RESUMO

Waste activated sludge (WAS) can become an important source of phosphorus (P). P speciation was examined under anaerobic conditions, with different pH (4, 6 and 8) and temperatures (10, 20 and 35 °C). Aqueous P was measured and an extraction protocol was used to find three solid phosphorus fractions. A pH of 4 and a temperature of 35 °C gave a maximum of 51% of total P solubilized in 22 days with 50% of total P solubilized in 7 days. Batch tests indicate that little pH depression is needed to release non-apatite inorganic P (including microbial polyphosphate), while a pH of 4 rather than 6 will release more apatite inorganic P, and that organic P is relatively more difficult to release from WAS. Fractionation analysis of P in WAS can aid in design of more efficient methods for P recovery from WAS.

Feruloyl, chlorogenate esterases, and tannases are enzymes useful in phenolic modifications of pharmaceutical relevance as protectors against several degenerative human diseases. Therefore, there is a growing interest in discovering new sources of these enzymes. However, traditional methods for their activity measurements are time-consuming and poorly adapted for high-throughput screening. In this study, a successful new microplate high-throughput screening method for the simultaneous quantification of all mentioned activities is demonstrated. This method allows the detection of activities as low as 1.7 mU ml(-1). Furthermore, reaction rates increased proportionally with the amount of enzyme added, and no interferences with the other commercial hydrolases tested were found. The utility of the method was demonstrated after simultaneously screening feruloyl, chlorogenate esterase, and tannase activities in solid state fermentation extracts obtained during the kinetics of production of 20 fungal strains. Among these, seven strains were positive for at least one of the esterase activities tested. This result shows the potential for the rapid routine screening assays for multiple samples of moderate low to high enzymatic levels.

The efficiency of a dark Fenton system (H2O2/Fe2+) to remove chlorothalonil from water, alone and in the presence of the organophosphorous pesticides methyl parathion and methamidophos, was evaluated. Here we show that the process is rapid and efficient. Nearly complete degradation of chlorothalonil and methyl parathion (98%) was observed in less than 10 min whereas methamidophos showed similar levels of efficiency only after 40 to 90 min. Formulating agents did not appear to impair the degradation of active ingredients. Interactions during the degradation of mixtures of the three pesticides only appeared to affect methamidophos degradation.

The ability of an anaerobic hybrid reactor, treating coffee wastewater, to achieve a quick start-up was tested at pilot scale. The unacclimatized seed sludge used showed a low specific methanogenic activity of 26.47 g CH4 as chemical oxygen demand (COD)/kg volatile suspended solids (VSS) x day. This strongly limited the reactor performance. After a few days of operation, a COD removal of 77.2% was obtained at an organic loading rate (OLR) of 1.89 kg COD/m3 x day and a hydraulic retention time (HRT) of 22 h. However, suddenly increasing OLR above 2.4 kg COD/m3 x day resulted in a deterioration in treatment efficiency. The reactor recovered from shock loads after shutdowns of 1 week. The hybrid design of the anaerobic reactor prevented the biomass from washing-out but gas clogging in the packing material was also observed. Wide variations in wastewater strength and flow rates prevented stable reactor operation in the short period of the study.
