BACKGROUND: This study aimed to establish the elemental profiling and origin combined with the genetic asset of coffee samples collected from major coffee-producing countries. A total of 76 samples were analysed for 41 elements using inductively coupled plasma-optical emission spectroscopy (ICP-OES), inductively coupled plasma-mass spectrometry (ICP-MS), and inductively coupled plasma-triple quadrupole mass spectrometry (ICP-MS/MS). The mineral composition of the silver skin detachment during the roasting process was also evaluated to verify the loss of minerals during roasting, differences in composition with beans, and between species. RESULTS: Application of linear discriminant analysis provided models with an accuracy of 93.3% for continents, 97.8% for countries of cultivation, and 100% for species. Discrimination between Arabica, Canephora coffee, and Eugenoides, and different varieties of Arabica species were identified in both models with calcium (Ca), barium (Ba), cadmium (Cd), rubidium (Rb), and strontium (Sr) as significant discriminant elements. Rb, Sr, sulphur (S), and thulium (Tm) were significant discriminant elements in both models for geographical distinction at different scales. Most of the elements had significantly higher values in silver skin than those in roasted coffee at different magnitudes, with exceptions of P and Rb. CONCLUSION: In summary, determination of mineral elements, processed by multivariate statistical analysis, was demonstrated to be discriminant for different coffee species. Linear discriminant analysis of the elemental analysis of samples from the seven major producing countries provided a reliable prediction model. Elemental analysis of major and minor elements is relatively easy and can be used together with other traceability systems and sensory evaluations to authenticate the origin of roasted coffee, different species, and varieties. © 2023 The Authors. Background: In 2017, the European Commission renewed the approval of glyphosate (GLY) but only for five years. GLY remains one of the most controversial and studied molecules. Method: A simplified method was tested for the determination of GLY in white rice (WR) and brown rice (BR), after extraction only with a methanol solution, by liquid chromatography coupled with inductively coupled mass triple quadrupole (HPLC-ICP-MS/MS) with a PRP-X100 anionic column. After performing a test on groundwater, the quantification of GLY in WR and BR was validated in terms of the LOD, LOQ, accuracy, precision, linearity, and the matrix effect. Results: The LOD was 0.0027 mg kg−1 for WR and 0.0136 mg kg−1 for BR. The LOQ was 0.0092 mg kg−1 for WR and 0.0456 mg kg−1 for BR. The mean recoveries were within 76−105% at three fortification levels. The relative standard deviation for the analysis (five replicates for three spike levels) was < 11% for both matrices. A linear response was confirmed in all cases in the entire concentration range (R2WR = 1.000 and R2BR = 0.9818). Conclusion: The proposed method could be considered useful for the determination of GLY in different types of rice and designed and adapted for other cereals. The matrix effect, quantified in BR matrix extraction, could be avoided by using a matrix-matched calibration line.

Plant Biostimulants (BSs) are a valid supplement to be considered for the integration of conventional fertilization practices. Research in the BS field keeps providing alternative products of various origin, which can be employed in organic and conventional agriculture. In this study, we investigated the biostimulant activity of the eluate obtained as a by-product from the industrial production of lactic acid bacteria on bare agricultural soil. Eluates utilization is in line with the circular economy principle, creating economical value for an industrial waste product. The research focused on the study of physical, chemical, biochemical, and microbiological changes occurring in agricultural soil treated with the biowaste eluate, applied at three different dosages. The final aim was to demonstrate if, and to what extent, the application of the eluate improved soil quality parameters and enhanced the presence of beneficial soil-borne microbial communities. Results indicate that a single application at the two lower dosages does not have a pronounced effect on the soil chemical parameters tested, and neither on the biochemical proprieties. Only the higher dosage applied reported an improvement in the enzymatic activities of ß-glucosidase and urease and in the chemical composition, showing a higher content of total, nitric and ammonia N, total K, and higher humification rate. On the other hand, microbial communities were strongly influenced at all dosages, showing a decrease in the bacterial biodiversity and an increase in the fungal biodiversity. Bioinformatic analysis revealed that some Operative Taxonomic Units (OTUs) promoted by the eluate application, belong to known plant growth promoting microbes. Some other OTUs, negatively influenced were attributed to known plant pathogens, mainly Fusarium spp. Finally, the ecotoxicological parameters were also determined and allowed to establish that no toxic effect occurred upon eluate applications onto soil.

