RESUMO

The influence of the chemical nature of the cluster constituents on the desorption/ionization process was investigated for desorption/ionization induced by neutral SO2 clusters (DINeC). The polar clusters act as a transient matrix in which the desorbed analyte molecules are dissolved during the desorption process. For drop-cast samples, the desorption/ionization efficiency was found to be largely independent of the pH value of the initial solution the samples were prepared from; positive ions were almost always dominant and no multiply charged negative ions were observed. The results were traced back to the interaction of SO2 with water present in the samples. Both H/D exchange experiments and surface charge measurements showed that SO2 from the cluster beam interacts with water on and in the sample forming sulfurous acid. The latter then acts as an efficient proton supply leading to an enhanced ionization efficiency. The results demonstrate the possibility to control the ionization efficiency when using reactive cluster constituents in desorption-based ionization methods such as DINeC and cluster-based secondary ion mass spectrometry.

Integrated pest management (IPM) is a method of reducing economic, human health, and environmental risks from pests and pest management strategies. There are questions about the long-term success of IPM programs in relation to continued use of pesticides in agriculture. Total pounds of pesticides applied is a mis-measure of the impact of IPM in agriculture. A more complete measurement of the long-term impact of IPM includes consideration of changes in agricultural production practices and productivity, toxicity of the pesticides used, risks from human exposure to pesticides, and environmental sampling for pesticides in air and water resources. In recent decades, agricultural IPM programs have evolved to address invasive pests, shifts in endemic pest pressures, reductions in pest damage tolerance in markets, and increases in crop yields. Additionally, pesticide use data from Arizona and California revealed reduced use of pesticides in some toxicity categories but increased use of pesticides in a couple of categories. Data from federal and California programs that monitored pesticide residue on food have documented low pesticide risk to consumers. Environmental monitoring programs documented decreased pesticide levels in surface water resources in agricultural watersheds in the western United States and low levels of pesticides in air resources in agricultural areas in California. The focus of IPM assessment should be on reducing economic, human health, and environmental risks, not on pounds of pesticides applied. More broadly, IPM programs have evolved to address changes in pests and agricultural production systems while continuing to reduce human health and environmental risk from pesticides.

Degeneration of the intervertebral disc (IVD) is a progressive and chronic process, and the high incidence of discogenic disorders calls for new therapeutic approaches, such as cell-based therapies using three dimensional cultures and mesenchymal stem cells (MSC), which can differentiate to chondrogenic- and IVD-lineages. Here, we investigated the growth and differentiation of human MSC culture on biodegradable collagen scaffolds in order to obtain an injectable suspension. Commercially available wound dressings were downsized to dimensions between 100 and 1500 µm and seeded with freshly isolated or early passages MSC. Proliferation rate and chondrogenic differentiation potential was tested at oxygenation levels of 2%, 5%, 10% and 21% in static and dynamic cultures. Evaluation methods included cell viability test, disc marker genes expression (aggrecan, collagen type I and type II), histological detection of proteoglycans and immunohistochemical analysis. On microcarriers, freshly isolated MSC had lower proliferation rate and chondrogenic differentiation potential compared with early passages MSC. Proliferation of MSC was significantly increased 1.7-fold at 5% oxygen level and in combination with dynamic culture was further increased to 2.3-fold, with respect to normoxia. Chondrogenesis was positively affected by 2% and 5% hypoxia, as shown by increased transcription levels and protein expression of collagen type II and proteoglycan accumulation in static cultures, while it was inhibited in dynamic cultures. Collagen type I and aggrecan expression were not affected by hypoxia. In conclusion, collagen based microcarriers are a suitable support for in vitro MSC growth and chondrogenesis especially when cultured at 5% oxygen level.

