RESUMO

The rhinebothriidean tapeworm family Escherbothriidae has recently been expanded to include the genus Ivanovcestus , species of which parasitise arhynchobatid skates. Similarities in morphology and host associations between Ivanovcestus and Semiorbiseptum - a genus yet to be assigned to one of the families in the order Rhinebothriidea - led us to explore the possibility that Semiorbiseptum might also belong in the Escherbothriidae. Morphological similarities with Scalithrium ivanovae , Scalithrium kirchneri and Rhinebothrium scobinae , all of which also parasitise arhynchobatid skates, raised questions regarding the generic placements of these species. In addition, new collections from the skate Sympterygia brevicaudata revealed two new species that morphologically resemble species of Ivanovcestus . A combination of morphological and molecular data were used to assess the generic placement of the newly discovered species and refine our understanding of the membership of the family Escherbothriidae. Sequence data for the D1-D3 region of the 28S rDNA gene were generated de novo for 14 specimens of 7 rhinebothriidean species and combined with comparable published data to represent all 6 families in the Rhinebothriidea in the analysis. The phylogenetic tree resulting from maximum likelihood analysis strongly supports the inclusion of the genus Semiorbiseptum in the family Escherbothriidae. Our work also suggests that the skate-hosted species previously assigned to Scalithrium and Rhinebothrium are also members of Semiorbiseptum and that Ivanovcestus is a junior synonym of Semiorbiseptum . Six species are transferred to Semiorbiseptum , bringing the total number of species in the genus to ten. The diagnosis of Semiorbiseptum is amended to accommodate the additional species. A second species in the previously monotypic type genus of the family, Escherbothrium , is described. The diagnosis of the Escherbothriidae is amended to include the new and transferred species. This study underscores the importance of integrating morphological and molecular data in bringing resolution to cestode systematics. We believe our findings provide a robust foundation for future research into the evolutionary history and host associations of cestodes within the order Rhinebothriidea and beyond. OBJECTIVE: The term source (or focus) control encompasses all those physical measures that can be used to reduce the inoculum and modify those factors in the infectious medium that promote microbial growth or foreign antimicrobial defenses of the host. The main objective of this systematic review (SR) is to know and compare whether early detection and control of the focus (in less than 6 hours) in adult patients treated in the ED for severe infection or sepsis, compared to not controlling the focus or delayed focus control (more than 12 hours) is more effective and safer (improves clinical evolution, mortality, complications, hospital stay or need for ICU admission). METHODS: A systematic review is carried out following the PRISMA regulations in the databases of PubMed, Web of Science, EMBASE, Lilacs, Cochrane, Epistemonikos, Tripdatabase and ClinicalTrials.gov from January 2000 to December 31, 2023 without language restrictions and using a combination of MESH terms: "Source Control", "Early" "Infection OR Bacterial Infection OR Sepsis", "Emergencies OR Emergency OR Emergency Department" and "Adults". Observational cohort studies were included. No meta-analysis techniques were performed, but results were compared narratively. RESULTS: A total of 1,658 articles were identified, of which 2 that met the inclusion criteria and were classified as high quality were finally analyzed. The included studies represent a total of 2,404 patients with 678 cases in which an intervention was performed to control the focus (28.20%). In the first study, 28-day mortality was lower in patients who underwent an intervention to control the focus (12.3% vs. 22.5%; P <0.001), with an adjusted HR of 0.538 (95% CI: 0.389-0.744; P<0.001). In the second, it was demonstrated that the time elapsed from when the patient was evaluated for the first time and was hemodynamically stabilized, until the start of surgery was associated with his survival at 60 days with an OR of 0.31 (95% CI: 0.19-0.45; P <0.0001). In fact, for each hour of delay an adjusted OR of 0.29 (95% CI: 0.16-0.47; P<0.0001) is established. So if the intervention is performed before 2 hours at 60 days, 98% of the patients are still alive, if it is performed between 2-4 hours it is reduced to 78%, if it is between 4-6 hours it drops to 55%, but if it is done for more than 6 hours there will be no survivors at 60 days. CONCLUSIONS: This review shows that source control carried out after the evaluation of patients attending the ED reduces short-term mortality (30-60 days) and that it would be advisable to implement any required source control intervention as soon as possible, ideally early (within 6 hours).

