Background: Given that epidemiological evidence suggests a potential protective role for Bacille-Calmette-Guerin against COVID-19, we aimed to explore whether pre-exposure of human monocyte-derived macrophages and dendritic cells to BCG could modulate their response to SARS-CoV-2 S-glycoprotein. Methods: Dual THP-1 cells containing 2 reporter plasmids for transcription factors NF-κB, and IRF were differentiated into macrophages over 3 days using phorbol 12-myristate 13-acetate, or into dendritic cells over 6 days using commercial monocyte-dencritic cell differentiation media and matured with recombinant tumor necrosis factor-α. Cells were exposed to BCG for 24 h and then stimulated with SARS-CoV-2 S-glycoprotein for 24 hours. Results: Pre-exposure of human macrophages and DCs to BCG increased IRF and NF-kb activation in response to the SARS-CoV-2 S-glycoprotein. Conclusions: Our results showed that pre-exposure of both types of cells to BCG exhibited an increase in inflammatory transcription factors upon stimulation with S-glycoprotein. BCG-induced trained immunity may be an important tool for reducing susceptibility to SARS-CoV-2 infection and severity of COVID-19. Our findings help in the design of future BCG-based therapeutic approaches in the treatment of diseases caused by viral infections.

Bisphenols such as bisphenol A (BPA), S (BPS), C (BPC), F (BPF), AF (BPAF), tetrabromobisphenol, nonylphenol, and octylphenol are plasticizers used worldwide to manufacture daily-use articles. Exposure to these compounds is related to many pathologies of public health importance, such as infertility. Using a protector compound against the reproductive toxicological effects of bisphenols is of scientific interest. Melatonin and vitamins have been tested, but the results are not conclusive. To this end, this systematic review and meta-analysis compared the response of reproductive variables to melatonin and vitamin administration as protectors against damage caused by bisphenols. We search for controlled studies of male rats exposed to bisphenols to induce alterations in reproduction, with at least one intervention group receiving melatonin or vitamins (B, C, or E). Also, molecular docking simulations were performed between the androgen (AR) and estrogen receptors (ER), melatonin, and vitamins. About 1234 records were initially found; finally, 13 studies were qualified for review and meta-analysis. Melatonin plus bisphenol improves sperm concentration and viability of sperm and increases testosterone serum levels compared with control groups; however, groups receiving vitamins plus bisphenols had lower sperm concentration, total testis weight, and testosterone serum levels than the control. In the docking analysis, vitamin E had the highest negative MolDock score, representing the best binding affinity with AR and ER, compared with other vitamins and melatonin in the docking. Our findings suggest that vitamins could act as an endocrine disruptor, and melatonin is most effective in protecting against the toxic effects of bisphenols.

Reactive oxygen species are frequently associated with various cancers including pancreatic ductal adenocarcinomas (PDACs). Superoxide dismutase 2 (SOD2) is an enzyme that plays an important role in reactive oxygen species (ROS) signaling. Investigating the molecular function and biological functions of SOD2 can help us develop new therapeutic options and uncover new biomarkers for PDAC diagnosis and prognosis. Here, we show that nimbolide (NB), a triterpene limonoid, effectively blocks the growth and metastasis of PDACs by suppressing the expression and activity of SOD2. To identify the role of SOD2 in NB-induced anticancer activity, we used RNA interference to silence and plasmid transfection to overexpress it. Silencing SOD2 significantly reduced the growth and metastatic characteristics like epithelial-to-mesenchymal transition, invasion, migration, and colony-forming capabilities of PDACs, and NB treatment further reduced these characteristics. Conversely, the overexpression of SOD2 enhanced these metastatic characteristics. ROS signaling has a strong feedback mechanism with the PI3K/Akt signaling pathway, which could be mediated through SOD2. Finally, NB treatment to SOD2-overexpressing PDAC xenografts resulted in significant inhibition of tumor growth and metastasis. Overall, this work suggests that NB, a natural and safe phytochemical that silences SOD2 to induce high levels of ROS generation, results in increased apoptosis and reduced growth and progression of PDACs. The role of SOD2 in regulating NB-induced ROS generation presents itself as a therapeutic option for PDACs.

