Gastrointestinal dysbiosis is a disturbance in mucosal homeostasis, producing low-grade chronic intestinal inflammation and impaired intestinal barrier function. It is induced by several factors, including nutrition and stress, which are both significant factors when considering current broiler breeder practices. A great grandparent (GGP) chicken meat line was identified displaying clinical signs characteristic of potential dysbiosis, including wet droppings and litter, in addition to reduced reproductive performance when compared to a consistently high performing line. This study aimed to determine whether the reduced reproductive performance observed in these hens was a result of dysbiosis and whether dietary supplementation with a Saccharomyces cerevisiae (SC) fermentation product would alleviate clinical signs. Dietary inclusion of SC did not influence intestinal permeability, WBC differentials, or corticosterone concentration in either the wet litter (WL) or high-performing (HP) breeder lines. Compared to hens from the HP line, WL line hens had a significant increase in intestinal permeability at 26 wk (onset of lay). WL hen heterophil counts were increased markedly at week 26 before declining. At weeks 26, 32, and 37 there were also significant increases in monocytes. Higher plasma corticosterone was also observed in WL hens at 37 wk. No significant differences in heterophil to lymphocyte (H:L) ratios or feather corticosterone were observed between lines. Dietary inclusion of SC supplementation to breeder diets had some benefit in regards to reducing hen mortality, improving egg production and hatchability but only in the WL line. Results from this study did not indicate that hens from the wet litter line were experiencing gut dysbiosis. Chronic intestinal inflammation may be a possible reason for the increase in intestinal permeability. These results do indicate that both breeder lines may be exhibiting physiological stress. Future investigation into the physiology and behavior around point of lay is required to find novel strategies to alleviate this stress and in turn, potentially improve welfare and production outcomes.

Current interventions targeting antimicrobial resistance (AMR), a major impact on commercial pork production, focus on reducing the emergence of AMR by minimising antimicrobial usage through antimicrobial stewardship and a range of alternative control methods. Although these strategies require continued advancement, strategies that directly aim to reduce or eliminate existing antimicrobial resistant bacteria, specifically bacteria resistant to critically important antimicrobials (CIAs), need to be investigated and established. This study established an in vivo model for examining the effects of postbiotics, in the form of Lactobacillus acidophilus fermentation products (LFP) and Saccharomyces cerevisiae fermentation products (SFP), on the shedding of extended-spectrum cephalosporin (ESC)-resistant E. coli. The model was successful in demonstrating the presence of ESC-resistant E. coli as evidenced by its detection in 62 of 64 pigs. There was a strong trend (p = 0.065) for the SFP postbiotics to reduce the shedding of ESC-resistant E. coli, indicating positive impacts of this additive on reducing the carriage of bacteria resistant to CIAs. Overall, this in vivo model enables future evaluation of strategies targeting ESC-resistant E. coli while increasing our knowledge on the carriage of ESC-resistant E. coli in pigs.

Younger broiler breeder flocks produce smaller eggs containing smaller yolks, with potentially lower energy reserves for the developing chick. Creatine is a naturally occurring energy source and is abundant in metabolically active tissues; providing this to chicks in ovo should provide additional energy to improve hatchability and post-hatch growth. Thus, post-hatch performance of male and female chicks hatched from younger breeder flocks supplemented with creatine monohydrate (CrM) in ovo was investigated. Four hundred eggs from Ross 308 breeder hens aged 27 to 29 wk were collected and at d 14 assigned to a treatment group and received 1) no injection, 2) 0.75% saline injection, or 3) 8.16 mg creatine monohydrate in 0.75% saline. At hatch 72 birds (24/treatment) were euthanized and BW, breast muscle, heart and liver weight were obtained, and breast muscle tissue was placed in 10% buffered formalin. Birds were then placed in raised metal pens (24 pens; 10-11 birds/pen; 8 replicates/treatment) and grown to d 42 with BW and pen feed intake measured once a week. At d 42, ninty-six birds were euthanized (2 male and 2 female/pen) and the process occurred as at hatch. Body composition was obtained for 48 birds (2/pen; 1 male,1 female) with a dual energy X-ray absorptiometry (DXA) scanner. Breast muscle tissue was processed for histological analysis and breast muscle fiber parameters were analyzed by ImageJ. While not statistically significant, the CrM treatment group saw an improved hatch rate (CrM: 93.5%, Saline: 88.6%, Control: 88.8%) and reduced early post hatch mortality. Chicks given in ovo CrM had significantly increased creatine concentrations in both liver and heart tissue at hatch compared to those in the saline and control groups. BW, BW gain, and final body composition parameters were not statistically different between treatments and in ovo CrM did not affect breast muscle fiber number or area. The creatine injection likely improved the energy status of the growing embryo resulting in the improved hatch rate but leaving little reserves for post-hatch growth.

