188 surface soil samples were collected in Shenzhen of China to determine the occurrence and spatial differentiation of polycyclic aromatic hydrocarbons (PAHs), based on which we studied the correlation between PAHs concentrations and urbanization levels, as well as the PAHs ecological risk. The total concentrations of 28 PAHs (sigma28 PAHs), 16 EPA PAHs (sigma 16 PAHs) and 7 carcinogenic PAHs (sigma7 CarPAHs) ranged from 5 to 7939 ng x g(-1), 2 to 6745 ng x g(-1) and not detected to 3786 ng x g(-1), respectively. 8 kinds of land use types according to sigma16 PAHs average levels in descending order were: transportation lands, commercial lands, industrial lands, agricultural lands, residential lands, urban green space, orchards and woodland. And sigma16 PAHs of construction and non-construction lands samples were mainly derived from combustion of various fossil fuels with contribution of 75.1% and 68.2%, respectively. Significant positive correlation was also found between PAHs concentrations of high molecular weight and urbanization levels. And PAHs pollution in the top soils of Shenzhen was at a low-end level of the world.

Although an important role for mast cells in several viral infections has been demonstrated, its role in the invasion of highly pathogenic H5N1 influenza virus is unknown. In the present study, we demonstrate that mast cells were activated significantly by H5N1 virus (A/chicken/Henan/1/2004) infection both in vivo and in vitro. Mast cells could possibly intensify the lung injury that results from H5N1 infection by releasing proinflammatory mediators, including histamine, tryptase, and gamma interferon (IFN-γ). Lung lesions and apoptosis induced by H5N1 infection were reduced dramatically by treatment with ketotifen, which is a mast cell degranulation inhibitor. A combination of ketotifen and the neuraminidase inhibitor oseltamivir protected 100% of the mice from death postinfection. In conclusion, our data suggest that mast cells play a crucial role in the early stages of H5N1 influenza virus infection and provide a new approach to combat highly pathogenic influenza virus infection.

This study examined polybrominated diphenyl ethers (PBDEs) in central air conditioner filter (CACF) dust from a new office building in Shenzhen, China. Human exposure to PBDE via dust inhalation and ingestion were also estimated. PBDEs level in CACF dust was lower than those in the other countries and regions. Approximately 0.671 pg/kg bw/day PM(2.5) (Particulate Matter up to 2.5 µm in size) bounded Σ(15)PBDEs can be inhaled deep into the lungs and 4.123 pg/kg bw/day PM(10) (Particulate Matter up to 10 µm in size) bounded Σ(15)PBDEs tend to be deposited in the upper parts of the respiratory system. The average total intake of Σ(15)PBDEs via dust inhalation and ingestion for adults reached ∼ 141 pg/kg bw/day in this building. This value was far below the reference dose (RfD) recommended by United States Environmental Protection Agency. Human exposure to PBDEs via dust inhalation and ingestion in the new building is less than the old ones.

To determine the incidence of organochlorine pesticides (OCPs) in soil in a rapid urbanization region, soil samples from various land use types in Shenzhen were collected in winter, 2007. The concentration of dichlorodiphenyltrichloroethanes (DDTs) and hexachlorocyclohexanes (HCHs) ranged from non-detected to 149 ng g(-1) and 19 to 88 ng g(-1), respectively. The highest levels of OCPs were observed in soil from traffic and industry areas, reflecting that intensive human disturbance make the soil pollution accumulation more disperse. HCHs and DDTs profiles revealed that the sources were associated mainly with lindane and technical DDTs, respectively, while HCHs in the soil of Shenzhen might originate from both recent and historical sources. The loss of OCPs by soil erosion will enter surface runoff and impose impact on the water environment. Non-dietary exposure estimation indicates that children were the most sensitive group. The average daily exposure to OCPs for males was more serious than for females. Non-dietary exposure to DDTs and HCHs in residential blocks of Shenzhen were far below the acceptable daily intake recommended by the Food and Agriculture Organization/World Health Organization.