(1) Background: This study focusses on establishing the relationship between quality of movement (based on the functional movement screen, or FMS) and foot posture (based on the foot posture index, or FPI-6). We hypothesised that a poor FMS test score may be derived from the foot position in the space measured by FPI-6. (2) Methods: a quasi-experimental and cross-sectional study was designed to analyse foot posture in 30 healthy football players, using the foot posture index and the functional movement screen. (3) Results: No significant relationships were found between movement quality and foot posture. Poor movement quality is present in more than half of all foot positions, supination, pronation, and neutral. Good quality seems to be more associated with a neutral foot position (23.3%) and supinated (16.6%) than a pronated foot position (6.6%). (4) Conclusions: this study found no relationship between the two tests; therefore, we cannot demonstrate that foot posture is relevant in the quality of the movement of the football players studied.

This work presents an inventory of the Polyplacophora species registered in December 2019 in the intertidal environment of 10 sampling sites of the Northern Gulf of California, México. Fifteen species were obtained from a total of 33 species previously reported throughout the Gulf. Acanthochitona avicula, Ischnochiton guatemalensis, Lepidozona serrata, and Dendrochiton lirulatus are rare with only 1 to 3 specimens recorded of each species. Stenoplax mariposa and Lepidozona subtilis were the most abundant with 162 and 284 specimens, respectively. Acanthochitona avicula, I. guatemalensis, Lepidozona serrata, and Lepidochitona beanii are rare and each was only recorded at one sampling site. For a better recognition of these species, we provide a dichotomous identification key, which is complemented with high-resolution photographs and images obtained by scanning electron microscopy. In addition, habitat data and taxonomic comments are included to facilitate a better understanding of the habitat preferences, relationships and current taxonomic position of these species.

Aluminum-based alloys have been considered candidate materials for cathodic protection anodes. However, the Al-based alloys can form a layer of alumina, which is a drawback in a sacrificial anode. The anodes must exhibit uniform corrosion to achieve better performance. Aluminum can be alloyed with Zn to improve their performance. In this sense, in the present research, the electrochemical corrosion performance of Al-xZn alloys (x = 1.5, 3.5, and 5 at.% Zn) exposed to 3.5 wt.% NaCl for 24 h was evaluated. Polarization curves, linear polarization resistance (LPR), and electrochemical impedance spectroscopy (EIS) were used to identify the electrochemical behavior. The microstructure of the samples before the corrosion assessment was characterized by means of X-ray diffraction analyses (XRD) and scanning electron microscopy (SEM). In addition, microstructures of the corroded surfaces were characterized using X-ray mappings via SEM. Polarization curves indicated that Zn additions changed the pseudo-passivation behavior from what pure Al exhibited in a uniform dissolution regime. Furthermore, the addition of Zn shifted the corrosion potential to the active side and increased the corrosion rate. This behavior was consistent with the proportional decrease in polarization resistance (Rp) and charge transfer resistance (Rct) in the EIS. The analysis of EIS was done using a mathematical model related to an adsorption electrochemical mechanism. The adsorption of chloride at the Al-Zn alloy surface formed aluminum chloride intermediates, which controlled the rate of the process. The rate constants of the reactions of a proposed chemical mechanism were evaluated.

Training simulators can facilitate the acquisition and development of basic surgical skills, and they constitute a safe and humane method that does not harm animals in the process. The objective of this work was to create and evaluate a low-cost simulator to help undergraduate students of veterinary medicine acquire and practise vascular ligation skills. A training model was made by using easily accessible and inexpensive materials. Fifteen students, without prior surgical experience, each performed the orchiectomy technique on the simulator seven times. The emphasis was on performing vascular ligatures in both testicles of the simulator (n = 14), in two different scenarios: firstly, with the contents of the blood vessels not under pressure; and secondly, with the syringe plunger depressed by 1 ml to pressurise the blood vessels. The outcomes of the procedure in the simulator were based on three qualitative criteria: Correct (no 'bleeding'), Sufficient (light 'bleeding') and Incorrect (heavy 'bleeding'). After the seventh attempt, all participants were able to perform vascular ligatures with at least a score of Sufficient in both scenarios. By the 10th ligature attempt, they were all able to perform the procedure with a score of Correct (p < 0.05). There was a trend toward a decrease in the time taken to carry out the procedure as learning progress was made during training, with this being significant from the 12th attempt (p < 0.05). The use of this low-cost simulator represents a useful didactic tool, which supports the acquisition and practise of manual skills by using methods that do not involve the use of animals. In addition, this training simulator could be useful in distance learning, in view of the ready accessibility of the materials required for its construction.

