RESUMO

The aim of this research was to investigate the variation regarding the chemical composition and biological activities of needles essential oils (EOs) of P. halepensis. Chemical profiles demonstrated a significant (P < 0.05) variability among the different EOs. The main identified compounds were caryophyllene (48.77 ± 2.26), phenyl isovalerate (22.22 ± 2.26), ß-myrcene (15.55 ± 5.65) and α-pinene (14.52 ± 2.26). Further, it was shown that EO from Tabouba (Tab) displayed the highest DPPH scavenging (IC50 = 73.03 mg/mL), anti-inflammatory (IC50 = 23.29 µg/mL) and α-glucosidase inhibition activities (IC50 = 254.45 µg/mL). While Elmahres (Elm) exhibited the most potent ABTS radical's inhibition (IC50 = 197.87 mg/mL). For the cytotoxic capacities, Kettana (Ket) was the most efficient against breast cancer MCF-7 cell line with IC50 value better than doxorubicin used as positive control. Obtained results suggest that EO of P. halepensis could be used as a source of bioactive compounds.

Artemisia herba-alba (Aha) is an aromatic and medicinal plant rich in health-promoting flavonoids compounds that play an important role as antioxidant. For better utilization of this resource, the ultrasound-assisted extraction (UAE) of flavonoids and antiradical activities of obtained extracts from Aha using response surface methodology was assessed. A four-factor five-level central composite rotatable design was employed to discuss these extracting parameters: ethanol concentration (X1), temperature (X2), extraction time (X3) and solvent to sample ratio (X4). The optimum extraction conditions for simultaneous maximization of total flavonoids (TF) and scavenging activity were solvent concentration of 50%, temperature of 55 °C, extraction time of 50 min and solvent to sample ratio of 90 mL/g. Under these conditions, the experimental yield of TF and DPPH-radical scavenging activity IC50% were respectively 102.49 mg CE/g DM and 7 µg/mL. Additionally, good and moderate linear correlation was observed between antiradical activity and TF content (R2 = 0.709) which indicated that these compounds are responsible for antioxidant activity of Aha. When compared to the most conventional extraction, antioxidant activity and amounts of major flavonoids are enhanced when using UAE with reduced extraction time.

This study was conducted to investigate potentially protective and curative effects of Curcuma longa root (turmeric) powder on CCl4-induced hepatotoxicity in rats. Turmeric was administered before (preventive effect) or after (curative effect) treatment with CCl4. Total phenolic and flavonoid levels were 26.35 mg GAE/g and 12.35 mg CE/g, respectively. Using HPLC analysis, turmeric powder was rich in curcumin (62.97%), demethoxycurcumin (20.86%) and bisdemethoxycurcumin (16.17%). Curcuma longa powder showed important in vitro antioxidant activities. Results showed that the activities of aspartate aminotransaminase and alanine aminotransaminase, and the levels of bilirubin and serum lipids were increased in CCl4-treated animals. However, total protein and albumin levels and antioxidant enzyme activities were decreased. Turmeric administration, before or after CCl4 treatment, significantly decreased the activities of marker enzymes and lipid levels in serum. Moreover, total protein and albumin contents were restored to nearly normal levels after turmeric administration accompanied with increase of antioxidant enzymes activities.

CONTEXT: Frankenia pulverulenta L. (Frankeniaceae) is a medicinal species with carminative, analgesic and antiviral properties. However, phytochemical investigations, antioxidant and neuroprotective capacities of this plant remain unclear. OBJECTIVE: This work assesses the phenolic composition of F. pulverulenta shoot and root and evaluates their antioxidant and neuroprotective capacities. MATERIALS AND METHODS: Successive fractionation of F. pulverulenta shoot and root using 6 solvents were used. Antioxidant capacity of these fractions was assessed through four in vitro tests (DPPH, ABTS, Fe-chelating activity and ORAC). Phenolic identification, purification as well as neuroprotective activity of ethyl acetate (EtOAc) fraction and purified molecules were assessed. RESULTS: Among the tested fractions, EtOAc shoot and root fractions possessed considerable phenolic contents (383 and 374 mg GAE/g E, respectively) because of their important ORAC (821 and 1054 mg of TE/g E), DPPH (586 and 750 mg of TE/g) and ABTS (1453 and 1319 mg of TE/g) results. Moreover, gallic acid, quercetin, quercetin galloyl glucoside, trigalloyl hexoside, procyanidin dimers and sulfated flavonoids were identified by LC-DAD-ESI-MS for the first time in this species. The relevant cytoprotective capacity (at 300 µg/mL) against ß-amyloid peptide induced toxicity in PC12 cells of EtOAc fractions were corroborated with the chemical composition. In addition, purified molecules were tested for their ORAC and neuroprotective activity. Quercetin showed the best ORAC value (33.55 mmol TE/g polyphenols); nevertheless, procyanidin dimer exhibited an exceptionally efficient neuroprotective activity (100% of viability at 50 µg/mL). DISCUSSION AND CONCLUSIONS: These findings suggest that this halophyte is a promising source of antioxidant and neuroprotective molecules for pharmaceutical purposes.

This work aimed to investigate the richness of a Tunisian xerohalophyte Frankenia thymifolia aerial and root parts on phenolics and to evaluate the antioxidant and neuroprotective properties of this medicinal species. After fractionation using increasing and different solvent polarities, results displayed five fractions, where ethyl acetate (EtOAc) shoot and root fractions possess considerable total phenolic contents (221 and 308 mg of GAE/g of E, resp.) related to their important antioxidant activities such as ORAC (918 and 713 mg of TE/g of E), DPPH (282 and 821 mg of TE/g), and ABTS (778 and 1320 mg of TE/g) tests. Then, the identification of the main compounds by HPLC-DAD-ESI-MS and neuroprotective property of the most active fraction EtOAc were assessed. A total of 14 molecules were identified, which have been described for the first time in F. thymifolia. The major compounds identified were pinoresinol and kaempferol glycoside in aerial parts and gallic acid and ellagitannin in roots. Neuroprotective capacity against ß-amyloid (Aß) peptide induced toxicity in PC12 cells of EtOAc fraction showed a significant protective activity at lower concentration (25 and 50 µM). The relevant antioxidant and neuroprotective activities of F. thymifolia EtOAc fraction corroborated their chemical compositions.
