RESUMO

Daphnia spinulata Birabén, 1917 is an endemic cladoceran species, frequent in the zooplankton communities of the shallow lakes of the Pampean region of Argentina. These lakes have varying salinity levels and, being located in agricultural areas, are frequently subject to pesticide pollution. This study aimed to determine the effects of the herbicide glyphosate (Panzer Gold®) in combination with different salinity levels on the biological parameters of D. spinulata and its recovery ability after a short exposure. Three types of assays were performed: an acute toxicity test, a chronic assessment to determine survival, growth and reproduction, and recovery assays under optimal salinity conditions (1 g L-1). The LC50-48 h of glyphosate was 7.5 mg L-1 (CL 3.15 to 11.72). Longevity and the number of offspring and clutches were significantly reduced due to the combined exposure of glyphosate and increased salinity. The timing of the first offspring did not recover after glyphosate exposure. Our results reveal that D. spinulata is sensitive to the herbicide Panzer Gold® at concentrations well below those indicated in the safety data sheet of this commercial formulation, which causes stronger negative effects in conditions of higher salinity. Further research is needed to shed light on the sensitivity of this cladoceran to glyphosate and its variability under other interactive stress factors.

The total phenolic, flavonoid, and anthocyanin contents were evaluated in 11 cultivars of Argentinian roses of different colors. HPLC-ESI-QTOF/MS was used to identify the components where ellagic and quinic acids, quercetin, and kaempferol glycosylated derivatives were found. The phenolic contents ranged from 78.8 ± 3.2 to 203.4 ± 3.1 mg GAE/g dw, the flavonoid content ranged from 19.1 ± 3.8 to 125.9 ± 6.5 mg QE/g dw, and the anthocyanin content ranged from less than 0.01 to 5.8 ± 0.1 mg CE/g dw. The dark red cultivars exhibited the greatest levels of the analyzed compounds and of the antioxidant activities, even higher than those of certain plants known for their high phenolic contents and antioxidant activity. Moreover, the addition of these extracts decreased the population of L. innocua and P. aeruginosa to undetectable levels 24 h after inoculation. Rose petal extracts, mainly those with a dark red color, can be used as natural additives in food, feed, and cosmetics, as they contain a high proportion of bioactive compounds with antioxidant and antimicrobial effects.

Steroids that take part in the pathways of human steroidogenesis are involved in many biological mechanisms where they interact with calcium. In the present work, the binding selectivities and affinities for calcium of progestagens, mineralocorticoids, androstagens, and estrogens were studied by Electrospray Ionization-Mass Spectrometry (ESI-MS). The adduct profile of each steroid was characterized by high resolution and tandem mass spectrometry. The relative stability of the most important adducts was studied by threshold collision induced dissociation, E1/2. Doubly-charged steroid-calcium complexes [nM + Ca]2+ with n = 1-6 were predominant in the mass spectra. The adduct [5M + Ca]2+ was the base peak for most 3-keto-steroids, while ligands bearing hindered ketones or α-hydroxy-ketones also yielded [nM + Ca + mH2O]2+ with n = 3-4 and m = 0-1. Principal component analysis allowed us to spot the main differences and similarities in the binding behavior of these steroids. The isomers testosterone and dehydroepiandrosterone, androstanolone and epiandrosterone, and 17-α-hydroxyprogesterone and 11-deoxycorticosterone showed remarkable differences in their adduct profiles. Computational modeling of representative adducts was performed by density functional theory methods. The possible binding modes at low and high numbers of steroid ligands were determined by calcium Gas Phase Affinity, and through modeling of the complexes and comparison of their relative stabilities, in agreement with the experimental results.

The host-microbiome associations occurring on the skin of vertebrates significantly influence hosts' health. However, the factors mediating their interactions remain largely unknown. Herein, we used integrated technical and ecological frameworks to investigate the skin metabolites sustaining a beneficial symbiosis between tree frogs and bacteria. We characterize macrocyclic acylcarnitines as the major metabolites secreted by the frogs' skin and trace their origin to an enzymatic unbalance of carnitine palmitoyltransferases. We found that these compounds colocalize with bacteria on the skin surface and are mostly represented by members of the Pseudomonas community. We showed that Pseudomonas sp. MPFS isolated from frogs' skin can exploit acylcarnitines as its sole carbon and nitrogen source, and this metabolic capability is widespread in Pseudomonas. We summarize frogs' multiple mechanisms to filter environmental bacteria and highlight that acylcarnitines likely evolved for another function but were co-opted to provide nutritional benefits to the symbionts.