This study aimed to establish the optimal operational conditions for hydrogen production using vermicomposting-tea and sugarcane molasses as substrate. The experiments were carried out by triplicate in 110 ml serological bottles, a Box-Behnken design of experiments was performed in anaerobic dark conditions. The maximal hydrogen production (HP), hydrogen production rate (HPR), and hydrogen yield (HY) attained were 1021.0 mlL-1, 5.32 mlL-1h-1, and 60.3 mlLH2-1/gTCC, respectively. The statistical model showed that the optimal operational conditions for pH, molasses concentration, and temperature were 6.5; 30 % (v/v) and 25 °C. The bioreactor run showed 17.202 L of hydrogen, 0.58 Lh-1, and 77.2 mlH2gTCC-1 For HP, HPR, and HY. Chemometric analysis for the volatile fatty acids obtained at the fermentation showed that only two principal components are required to explain 90 % of the variance. The representative pathways for hydrogen production were acetic and butyric acids. This study established the operational conditions for the upstream processing amenable to pilot and industrial-scale operations. Our results add value to molasses within the circular economy for hydrogen production using a novel consortium from vermicompost.

BACKGROUND: Psoriasis (Ps) is an autoimmune dermatosis. Previous studies have shown an association between KIR genes and susceptibility to some clinical variants of Ps. Therefore, we conducted an exhaustive systematic review with meta-analysis to evaluate the relationship between KIR genes and susceptibility to clinical variants of Ps in the overall population and according to ethnicity. METHODS: According to PRISMA guidelines, we performed a systematic review through PubMed and Web of Science to identify relevant available scientific publications about KIR genes and Ps. The quality of the studies was evaluated using the Newcastle-Ottawa scale. Odds ratios (OR) and 95% confidence intervals (95%CI) were estimated using random and fixed effect models for the analyzed genes. Heterogeneity was tested using Cochran's Q-Statistic and I2, and the risk of bias was tested using the Begg test and Egger linear regression. RESULTS: A total of 10 case-control studies were included, comprising a variable number of KIR typified genes and psoriasis vulgaris (PsV) as the main clinical variant studied. In the total pooled results, the KIR2DS1 gene (OR = 1.518, p = .010, 95%CI: 1.105 to 2.086) was related to higher susceptibility to PsV, while the KIR2DS4 (OR = 0.563, p = .005, 95%CI: 0.376 to 0.842) and KIR3DL1 (OR = 0.602, p = .040, 95%CI: 0.370 to 0.977) genes were related to protection against PsV. CONCLUSION: This meta-analysis demonstrates that subjects that carry the KIR2DS1 gene could have a potential risk factor for the development of PsV. Conversely, KIR2DS4 and 3DL1 genes appear to confer protection against PsV.

BACKGROUND AND AIMS: Many endogenous and exogenous risk factors are associated with multiple sclerosis (MS), but recent studies suggest that microbiome-derived ligands, play a role in the disease process. The goal of this study was to characterize the cellular response elicited in human microglia upon treatment with IFN-ß and Fingolimod, two first line medications for the management of MS, and determine whether these treatments affect the response of microglial cells to an MS-associated bacterial ligand, Lipid 654. MATERIALS AND METHODS: HMC3 human microglial cells were treated with IFN-ß or Fingolimod. Cytokine secretion was evaluated using a multiplex system, and microglia polarization was assessed by flow cytometry. RESULTS: We observed that treatment with IFN-ß or Fingolimod induced differential secretion of various pro-inflammatory cytokines. Upon cell stimulation with Lipid 654, we observed that IFN-ß and Fingolimod decreased the secretion of M1-associated cytokines. Using flow cytometry, we observed that the decrease in inflammatory cytokine secretion was likely due to a containment of M1 phenotype of microglia after stimulation with Lipid 654. CONCLUSIONS: Our findings provide new clues of still unknown mechanisms of action of IFN-ß and Fingolimod in human microglia, which will prompt new avenues of research on the use of these therapies in the regulation of the inflammatory response in MS.

