RESUMO

Radioisotopic assay has shown that in vitro histamine increases the metabolism of cyclic adenosine monophosphate (cAMP) through H-2 receptors in the peripheral blood lymphocytes of healthy individuals and patients with atopic bronchial asthma. In patients who have asphyxia the histamine-induced stimulation of the activity of cAMP phosphodiesterase metabolizing cAMP is considerably decreased. In is suggested that high cAMP metabolism may be a cause of desensitization of histaminic and adrenergic receptors in atopy whose aggravation is accompanied by histaminemia.

RESUMO

A synthetic carbon sorbent (SKNP-2) was included in the treatment of 197 patients with relapsing urticaria. Courses lasted 7, 14, 20 days. The clinical effect was favourable and its efficacy increased with prolongation of the therapy course time using the enterosorbent. Immunological parameters did not change in the course of treatment.

RESUMO

Alternative principles of regional IL-2-dependent adoptive immunocytotherapy of malignant tumours are suggested. Regional adoptive immunocytotherapy consists in the administration of the short-term IL-2 treated lymphocytes of the lymph nodes removed from the tumour which have tumour-associated suppressor activity and spleen cells after elimination of cells with the immunosuppressive activity.

RESUMO

The level of endogenous production of IL-2 by lymphocytes of lymph nodes regional to tumour and by mononuclear cells of peripheral blood, proliferative response of these cell to recombinant IL-2, as well as a modifying influence of autologous serum and actively proliferating bioptats of autologous tumours on enumerated parameters have been studied in cancer patients (tumours of the head and neck and locomotor system). Regional IL-2-dependent immunotherapy of malignant tumors with obligatory preliminary testing for individual sensitivity of the tumor bioptat to the influence of the RIL-2 and RIL-2 activated lymphocytes is shown to be promising.

RESUMO

Interleukin-2 (IL-2) production by lymphocytes of a peripheral blood (LPB) in patients with soft tissue sarcomas is considerably lower in comparison with that in donors. Autological serum had either suppressive or stimulative effect on LPB IL-2 production in the observed patients. The investigated serum affected LPB of donors in the same way. Serum of patients with soft tissue sarcomas had the same dual effect on the ability of the active drug IL-2 to proliferate human T-lymphoblasts. Moreover serum-inhibitors of production acted as IL-2 activity stimulators, while serum-stimulators of IL-2 production inhibited the mediator effects.

The data on biological properties and effects of interleukin-2, one of the main mediators of the immunity are presented. The results of its use in the tumour immunotherapy are analyzed, the used approaches and mechanisms of action of this kind of immunotherapy being estimated. The ways to increase the efficiency of interleukin-2 application in clinics are under discussion.

RESUMO

The ability of lymph node and spleen cells to secrete and sorb interleukin-2 (IL-2) during the growth of sarcomas induced by 20-methylcholanthrene is studied. Lymph node cells show an increased level of IL-2 production at early stages of the tumour growth whereas spleen cells produced IL-2 in minimal amounts. A decrease in the IL-2 production by lymph node cells with the tumour growth and a relative increase of IL-2 production in the spleen cell pool are observed. At all stages of the tumour growth the ability of lymphoid cells to sorb IL-2 decreased strongly in comparison with the analogous cells of normal animals activated by mitogen. Lymph node cells of animals with tumour sorb IL-2 more effectively than splenocytes.

RESUMO

It is shown that under conditions of IgE-induced reaction the tumour growth is altered, which may be explained by different degree of expression of functional activities of immunocompetent cells in mice with transplanted tumours and marked immediate-type reaction. Changes in the immunological state manifest in stimulation of the proliferative response primarily to B-cell mitogens under conditions of increased synthesis of IgE, which permits suggesting that under these conditions B-cells play an important role in antitumour immunity.

RESUMO

Interleukin-2 (IL-2) production of phytohemagglutinin-stimulated peripheral blood lymphocytes (PBL) was studied in 9 healthy subjects and 19 patients with soft tissue sarcomas. Mean IL-2 production by PBL in 19 patients was significantly diminished as compared with the control. Surgery leads to an increase of IL-2 production, however, the levels observed in the control do not restore completely.

RESUMO

The use of two independent test-systems (intravenous adoptive transfer of immune response on tumour cells by intact syngenic recipient and mixed cultivation of immunocompetent and tumour cells in diffusion chambers) made it possible to establish that exogenous IL-2 possesses an expressive capacity both to the positive and to the negative modulation of immunological mechanisms during the tumour growth.

RESUMO

Interleukin-2 is studied for its effect on proliferation of cultured tumour cells in vivo and on the nature of infiltration of malignant tumours explants of human soft tissues. On the basis of spheroid formation phenomenon and a number of morphological criteria three main types of reaction differing in inhibition or stimulation of the tumour growth, time of formation and the number of spheroids formed, manifestation of lymphocytic infiltration and nature of lymphocytes location on the filter have been distinguished.

RESUMO

The application of a complex of methods for fractionation of mouse spleen cells with transplanted rhabdomyosarcomas has shown that optimal conditions for adoptive intravenous transfer of the tumour-associated suppressor activity are created under joint injection of the primed macrophages and lymphoid cells of spleen (from animals with the progressively growing tumours) interacting with the macrophages to syngenic recipients.

RESUMO

Peculiarities of interaction of tumour cells and lymphocytes were studied in cultivating explants of 19 human lung tumours in diffusion chambers. Results were analyzed with due regard for the histological structure of tumours, level of tissue differentiation and clinical stage of the disease. Criteria are developed for evaluating the interaction of tumour cells and lymphocytes, namely: the relative content of lymphocytes in culture, the number of spheroids and a way of their destruction (edge one or with lymphocytes), presence of lymphocytic clusters and aggregates of blast-like cells, presence of tumour cell-lymphocyte complexes. The obtained results may be used for estimation of the tumour malignancy degree.

RESUMO

A functional state of neutrophils was studied by their ability to chemotaxis and phagocytosis as well as their enzymatic activity (myeloperoxidase, acid and alkaline phosphatase, glycogen, cation proteins) of 13 patients with malignant neoplasms in dynamics (before and 2 weeks after the radical operation) was studied. Decreasing of all investigated parameters before the operation and significant increasing of chemotaxis and phagocytosis after the operation were revealed.

RESUMO

The data based on the comparative evaluation of changes in immunological processes due to the effect of decaris, autovaccine and standard staphylococcal allergens are presented. These data indicate that such changes resulting from the clinical effectiveness of different kinds of immunotherapy are accompanied by identical changes in the immune system of patients with infectious allergic bronchial asthma. At the same time the results of this study point out that the therapeutic effectiveness of autovaccine is higher than that of decaris.

RESUMO

The work contains the results obtained in the study of immunological processes in the dynamics of specific hyposensitization with microbial allergens, prepared from Proteus and staphylococci, in 22 infectious allergic bronchial asthma patients sensitized to these microorganisms. The therapeutic effect of this treatment was accompanied by the activation of the processes of cellular and humoral immunity, while the aggravation of the main disease was observed in patients with a pronounced decrease in the activity of immunological processes which were unaffected by the stimulating influence of specific hyposensitization.
