numerical model was used to analyze the effect of three buffer osmotic pressures on the on-chip electrofusion in terms of membrane tension and cell size. Compared to hypertonic and isotonic buffers, hypotonic buffer not only increased the reversible electroporation area in the cell-cell contact zone by 1.7 times by inducing a higher membrane tension, but also significantly reduced the applied voltage required for cell electroporation by increasing the cell size. Finally, the microfluidic chip with arc micro-cavities was fabricated and tested for electrofusion of SP2/0 cells. The results showed that no cell fusion occurred in the hypertonic buffer. The fusion efficiency in the isotonic buffer was about 7%. In the hypotonic buffer, the fusion efficiency was about 60%, which was significantly higher compared to hypertonic and isotonic buffers. The experimental results were in good agreement with the numerical analysis results.

This paper presents a microfluidic device with 200 cell "cage" structures. Based on this microfluidics device, a new simple and stable electrofusion method was developed. Under hydrodynamic force, the cells moved to the desired "cage" cell capture structure and efficiently formed cell pairs (∼80.0 ± 4.6%). Intimate intercellular connectivity was induced by the precise modulation of hypotonic solution substitution and the microstructure, which ensured no cell movement or displacement during the cell electroporation/electrofusion process. It also guaranteed repeated electroporation occurring in the same contact region and provided a stable cell membrane recombination and a cell fusion microenvironment. When the pulse signal was applied, a high fusion efficiency of ∼88.3 ± 0.6% was demonstrated on the microfluidic device.

Background: The causal relationship between certain lifestyle factors and erectile dysfunction (ED) is still uncertain. Aim: The study sought to investigate the causal effect of 9 life factors on ED through 2-sample single-variable Mendelian randomization (SVMR) and multivariable Mendelian randomization (MVMR). Methods: Genetic instruments to proxy 9 risk factors were identified by genome-wide association studies. The genome-wide association studies estimated the connection of these genetic variants with ED risk (n = 223 805). We conducted SVMR, inverse variance-weighting, Cochran's Q, weighted median, MR-Egger, MR-PRESSO (Mendelian Randomization Pleiotropy RESidual Sum and Outlier), and MVMR analyses to explore the total and direct relationship between life factors and ED. Outcomes: The primary outcome was defined as self or physician-reported ED, or using oral ED medication, or a history of surgery related to ED. Results: In SVMR analyses, suggestive associations with increased the risk of ED were noted for ever smoked (odds ratio [OR], 5.894; 95% confidence interval [CI], 0.469 to 3.079; P = .008), alcohol consumption (OR, 1.495; 95% CI, 0.044 to 0.760; P = .028) and body mass index (BMI) (OR, 1.177; 95% CI, 0.057 to 0.268; P = .003). Earlier age at first intercourse was significantly related to reduced ED risk (OR, 0.659; 95% CI, -0.592 to -0.244; P = 2.5 × 10-6). No strong evidence was found for the effect of coffee intake, time spent driving, physical activity, and leisure sedentary behaviors on the incidence of ED (All P > .05). The result of MVMR analysis for BMI (OR, 1.13; 95% CI, 1.01 to 1.25; P = .045) and earlier age at first intercourse (OR, 0.77; 95% CI, 0.56 to 0.99; P = .018) provided suggestive evidence for the direct impact on ED, while no causal factor was detected for alcoholic drinks per week and ever smoked. Clinical implications: This study provides evidence for the impact of certain modifiable lifestyle factors on the development of ED. Strengths and limitations: We performed both SVMR and MVMR to strengthen the causal relationship between exposures and outcomes. However, the population in this study was limited to European ancestry. Conclusion: Ever smoked, alcoholic drinks per week, BMI, and age first had sexual intercourse were causally related to ED, while the potential connection between coffee intake, physical activity, recreational sedentary habits, and increased risk of ED needs to be further confirmed.