The aim of this study was to determine the potential fertilizing of spermatozoa from the epididymal tail in different periods of time post-orchiectomy (P-OQ). Therefore, the study was approached in two stages. In the first stage, the orchiectomy was performed in 30 adult pigs. The testicles were stored at 5.00 ˚C in physiological saline solution for 5, 24, 48, 72, 96 and 120 hr. The spermatozoa were obtained by backflushing the vas deferens. The spermogram and fluorometric study were performed for each sample to evaluate the exposure of phosphatidyl-serine (PS) and acrosome reaction (AR). The second stage included the fertilization test, 16 prepubertal sows were selected, after synchronizing the oestrous cycle and the post-cervical artificial insemination was performed with the refrigerated sperm samples from each P-OQ time. The percentage of live sperm remained without significant changes until 96 hr P-OQ. An increase in the percentage of spermatozoa that showed a PS exposure was observed. The premature AR was evident after 72 hr. Considering that the artificial insemination was performed ensuring a minimum number of live sperms, no significant differences were observed in the number of embryos and corpora lutea. The results indicated that pig sperm collected from the epididymal tail P-OQ and stored for 5 and up to 72 hr at 5.00 ˚C had viable characteristics and maintained their fertilization ability. However, there was an increase in the loss of phospholipid asymmetry of the plasma membrane as time increased (72 and 96 hr), therefore, sperm viability was decreased.

With the appearance of the SARS-CoV-2 virus in December 2019, all countries in the world have implemented different strategies to prevent its spread and to intensively search for effective treatments. Initially, severe cases of the disease were considered in adult patients; however, cases of older school-age children and adolescents who presented fever, hypotension, severe abdominal pain and cardiac dysfunction, positive for SARS-CoV-2 infection, have been reported, with increased pro-inflammatory cytokines and tissue damage, condition denominated multisystemic inflammatory syndrome (MIS-C); The emerging data from patients with MIS-C have suggested unique characteristics in the immunological response and also clinical similarities with other inflammatory syndromes, which can support as a reference in the search for molecular mechanisms involved in MIS-C. We here in propose that oxidative stress (OE) may play a very important role in the pathophysiology of MIS-C, such as occurs in Kawasaki disease (KD), severe COVID-19 in adults and other processes with characteristics of vascular damage similar to MIS- C, for which we review the available information that can be correlated with possible redox mechanisms.

BACKGROUND: Psoriasis (Ps) is a chronic dermatosis characterized by erythematous-squamous plaques derived from an inflammatory response. The effect of polymorphisms in the genes that encode the members of the IL-17 family and their receptors has been studied to find an association with the susceptibility to Ps. However, the findings have not been conclusive. OBJECTIVES: To describe the association between IL-17A, IL-17F and IL-17RA gene polymorphisms and susceptibility to Ps. METHOD: A systematic review was conducted using the PubMed and Scopus databases to identify studies that evaluated the association between IL-17A, IL-17F, and IL-17RA gene polymorphisms and Ps susceptibility. This meta-analysis included reports published until June 2021. Heterogeneity was assessed using Cochran's Q-statistic test and I2 statistics. The associations between polymorphisms and Ps susceptibility were determined by pooled OR with a 95% CI. RESULTS: Fifteen studies were included. The frequency of the T allele of the IL-17F rs763780 polymorphism was significantly lower in patients with vulgar Ps (OR = 0.732, p = 0.026). The TT genotype of the IL-17F rs763780 polymorphism was significantly associated with a decreased frequency in individuals with Ps and psoriatic arthritis (PsA) (TT:TC + CC OR = 0.664, p = 0.046). Regarding IL-17RA polymorphisms, the AG genotype of the rs4819554 polymorphism showed a near-significant decrease in psoriasis risk compared to the GG genotype (AG:GG OR = 0.604, p = 0.050). Other polymorphisms in IL-17A, IL-17F and IL-17RA showed no association with Ps. CONCLUSIONS: The T allele and TT genotype of the IL-17F rs763780 polymorphism may be associated with a decreased risk of psoriasis. Therefore, the implications of this variant on psoriasis pathogenesis and treatment require further investigation.