Entomopathogenic nematodes (EPNs) and predatory mirids are among the most effective natural enemies for the control of Tuta absoluta, a key pest of tomato worldwide. In the case of South America, the native EPN Heterorhabditis amazonensis and the predator Macrolophus basicornis (Stal) coexist in tomato. The intraguild interactions of two ENPs, H. amazonensis JPM4 and the widely used Steinernema carpocapsae, with the predator M. basicornis, were studied in laboratory setups with the objective of evaluating the potential interactions between them. We hypothesized that the two natural enemies do not interact negatively. The first nymphal stage of the predator was not affected when exposed to tomato leaves directly sprayed with both EPNs, which we attributed to the small size of its stylet. Individuals treated with H. amazonensis JPM4 had low mortality rates in all developmental stages with a maximum of 1.3%. Mortality was higher when the predator was exposed to S. carpocapsae (28% for adults) and correlated positively with increasing developmental stages. In prey choice tests between larvae infected with H. amazonensis and uninfected larvae, M. basicornis fed indistinctively showing no clear preference. When presented with choice larvae infected with S. carpocapsae, the consumption of healthy larvae was higher. In a no-choice prey acceptance experiment, M. basicornis preyed on both healthy and infected larvae with no difference in larvae consumed. Predators that had fed from infected larvae did not exhibit signs of nematode infection and survived. Our results indicate that the South American native H. amazonensis JPM4 and M. basicornis do not show a negative interaction and are promising candidates for combined use in T. absoluta biological control.

Scanning small-angle x-ray scattering (SAXS) measurements were performed on 36 formalin-fixed breast tissue biopsies obtained from two patients. All samples contained microcalcifications of type II, i.e. formed by hydroxyapatite. We demonstrate the feasibility of classifying breast lesions by scanning SAXS of tissues containing microcalcifications with a resolution of 35 [Formula: see text]m [Formula: see text] 30 [Formula: see text]m. We report a characteristic Bragg peak found around q = 1.725 nm-1 that occurs primarily for malignant lesions. Such a clear SAXS fingerprint is potentially linked to structural changes of breast tissue and corresponds to dimensions of about 3.7 nm. This material property could be used as an early indicator of malignancy development, as it is readily assessed by SAXS. If this fingerprint is combined with other known SAXS features, which also indicate the level of malignancy, such as lipid spacing and collagen periodicity, it could complement traditional pathology-based analyses. To confirm the SAXS-based classification, a histopathological workup and a gold standard histopathological diagnosis were conducted to determine the malignancy level of the lesions. Our aim is to report this SAXS fingerprint, which is clearly related to malignant breast lesions. However, any further conclusion based on our dataset is limited by the low number of patients and samples. Running a broad study to increase the number of samples and patients is of great importance and relevance for the breast-imaging community.

Tuta absoluta (Meyrick), a key pest of tomato, is quickly spreading over the world and biological control is considered as one of the control options. Worldwide more than 160 species of natural enemies are associated with this pest, and an important challenge is to quickly find an effective biocontrol agent from this pool of candidate species. Evaluation criteria for control agents are presented, with the advantages they offer for separating potentially useful natural enemies from less promising ones. Next, an aggregate parameter for ranking agents is proposed: the pest kill rate km. We explain why the predator's intrinsic rate of increase cannot be used for comparing the control potential of predators or parasitoids, while km can be used to compare both types of natural enemies. As an example, kill rates for males, females and both sexes combined of three Neotropical mirid species (Campyloneuropsis infumatus (Carvalho), Engytatus varians (Distant) and Macrolophus basicornis (Stål)) were determined, taking all life-history data (developmental times, survival rates, total nymphal and adult predation, sex ratios and adult lifespan) into account. Based on the value for the intrinsic rate of increase (rm) for T. absoluta and for the kill rate km of the predators, we predict that all three predators are potentially able to control the pest, because their km values are all higher than the rm of the pest. Using only km values, we conclude that E. varians is the best candidate for control of T. absoluta on tomato, with C. infumatus ranking second and M. basicornis last.

