RESUMO

The presence of anti-polyethylene glycol (PEG) antibodies can limit the clinical efficacy of PEGylated drugs and cause anaphylactic reactions in patients. Monocytes/macrophages are effector cells involved in IgG-mediated passive systemic anaphylaxis. We studied the influence of human blood serum on the efficiency of uptake of PEGylated nanoparticles by human blood monocytes. It has been shown that magnetic nanoparticles modified with PEG-3000 and solid lipid nanoparticles containing PEG-2000 are avidly internalized by human blood monocytes in vitro, the uptake efficiency depends on the features (composition) of donor blood serum, but does not correlate with the level of the IgG antibody against PEG.

RESUMO

We studied an original radiolabeled complex of antimicrobial peptides UBI29-41 and UBI18-35, ubiquicidin derivatives, for distinguishing between bacterial and aseptic inflammation. For radiolabeling of the peptides with technetium-99m, a bifunctional chelating agent succinimide-1-yl 6-(bis(pyridin-2-ylmethyl)amino)hexanoate was used. The obtained complexes 99mТс-DPAH-UBI29-41 and 99mТс-DPAH-UBI18-35 had radiolabeling yield >80% and radiochemical purity >96%. Accumulation of the complexes in the focus of bacterial inflammation in bone structures and the absence of this complex in the site of aseptic inflammation was confirmed in a rat model of traumatic osteomyelitis by single-photon emission computed tomography.

BACKGROUND: Heavy bleeding is developed in case of spleen injury. It often leads to death. At present the search and development of new methods for hemostasis of spleen bleeding continues. An innovative method is coagulation of nonequilibrium plasma. OBJECTIVE: Our aim was to study the effect of nonequilibrium plasma on the morphofunctional state of the spleen. METHODS: The non-randomized study by type of "case-control" was conducted. The experiment was carried out on 45 male rabbits weighing 3000-3200 g. 1st group (control; n = 5)--intact animals without surgery; 2nd group (experimental, n = 40)--animals after atypical resection of the spleen and hemostasis by nonequilibrium plasma. The volume of blood loss, FBC, histological data, splenogram analysis data were assessed at defined periods (60 min, 3rd, 5th, 7th, 14th, 30th, 90th, 180th days) after surgery. They were compared with data of the control group. RESULTS: The volume of blood loss during the resection of the spleen with hemostasis by nonequilibrium plasma was 16.6 [15.98; 17.22] ml. Increase of neutrophil to 38 [24; 39] (control group 17[13, 18], p = 0.009), monocytes to 15 [14, 18] (control group 8 [6, 11], p = 0.036) is revealed in the FBC in the early period after hemostasis with nonequilibrium plasma. It is reduced to the norm on 30 day. Histological examination revealed leukocyte infiltration, edema, microvascular stasis, dilatation of vessels in the early period. At long-term period structure of the organ is normalized. Splenogramm analysis revealed a statistically significant (p = 0.023) decrease in the relative number of small lymphocytes by 28% in animals after hemostasis with nonequilibrium plasma as compared to control (23.3 [179; 26.7] versus 30.8 [29.25; 34.3] respectively). CONCLUSION: treatment of bleeding surface by plasma flow for 1.5-2 min is required order to achieve effective hemostasis during surgery of spleen injuries in the experiment. After coagulation there is minimal damage of the spleen parenchyma, which manifests itself as activation reaction. At long-term period parenchyma ofthe spleen completely regenerates with theformation of cicatrical tissue that does not affect thefunctioning of organ.

Authors submitted an analysis of papers given up an involvement of protein kinases in heart ischemic postconditioning. This analysis of literature source allowed to authors affirms that signaling system of postconditioning can involve kinases: PKC, PI3K, Akt, MEKl/2, ERK1/2, MTOR, p70s6K, GSK3b, PKG and also eNOS, NO, GC, motoKATP channel, ROS, MPT pore. At the same time it is unclear a real contributions of kinases mTOR, p70s6, AMPK and GSK3b in the mechanism of infarct limiting impact of postconditioning. It is required a further study of the chain of signaling events following JAK2 and p38 kinase activation. The knowledge of Ras and Raf-1 role in postconditioning has hypothetical character. The tyrosine kinase significance in postcondi-tioning is unclear, particular Src kinase, which plays an important role in the regulation of cardiac tolerance to an impact of ischemia and reperfusion.