In this study, the in vitro effects of different Se concentrations (5, 10, 15, 20, and 100 mg kg-1) from different Se forms (sodium selenite, sodium selenate, selenomethionine, and selenocystine) on the development of a Fusarium proliferatum strain isolated from rice were investigated. A concentration-dependent effect was detected. Se reduced fungal growth starting from 10 mg kg-1 and increasing the concentration (15, 20, and 100 mg kg-1) enhanced the inhibitory effect. Se bioactivity was also chemical form dependent. Selenocystine was found to be the most effective at the lowest concentration (5 mg kg-1). Complete growth inhibition was observed at 20 mg kg-1 of Se from selenite, selenomethionine, and selenocystine. Se speciation analysis revealed that fungus was able to change the Se speciation when the lowest Se concentration was applied. Scanning Electron Microscopy showed an alteration of the fungal morphology induced by Se. Considering that the inorganic forms have a higher solubility in water and are cheaper than organic forms, 20 mg kg-1 of Se from selenite can be suggested as the best combination suitable to inhibit F. proliferatum strain. The addition of low concentrations of Se from selenite to conventional fungicides may be a promising alternative approach for the control of Fusarium species.

Multielement analysis and chemometric methods were proposed to discriminate the Sorrento lemon (PGI) juices according to geographical origin. In 2018 and 2019, 169 fruits from three farms in PGI area and two in not-PGI area were collected and analysed for essential and not-essential elements by ICP-MS. The PCA of multielement fingerprinting grouped lemon juices from PGI farms revealing a strong differentiation at small geographical scale. The S-LDA discriminated lemon juices for Mo, Ba, Rb, Mg, Co, Ca, Fe, Sr on the two production years, giving 97.7% correct classification, 98.5% accuracy and 93.8% external validation. The good correlation lemon juice vs cultivation soil and the soil discrimination by not-essential elements suggested the use of these elements as reliable indicators of lemon juice provenances. Despite lowering the number of variables, constituted by not-essential elements Ba, Rb, Ti, Co, the use of S-QDA discriminated the lemons juices with 87.5% accuracy and 83.9% validation.

We assessed the effects of EDTA and selected plant growth-promoting rhizobacteria (PGPR) on the phytoremediation of soils and sediments historically contaminated by Cr, Ni, and Cu. A total of 42 bacterial strains resistant to these heavy metals (HMs) were isolated and screened for PGP traits and metal bioaccumulation, and two Enterobacter spp. strains were finally selected. Phytoremediation pot experiments of 2 months duration were carried out with hemp (Cannabis sativa L.) and giant reed (Arundo donax L.) grown on soils and sediments respectively, comparing in both cases the effects of bioaugmentation with a single PGPR and EDTA addition on plant and root growth, plant HM uptake, HM leaching, as well as the changes that occurred in soil microbial communities (structure, biomass, and activity). Good removal percentages on a dry mass basis of Cr (0.4%), Ni (0.6%), and Cu (0.9%) were observed in giant reed while negligible values (<100‰) in hemp. In giant reed, HMs accumulated differentially in plant (rhizomes > > roots > leaves > stems) with largest quantities in rhizomes (Cr 0.6, Ni 3.7, and Cu 2.2 g plant-1). EDTA increased Ni and Cu translocation to aerial parts in both crops, despite that in sediments high HM concentrations in leachates were measured. PGPR did not impact fine root diameter distribution of both crops compared with control while EDTA negatively affected root diameter class length (DCL) distribution. Under HM contamination, giant reed roots become shorter (from 5.2 to 2.3 mm cm-3) while hemp roots become shorter and thickened from 0.13 to 0.26 mm. A consistent indirect effect of HM levels on the soil microbiome (diversity and activity) mediated by plant response (root DCL distribution) was observed. Multivariate analysis of bacterial diversity and activity revealed not only significant effects of plant and soil type (rhizosphere vs. bulk) but also a clear and similar differentiation of communities between control, EDTA, and PGPR treatments. We propose root DCL distribution as a key plant trait to understand detrimental effect of HMs on microbial communities. Positive evidence of the soil-microbe-plant interactions occurring when bioaugmentation with PGPR is associated with deep-rooting perennial crops makes this combination preferable over the one with chelating agents. Such knowledge might help to yield better bioaugmented bioremediation results in contaminated sites.

