RESUMO

Objective To explore the value of mast cell tryptase and brain natriuretic peptide(BNP)in the differential diagnostic of sudden death due to hypersensitivity and coronary atherosclerotic heart dis-ease.Methods Totally 30 myocardial samples were collected from the autopsy cases in the Department of Forensic Pathology, Shanxi Medical University during 2010—2015. All samples were divided into three groups:death of craniocerebral injury group, sudden death of hypersensitivity group and sudden death of coronary atherosclerotic heart disease group, 10 cases in each group. Mast cell tryptase and BNP in myocardium were detected by immunofluorescence staining and Western Blotting.Results Immunofluo-rescence staining showed that the positive staining mast cell tryptase appeared in myocardium of sudden death of hypersensitivity group and coronary atherosclerotic heart disease group. Among the three groups, the expression of mast cell tryptase showed significantly differences through pairwise comparison(P0.05).Conclusion The combined detection of the mast cell tryptase and BNP in myocardium is expected to provide help for the forensic differential diagnosis of sudden death due to hypersensitivity and coronary atherosclerotic heart disease.

RESUMO

Objective To analyze the characteristics in the incarcerated inm ate’s death, investigate the m ain cause of death of the incarcerated inm ate and provide som e inform ation for forensic investigation. Methods The cases from the forensic m edical center of Shanxi Medical U niversity from 2005 to 2013 were selected. The statistical analysis w as perform ed by using the incarcerated inm ate’s gender, age, cause of death, m anner of death, and disease as the m arkers. Results There were 100 men, 5 w omen in the 105 incarcerated inm ates;the age range w as from 16 to 65 years;Inm ates were mostly died of nat-ural diseases, m ainly in the respiratory and cardiovascular diseases;the m ain unnatural death w as suicide w ith a rate of 54.5%. Conclusion Atpresent, most incarcerated inm ate’s death are due to natural dis-eases. The prison should im prove incarcerated inm ate’s lives, w ork and health care conditions, and strengthen supervision of law enforcement.

RESUMO

Objective In order to understand which kind of function genes play an important role for es-timating wound age, the variation of difference genes’ mRNA expression were compared after injury. Methods T he mRNA expression levels of seven candidate genes (ICAM-1, NF-κB, MX2, MT1, MT2, sTnI, and Cox6c) were analyzed in contused rat skeletal muscle at different time points using real-time fluorescent quantitative PC R (R T-qPC R ). T he rawC t values were normalized relative to that of RPL32 mRNA , and converted to standard C t values. A t each time point after injury, the standard deviations (SD ) of the standard C t values were calculated by SPSS. Results T he expression trends of the seven genes were all found to be related to wound age, but there were lower variation coefficients and greater reliability of sTnI and Cox6c when compared with other genes. Conclusion T he genes encoding struc-tural proteins or proteins that performbasic functions can be suitable for wound age estimation.

RESUMO

Objective To investigate the diagnostic significance of basophil activation test (B A T) in ana-phylaxis to non-ionic contrast media through testing the content of CD 63, m ast cell-carboxypeptidase A 3 (M C-CPA 3), and term inal com plem ent com plex SC5b-9 of the individuals by testing their levels in the norm al im m une group and the anaphylaxis groups to β-lactam drugs and non-ionic contrast media. Methods The CD 63 expression of basophilic granulocyte in blood w as detected by flow cytom etry. The levels of M C-CPA 3 in blood serum and SC5b-9 in blood plasm a w ere detected by ELISA . Results The CD 63 expression of basophilic granulocyte in blood, the levels of M C-CPA 3 and SC5b-9 of anaphylaxis to non-ionic contrast media and β-lactam drugs w ere significantly higher than that in norm al im m une group (P<0.05). Conclusion There is activation of basophilic granulocytes, m ast cells and com plem ent system in anaphylaxis to non-ionic contrast media. B A Tcan be used to diagnose the anaphylaxis to non-ionic contrast media.

