RESUMO

The extraction process, structural characterization and free radical scavenging ability of polysaccharides from Camellia oleifera have already been widely studied. However, the antioxidant activities are still lack of systematic experiments. In this study, we used Hep G2 cells and Caenorhabditis elegans to evaluate the antioxidant potential of polysaccharides that from C. oleifera flowers (P-CF), leaves (P-CL), seed cakes (P-CC) and fruit shells (P-CS). The results showed all these polysaccharides could protect cells from oxidative damage induced by t-BHP. The highest cell viabilities were 66.46 ± 1.36 % (P-CF), 55.2 ± 2.93 % (P-CL), 54.49 ± 1.29 % (P-CC) and 61.45 ± 1.67 % (P-CS), respectively. Studies have shown that four polysaccharides may protect cells from apoptosis by reducing ROS levels and maintaining MMP balance. Moreover, P-CF, P-CL, P-CC and P-CS increased the survival rate of C. elegans under thermal stress, which reduced the production of ROS by 56.1 ± 0.67 %, 59.37 ± 1.79 %, 16.63 ± 2.51 % and 27.55 ± 2.62 %, respectively. P-CF and P-CL showed stronger protective effects on C. elegans by increasing the nuclear entry rate of DAF-16 and stimulating the expression of SOD-3. Our study suggested that C. oleifera polysaccharides have the potential to develop into a natural supplement agent.

This work describes the first example of semicrystalline poly(thiocarbonate)s from carbon disulfide (CS2 ) and ethylene oxide (EO), two mass producible low-cost monomers. Lewis acid/base pairs (LPs) exhibit high activity (EO conversion up to >99%, 8 h) in catalyzing the copolymerization under low Lewis pair/monomer ratio of 1:1500. Oxygen-sulfur exchange reaction (O-S ER) during the copolymerization of CS2 and EO, the generation and mutual copolymerization with COS, CO2 , and episulfide, is harnessed to introduce crystallizable segments [SC(O)O and SC(S)S] in the copolymer. The type of Lewis base is found to have a great impact on the chain microstructure and the crystalline properties. The formed copolymers with melting point from 117.7 to 245.3 °C are obtained. The maximum crystallinity is estimated to be 78% based on the powder wide-angle X-ray diffraction pattern. This work provides a general method to prepare semicrystalline sulfur-containing polymers.

The most daunting challenge of solid polymer electrolytes (SPEs) is the development of materials with simultaneously high ionic conductivity and mechanical strength. Herein, SPEs of lithium bis-(trifluoromethanesulfonyl)imide (LiTFSI)-doped poly(propylene monothiocarbonate)-b-poly(ethylene oxide) (PPMTC-b-PEO) block copolymers (BCPs) with both blocks associating with Li+ ions are prepared. It is found that the PPMTC-b-PEO/LiTFSI electrolytes with double conductive phases exhibit much higher ionic conductivity (2 × 10-4 S cm-1 at r.t.) than the BCP electrolytes with a single conductive phase. Concurrently, the storage moduli of PPMTCn -b-PEO44 /LiTFSI electrolytes are ≈1-4 orders of magnitude higher than that of the neat PEO/LiTFSI electrolytes. Therefore, simultaneous improvement of ionic conductivity and mechanical properties is achieved by construction of a microphase-separated and disordered structure with double conductive phases.

The synthesis of poly(thioether), a highly desired sulfur-containing polymer, is still a key challenge. Herein, we report a simple and facile approach to poly(thioether)s by closed-system one-pot reaction of carbonyl sulfide (COS) and epoxides. This route underwent the coupling reaction of COS with epoxides, followed by decarboxylative ring-opening polymerization (ROP) of the generated mixed cyclic thiocarbonates with releasing of CO2 and a little bit of COS. Organic base was used as catalyst and initiator in the two steps, respectively. The oxygen/sulfur exchange reaction was driven by successive regioselective elementary reactions and spontaneous releasing of CO2 (COS), leading to the sulfur atom of COS transferring to poly(thioether)s, which was well demonstrated by DFT studies. This work provides an easy-to-handle, metal-free route to poly(thioether)s bearing diverse structures by using readily available chemicals.

A one-pot synthesis of block copolymer with regioregular poly(monothiocarbonate) block is described via metal-free catalysis. Lewis bases such as guanidine, quaternary onium salts, and Lewis acid triethyl borane (TEB) were equivalently combined and used as the catalysts. By using polyethylene glycol (PEG) as the macromolecular chain transfer agent (CTA), narrow polydispersity block copolymers were obtained from the copolymerization of carbonyl sulfide (COS) and propylene oxide (PO). The block copolymers had a poly(monothiocarbonate) block with perfect alternating degree and regioregularity. Unexpectedly, the addition of CTA to COS/PO copolymerization system could dramatically improve the turnover frequency (TOF) of PO (up to 240 h-1), higher than that of the copolymerization without CTA. In addition, the conversion of CTA could be up to 100% in most cases, as revealed by ¹H NMR spectra. Of consequence, the number-average molecular weights (Mns) of the resultant block copolymers could be regulated by varying the feed ratio of CTA to PO. Oxygen-sulfur exchange reaction (O/S ER), which can generate randomly distributed thiocarbonate and carbonate units, was effectively suppressed in all of the cases in the presence of CTA, even at 80 °C. This work presents a versatile method for synthesizing sulfur-containing block copolymers through a metal-free route, providing an array of new block copolymers.