Organic and mineral fertilizers are important sources of ammonia (NH3 ) emissions from agricultural fields. The objectives of this study were (a) to evaluate how different cover crop (CC) residues (i.e., rye [Secale cereale L.], white mustard [Sinapis alba L.], and bare soil as control) in combination with different application methods of digestate (surface broadcast vs. shallow injection) affect NH3 volatilization before planting maize (Zea mays L.) and (b) to assess the residual effect of previous CCs on NH3 volatilization after urea top-dress application at the V5-V6 phonological stage of maize. Ammonia volatilization was measured using semi-static chambers for 14 d (335 h) after planting and for 6 d (150 h) at the V5-V6 stage. Overall, NH3 emissions decreased by 67-77% with digestate injection compared with surface broadcasting. However, the reduction in NH3 volatilization using the injection method was significantly lower with mustard residue (6.72 kg NH3 -N ha-1 ) than with rye residue (14.15 kg NH3 -N ha-1 ), which allowed for more volatilization by increasing the exposure of digestate to the air. Broadcast digestate method did not affect the cumulative NH3 -N losses obtained with different CC types. After urea top-dressing at the V5-V6 stage of maize, the cumulative losses of NH3 (during 150 h) were 2.99 kg NH3 -N ha-1 with rye as previous CC and 2.49 kg NH3 -N ha-1 with mustard. Our study shows that digestate injection before maize planting and urea top-dressing application followed immediately by irrigation (15 mm) could be considered as useful strategies to mitigate NH3 volatilization and increase N use efficiency in maize.

The phytoremediation potential of four crop species cultivated on carbonation lime coming from the sugar industry with water-soluble nickel (Ni) exceeding the Italian legal limit of 10 µg L-1 was assessed. Two autumn-winter species (spinach and canola) were tested with and without the addition of bentonite in a greenhouse experiment in order to overcome prolonged unfavourable weather conditions. Two spring-summer species (sunflower and sorghum) were grown in outdoor boxes. Plant species were selected among crops of interest for phytoremediation and their rotation throughout the year enable to maintain a permanent vegetation cover. Nickel concentration in different plant tissues and the concentrations of soluble and bioavailable Ni in lime were measured. In the greenhouse study, soluble Ni decreased below the legal limit in all the tests, and the combined effect of bentonite and plants reduced Ni in lime mainly in the bioavailable fraction. Spinach and sunflower emerged to be more suitable for phytoextraction than canola and sorghum, because of the higher concentration of the metal in the epigeal portions. The results from the outdoor experiment highlighted that sorghum has a good phytostabilisation potential since its ability to accumulate Ni mainly at the root level and to attract a significant amount of bioavailable Ni in the rhizosphere. This study arose from a real scenario of environmental contamination and investigated the potential of different approaches on the bioremediation of a specific industrial waste product.

This study investigated the effect of a dietary combination of selenium and olive leaves on rabbit health status in order to evaluate the potential use of these combinations as functional ingredients in feed and food. Sixty weaning rabbits were fed with three diets: control feed (C), control feed + 10% normal olive leaves (OL), or olive leaves enriched in Se (2.17 mg Se/kg d.m.; SeOL). The plasma mineral profile, antioxidant status, and leukocyte DNA damage were determined. Inorganic Se was the most abundant form in the OL diet, while the organic one was higher in SeOL than C and OL. A similar trend was found in the plasma. Protein oxidation showed higher values in both supplemented groups; in addition, dietary Se led to a significant improvement (+ 40%) in ferric reducing ability of plasma (FRAP). A marked reduction in DNA damage (9-fold) was observed in the SeOL group compared to C. The combination of selenium and olive leaves in the diet of growing rabbits increased plasma SeMet and FRAP and reduced leukocyte DNA damage.