BACKGROUND: "Nubiotics" are synthetic oligonucleotides and nucleotides with nuclease-resistant backbones, and are fully protonated for enhanced ability to be taken up by bacterial cells. Nu-3 [butyl-phosphate-5'-thymidine-3'-phosphate-butyl], one of the family members of Nubiotics was efficacious in the treatment of burn-wound infections by Pseudomonas aeruginosa in mice. Subsequent studies revealed that Nu-3 had a favorable toxicological profile for use as a pharmaceutical agent. This study evaluated the antibacterial activity of Nu-3 in vitro and its efficacy as a topical antibiotic. In addition, we investigated the possible mechanisms of Nu-3 action at the levels of DNA synthesis and bacterial membrane changes. METHODS: Antimicrobial minimum inhibitory concentrations (MIC) experiments with Nu-3 and controls were measured against a range of Gram-positive and Gram-negative bacteria, including some hospital isolates according to Clinical and Laboratory Standards Institute (CLSI) guidelines. Analysis of the killing kinetics of Nu-3 was also performed against two strains (Staphylococcus aureus cvcc 2248 and Pseudomonas aeruginosa cvcc 5668). The mouse skin suture-wound infection model was used to evaluate the antibacterial activity of Nu-3. We used a 5-Bromo-2'-deoxy-uridine Labeling and Detection Kit III (Roche, Switzerland) to analyze DNA replication in bacteria according to the manufacturer's instruction. The BacLight™ Bacterial Membrane Potential Kit (Invitrogen) was used to measure the bacterial membrane potential in S. aureus. RESULTS: Nu-3 had a wide antibacterial spectrum to Gram-positive, Gram-negative and some resistant bacteria. The MIC values of Nu-3 against all tested MRSA and MSSA were roughly in a same range while MICs of Oxacillin and Vancomycin varied between the bacteria tested. In the mouse model of skin wound infection study, the treatment with 5% Nu-3 glycerine solution also showed comparable therapeutic effects to Ciprofloxacin Hydrochloride Ointment. While Nu-3 had no effect on DNA synthesis of the tested bacteria as demonstrated in a BrdU assay, it could cause bacterial cell membrane depolarization, as measured using a BacLight™ Bacterial Membrane Potential Kit. CONCLUSIONS: These results provide additional experimental data that are consistent with the hypothesis that Nu-3 represents a new class of antibacterial agents for treating topical infections and acts via a different mechanism from conventional antibiotics.

Our previous study indicated that the current level of polycyclic aromatic hydrocarbons (PAHs) in Shenzhen soil is in the low-end of world soil PAH pollution. In this study, the fate of PAHs in the soil of Shenzhen was investigated. The mass inventories of Σ(27)PAHs and Σ(15)PAHs (defined as the sum of the 27 or 15 PAH compounds sought) in topsoil of Shenzhen were ∼204 and ∼152 metric tons, respectively. Fate estimation of Σ(15)PAHs shows that air-soil gaseous exchange is the primary environmental process with ∼10,076 kg/year diffusing from soil to air. Rain washing (∼1131 kg/year from air to soil) is the most important input pathway followed by wet (∼17 kg/year) and dry deposition (∼8 kg/year) to soils in Shenzhen. The transport of Σ(15)PAHs by soil erosion is a crucial loss process for soil PAHs in Shenzhen (1918 kg/year for water runoff and 657 kg/year for solid runoff from soil). Moreover, degradation is not ignorable at present (95 kg/year). Comparison of inventory and residue (defined as Σ(15)PAHs left in topsoils after all environmental loss processes) suggested that input and loss of high molecular weight PAHs for Shenzhen's soil reached apparent equilibrium. Soil PAH pollution in Shenzhen will stay in a quasi-steady state for a long period and the natural environmental processes can not significantly reduce the pollution.

Nu-3 [butyl-phosphate-5'-thymidine-3'-phosphate-butyl] is a modified nucleotide that has been shown to have antimicrobial activity against a range of bacteria including Pseudomonas aeruginosa. However, data on the toxicological profile of Nu-3 are still lacking. In the present study, the toxicity of Nu-3 was evaluated by the following studies: acute oral toxicity, dermal and mucous membrane irritation, multiple-dose toxicity and genotoxicity in vivo and vitro. The acute oral toxicity test in mice showed that Nu-3 had an LD(50) of 2001 mg/kg body weight. The irritation tests on rats revealed that Nu-3 was not irritant, with an irritation scoring of 0. The multiple-dose toxicity study in rats showed that Nu-3 did not cause significant changes in histology, selected serum chemistry, and hematological parameters compared to the controls. Rats administrated with multiple-doses of Nu-3 showed no visible toxic symptoms. Both in vitro and in vivo, Nu-3 exhibited no notable genetic toxicity. Overall, the data suggest that Nu-3 is hypotoxic or nontoxic antimicrobial compound that warrants being further developed for treating Pseudomonas aeruginosa infection.

Twenty eight polycyclic aromatic hydrocarbons (PAHs) were determined for a total of 203 top soil samples collected from eight different land categories in Shenzhen, China during the winter of 2007. The concentrations, compositional profile, and the potential sources of PAHs in soil were discussed. Overall, PAH pollution in the soil of Shenzhen is still in the low-end of the world after 30 years rapid urbanization. Based on that, human exposure to soil PAHs via inhalation and soil particle intake were estimated as well. The concentrations of Sigma(28)PAHs in Shenzhen soil ranged from 2.68 to 17,767 ng g(-1) (average: 546 ng g(-1)). The highest levels of PAHs were found in the traffic land (average: 2104 ng g(-1)) and the lowest concentrations were detected in forest land (average: 144 ng g(-1)) in eight land categories. PAH isomeric ratios indicated that PAHs in Shenzhen soil were mainly derived from combustion. In the current pollution levels in Shenzhen, children aged 0 to 8 were the most sensitive sub-group of exposure to PAHs (59.2 ng (kg d)(-1)), and the exposure to PAHs for males (36.2 ng (kg d)(-1)) was more serious than for females (32.7 ng (kg d)(-1)). Inhalation was the major way of non-dietary exposure (over 96%).