BACKGROUND: Lung-protective mechanical ventilation during general surgery including the application of PEEP can reduce postoperative pulmonary complications. In a prospective clinical observation study, we evaluated volume-dependent respiratory system compliance in adult patients undergoing ear-nose-throat surgery with ventilation settings chosen empirically by the attending anaesthetist. METHODS: In 40 patients, we measured the respiratory variables during intraoperative mechanical ventilation. All measurements were subdivided into 5 min intervals. Dynamic compliance (CRS) and the intratidal volume-dependent CRS curve was calculated for each interval and classified into one of the six specific compliance profiles indicating intratidal recruitment/derecruitment, overdistension or all. We retrospectively compared the occurrences of the respective compliance profiles at PEEP levels of 5 cm H2O and at higher levels. RESULTS: The attending anaesthetists set the PEEP level initially to 5 cm H2O in 29 patients (83%), to 7 cm H2O in 5 patients (14%), and to 8 cm H2O in 2 patients (6%). Across all measurements the mean CRS was 61 (11) ml cm H2O(-1) (40-86 ml cm H2O(-1)) and decreased continuously during the procedure. At PEEP of 5 cm H2O the compliance profile indicating strong intratidal recruitment/derecruitment occurred more often (18.6%) compared with higher PEEP levels (5.5%, P<0.01). Overdistension was practically never observed. CONCLUSIONS: In most patients, a PEEP of 5 cm H2O during intraoperative mechanical ventilation is too low to prevent intratidal recruitment/derecruitment. The analysis of the intratidal compliance profile provides the rationale to individually titrate a PEEP level that stabilizes the alveolar recruitment status of the lung during intraoperative mechanical ventilation. UNLABELLED: Contemporary femur fracture rates were examined in northern California women and compared by race/ethnicity. During 2006-2012, hip fracture rates declined, but diaphyseal fracture rates increased, especially in Asians. Women with diaphyseal fracture were younger and more likely to be bisphosphonate-treated. These disparities in femur fracture should be further examined. INTRODUCTION: The epidemiology of diaphyseal femur fracture differs from proximal femur (hip) fracture, although few studies have examined demographic variations in the current era. This study examines contemporary differences in low-energy femur fracture by race/ethnicity in a large, diverse integrated health-care delivery system. METHODS: The incidence of hip and diaphyseal fracture in northern California women aged ≥50 years old during 2006-2012 was examined. Hip (femoral neck and pertrochanteric) fractures were classified by hospital diagnosis codes, while diaphyseal (subtrochanteric and femoral shaft) fractures were further adjudicated based on radiologic findings. Demographic and clinical data were obtained from health plan databases. Fracture incidence was examined over time and by race/ethnicity. RESULTS: There were 10,648 (97.3 %) hip and 300 (2.7 %) diaphyseal fractures among 10,493 women. The age-adjusted incidence of hip fracture fell from 281 to 240 per 100,000 women and was highest for white women. However, diaphyseal fracture rates increased over time, with a significant upward trend in Asians (9 to 27 per 100,000) who also had the highest rate of diaphyseal fracture. Women with diaphyseal fracture were younger than women with hip fracture, more likely to be of Asian race and to have received bisphosphonate drugs. Women with longer bisphosphonate treatment duration were also more likely to have a diaphyseal fracture, especially younger Asian women. CONCLUSION: During 2006 to 2012, hip fracture rates declined, but diaphyseal fracture rates increased, particularly among Asian women. The association of diaphyseal fracture and bisphosphonate therapy should be further investigated with examination of fracture pattern.

Degeneration of intervertebral discs (IVDs) is associated with back pain and elevated levels of inflammatory cells. It has been hypothesised that discogenic pain is a direct result of vascular and neural ingrowth along annulus fissures, which may expose the avascular nucleus pulposus (NP) to the systemic circulation and induce an autoimmune reaction. In this study, we confirmed our previous observation of antibodies in human degenerated and post-traumatic IVDs cultured in vitro. We hypothesised that the presence of antibodies was due to an autoimmune reaction against specific proteins of the disc. Furthermore we identified antigens which possibly trigger an autoimmune response in degenerative disc diseases. We demonstrated that degenerated and post-traumatic IVDs contain IgG antibodies against typical extracellular proteins of the disc, particularly proteins of the NP. We identified IgGs against collagen type II and aggrecan, confirming an autoimmune reaction against the normally immune privileged NP. We also found specific IgGs against collagens types I and V, but not against collagen type III. In conclusion, this study confirmed the association between disc degeneration and autoimmunity, and may open the avenue for future studies on developing prognostic, diagnostic and therapy-monitoring markers for degenerative disc diseases.

