RESUMO

NaClO effect on rat central neuron morphology and transcription after injection of high doses (500 mg/kg b.w.) of nembutal (NB) was studied. Cortical pyramids, Purkinje, basket and granule cells were examined. NaClO (3 mg/kg b.w) was injected 45 min after NB injection, and 45 min later animals were sacrificed. Intact rats and rats injected NB or NaClO alone were controls. NB treatment resulted in toxic-hypoxic injury of cerebral and cerebellar neurons. Transcription in all types of neurons was lower than in control cells. NaClO injected alone had virtually no effect on neuronal morphology while transcription in the middle pyramids and Purkinje cells was activated. NaClO injected after NB normalized morphology and transcription in all neuronal populations under study. In cortical pyramids transcription was even higher than in control group.

RESUMO

The effect of different regimens of hyperbaric oxygenation (HBO) on cortical pyramids morphology and transcription were studied under occlusion of common arteria carotis. The effect of early (3 hours after occlusion) and late (14 hours after occlusion) use of 1.2 and 2.0 ata of HBO was estimated. The rats were killed 15 minutes or 24 hours after HBO session. The effect of early use of HBO was normalizing, stabilizing and independent on the dose. Protecting effect of the late HBO on morphology and transcription activity of nuclear chromatin of pyramids was higher in the 1.2 ata regimen. In case of late HBO both pressures produced depressing effect on the nucleoplasmic chromatin, transcription.

RESUMO

Its effect of hyperbaric oxygenation (the doses 1.2 and 2 ata) on cortical pyramids of the rats with one-or both-side ligation of common arteria carotis 2.5 or 24 hours after operation was studied. The neuron survival and transcription activity were estimated. In all experimental situations except the most serious, e.g. 24 hours after the both-side artery ligation, the dose 2 ata was more effective. On the contrary, in most serious cases the dose 1.2 ata or applying in the baro-camera the air instead of oxygen provided the better result.

RESUMO

Transcriptional activity of the dark and light Purkinje cells' nuclei in cerebellar cortex was studied in rats 1h, 24h, 96h and 7 days post a systemic blood circulation arrest. The study was performed with the help of the histoautoradiographic method of Moore. Some substantial changes in template activity of the Purkinje cells' chromatin were observed in the postresuscitation period. Those began to occur immediately after resuscitation and were of phasic nature. The shifts in transcriptional activity of nucleolar and extranucleolar chromatin appeared to be analogous and of similar dynamics. The changes were more significant in the light neurons than in the dark ones. It has been supposed that the changes revealed might be one of the causes of development of neurologic disorders induced by severe hypoxia.

RESUMO

We report findings about age-related changes in the chromatin structure detected using ammoniacal silver and changes in the transcription rate in neurons by layers II-V of the sensomotor cortex of rats at days 7, 14 and 60 of postnatal life. Different types of neurons showed different average template activity but similar patterns of its age-related changes. The rearrangement of the chromatin structure in various types of cortical cells showed different orientation after day 14 of postnatal life.

RESUMO

Structuro-functional state of chromatin of the neurons' nuclei of the third, fourth and fifth layers of the sensorimotor cortex was investigated by Moore's method and with the help of ammonia silver colouring an hour and 24 hr post resuscitation in rats who had endured a systemic arrest of blood circulation. It was determined, that the level of labelling of nucleoli of light neurons increased in the early postresuscitation period, while that of dark neurons decreased as compared to check samples. The level of labelling of extranucleolar chromatin increased in both light and dark neurons in the cortex layers under investigation during the 1st hour post resuscitation. The transcriptional activity of extranucleolar chromatin of neurons of both types remained increased in the third and fourth cortex layers 24 hr post resuscitation, while in the fifth one it tended to normalize during the same period. Essential restructuring of the neurons's chromatin was determined with the help of ammonia silver in all cortex layers, which was indicative of a possible change in the spectrum of the synthesized proteins.

On the 4th day after revivification of rats, subjected to blood circulation arrest, transcriptional activity of chromatin has been evaluated after Moor's method and the state of histones has been investigated in neuronal nuclei of the III, IV and V layers of the cerebral cortex sensomotor area. Both in intact animals and those, subjected to clinical death, the neurons of the cortical layers examined sharply differ in their transcriptional activity of chromatin. The lowest level of labelling is revealed in the IV, the highest--in the V layer. After reviving the animals the transcriptional activity of chromatin in the neuronal nuclei of the III and V layers increases, and that of extranuclear chromatin--in all cortical layers. Increasing matrix activity occurs mostly in light cells; this demonstrates their greater reactivity than in dark cells. During the postresuscitation period essential changes of chromatin histone component are observed in neuronal nuclei of all the cortical layers studied.