The effect of freeze and hot air drying methods on the retention of total phenolics, antioxidant activity (AA), and color of different cultivars of rose petals was analyzed. Both methods similarly preserved the phenolic content and AA, while freeze drying showed better red color retention. Furthermore, the conditions of total phenolics and AA extraction from two rose cultivars, Lovely Red and Malu, were optimized by response surface methodology through a Box-Behnken design. The solvent exhibited a major effect on the total phenolic content (TPC) and AA. The selected parameters were ethanol 38%, 75 °C, and 30 min. Under these conditions, the predicted values for Lovely Red were 189.3 mg GA/g dw (TPC) and 535.6 mg Trolox/g dw (AA), and those for Malu were 108.5 mg GA/g dw (TPC) and 320.7 mg Trolox/g dw (AA). The experimental values were close to the predicted values, demonstrating the suitability of the model. Ultrasound-assisted extraction increased the AA of the extracts but not the TPC. Fifteen compounds were identified in the Lovely Red cultivar, with no differences between the two drying methods. The results obtained suggest that the analyzed cultivars, particularly the red ones, can be considered a natural source of powerful antioxidant compounds.

Chemical investigation of the extract of the macroscopic fungus Beenakia informis led to the isolation of a previously unreported γ-pyrone and two new isoprenylated cyclohexanoids, together with speciocin N. Their structures were elucidated spectroscopically and the absolute configuration was determined by comparison of the experimental vs. calculated ECD curves. Three of the compounds showed very good to moderate activity against phytopathogenic fungi.

Amphibians have a great diversity of bioactive peptides in their skin. The cDNA prepro-peptide sequencing allowed the identification of five novel mature peptides expressed in the skin of Boana pulchella, four with similar sequences to hylin peptides having a cationic amphipathic-helical structure. Whole mature peptides and some of their fragments were chemically-synthesized and tested against Gram-positive and Gram-negative bacterial strains. The mature peptide hylin-Pul3 was the most active, with a MIC= 14 µM against Staphylococcus aureus. Circular dichroism assays indicated that peptides are mostly unstructured in buffer solutions. Still, adding large unilamellar vesicles composed of dimyristoyl phosphatidylcholine and dimyristoylphosphatidylglycerol increased the α-helix content of novel hylins. These results demonstrate the strong influence of the environment on peptide conformation and highlight its significance while addressing the pharmacology of peptides and their biological function in frogs.

The Antarctic fungus Cadophora malorum produces previously undescribed cyclic heptapeptides (cadophorin A and B) containing an anthranilic acid residue. The planar structure of these peptides was determined by high-resolution mass spectrometry combined with extensive 1D and 2D NMR spectroscopy. The absolute configuration of the amino acids was determined by Marfey's method, with HPLC analysis of FDVA (Nα-(2,4-dinitro-5-fluorphenyl)-L-valinamide) derivatives making use of a PFP column. Remarkably, cadophorin 2 possesses both the uncommon D-Ile and D-allo-Ile in its structure. The peptides have metal binding properties as shown by LCMS with post column addition of metal salt solutions. These results were supported by DFT calculations.

ABSTRACT Objective: To evaluate and compare anatomical measurements of C7, T1 and T2 vertebrae in children from 3 to 12 years of age to provide useful epidemiological data for determining the safe anatomical margin for transpedicular and translaminar fixation with screws in this population. Methods: This observational retrospective cross-sectional study evaluated 76 computed tomography scans obtained over 6 months, analyzing the following parameters: the angle of attack, length, thickness and diameter of the pedicle; and the angle of attack, length and thickness of the lamina. Results: The lamina length and thickness, as well as pedicle length varied in size according to age. Although the angle of attack was similar across different ages, age-dependent variation occurred in the T1 vertebra. Conclusion: Screws with a 3.5 mm diameter are safe to use in the C7 and T2 pedicles, while the T1 pedicle allows the introduction of larger screws ranging from 3.5-4.5 mm in diameter. In the lamina, 3.5 mm screws are safe for use only in children older than 7 years. However, each case should be analyzed individually, with the present study not aiming to replace the preoperative use of CT. Level of Evidence III, Retrospective comparative study.