Engineered macroscopic quantum systems based on superconducting electronic circuits are attractive for experimentally exploring diverse questions in quantum information science. At the current state of the art, quantum bits (qubits) are fabricated, initialized, controlled, read out and coupled to each other in simple circuits. This enables the realization of basic logic gates, the creation of complex entangled states and the demonstration of algorithms or error correction. Using different variants of low-noise parametric amplifiers, dispersive quantum non-demolition single-shot readout of single-qubit states with high fidelity has enabled continuous and discrete feedback control of single qubits. Here we realize full deterministic quantum teleportation with feed-forward in a chip-based superconducting circuit architecture. We use a set of two parametric amplifiers for both joint two-qubit and individual qubit single-shot readout, combined with flexible real-time digital electronics. Our device uses a crossed quantum bus technology that allows us to create complex networks with arbitrary connecting topology in a planar architecture. The deterministic teleportation process succeeds with order unit probability for any input state, as we prepare maximally entangled two-qubit states as a resource and distinguish all Bell states in a single two-qubit measurement with high efficiency and high fidelity. We teleport quantum states between two macroscopic systems separated by 6 mm at a rate of 10(4) s(-1), exceeding other reported implementations. The low transmission loss of superconducting waveguides is likely to enable the range of this and other schemes to be extended to significantly larger distances, enabling tests of non-locality and the realization of elements for quantum communication at microwave frequencies. The demonstrated feed-forward may also find application in error correction schemes.

RATIONALE: Desorption-and-ionization induced by neutral cluster impact is a soft and matrix-free method, which leads to the formation of free ions of oligopeptides and smaller proteins without fragmentation. As a prerequisite for its successful application in bioanalytics, especially with respect to sensitivity, the ionization efficiency, i.e., the ion-to-neutral ratio of the desorbing molecules, was determined. METHODS: Neutral SO2 clusters of 10(3) to 10(4) molecules in size were seeded in a pulsed He beam and used to desorb and ionize oligopeptides by means of cluster surface impact. The samples were prepared by drop casting a well-defined amount of substance on the substrate surface; the desorbing ions were identified by means of time-of-flight mass spectrometry. Furthermore, the ion current leaving the surface was determined for positive ions, which predominate in the investigated oligopeptides. RESULTS: For angiotensin II, bradykinin (1-7), and adrenocorticotropic hormone (34-39), the number of ions desorbed from the respective samples was compared with the amount of substance applied on the substrate. Assuming that all biomolecules were desorbed during the experiment, the ion-to-neutral ratio or ionization efficiency η was determined. For the tested molecules, values of η between 0.5% and 3% were observed; the substrate material and the total amount of substance applied were shown to have a minor effect on the results. CONCLUSIONS: The ion-to-neutral ratio in desorption/ionization of oligopeptides induced by neutral cluster impact was determined to be of the order of 10(-3) to 10(-2). The soft and matrix-free nature of the method in combination with this value of η might be interesting for applications in bioanalytics.

Prostate cancer is the most common malignant tumor in men. Radical prostatectomy, the most common surgical therapy, is typically accompanied by erectile dysfunction and incontinence due to severing of the axons of the plexus prostaticus. To date, no reconstructive therapy is available as the delicate network of severed nerve fibers preclude the transplantation of autologous nerves or synthetic tube implants. Here, we present an injectable hydrogel as a regenerative matrix that polymerizes in situ and thus, adapts to any given tissue topography. The two-component hydrogel was synthesized from a hydrolyzed collagen fraction and stabilized by enzymatic crosslinking with transglutaminase. Physical analysis employing osmolarity measurements and cryosectioning revealed an isotonic, microstructured network that polymerized within 2min and displayed pronounced adhesion to abdominal tissue. Cell culturing demonstrated the biocompatibility of the gel and a general permissiveness for various neuronal and non-neuronal cell types. No effect on cell adhesion, survival and proliferation of cells was observed. A chemotherapeutic drug was integrated into the hydrogel to reduce the risk of fibrosis and tumor relapse. Significantly, when the hydrogel was employed as a drug release depot in vitro, aversive fibroblast- and prostate carcinoma cell growth was inhibited, while axonal outgrowth from peripheral nervous system explants remained completely unaffected. Taken together, these results suggest that the gel's adequate viscoelastic properties and porous microstructure, combined with its tissue adhesion and neuritotrophic characteristics in the presence of a cell type-specific cytostatic, may constitute an appropriate hydrogel implant applicable to patients suffering from prostatectomy associated side effects.

RESUMO

Experimental implementations of quantum information processing have now reached a level of sophistication where quantum process tomography is impractical. The number of experimental settings as well as the computational cost of the data postprocessing now translates to days of effort to characterize even experiments with as few as 8 qubits. Recently a more practical approach to determine the fidelity of an experimental quantum process has been proposed, where the experimental data are compared directly with an ideal process using Monte Carlo sampling. Here, we present an experimental implementation of this scheme in a circuit quantum electrodynamics setup to determine the fidelity of 2-qubit gates, such as the CPHASE and the CNOT gate, and 3-qubit gates, such as the Toffoli gate and two sequential CPHASE gates.