RESUMO

Objective To observe the dynamic changes in plasma coagulation indexes in the rabbit's amniotic fluid embolism. Method The experimental amniotic embolism model was established by injecting the amniotic fluid or placental extract-amniotic fluid through the rabbit's marginal vein of the ear. Blood samples were taken from the heart before and after infusion. Fibrinogen (FIB), prothrombin time (PT), thrombin time (TT), and activated partial thromboplastin time (APTT) were measured with coagulation analyzer. RESUMO

Transmission electron microscopy, histochemstry, immunohistochemistry and morphometry were used to study ultrastructural changes, succinic dehydrogenase (SDH) activity and myoglobin (Mb )content of the rabbit's myocardium bleeding to death and to hemorrhagic shock. The results were as follows: the myocardium showed different degrees of ultrastructural changes; various degrees of decreasing of SDH activity and deletion of Mb. These changes gradually become more serious with lapse of time after shock. The mechanism of the changes and the significance in forensic medicine are discussed.

Objective Carry out the mast cell tryptase (MCT) granule count in throat, lung and small intestine mast cells in persons died of anaphylactic shock.Method Control group: 10 cases died from severe brain injury. Research group:15 cases died from anaphylactic shock(group A)and 8 cases died of amniotic fluid embolism (group B). H & E staining and immunohistochemical staining were employed and the MCT granule count performed on the throat, lung and small intestine tissue.Results There was marked increase of MCT granule count in the throat (average MCT-positive number of group A 28.48 and of group B 24.35),lung (average MCT-positive number of group A 46.98 and of group B 43.50) and small intestine (average MCT-positive number of group A 48.23 and of group B 42.72) in specimens of group A and group B as compared with the control group (average number in throat 7.97,lung 12.94 and small intestine 20.25).Conclusion Tryptase granular count of MCT increased in throat, lung and small intestine tissue in persons died from anaphylactic shock and amniotic fluid embolism.

RESUMO

Objective To investigate a method for postmortem diagnosis of the sudden death from hypersensitivity by detection of serum tryptase and IgE.Methods Venous blood samples were collected from 9 cases of sudden death from hypersensitivity,19 cases of death from other causes and 10 health living adult individuals.The serum levels of the mast cell tryptase(MCT)and IgE were detected with immunofluorescent enzyme assay and enzyme linked immunosorbent assay(ELISA)respectively.MCT in the lungs was detected by immunohistochemistical method.Results The serum level of both MCT and IgE in cases of sudden death from hypersensitivity were higher than those in health living adults and cases of death from other causes(P﹤0.01).However,there was significant difference(P﹥0.05)in the serum level of IgE and MCT between the health living individuals and cases of death from other cause.More cells containing MCT-positive granules were detected in the lung in cases of sudden death from hypersensitivity than in cases of death from other causes(P

Objective To explore the pathognomonic findings of anaphylactic shock in forensic medicine. Methods Animal model of anaphylactic shock was developed in guinea pigs. Substance P level in plasma was determined radioimmunoassay, and detected in laryngopharyneal tissue and the lungs by immunohistochemical SABC method followed by image analysis using BI - 2000 image analyzing system. Objective To observe the number of pulmonary mast cell tryptase(MCT) after amniotic fluid embolism(AFE) in experimental rabbits.Method The experimental amniotic fluid embolism rabbits model was established by injecting the amniotic fluid(AF) or placental extracts mixed with amniotic fluid(PAF)into the marginal vein of the pregnant rabbits.Immunohistochemistry and microscopy were employed to observe the number of pulmonary MCT in AFE rabbits.Results A marked increase in MCT positive cells was observed in lungs in both AF and PAF groups(average cell number were 32 98 and 45 53respectively)as compared with the control group(average cell number was 11 25).Conclusion The results indicated that the number of MCT cells were marked increased in case of the AFE.

RESUMO

To study the pathological process and renal patho morphology of crush syndromed.The rabbit model of crush injury crush syndrome was established.Transmission electron microscopy(TEM),biochemistry,radioimmunoassay and immuohistochemistry were employed.The results demonstrated that the serum K + and BUN increased,The serum CO 2CP decreased after crush injury.The degeneration and necrosis of the epithelial cells of the renal tubule and myoglobin casts in the renal tubules were observed in crush syndrome,the serum K + increase was absent.Our results may be regarded as criteria for the diagnosis of the crush injury crush syndrome.The serum K + increase was the cause of death in early stage after crush injury.