Harris hawks (Parabuteo unicinctus) are the raptors with more anthropogenic use, which makes it necessary to improve their reproduction either natural or assisted for sustainable use even though they are not endangered. The aim of this study was to determine fresh and thawed P. unicinctus sperm capacitation and acrosome reaction parameters as indicators of sperm viability in ejaculates of birds with injuries. A total of 30 ejaculates were analyzed as fresh and thawed samples. Basic sperm evaluations as well as in vitro acrosomal reaction capability through the presence and distribution of Ca2+ and N-acetylglucosamine and sialic acid using chlortetracycline and agglutinin-fluorescein isothiocyanate Triticum vulgaris were performed, respectively. Tagged samples were observed using epifluorescence microscopy. The results indicated 30.00% decrease in sperm viability following freeze-thaw, while rate of capacitated sperms (10.00%) and the percentage of sperm with acrosomal reaction (40.00%) were increased. The results of the experiment demonstrated that the post-thaw viability of the obtained sperm may be an alternative for use in artificial insemination.

The aim of this study is to design and develop an efficient leaching process based on a fundamental and theoretical thermodynamic analysis and the optimization of the operation parameters via the response surface methodology (RSM). Using this methodology, the design of a leaching process for the recovery of copper, silver, and lead from highly metal-concentrated fractions of e-waste is presented. Thermodynamic predictions were performed through the construction and analysis of Pourbaix diagrams for the specific conditions of the leaching system. From this analysis, it was possible to determine the values of potential (E vs NHE) and pH at which the leaching reactions occur spontaneously. Additionally, RSM was useful to deduce a quadratic semiempirical model that predicts the copper leaching efficiencies as a function of two parameters involved in the leaching procedure, the stirring speed and the solid/liquid ratio, by which the response variable, the leaching efficiency, can be optimized.

RESUMO

A new a species of pinnotherid crab in the eastern Pacific coasts is presented. Twenty males and 21 females of small crabs were collected from burrows, presumably from ghost shrimp (Neotrypaea spp.), in sand-mud substrata from the Santa María-La Reforma coastal lagoon, SE Gulf of California. The new species was assigned to the genus Glassella because its morphological characteristics clearly match the amended diagnosis of the genus Glassella by Palacios Theil and Felder (2020) and Felder Palacios Theil (2020), including the presence of a gonopodal plate (GP) inserted in the internal part of the male pleon, similar to that described for most of the species currently grouped into the genus Glassella. The new species is similar to G. miamiensis (McDermott, 2014) from western Atlantic, but it can be distinguished from this and the rest of the species of Glassella by differences in carapace margins and ridges, male pleon outline, and the shape of the GP.

ABSTRACT Objective. The sex ratio, proportion of ovigerous females, length at sexual maturity and fecundity of Grapsus grapsus crabs in the Lobos, Venados and Pajaros islands (southeastern Gulf of California) were analyzed. Materials and methods. Sampling was conducted monthly between March 2011 and February 2012, during the night at low tide. Thirty crabs were collected in a quadrant (25 m2) at each sampling site on each island. cw (mm) and w (g) were determined. The sex ratio and size at sexual maturity (cw50%) were estimated, and for ovigerous females, embryonic stages and fecundity (gravimetric method) were determined. Results. The sex ratio (M:F) was 1:1.3. The average size at sexual maturity (cw50%) was 34.9 mm. The majority of females were ovigerous (71.3%), and 48% of the embryos of ovigerous females were at the red-orange phase. Egg diameter ranged from 1.1 to 5 µm, with an average of 2.05 µm. The mean fecundity was 24339.3 eggs. The maximum and minimum weight of ovigerous females was 69.9 and 15.2 g. Conclusions. The studied characteristics of sex ratio, proportion of ovigerous females, length at sexual maturity and fecundity of G. grapsus, indicate the effective administration and management of this resource in this area.