Abdominal aortic and popliteal artery aneurysms are vascular diseases which show massive degeneration, weakening of the vascular wall and loss of the vascular tissue functionality. They are driven by inflammatory, hemodynamical factors and biological alterations that may lead, in the case of an abdominal aortic aneurysm, to sudden and dangerous ruptures of the arteries. Here, human aortic and popliteal aneurysm tissues were obtained during surgical repair, and studied by synchrotron radiation X-ray scanning microdiffraction and small-angle scattering, to investigate the microcalcifications present in the tissues. Data collected during the experiments were transformed into quantitative microscopy images through the combination of statistical approaches and crystallographic methods. As a result of this multi-step analysis, microcalcifications, which are markers of the pathology, were classified in terms of chemical and structural content. This analysis helped to identify the presence of nanocrystalline hy-droxy-apatite and microcrystalline cholesterol, embedded in myofilament, and elastin-containing tissue with low collagen content in predominantly nanocrystalline areas. The generality of the approach allows it to be transferred to other types of tissue and other pathologies affected by microcalcifications, such as thyroid carcinoma, breast cancer, testicular microli-thia-sis or glioblastoma.

In most cases, the analysis of small-angle and wide-angle X-ray scattering (SAXS and WAXS, respectively) requires a theoretical model to describe the sample's scattering, complicating the interpretation of the scattering resulting from complex heterogeneous samples. This is the reason why, in general, the analysis of a large number of scattering patterns, such as are generated by time-resolved and scanning methods, remains challenging. Here, a model-free classification method to separate SAXS/WAXS signals on the basis of their inflection points is introduced and demonstrated. This article focuses on the segmentation of scanning SAXS/WAXS maps for which each pixel corresponds to an azimuthally integrated scattering curve. In such a way, the sample composition distribution can be segmented through signal classification without applying a model or previous sample knowledge. Dimensionality reduction and clustering algorithms are employed to classify SAXS/WAXS signals according to their similarity. The number of clusters, i.e. the main sample regions detected by SAXS/WAXS signal similarity, is automatically estimated. From each cluster, a main representative SAXS/WAXS signal is extracted to uncover the spatial distribution of the mixtures of phases that form the sample. As examples of applications, a mudrock sample and two breast tissue lesions are segmented.

Tuta absoluta (Meyrick; Lepidoptera: Gelechiidae) is a key pest of tomato and is quickly spreading over the world. We conducted an experiment aimed at evaluating the control capacity and risk for plant damage of three Neotropical mirid species, Campyloneuropsis infumatus (Carvalho; Hemiptera: Miridae), Engytatus varians (Distant; Hemiptera: Miridae) and Macrolophus basicornis (Stal; Hemiptera: Miridae) on T. absoluta infested tomato plants in large cages in an experimental greenhouse. During three successive periods of 9 wk each, we followed population development of the three mirids when exposed to T. absoluta, and of T. absoluta alone in separate cages in the greenhouse. We determined weekly the numbers of T. absoluta eggs and larvae per leaf, the number of mirid predators per leaf, the percentage of damaged leaves and fruits by T. absoluta, and the weight of fruits. Two of the mirid predators, C. infumatus and M. basicornis, successfully established on T. absoluta infested tomato plants and significantly reduced T. absoluta numbers, which ultimately resulted in an increased yield. These two mirid species hardly injured tomato plants or fruits as a result of plant feeding. Surprisingly, the species E. varians, which showed high predation rates in laboratory experiments, did not establish and reduce pest populations in any of the tests.

The predators Macrolophus basicornis (Stal), Engytatus varians (Distant) and Campyloneuropsis infumatus (Carvalho) consume large numbers of tomato pests such as Bemisia tabaci (Gennadius) and Tuta absoluta (Meyrick). However, they are zoophytophagous and feed on plant parts as well. We evaluated the type and effect of injury caused by nymphs and adults of these mirids on tomato seedlings and fruit in the absence of prey. For each mirid species, seedlings were exposed to groups of 20 nymphs or adults for 72 h, and fruits were exposed for 48 h to groups of four nymphs or adults. Type and the number of injury on stems, petioles and leaflets of tomato seedlings and fruits were recorded after removal of insects. Nymphs and adults of these mirids caused necrotic rings on the leaflets, but no injury was observed on stem and petioles. The necrotic rings on leaflets consisted of blemishes, characterized by feeding punctures surrounded by a yellowish, bleached area. The number of necrotic rings did not exceed one per individual mirid and seedlings developed normally. Nymphs also caused feeding punctures on tomato fruit, but in even lower numbers than on leaflets. Two weeks after the start of the experiment the tomato fruit still looked fresh and feeding punctures had disappeared. Adults did not cause any injury to tomato fruit. The results indicate that nymphs and adults of these zoophytophagous mirids cause little injury to tomato seedlings and fruit, even when present in high densities and in the absence of prey, making them interesting candidates for biological control.