Domesticated animals play an important role in the life of humanity. All these domesticated animals undergo same process, first domesticated from wild animals, then after long time natural and artificial selection, formed various breeds that adapted to the local environment and human needs. In this process, domestication, natural and artificial selection will leave the selection signal in the genome. The research on these selection signals can find functional genes directly, is one of the most important strategies in screening functional genes. The current studies of selection signal have been performed in pigs, chickens, cattle, sheep, goats, dogs and other domestic animals, and found a great deal of functional genes. This paper provided an overview of the types and the detected methods of selection signal, and outlined researches of selection signal in domestic animals, and discussed the key issues in selection signal analysis and its prospects.

GnRH sterilization vaccines have been developed for various practical and clinical reasons. However, conjugation of GnRH peptide to carrier protein has many drawbacks, hampering the further commercialization of GnRH vaccines. In this study, a new nonconjugated GnRH vaccine, D-Lys6-GnRH-tandem-dimer peptide (TDK), emulsified in Specol adjuvant was investigated for its immunocastration efficacy in young male rats. Prepubertal male rats were randomly allocated into three groups (n = 12): control (no treatment), surgically castrated or immunized against 100 µg TDK in Specol adjuvant at 6 weeks of age (with a booster 8 weeks later). Blood samples (for antibody titers and hormone concentrations) were collected at 2-week intervals until rats were killed (18 weeks of age). Compared to intact controls, active immunization against TDK reduced (P < 0.05) serum concentrations of testosterone, inhibin B, LH and FSH, prevented the onset of spermatogenesis at puberty. Furthermore, mRNA expressions of GnRH receptor, LH-ß and FSH-ß in the pituitary, LH receptor, FSH receptor, inhibin α, ßA and ßB subunit in the testes were decreased in immunocastrated rats compared to intact controls (P < 0.05). These results demonstrate for the first time that GnRH-tandem-dimer peptide emulsified in Specol is a promising veterinary sterilization medicine.

The purpose of the study was to construct mucosal vaccine of a recombinant Lactococcus lactis expressing PRRSV ORF6 gene and evaluate mucosal and systemic immune response against PRRSV in mice after intranasal immunization. The result show that the vaccine can stimulate mice to produce specific IgG in serum and remarkable special s-IgA in lung lavage fluid, at the same time, the contents of cytokines IL-2 and IFN-γ of the experimental group were significant higher than those of the control group (P < 0.01), however, the contents of cytokines IL-4 was not different to the all groups. In summary, the constructed mucosal vaccine can significantly induce mucosal immune, humoral immunity and cellular immunity involved Th1 type cytokines, which will lay a theoretical foundation on immune mechanism and new efficient vaccines for PRRSV.

We sought to determine the effects of active anti-GnRH immunization on GnRH synthesis in the hypothalamus. Adult male rats (n = 36) were randomly and equally allocated into three groups: Control (no treatment), surgically castrated, or immunized against 50 µg D-Lys6-GnRH-tandem-dimer peptide conjugated to ovalbumin in Specol adjuvant at 12 week of age (with a booster 8 week later). Blood samples (for antibody titers and hormone concentrations) were collected at 2-week intervals until rats were killed (20 week). Compared with intact controls, immunocastration reduced (P < 0.05) serum concentrations of testosterone, LH, and FSH, and GnRH content in the median eminence, reduced the weight of the hypohysis (P < 0.01), and induced testicular atrophy (suppression of spermatogenesis). Furthermore, mRNA expression of GnRH in the hypothalamus, GnRH receptor, LH-ß and FSH-ß in the pituitary, LH receptor and FSH receptor in the testes, and genes in sex steroid feedback loops (androgen receptor [AR], kisspeptin encoded gene (Kiss-1), and kisspeptin receptor (GPR54) in the hypothalamus were decreased in immunocastrated rats compared with intact controls (P < 0.05). Similarly, surgical castration reduced GnRH in the median eminence as well as mRNA expression of GnRH, AR, Kiss-1, and GPR54 in the hypothalamus (P < 0.05). We concluded that anti-GnRH immunization in adult rats reduced synthesis of hypothalamic GnRH by decreasing androgen-AR-Kisspeptin-GPR54 signaling pathways, and caused dysfunction of the pituitary-testicular axis, thereby suppressing spermatogenesis, resulting in testicular atrophy.