Teleportation of a quantum state may be used for distributing entanglement between distant qubits in quantum communication and for quantum computation. Here we demonstrate the implementation of a teleportation protocol, up to the single-shot measurement step, with superconducting qubits coupled to a microwave resonator. Using full quantum state tomography and evaluating an entanglement witness, we show that the protocol generates a genuine tripartite entangled state of all three qubits. Calculating the projection of the measured density matrix onto the basis states of two qubits allows us to reconstruct the teleported state. Repeating this procedure for a complete set of input states we find an average output state fidelity of 86%.

OBJECTIVE: The objective of this study was to analyze the effect of a fructose-restricted diet in otherwise healthy children with abdominal pain and pathologic fructose hydrogen breath test. SUBJECTS AND METHODS: 75 children (aging 3-14 years) with recurrent abdominal pain without gastrointestinal disease and fructose malabsorption followed a fructose restricted diet for 4 weeks. RESULTS: A median decline of weekly pain frequency from 4 (mean 3.64+1.6) before diet to 1 (mean 1.46+1.4; p<0.001) under fructose restriction was documented. The intensity of pain decreased from median 6 (mean 5.83+2.0) before intervention to median 3 (mean 3.4+2.5; p<0.001) with diet. Several additional life quality-influencing parameters such as daily stool frequency, nausea, problems to fall asleep, missed school days also improved significantly. CONCLUSIONS: Our study provides evidence that dietary fructose restriction represents a useful approach to address recurrent abdominal symptoms in children with fructose malabsorption.

The Toffoli gate is a three-quantum-bit (three-qubit) operation that inverts the state of a target qubit conditioned on the state of two control qubits. It makes universal reversible classical computation possible and, together with a Hadamard gate, forms a universal set of gates in quantum computation. It is also a key element in quantum error correction schemes. The Toffoli gate has been implemented in nuclear magnetic resonance, linear optics and ion trap systems. Experiments with superconducting qubits have also shown significant progress recently: two-qubit algorithms and two-qubit process tomography have been implemented, three-qubit entangled states have been prepared, first steps towards quantum teleportation have been taken and work on quantum computing architectures has been done. Implementation of the Toffoli gate with only single- and two-qubit gates requires six controlled-NOT gates and ten single-qubit operations, and has not been realized in any system owing to current limits on coherence. Here we implement a Toffoli gate with three superconducting transmon qubits coupled to a microwave resonator. By exploiting the third energy level of the transmon qubits, we have significantly reduced the number of elementary gates needed for the implementation of the Toffoli gate, relative to that required in theoretical proposals using only two-level systems. Using full process tomography and Monte Carlo process certification, we completely characterize the Toffoli gate acting on three independent qubits, measuring a fidelity of 68.5 ± 0.5 per cent. A similar approach to realizing characteristic features of a Toffoli-class gate has been demonstrated with two qubits and a resonator and achieved a limited characterization considering only the phase fidelity. Our results reinforce the potential of macroscopic superconducting qubits for the implementation of complex quantum operations with the possibility of quantum error correction.

Continuous variable entanglement between two modes of a radiation field is usually studied at optical frequencies. Here we demonstrate experiments that show the entanglement between microwave photons of different energy in a broadband squeezed beam. We use a Josephson parametric amplifier to generate the two-mode correlated state and detect all four quadrature components simultaneously in a two-channel heterodyne setup using amplitude detectors. Analyzing two-dimensional phase space histograms for all possible pairs of quadratures allows us to determine the full covariance matrix, which is in good agreement with the one expected for a two-mode squeezed state.

Creating a train of single photons and monitoring its propagation and interaction is challenging in most physical systems, as photons generally interact very weakly with other systems. However, when confining microwave frequency photons in a transmission line resonator, effective photon-photon interactions can be mediated by qubits embedded in the resonator. Here, we observe the phenomenon of photon blockade through second-order correlation function measurements. The experiments clearly demonstrate antibunching in a continuously pumped source of single microwave photons measured by using microwave beam splitters, linear amplifiers, and quadrature amplitude detectors. We also investigate resonance fluorescence and Rayleigh scattering in Mollow-triplet-like spectra.