Métodos: Estudo transversal retrospectivo observacional. Um total de 76 tomografias computadorizadas foram analisadas em um período de 6 meses. Os seguintes parâmetros foram analisados: ângulo de ataque, comprimento, espessura e diâmetro do pedículo, comprimento e espessura da lâmina. Resultados: O comprimento e espessura da lâmina bem como o comprimento do pedículo aumenta em tamanho conforme a idade. Enquanto o ângulo de ataque permanece estável conforme variação de idade; variação dependente da idade ocorre somente na vértebra T1. Conclusão: Parafusos com diâmetro de 3.5mm podem ser inseridos de maneira segura nos pedículos de C7 e T2. Já no pedículo de T1 pode-se inserir parafusos com medidas de 3.5 a 4.5mm de diâmetro de maneira segura. Na lâmina, parafusos de 3.5mm podem ser usados de maneira segura somente em crianças maiores de 7 anos. No entanto, cada caso deve ser analisado de maneira individualizada, e o presente estudo não objetivo substituir o uso de tomografia computadorizada no pré-operatório. OBJECTIVES: To determine the rate of and main risk factors for postoperative infection in cancer patients who underwent spine surgery in the last 5 years in order to determine whether there is an association between postoperative infection and increased mortality during hospitalization. METHODS: All cancer patients who underwent surgical procedures between January 2015 and December 2019 at a single hospital specializing in spine cancer surgery were analyzed. The primary outcome of interest was postoperative infection. Bivariate logistic regression was used to estimate the odds ratio and 95% confidence interval for each variable in relation to the occurrence of infection. RESULTS: We evaluated 324 patients, including 176 men (54.3%) and 148 women (45.7%) with a mean age of 56 years. The incidence of postoperative infection was 20.37%. Of the 324 patients, 39 died during hospitalization (12%). CONCLUSIONS: Surgical time greater than 4 hours, surgical instrumented levels greater than 6, and an Eastern Cooperative Oncology Group of 3 or 4 were associated with an increased risk of postoperative infection, but these factors did not lead to an increase in mortality during hospitalization.

In this work, we describe how stereochemically complex polycyclic compounds can be generated by applying a synthetic sequence comprising an intramolecular Ugi reaction followed by a Pictet-Spengler cyclization on steroid-derived scaffolds. The resulting compounds, which combine a fragment derived from a natural product and a scaffold not found in nature. are both structurally distinct and globally similar to natural products at the same time, and interrogate an alternative region of the chemical space. One of the new compounds showed significant antiproliferative activity on HepG2 cells through a caspase-independent cell-death mechanism, an appealing feature when new antitumor compounds are searched.

RESUMO

Five new lanostanoid triterpenes were isolated from the extract of R. microporus. Three of the metabolites (1-3) present a Δ8,9 skeleton with an uncommon keto functionality at C-1. Another compound (4) has an unprecedented rearranged skeleton in which methyl-19 was transposed to C-1, with conjugated double bonds at Δ1-10 and Δ8-9. All of the compounds have hydroxylated or furane-cyclized side-chains. The structures were elucidated by spectroscopic methods, and the absolute configuration of the hydroxyl-bearing carbon in the side chain of compound 5 was established in silico. The metabolites were evaluated for their antifungal activity and the bioactivity as agonist/antagonists of the liver X receptors (LXRs). Compound 4 presents antifungal activity and compounds 3 and 5 are the agonists of LXRs.

INTRODUCTION: An efficient characterisation of metabolites is a crucial task in many aspects of basic research, such as the de-replication of crude extracts in natural products chemistry or the tentative identification of compounds in untargeted metabolomics. OBJECTIVE: The goal of this work is the evaluation of the reaction with phenylhydrazine for the derivatisation post-column in situ of carbonyl-containing compounds in liquid chromatography-mass spectrometry (LC-MS). MATERIALS AND METHODS: LC-MS was performed using electrospray, Atmospheric Pressure Chemical Ionisation (APCI) or Atmospheric Pressure Photoionization (APPI) as ionisation techniques. The post-column addition of phenylhydrazine was done through a syringe pump via a T-junction before entrance to the ion source. RESULTS: A variety of natural products having carbonyl groups, such as cycloartanes, steroids, cardenolides and other terpenoids, were analysed by this method. In the case of compounds with non-hindered aldehyde or keto groups, the main signals of the mass spectra were those corresponding to the phenylhydrazones. However, the spectra of compounds with hindered carbonyl groups displayed mainly those signals corresponding to the product of the nucleophilic addition adduct of phenylhydrazine to the carbonyl, which is the first step of the derivatisation process. Finally, those compounds with conjugated ketones did not react with phenylhydrazine. This methodology was applied in the analysis of crude natural extracts. CONCLUSION: The results show that in situ derivatisation of carbonyl compounds in the ionisation source was achieved, yielding the typical derivatives of carbonyl compounds with phenylhydrazine.