The occurrence of a weak auditory warning stimulus increases the speed of the response to a subsequent visual target stimulus that must be identified. This facilitatory effect has been attributed to the temporal expectancy automatically induced by the warning stimulus. It has not been determined whether this results from a modulation of the stimulus identification process, the response selection process or both. The present study examined these possibilities. A group of 12 young adults performed a reaction time location identification task and another group of 12 young adults performed a reaction time shape identification task. A visual target stimulus was presented 1850 to 2350 ms plus a fixed interval (50, 100, 200, 400, 800, or 1600 ms, depending on the block) after the appearance of a fixation point, on its left or right side, above or below a virtual horizontal line passing through it. In half of the trials, a weak auditory warning stimulus (S1) appeared 50, 100, 200, 400, 800, or 1600 ms (according to the block) before the target stimulus (S2). Twelve trials were run for each condition. The S1 produced a facilitatory effect for the 200, 400, 800, and 1600 ms stimulus onset asynchronies (SOA) in the case of the side stimulus-response (S-R) corresponding condition, and for the 100 and 400 ms SOA in the case of the side S-R non-corresponding condition. Since these two conditions differ mainly by their response selection requirements, it is reasonable to conclude that automatic temporal expectancy influences the response selection process.

Leafminers (Diptera: Agromyzidae) are pests of various crops, mainly in greenhouses, and have Diglyphus spp. as important leafminer larval parasitoids. Until recently, only Diglyphus insularis (Gahan) had been reported in Brazil. In here we report the first records of Diglyphus begini (Ashmead), D. intermedius (Girault) and D. isaea (Walker) in Brazil. These parasitoids were found parasitizing leafminer larvae on cultivated and spontaneous plants in some areas of Minas Gerais state, Brazil.

Ischemia and reperfusion injury (IR) is an antigen independent inflammatory process that causes tissue damage. After IR, kidneys up-regulate leukocyte adhesion molecules and toll-like receptors (TLRs). Moreover, injured kidneys can also secrete factors (i.e. heat shock protein) which bind to TLRs and trigger intracellular events culminating with the increase in the gene expression of inflammatory cytokines. FTY720 is an immunomodulatory compound and protects at least in part kidneys submitted to IR. The mechanisms associated with FTY720's beneficial effects on kidneys after IR remain elusive. We investigated whether FTY720 administration in mice submitted to kidney IR is associated with modulation of TLR2 and TLR4 expression. C57BL/6 mice submitted to 30min of renal pedicles clamp were evaluated for serum parameters (creatinine, urea and nitric oxide), kidney histology, spleen and kidney infiltrating cells expression of TLR2 and TLR4, resident kidney cells expression of TLR2 and TLR4 and IL-6 protein expression in kidney. FTY720-treated mice presented decrease in serum creatinine, urea and nitric oxide, diminished expression of TLR2 and TLR4 both in spleen and kidney infiltrating cells, and reduced kidney IL-6 protein expression in comparison with IR non-treated mice. However, acute tubular necrosis was present both in IR non-treated and IR+FTY720-treated groups. Also, FTY720 did not prevent TLR2 and TLR4 expression in kidney resident cells. In conclusion, FTY720 can promote kidney function recovery after IR by reducing the inflammatory process. Further studies are needed in order to establish whether TLR2 and TLR4 down regulation should be therapeutically addressed as protective targets of renal function and structure after IR.