Guanidinoacetic acid (GAA) is the direct precursor of creatine and can spare arginine (Arg) for creatine synthesis in low crude protein (CP) broiler diets. This study aimed to determine the extent GAA could spare Arg in broilers offered low CP diets and if supplemental betaine provides additional benefits. Seven hundred twenty-day-old Ross 308 male broilers were assigned into 9 dietary treatments with 8 replicates of 10 birds each. The treatments were; normal CP diet, a low CP (-15 g/kg) diet deficient in Arg, a low CP diet sufficient in Arg, and low CP diets with GAA, where 0.1% added L-Arg was spared by GAA at 50, 100, and 150% with and without 0.1% betaine. The treatments were offered during grower (d 10-24) and finisher (d 25-42) phases. The birds offered a low CP Arg deficient diet had 7.8% lower weight gain, 10 points higher FCR, 8.5% lower breast meat yield, 27.2% lower breast meat creatine concentration and 30.4% more abdominal fat pad compared to those offered a normal CP diet. When Arg was added back to the Arg deficient diet, growth performance, breast meat yield and creatine concentration loss were restored. When GAA spared Arg at 150%, feed intake, weight gain, FCR, breast and abdominal fat yields, breast meat moisture, drip loss, and breast meat creatine concentration became comparable to Arg sufficient low CP and normal CP treatments. When GAA spared Arg at 100 and 50%, FCR was 3 and 5 points lower than the normal CP treatment. Breast meat creatine concentration was positively correlated to feed efficiency (r = 0.70, P < 0.001) and breast meat moisture (r = 0.33, P < 0.01), and negatively correlated to relative weight of abdominal fat (r = -0.37, P < 0.01) and breast meat pH (r = -0.49, P < 0.001). There were no benefits of adding betaine with GAA on the parameters measured but the results with GAA were consistent in the presence or absence of betaine.

Broiler chickens grow rapidly, and their nutrient requirements change daily. However, broilers are fed three to five diet phases, meaning nutrients are under or oversupplied throughout production. Increasing diet phases improves production efficiency as there is less time in the production cycle that nutrients are in under or over-supply. Nevertheless, the process of administering four or more diets is costly and often impractical. New technologies are now available to blend feed to match the daily nutrient requirements of broilers. Thus, the aim of this review is to evaluate previous studies measuring the impact of increasing feed phases on nutrient utilisation and growth performance, and review recent studies taking this concept to the extreme; precision nutrition - feeding a new diet for each day of the production cycle. This review will also discuss how modern precision feeding technologies have been utilised and the potential that new technologies may bring to the poultry industry. The development of a precision nutrition regime which targets daily requirements by blending dietary components on farm is anticipated to improve the efficiency of production, reduce production cost and therefore improve sustainability of the industry. There is also potential for precision feeding technology along with precision nutrition strategies to deliver a plethora of other management and economic benefits. These include increased fluidity to cope with sudden environmental or market changes, and the ability to alter diets on a farm by farm level in a large, integrated operation. Thus, the future possibilities and practical implications for such technologies to generate a paradigm shift in feed formulation within the poultry industry to meet the rising demand for animal protein is also discussed.