El 81,3% de una encuesta realizada a 96 ortodoncistas y ortopedistas calificados y técnicos radiólogos consultados informan que ubican la cabeza del paciente en una posición ideal, subjetiva del profesional instruido que lo asiste, ya que muchas de sus fotografías y telerradiografías no son posiciones naturales de la cabeza genuinas. Cryptococcus species are responsible for important systemic mycosis and are estimated to cause millions of new cases annually. The available therapy is limited due to the high toxicity and the increasing rates of yeast resistance to antifungal drugs. Popularly known as "sucará," Xylosma prockia (Turcz.) Turcz. (Salicaceae) is a native plant from Brazil with little information on its pharmacological potential. In this work, we evaluated in vitro anticryptococcal effects of the leaf ethanolic extract of X. prockia and its fractions against Cryptococcus gattii and Cryptococcus neoformans. We also evaluated phenotypic alterations caused by ethyl acetate fraction (EAF) (chosen according to its biological results). The liquid chromatography-mass spectrometry (LC-MS) analysis of EAF demonstrated the presence of phenolic metabolites that belong to three structurally related groups as majority compounds: caffeoylquinic acid, coumaroyl-glucoside, and caffeoyl-glucoside/deoxyhexosyl-caffeoyl glucoside derivatives. The minimum inhibitory concentration (MIC) values against C. gattii and C. neoformans ranged from 8 to 64 mg/L and from 0.5 to 8 mg/L, for ethanolic extract and EAF, respectively. The EAF triggered an oxidative burst and promoted lipid peroxidation. EAF also induced a reduction of ergosterol content in the pathogen cell membrane. These effects were not associated with alterations in the cell surface charge or in the thermodynamic fingerprint of the molecular interaction between EAF and the yeasts evaluated. Cytotoxic experiments with peripheral blood mononuclear cells (PBMCs) demonstrated that EAF was more selective for yeasts than was PBMCs. The results may provide evidence that X. prockia leaf extract might indeed be a potential source of antifungal agents.

Peptides from skin secretions of amphibians are considered important components of their immune system and also play a relevant role in their defense mechanism against predators. Herein, by using mass spectrometry (MS), we characterize the sequence of 13 peptides from the gland secretion of the hylid tree frog, Boana punctata. Using in situ matrix-assisted laser desorption ionization imaging MS of a transverse section of the skin tissue, we show that some peptides are stored as longer molecules that are cleaved after being secreted, whereas others do not undergo any modification. Sequence comparison with peptides from other Boana species and analysis of the three-dimensional theoretical structure indicate that this cleavage depends on both the presence of a specific sequence motif and the secondary structure. The fact that peptides undergo a rapid cleavage upon secretion suggests that stored and secreted peptides may have distinct roles for anuran survival, including defense against pathogens and predators.

Secondary metabolites from the cultures of the dark septate fungal endophyte (DSE) Drechslera sp., isolated from the roots of rye grass (Lollium sp.) and cultured under different experimental conditions, are described here for the first time. The use of suberoylanilidehydroxamic acid (SAHA) and other histone deacetylase inhibitors as epigenetic modifiers in the culture medium was evaluated by LC/MS and LC/MS/MS. Several differences in the metabolite production were detected by means of supervised principal component analysis (PCA) of LC/MS data. The presence of the compounds in the culture medium or in the mycelium was compared. In order to confirm their structure, many of these natural products were isolated from a larger scale culture. These metabolites were characterized as prenylhydroxybenzoic acids and chromans, two compounds, one of each class were previously undescribed, prenylquinoids, diketopiperazines and macrosphelides. Some of the compounds, which were released to the medium, showed good antifungal activity, suggesting that these compounds could protect Lollium from fungal phytopatogens. The use of SAHA as an additive of the cultures also induced the release of hexosylphytosphyngosine to the culture medium. The biotransformation of the inhibitors was observed in addition to the production of antifungal metabolites, showing the ability of this endophytic strain to control xenobiotics.