Analysis of literature source indicates that main pretenders to the role of end-effectors of ischemic postconditioning of the heart are: 1) Ca(2+)-dependent K+ channel of BK-type (big conductance K+ channel), 2) mitoK(ATP) channel (mitochondrial ATP-sensitive K(+)-channel), 3) MPT pore (mitochondrial permeability transition pore). At the same time, some investigators consider that mitoK(ATP) channel is only an intermediate link in the series of signaling events ensured an increase in cardiac tolerance to impact of ischemia-reperfusion. The most likely end-effector of the three structures is MPT pore. Alternatively, it is possible, that unique molecular complex appearing a single end-effector of postconditioning does not exist. Perhaps, that there are several effectors ensured cardioprotective effect of adaptive phenomenon of postconditioning.

RESUMO

The effects of recombinant IL-2 on apoptosis of lymphocytes of healthy donors were studied in in vitro experiments. It was shown that the inductive and inhibitory effects of IL-2 on apoptotic process depend on the dose of the cytokine and cell microenvironmental conditions. Culturing of lymphocytes with recombinant IL-2 increases the percent of cells with reduced transmembrane potential, reduces the content of intracellular proteins Bcl-2, Bcl-X(L) and и Bax, and increases the level of Bad. The proapoptotic effect of this cytokine is realized with participation of nuclear transcription factor NF-κB and transcription factor P53.

RESUMO

The ultradispersed piezoceramic powder containing lead oxides, zirconium, titanium, bismuth, cadmium, manganese, strontium, and chromium was tested for its effect on the health status of the staff serving pilot industrial units producing the powder. There was a significant increase in the incidence of digestive disease, a tendency for respiratory and osteomuscular diseases to increase. It is concluded that it is necessary to use individual means of the protection and prophylaxis of the observed diseases.

The essential decrease of the rate of turnover of phosphatidic acids (PA), polyglycerophosphates (PGP) and their lysoderivatives (LPGP), sphinigomyelin (SM) and lysophosphatidylcholine (LPC), phosphatidyletnanolamine (PEA) was found in male mice of BALB/c strain with subacute alloxan diabetes (glucose concentration in the blood was not less than 14 mmol/l) during examination of phospholipid metabolism in the liver by the rate of 3H-acetate and 14C-palmitate incorporation into corresponding chromatographic phospholipid fractions. On the contrary, turnover of phosphatidilcholine (PC) significantly increased. The of main structural phospholipid components of hepatocyte cellular membranes--PEA and PC--were found to be decreased by 41 and 32%, respectively. LPGP and PA levels decreased to a lesser extent. The level of PGP, SM, phosphatidylinositol and LPG did not differ from the control level.

RESUMO

The authors studied morphological changes in renal tissue in terminal hydronephrosis and in exposure of freezing, regulation of arterial pressure and erythropoiesis in cryogenic effects on hydronephrotic kidney in experiment. Reparative changes in the parenchyma appear on day 30 after freezing, systemic arterial pressure and bone marrow erythropoietic activity returned to normal. Freezing is thought more physiological compared to nephrectomy in respect to prevention of nephrogenic hypertension and anemia.

The paper presents experimental evidence on morphologic changes in intact kidney, tissue temperature, renal blood supply and glomerular filtration in response to freezing. Cryogenic effects manifested as necrotic and dystrophic processes. Damage to the tubular and glomerular cells can be corrected. Blood flow in the kidney was not interrupted but was directed through the bypass routes.

RESUMO

Experiments on animals have been carried out to investigate the hemodynamic changes during manual abdominal compression (AC) under conditions of heart fibrillation and acute heart failure. The possibility to increase the efficacy of left ventricular bypass (LVB) has been studied. Manual AC enhanced the pressure and blood flow in low-pressure compartments and increased aortic systolic and diastolic pressure. It has been also noted that LVB efficacy was increased in simultaneous assisted perfusion and abdominal compression. The first clinical data confirm the experimental findings.