Increased intestinal permeability (IP) and inflammation are both linked with functionality of the intestinal barrier and in particular enterocytes. Currently, almost all assessment methods of the intestinal barrier function are invasive. The present study aimed to quantify selected proteins as novel biomarkers in excreta of broiler chickens to facilitate non-invasive assessment of gut barrier function using enzyme-linked immunosorbent assays (ELISA). It was further hypothesised that probiotics as feed additives may counteract gut barrier dysfunction. A 3 × 2 factorial arrangement of treatments was used with the main factors being gut barrier dysfunction models (control, rye-based diet, and dexamethasone-DEX) with and without probiotic supplementation (a three-strain Bacillus) using 72 male Ross 308 day-old chickens. Each of the 6 experimental treatments was replicated 12 times. On d 21 of age, fluorescein isothiocyanate dextran (FITC-d) uptake into serum was examined to test IP. Fresh excreta samples were collected on d 20. The biomarkers included alpha-1 antitrypsin (A1AT), intestinal fatty acid binding protein (I-FABP), lipocalin-2 (LCN2), fibronectin (FN), intestinal alkaline phosphatase (IAP), ovotransferrin (OVT) and superoxide dismutase [Cu-Zn] (SOD1). Only DEX increased (P<0.001) FITC-d passage to the blood on d 21 of age, indicating a greater IP. The excreta concentrations of A1AT, I-FABP and SOD1 were unaltered by the experimental treatments. DEX increased (P<0.05) FN concentration in excreta compared with control birds. Conversely, inclusion of rye in the diet reduced (P<0.05) FN but increased (P<0.001) OVT in excreta. Independently, DEX decreased IAP (P<0.05) in excreta compared with control and rye-fed birds. The excreta concentration of LCN2 tended (P = 0.086) to increase in birds injected by DEX. There was no demonstrable effect of probiotic addition on any of the studied parameters. Among the tested biomarkers, FN, IAP, and LCN2 revealed promise as biomarkers of intestinal barrier function quantified by ELISA kits.

Global consumption of chicken meat has increased at a faster rate than any other animal protein source, and thus refinements in energy formulation techniques for feed have continued to gain importance. Formulation of animal feed based on net energy (NE) has been implemented in ruminants and pigs but not in poultry. A closed-circuit respiratory calorimetry system was employed on 25- to 28-day-old broilers fed 19 diets formulated with varying nutrient composition to produce equations to predict NE and apparent metabolizable energy (AME) efficiency of feed for broiler chickens. Performance, energy and N balance, respiratory quotient, and energy utilization were measured in the birds. Linear regression analysis was performed to generate prediction equations for dietary energy content and AME efficiency. The NE content was positively related to AME and ether extract, but negatively to crude protein. The study generated equations that can accurately predict NE, and NE/AME using AME value and chemical composition of feeds. The NE prediction equations were further validated on a separate set of diets with high correlation (r = 0.99) and accuracy. The outcomes are an important step for the broiler industry to adapt to an NE system in place of AME systems for the formulation of broiler chicken feeds following robust validation experiments.

This review is an outlook for sorghum as a feed grain for broiler chickens based on a survey of relevant stake-holders and recent research outcomes. Australian grain sorghum production will probably continue to generate a harvest in the order of 2.5 × 106 t of which some 7.9 × 105 t will be used as a feed grain for poultry and pigs. Feed grains are included primarily to provide energy from starch, but energy utilisation by broiler chickens offered sorghum-based diets is relatively inferior, because of incomplete starch digestion. Kafirin, the dominant protein fraction, 'non-tannin' phenolic compounds and phytate are 3 'starch extrinsic' factors in sorghum that compromise starch digestibility and energy utilisation in broiler chickens offered sorghum-based diets. Kafirin concentrations in 6 sorghum varieties were negatively correlated with metabolizable energy to gross energy (ME:GE) ratios (r = -0.891; P < 0.02) or the efficiency of energy utilisation in broiler chickens. Importantly, kafirin proportions of sorghum protein may be increasing with time in Australia. If so, this represents a fundamental challenge to sorghum breeders which presumably could be met by the development of sorghum varieties with different characteristics, especially in relation to the γ- and ß-kafirin fractions. White sorghum varieties contain lower polyphenol concentrations which should be advantageous as concentrations of total phenolic compounds were negatively correlated to ME:GE ratios (r = -0.838; P < 0.04) in 6 sorghum varieties. It would be desirable if more white varieties were to become available. It is suggested that responses to exogenous phytase in birds offered sorghum-based diets would be more robust if sorghum were to contain lower concentrations of kafirin and phenolic compounds. Paradoxically, while better sorghum varieties almost certainly could be developed, it may not necessarily follow that they will command a price premium from poultry and pig producers.

A total of ten experimental diets with protein concentrations ranging from 154 to 400 g/kg and two lipid levels (46 and 85 g/kg) with identical energy densities were offered to 240 male Ross 308 broilers from 7 to 28 d post-hatch. Growth performance was monitored and nutrient utilisation (apparent metabolisable energy (AME), N-corrected AME (AMEn), AME daily intake, AME:gross energy ratios, N retention) was determined. The weight gain response of broiler chickens to dietary protein concentrations in diets containing high and low lipid levels was diverse, with the relevant quadratic regressions being significantly different (P0·05). AMEn was also linearly (P<0·0001) increased with dietary protein concentrations but regressions in diets with low and high lipid content were significantly different (P<0·03). Carcass protein content increased linearly with dietary protein content in diets containing high lipid concentrations (r 0·933, P<0·0001); by contrast, this relationship was quadratic (R 2=0·93, P<0·0001) in diets with low lipid levels. In conclusion, predictably, the effects of dietary protein concentrations on broiler performance were profound; however, the impact of dietary protein on performance in broiler chickens was modified by dietary lipid concentrations.