Over the last decades, much effort has been devoted to the design of the "ideal" library for screening, the most promising strategies being those which draw inspiration from biogenic compounds, as the aim is to add biological relevance to such libraries. On the other hand, there is a growing understanding of the role that molecular complexity plays in the discovery of new bioactive small molecules. Nevertheless, the introduction of molecular complexity must be balanced with synthetic accessibility. In this work, we show that both concepts can be efficiently merged-in a minimalist way-by using very simple guidelines during the design process along with the application of multicomponent reactions as key steps in the synthetic process. Natural phenanthrenoids, a class of plant aromatic metabolites, served as inspiration for the synthesis of a library in which complexity-enhancing features were introduced in few steps using multicomponent reactions. These resulting chemical entities were not only more complex than the parent natural products, but also interrogated an alternative region of the chemical space, which led to an outstanding hit rate in an antiproliferative assay: four out of twenty-six compounds showed in vitro activity, one of them being more potent than the clinically useful drug 5-fluorouracil.

Boeckella poopoensis Marsh, 1906 es uno de los copépodos más frecuentes en el zooplancton de los lagos salinos sudamericanos, donde generalmente es la especie que más contribuye a la biomasa de la comunidad zooplanctónica. A pesar de su amplia distribución geográfica y su importancia ecológica, las características que distinguen sus estadios postembrionarios no son conocidas, lo que no permite conocer detalladamente la estructura y dinámica de sus poblacionales naturales. El objetivo de este trabajo fue describir la morfología de los estadios postembrionarios y compararlos con los de otras especies de Centropagidae. Los especímenes estudiados fueron obtenidos de un cultivo monoespecífico aclimatado en laboratorio. El linaje proviene del lago El Carancho (65°03W, 37º27S). La identificación de los estadios se hizo de acuerdo a los caracteres establecidos para los copépodos calanoideos. En base a todos los estados de desarrollo, algunas de las características que distinguen a B. poopoensis de otras especies del género son: lóbulos de los nauplios con el mismo número de sedas de igual longitud; la segmentación del cuerpo se inicia en el estadio de nauplio III; el dimorfismo sexual empieza a notarse a partir del copepodito IV (aunque sólo es evidente en la pata V); el endopodito de la pata V de los copepoditos V machos y hembras es biarticulado pero, mientras las hembras adquieren sedas con respecto al estadio anterior, los machos las pierden, llegando a ser glabras como en los adultos. Los copepoditos V no presentan características típicas de los adultos, tales como, el ala toracal izquierda más desarrollada en las hembras y la antena derecha geniculada en los machos. Todos los estadios de B. poopoensis son más grandes que los correspondientes a otros centropágidos y los estadios inmaduros muestran su propio patrón de sedas, con un número ligeramente menor que el de otras especies de la familia.


Boeckella poopoensis Marsh, 1906 is one of the most common copepods in the saline lakes of South America, where generally is the species that greater contributes to the zooplankton community biomass. Despite of its wide geographic distribution and ecological importance, the characteristics of the postembryonic stages are unknown, which prevents a detailed understanding of the structure and dynamics of natural populations. The objective of this work was to describe the morphology of the postembryonic stages and to compare it with other species of Centropagidae. The specimens were obtained from a monospecific culture acclimatized in the laboratory. The lineage comes from El Carancho shallow lake (65°03W, 37°27S). The identification of the postembryonic stages was performed according to the characteristics established for copepod calanoids. Based in all developing stages, some characteristics that distinguish B. poopoensis from another species of the genus are: lobes of nauplii with the same number of setae of equal length; beginning of body segmentation from nauplii III; sexual dimorphism from copepodite IV (although only evident in the V leg); the endopod of the V leg of copepodites V males and females are biarticulate, but while females endopod add setae respect to the previous stage, males endopod lose them and remain glabrous as in the adults. Copepodites V do not present characteristics typical of adults, such as, the most developed thoracic left wing of females and in the geniculate right antenna of males. All stages of B. poopoensis are larger than those corresponding to other species of centropagids, and in the immature stages, the appendages show their own patterns of setae and have a slightly smaller number of setae than that of the other species of the family.