Iron (Fe) is an essential micronutrient for plant life and development. However, in soil, Fe bioavailability is often limited and variable in space and time, thus different regions of the same root system might be exposed to different nutrient provisions. Few studies showed that the response to variable Fe provision is controlled at local and systemic levels, albeit the identity of the signals involved is still elusive. Iron itself was suggested as local mediator, whilst hormones were proposed for the long-distance signalling pathway. Therefore, the aim of this work was to assess whether Fe, when localized in a restricted area of the root system, might be involved in both local and systemic signaling. The combination of resupply experiments in a split-root system, the use of 57Fe isotope and chemical imaging techniques allowed tracing Fe movement within cucumber plants. Soon after the resupply, Fe is distributed to the whole plant, likely to overcome a minimum Fe concentration threshold aimed at repressing the deficiency response. Iron was then preferentially translocated to leaves and, only afterwards, the root system was completely resupplied. Collectively, these observations might thus highlight a root-to-shoot-to-root Fe translocation route in cucumber plants grown on a patchy nutrient substrate.

Chianti is a precious red wine and enjoys a high reputation for its high quality in the world wine market. Despite this, the production region is small and product needs efficient tools to protect its brands and prevent adulterations. In this sense, ICP-MS combined with chemometrics has demonstrated its usefulness in food authentication. In this study, Chianti/Chianti Classico, authentic wines from vineyard of Toscana region (Italy), together samples from 18 different geographical regions, were analyzed with the objective of differentiate them from other Italian wines. Partial Least Squares-Discriminant Analysis (PLS-DA) identified variables to discriminate wine geographical origin. Rare Earth Elements (REE), major and trace elements all contributed to the discrimination of Chianti samples. General model was not suited to distinguish PDO red wines from samples, with similar chemical fingerprints, collected in some regions. Specific classification models enhanced the capability of discrimination, emphasizing the discriminant role of some elements.

In the present study the use of Selenium-fortified olive leaves as potential dietary source of Se in rabbit nutrition was evaluated. Sixty New Zealand White rabbits (35 days of age) were randomly assigned to the following dietary treatments: standard diet (C), and C supplemented with either 10% olive leaves (OL) or 10% Selenium-fortified olive leaves (SeOL; 100 mg/L of foliar spray sodium selenate solution). At 70 days of age, 10 rabbits per group were slaughtered and the oxidative status, mineral profile and Selenium speciation of Longissimus dorsi meat was analyzed. Meat of the SeOL group exhibited better oxidative status (lower TBARS, higher GPx and α-tocopherol values) and a 5-fold higher Se content compared to that of the other treatments. The main Se form was SeMet (7-fold higher in the SeOL group), followed by SeCys2. The present trial demonstrates the possibility of using agro-industrial by-products as ingredients in rabbit feeds, thereby enriching meat bioactive compound content.

The contents of total Se and of inorganic and organic Se species, as well as the contents of proteins, chlorophylls, carotenoids, and phenolic acids, were measured in 10-day old sprouts of rice ( Oryza sativa L.) obtained with increasing levels (15, 45, 135, and 405 mg Se L-1) of sodium selenite and sodium selenate and with distilled water as control. Increasing Se levels increased organic and inorganic Se contents of sprouts, as well as the content of phenolic acids, especially in their soluble conjugated forms. Moderate levels of sodium selenite (i.e., not higher that 45 mg L-1) appeared the best compromise to obtain high Se and phenolic acid yields together with high proportion of organic Se while limiting residual Se in the germination substrate waste. Se biofortification of rice sprouts appears a feasible and efficient way to promote Se and phenolic acid intake in human diet, with well-known health benefits.