The right-angled triangle mixture experiment was designed to include fourteen diets with different concentrations of starch, protein and lipid. Experimental diets were offered to male Ross 308 broiler chickens from 10 to 23 d after hatching, and response curves and surfaces were generated to illustrate the influence of macronutrients on growth performance and nutrient utilisations. Despite the primary function of macronutrients, especially protein, may not be providing energy, macronutrients were expressed as energy derived from starch, protein and fat for statistical purposes in the mixture design. Energy derived from lipid had a greater impact on feed intake than energy derived from starch and protein. When we compared the influence of starch and protein on feed intake, 'equal distance rule' was observed, which means the animal consumes feed to the point on its respective nutritional rails where the shortage of starch exactly equals the surplus of consumed protein. Increasing the protein-derived energy intake increased weight gain in broiler chickens, whereas energy intake derived from starch and lipid had little impact on weight gain. Feed conversion ratio (FCR) may be reduced by either increasing protein energy intake or decreasing starch energy intake. As the slope of the contours was less than 1, the influence of starch energy intakes on FCR exceeded that of protein energy intakes. In conclusion, energy derived from protein is more important than non-protein energy in terms of weight gain, and a balance between protein and energy supplies is required for efficient muscle protein deposition.

We investigated the pattern of non-starch polysaccharide (NSP) digestion along the gut of pigs fed two different wheats, which were offered with or without xylanase supplementation. The two wheats used were pre-characterised before the experiment on the basis of low and normal feed intake of young pigs. Wheat type significantly influenced feed intake and growth rate in the first 7 days, however, by day 14 the only significant effect of wheat type was on growth rate. Xylanase supplementation increased the growth performance of pigs fed the poor quality wheat to a level similar to those fed the normal wheat. It also increased the daily gain of pigs fed the normal wheat. Wheat type had no significant effect on the digestibility of dry matter (DM), energy, free sugars or the different fractions of NSP in the duodenum, ileum or in the faeces. The duodenal gross energy digestibility values for the low and high performance diets were -27.4 and -47.5%, respectively, and xylanase supplementation significantly increased the digestibility of energy back to positive levels. Dry matter digestibility values followed a similar pattern. In the duodenum, xylanase increased (P < 0.05) the digestibility values of both soluble and insoluble NSP, whereas in the ileum, xylanase had a significant effect only on the digestibility of the soluble NSP fraction. Xylanase did not affect free sugar digestibility. The reduction in soluble NSP level coincided with a marked reduction in the amount of fucose, a prominent component of mucosal polysaccharides. This suggests that soluble NSP substantially increase endogenous losses. The absence of differences in the digestibility of the measured NSP between the two wheat samples suggests that the structures of the NSP, rather than just their amount and solubility, are important for the anti-nutritional properties of NSP in pig diets.

The European Centre for Ecotoxicology and Toxicology of Chemicals (ECETOC) convened a workshop in June 2005 to address the speculation that exposure to specific chemicals, and/or chemical pollutants in general, may play an important role in the increased prevalence of allergy and asthma in 'westernized' societies. This paper summarises one perspective arrived at during this workshop. It was acknowledged that certain chemicals and certain types of pollution might trigger or exacerbate asthmatic reactions in sensitised subjects. However, overall levels of pollution appear not to have had a major impact upon the prevalence of atopic allergy. Epidemiological studies suggest that pollution may in some circumstances protect from acquisition of sensitisation. Increasing exposure to household chemicals may enhance pre-existing allergies, but evidence for their causation of allergy is lacking. Other risk factors considered included societal dietary changes and exposure to endotoxins. Future research needs were identified which included epidemiological studies employing exposure and biomonitoring data, studies on domestic exposure to chemicals and their association with the incidence of allergy and asthma, and prospective birth cohort studies employing well-defined aspects of lifestyle, diet, chemical and endotoxin exposure as factors that may drive susceptibility to allergy and asthma.