Se analizó la proporción de sexos, hembras ovígeras, talla de primera madurez sexual y fecundidad del cangrejo roca Grapsus grapsus en islas Lobos, Venados y Pájaros (sureste del Golfo de California). Material y métodos. Los muestreos fueron mensuales entre marzo 2011 y febrero 2012, las colectas fueron nocturnas durante la bajamar, se obtuvieron en un cuadrante (25 m2) por isla 30 organismos al azar, se les determinó el AN (mm) y PT (g). Se estimó la proporción de sexos y talla de primera madurez sexual (AN50%), se analizaron en hembras grávidas, las fases embrionarias y la fecundidad (método gravimétrico). Resultados. La proporción de M:H fue 1:1.3. La talla media de primera madurez fue AN50% 34.9 mm. Es evidente la presencia de hembras ovígeras (71.3%) y todas las fases embrionarias, la fase rojo-naranja fue la mayor representada en 48%. La variación del diámetro del huevo fue 1.1 a 5 µm y el promedio de 3.05 µm. La fecundidad media fue 24339.3 cigotos. El máximo y mínimo peso de hembras ovígeras fue 69.9 y 15.2 g, respectivamente. Conclusiones. Con base a las características biológicas del recurso tales como la proporción de sexos, hembras ovígeras, talla de primera madurez sexual y fecundidad en la población de G. grapsus, representa un efecto favorable en su posterior administración y manejo de este recurso en esta zona.

Background: Psoriatic Arthritis (PsA) is a seronegative spondyloarthropathy frequently associated with psoriasis. Studies have shown different members of the KIR (Killer Immunoglobulin-like Receptor) gene family may act as potential susceptibility factors; however, data have been inconsistent or with a reduced sample size. Therefore, the objective of this investigation was to determine associations between KIR genes and PsA susceptibility a meta-analysis approach. Methods: We performed a systemic search on PubMed, Scopus, and Web of Science to identify association studies linking KIR genes with PsA susceptibility. The search cut-off was May 2019. Odds Ratio (OR), 95% Confidence Intervals (95% CI), and forest plots were obtained for each KIR gene. Publication bias was evaluated by Begg and Egger linear regression tests. Results: Five articles were included in this meta-analysis. The KIR2DL2, 2DS1, 2DS2, and 2DS3 genes were positively associated with susceptibility to PsA (OR = 1.269, p = .003; OR = 1.392, p < .001; OR = 1.279, p = .002; and OR = 1.230, p = .038, respectively). In Caucasians, positive association with susceptibility to PsA were maintained by KIR2DL2, 2DS1, and 2DS2 genes (OR = 1.257, p = .005; OR = 1.535, p = .003; and OR = 1.267, p = .004, respectively). Conclusion: These associations suggest that KIR2DL2, 2DS1, 2DS2, and 2DS3 genes are susceptibility factors for PsA.

Organic waste from greens of tomato plants, gardening substrate, rice husks and shrimp-derived chitin were pyrolyzed at 400 °C and 500 °C for 3 h, with the aim to elucidate the feasibility of using such products as replacement of peat in soilless gardening substrates. Characterization of the carbonized organic matter (COM) and the gardening substrate indicated that neither the peat nor the COMs provided the recommended levels of nutrients for the cultivation of tomato plants, although improvements could be obtained using COM/substrate mixtures. The toxicity thresholds for Zn were exceeded significantly by the COMs of the tomato greens and high boron levels were found for all the COMs except for those derived from chitin. In a 40-days pot experiment, germination and development of tomato seeds and plants (Solanum lycopersicum L.) were tested on COM/peat mixtures at 30%, 60% and 100% COM substitution rate. The lack of seed germination on the mixtures with COM from tomato greens is best explained with the high salinity of the COM. Best plant growth was obtained with COM from chitin at 60%, most likely because its high N content satisfied best the N-needs of the growing tomato plants without increasing the pH of the growing media. Moreover, an increase of water retention was evidenced for COM/substrate mixtures. Although the use of COM from chitin and rice husks showed promising results, the proposed recycling of organic waste from agriculture or fishery as soilless gardening substrate requires the development of formulations of COM/peat/and added nutrients with ready-to-use characteristics to increase its feasibility.