Rice and rice-based foodstuffs are important pathways for inorganic As dietary intake. This work shows a detailed picture of As content and speciation in Italian rice, which contributes to more than one-half of the European production, and addresses the role of soil chemistry and agronomic management on As concentration in rice grain, in view of ameliorative strategies. The mean total As content in Italian white rice was 155 ± 65 µg kg-1 with significant differences among producing areas, while the mean inorganic As was 102 ± 26 µg kg-1, largely below the E.U. limit of 200 µg kg-1 for white rice, although part of the production would not be suitable for baby food production, which requires less than 100 µg kg-1 of inorganic As. The differences in As content and speciation in rice among the studied areas resulted from the complex interactions of soil, plant, and anthropic factors. Among others, Si nutrition seemed to play a key role in regulating As transfer from soil to plant.

Biofortification is an agronomic-based strategy, utilized by farmers, to produce selenium (Se)-enriched food products that may help reduce dietary deficiencies of Se occurring throughout susceptible regions of the world. The foliar exposure route application ensures a high efficiency of Se assimilation by the plant since it does not depend on root-to-shoot translocation. In this study we treated grapevines of Sangiovese variety in the pre-flowering period with sodium selenate (100mg Se L-1). Se content was measured in leaves, fruit at harvest time and in wine respectively in treated and not treated samples with ICP-MS. At harvest, a higher amount of Se in the treated leaves compared to untreated ones was found, 16.0±3.1mgkg-1 dry weight (dw) against 0.17±0.006mgkg-1 dw in the untreated ones. The treated grapes had a content of Se of 0.800±0.08mgkg-1 dw, while that untreated one 0.065±0.025mgkg-1 dw. Immediately after the malolactic fermentation, the wine obtained from treated and untreated vines had a Se content of 0.620±0.09mg Se L-1 and 0.024±0.010mg Se L-1 respectively. In our case the percentage of inorganic Se is 26% of the total Se in the untreated wine, while in Se enriched wine this percentage increase to 47.5% of the total Se. The Se(VI) was the inorganic chemical form more present in enriched wine, probably due to foliar application with selenate. Distributions of Se species suggested being careful to the choice of the enrichment solutions to promote a balanced distribution of different chemical forms, perhaps favouring the accumulation of organic forms.

This work provides a method with application of valid techniques to extract and determinate inorganic species of antimony (Sb) for water. The procedure involves•the simultaneous accumulation of Sb(III) and Sb(V) on passive samplers like Diffusive Gradient in Thin Films (DGT) with iron (Fe) oxide gel, eliminating the risk of speciation changes due to transport and storage;•application of less concentrated acid (50 mM Na2EDTA) for elution and preservation of Sb species from DGT resin;•subsequent analytical determination of inorganic species with High Performance Liquid Chromatography-Isotopic Dilution-Inductively Coupled Plasma Mass Spectrometer (HPLC-ID-ICP MS) based on determination of the isotope ratio ((123)Sb/(121)Sb) of isotopes in the samples after spiking with 123Sb enriched standard solution, reducing the effect of signal drift and matrix effect on the final value.

Mangroves are critically threatened by human activities, despite the important ecosystem functions and services they provide. Mangroves in Cameroon represent no exception to the worldwide trend of mangrove destruction, especially around Douala, on the Wouri river estuary. In two sites around Douala, we assessed the presence of sterols, PAHs, PCBs, DEHP, DDT and its metabolite p,p'-DDE and potentially toxic metals in sediment samples. As a proxy of ecological quality, we measured the diversity and abundance of macrobenthos assemblages. We detected p,p'-DDE contamination, with concentrations higher than 3µgkg(-1) in 16 out of 26 samples which were attributed to recent widespread use of DDT. The detection of sterols revealed faecal contamination. Significant sensitivity of the macrobenthos to contaminants was revealed, with possible implications on the overall mangrove vulnerability to climate change and on the provision of ecosystem services to local populations.