The metabolism of neutral lipids (free and ester-bound cholesterols, mono-, di- and triacylglycerides) was greatly impaired in liver, spleen, thymus tissues and ileocecal lymph nodes of BALB/c mice with subacute alloxan diabetes (content of sugar in blood was no less than 14 mmol/l) as shown by 3H-acetate and 14C-palmitate incorporation into the corresponding lipid fractions. The rate of neutral lipid turnover was specifically decreased in all the tissues studied. However, a considerable accumulation of these lipid fractions was found in thymus tissue. Incorporation of labelled palmitate into triacylglycerides was increased in the spleen but without obesity of the organ.

RESUMO

Rates of 3H-thymidine, 3H-uridine and 14C-glycine incorporation in nucleic acids and total proteins were studied in cells of bone marrow, spleen, thymus and ileocecal lymph nodes tissues of BALB/c mice with subacute alloxan diabetes (content of blood sugar was higher than 14 mmol/l). Synthesis of DNA, RNA and protein was not considerably inhibited in the thymus tissue of the impaired animals which may correlate with pronounced dysfunction of cytotoxic T lymphocytes typical for the disease.

RESUMO

The experience of the use of noninvasive assisted circulation in patients with cardiac pathology is summarized. It has been demonstrated that rhythmic pneumo-compression of the lower extremities, abdominal compression, muscular electrical stimulation of the lower extremities and the abdomen have a pronounced effect on circulation compartments with low pressure. All the above pre-determines the role of these techniques in the management of cardiac failure and in intensive care.

A rate of incorporation of 3H-thymidine, 3H-uridine and 14C-glycine into the nucleoproteins of bone marrow, spleen, thymus and ileocecal lymph nodes was studied in BALB/c mice with hypothyrosis developed after administration of 1% KClO4 within a month. Distinct hypoplasia of bone marrow and spleen was found in the experimental animals; the synthesis of nucleoproteins was most impaired in bone marrow. The dependence of hemopoietic tissues on thyroid hormones was decreased as follows: bone marrow > spleen > thymus > lymph node. After subcutaneous administration of triiodothyronine (T3) in a dose of 50 mg/kg daily within 3 days of synthesis of nucleoproteins was considerably stimulated and the amount of cells was increased in bone marrow and spleen. Less distinct stimulating effect of T3 was observed in the thymus and lymph nodes of these animals.

RESUMO

Thirty experiments were conducted on calves for hemodynamic study of mechanical aid to the heart (biventricular bypass by two artificial ++"Module"+ ventricles of the heart) in acute cardiac failure. The cavities of the heart were cannulated with a special perforating device which, when necessary, is an obturator of the main line. Analysis of the hemodynamic values showed a sufficiently high efficacy of biventricular shunting in acute cardiac failure. The device for draining the heart cavities has advantages over the traditional main lines, in particular the number of complications from bleeding and air embolism reduces with its use.

Experiments were performed on 15 mongrel dogs to evaluate the influence of electrostimulated contractions of the hind limb and abdominal skeletal muscles on the efficacy of assisted circulation (AC). The influence of massage of the abdomen on AC efficacy was studied in 10 experiments. In electrostimulation of the hind limb muscles the drainage along the femoral vein, measured by the direct method, increased threefold: from 58 +/- 9 to 162 +/- 7 ml/min under conditions of an intact heart and from 5.8 +/- 0.8 to 15 +/- 1.2 ml/min in cardiac insufficiency. The efficacy of AC by the method of left-ventricular bypass by means of the Module artificial heart ventricle (AHV) increased through the flow of blood to the heart, which was manifested by greater AHV performance with rise of systolic and diastolic pressure to 7-8 mm Hg. In cardiovascular insufficiency the influence of electrostimulation on AC efficacy was weaker, the systolic and diastolic pressure increased only by 2-4 mm Hg, in massage of the abdomen both indices increased by 8-10 mm Hg.