Óleos essenciais de plantas têm sido formulados de forma caseira ou colocados no mercado de forma artesanal para repelir mosquitos hematófagos. O presente trabalho avaliou o possível efeito repelente de alguns excipientes normalmente usados em formulações comerciais e de óleos essenciais de nove plantas, sobre fêmeas de Aedes albopictus. Os testes foram realizados em caixas contendo mosquitos, oferecendo-se uma área definida de dedos da mão tratados ou não. Exceto pelo creme Lanatte e os géis de carbopol, as substâncias excipientes avaliadas mostraram ação repelente. Apenas os óleos provenientes de citronela (5 e 10 por cento) resultaram em repelência expressiva, com índices médiosde proteção acimade 98 por cento. Os resultados indicam que a metodologia empregada é adequada para a avaliação preliminar de produtos com potencial para a repelência de mosquitos.


Plant essential oils have been domestically formulated or marketed as handcraft products to repel hematophagous mosquitoes. This work evaluated the possible repellent effect of both excipients normally used in commercial formulations and essential oils from nine plants against Aedes albopictus females. The assays were carried out in boxes containing mosquitoes, where a defined area of treated or non-treated fingers was offered. Except for Lanatte cream and carbopol gels, the evaluated excipients had repellent action. Only the oils from citronella (5 and 10 percent) presented expressive repellent action, with mean protection indexes above 98 percent. The results indicate that the used methodology is appropriate for the preliminary evaluation of products with potential to repel mosquitoes.

Cell phenotype evaluation enables better understanding of the rejection process in experimental transplantation. We studied allograft survival and the mechanisms associated with rejection in a murine model of skin transplantation in the absence of immunosuppression or after FTY720 or sirolimus (SRL) administration for 21 days. Leukocyte phenotype was evaluated in the peripheral blood, spleen, axillary lymph nodes, thymus gland, and skin graft using flow cytometry at 5 days posttransplantation. Treatment with FTY720 plus SRL increased skin allograft survival in association with lymphopenia, reduced CD11b+ and CD3+CD4+ cell percentages in the graft, decreased CD3+CD4+, CD3+CD8+, and CD11b+ cell counts in lymphoid organs, and decreased CD4+CD8+ cell count in the thymus. These results suggest that increased allograft survival in animals treated with FTY720 plus SRL is due to possible impairment of antigen presentation or recognition in the graft and secondary lymphoid organs, and decreased emigration of mature thymocytes to the periphery.

In transplantation, parasite diseases are transmitted from the donor, or appear as de novo infections, or activate from a dormant insource as a consequence of immunosuppression. Clinical findings have shown that an intact immune system is crucial to prevent recurrence of Leishmania infection. We used BALB/c and C57BL/6 mice to evaluate the role of FTY720 in leishmaniasis. Mice inoculated with Leishmania (Leishmania) amazonensis were followed over 7 weeks for foot thickness measurements after initiation of FTY720 treatment. After 10 days of treatment, spleen, blood, and the foot were harvested for evaluation. BALB/c showed greater evident foot thickness than C57BL/6 mice. Oral treatment with FTY720 (1 mg/kg/d) over 10 days produced the same outcome. Increases in CD4(+) and CD8(+) T cells were observed after infection; FTY720 treatment was associated with a decrease in CD4(+) T cells only in BALB/c mice, whereas CD8(+) T cells were decreased in both mice strains. CD11b(+) expression decreased after infection with a discrete increase after FTY720 treatment. Lymphopenia was observed among all FTY720-treated mice. In conclusion, we observed that FTY720 produced no worse an outcome as monotherapy in established infections with L (L) amazonensis.

The goal in transplantation is to obtain immunosuppressant combinations that decrease the incidence of acute and chronic rejection but cause fewer side effects. FTY720 is a new immunomodulator that prevents experimental allograft rejection without inhibiting T-cell activation. It is currently under clinical investigation for multiple sclerosis. We investigated whether FTY720 in combination with sirolimus (SRL) could cause renal toxicity in C57BL/6 mice when administered for 21 days. Serum creatinine and 24-hour urinary creatinine concentrations were assessed by enzymatic colorimetric assays. Urinary protein concentration was measured by the Bradford protein assay. Whereas serum creatinine levels were increased in FTY720 + SRL-treated animals, there were no changes in urinary volume, urinary protein levels, serum urea concentration, creatinine clearance, and kidney structure. Our findings suggested that FTY720 monotherapy for multiple sclerosis and other diseases could play an important immunomodulatory role without causing the side effects frequently observed with other transplantation regimens.