The aims of the work were to detect and quantify hexavalent chromium in 14 soil samples from an area in Lombardia (northern Italy) contaminated by two polluted water plumes. Cr(VI) was extracted from the solid samples by applying focused microwaves in an alkaline medium after Cr(III) complexation with EDTA. Cr(VI) was reduced to Cr(III) when previously reported extraction conditions for the analysis of certified reference materials were used, and Cr(VI) could not be reliably quantified in the soil samples. The influence of organic matter and iron contents in the samples on the reduction of Cr(VI) was subsequently studied using a new set of soil samples with different iron and organic matter concentrations. Isotope dilution mass spectrometry (IDMS) measured two different enriched stable isotopes of Cr (54 and 53) to evaluate the reduction extent of hexavalent chromium during the analytical procedure. The extraction conditions were optimized to obtain the lowest amount of Cr(VI) reduction and quantify Cr(VI) in the polluted soil samples from Lombardia.

BACKGROUND: For the past century, ferroalloy industries in Brescia province, Italy produced particulate emissions enriched in manganese (Mn), lead (Pb), zinc (Zn), copper (Cu), cadmium (Cd), chromium (Cr), iron (Fe), and aluminum (Al). This study assessed metal concentrations in soil and vegetables of regions with varying ferroalloy industrial activity levels. METHODS: Home gardens (n=63) were selected in three regions of varying ferroalloy plant activity durations in Brescia province. Total soil metal concentration and extractability were measured by X-Ray Fluorescence (XRF), aqua regia extraction, and modified Community Bureau of Reference (BCR) sequential extraction. Unwashed and washed spinach and turnips cultivated in the same gardens were analyzed for metal concentrations by flame atomic absorption spectrometry. RESULTS: Median soil Al, Cd, Fe, Mn, Pb, and Zn concentrations were significantly higher in home gardens near ferroalloy plants compared to reference home gardens. The BCR method yielded the most mobile soil fraction (the sum of extractable metals in Fractions 1 and 2) and all metal concentrations were higher in ferroalloy plant areas. Unwashed spinach showed higher metal concentrations compared to washed spinach. However, some metals in washed spinach were higher in the reference area likely due to history of agricultural product use. Over 60% of spinach samples exceeded the 2- to 4-fold Commission of European Communities and Codex Alimentarius Commission maximum Pb concentrations, and 10% of the same spinach samples exceeded 2- to 3-fold maximum Cd concentrations set by both organizations. Turnip metal concentrations were below maximum standard reference values. CONCLUSIONS: Prolonged industrial emissions increase median metal concentrations and most soluble fractions (BCR F1+F2) in home garden soils near ferroalloy plants. Areas near ferroalloy plant sites had spinach Cd and Pb metal concentrations several-fold above maximum standard references. We recommend thorough washing of vegetables to minimize metal exposure.

This work is part of a wider monitoring project of the agricultural soils in Lombardia, which aims to build a database of topsoil properties and the potentially toxic elements, organic pollutants and gamma emitting radionuclides that the topsoils contain. A total of 156 agricultural soils were sampled according to the LUCAS (Land Use/Cover Area frame statistical Survey) standard procedure. The aim was to provide a baseline to document the conditions present at the time of sampling. The results of the project concerning soil radioactivity are presented here. The aim was to assess the content of (238)U, (232)Th, (137)Cs and (40)K by measuring soil samples by gamma spectrometry. (238)U, (232)Th and (40)K activities range 24-231, 20-70, and 242-1434 Bq kg(-1) respectively. The geographic distribution of (238)U reflects the geophysical framework of the Lombardia region: the soils with high content of uranium are distributed for the most part in the South Alpine belt, where the presence of magmatic rocks is widespread. These soils show an higher activity of (238)U than of (232)Th. The (238)U activities become lower than (232)Th when soils are located in the plain, originating from basic sedimentary rocks. (137)Cs activity ranges 0.4-86.8 kBq m(-2). The lowest activity of (137)Cs is in the plain, whereas the highest is in the North on soils kept as lawn or pasture. The (137)Cs activity of some samples suggests the presence of accumulation processes that lead to (137)Cs enriched soils. This is the first survey of gamma emitting radionuclides in Lombardia that is based on the LUCAS standard sampling. The results from this monitoring campaign are important for the human radiation exposure and provide the zero point, which will be useful for assessing future effects due to external factors such as human activities.