There is evidence that mesenchymal stem cells (MSCs) can differentiate towards an intervertebral disc (IVD)-like phenotype. We compared the standard chondrogenic protocol using transforming growth factor beta-1 (TGFß) to the effects of hypoxia, growth and differentiation factor-5 (GDF5), and coculture with bovine nucleus pulposus cells (bNPC). The efficacy of molecules recently discovered as possible nucleus pulposus (NP) markers to differentiate between chondrogenic and IVD-like differentiation was evaluated. MSCs were isolated from human bone marrow and encapsulated in alginate beads. Beads were cultured in DMEM (control) supplemented with TGFß or GDF5 or under indirect coculture with bNPC. All groups were incubated at low (2 %) or normal (20 %) oxygen tension for 28 days. Hypoxia increased aggrecan and collagen II gene expression in all groups. The hypoxic GDF5 and TGFß groups demonstrated most increased aggrecan and collagen II mRNA levels and glycosaminoglycan accumulation. Collagen I and X were most up-regulated in the TGFß groups. From the NP markers, cytokeratin-19 was expressed to highest extent in the hypoxic GDF5 groups; lowest expression was observed in the TGFß group. Levels of forkhead box F1 were down-regulated by TGFß and up-regulated by coculture with bNPC. Carbonic anhydrase 12 was also down-regulated in the TGFß group and showed highest expression in the GDF5 group cocultured with bNPC under hypoxia. Trends in gene expression regulation were confirmed on the protein level using immunohistochemistry. We conclude that hypoxia and GDF5 may be suitable for directing MSCs towards the IVD-like phenotype.

Limited information exists on the insecticide susceptibility of redbanded stink bug, Piezodorus guildinii (Westwood) (Heteroptera: Pentatomidae), despite its impact on soybean, Glycine max (L.) Merr., production in Brazil and the United States. Therefore, this study set out to 1) determine baseline levels of susceptibility to currently recommended pesticides using topical and vial bioassays, 2) determine the levels of esterase activity in populations in the United States and Brazil, and 3) compare control among products in field trials. In topical bioassays conducted in the United States using technical grade materials, the LC50 values of lambda-cyhalothrin, acephate, and methamidophos were 4-25, 141-295, and 40-151 ng per insect, respectively. The LC50 values of imidacloprid and thiamethoxam were 11 and 27 ng per insect, respectively. In vial bioassays conducted in the United States using technical grade materials, the LC50 values of cypermethrin, acephate, and methamidophos were 0.4-0.9, 3.8, and 1.6 microg per vial, respectively. In topical bioassays conducted in Brazil by using commercially formulated products, the LC50 values of acephate, methamidophos, endosulfan, and imidacloprid were 0.90-1.9, 0.4-0.6, 1.5-6.6, and 0.2-0.3 microg per insect, respectively. In vial bioassays conducted in Brazil using commercially formulated products, the LC50 values of endosulfan, methamidophos, and lambda-cyhalothrin were 4-32 and 2-24 microg/cm2 for thiamethoxam and imidacloprid. Esterase activity in Louisiana (United States) populations ranged from 251 to 658 nmol alpha-naphthol formed/min/mg protein. Esterase activity levels in Londrina (Brazil) populations averaged 163 nmol/min/mg. In field tests, P. guildinii in Louisiana were controlled by organophosphates thiamethoxam and imidacloprid and in Brazil, with combinations of neonicotinoids and pyrethroids.

RESUMO

We present the realization of a cavity quantum electrodynamics setup in which photons of strongly different lifetimes are engineered in different harmonic modes of the same cavity. We achieve this in a superconducting transmission line resonator with superconducting qubits coupled to the different modes. One cavity mode is strongly coupled to a detection line for qubit state readout, while a second long lifetime mode is used for photon storage and coherent quantum operations. We demonstrate sideband-based measurement of photon coherence, generation of n photon Fock states and the scaling of the sideband Rabi frequency with square root of n using a scheme that may be extended to realize sideband-based two-qubit logic gates.

The aim of this study was to demonstrate a pathologic complete response (pCR) rate of at least 10% with an acceptable toxicity achieved by preoperative chemoradiotherapy with 5-fluorouracil (5-FU)/leucovorin in patients with locally advanced rectal cancer. Patients were treated by radiotherapy targeting 50 Gy and 5-FU/leucovorin intravenously during the 1st, 4th and 7th week after start of radiotherapy followed by surgery and adjuvant chemotherapy. In 71 evaluable patients, the pCR rate was 14.1% (95% CI, 6.0-22.2); the local relapse rate, 6.1%; the 5-year disease-free survival, 54% and the overall 5-year survival, 68%. The most severe adverse events were neutropenia (17%), diarrhoea (17%), infection (8%) and fatal cardiovascular function (1%). This therapy yielded a high rate of pCR, a low rate of local relapse and a long disease-free and overall survival. To increase its feasibility, radiation dose reduction to 45 Gy and administration of only two preoperative cycles of chemotherapy is recommended.
