OBJECTIVE: To investigate the effect of acupotomy, on mitophagy and the Pink1-Parkin pathway in chondrocytes from rabbits with knee osteoarthritis (KOA). METHODS: A KOA model was established via the modified Videman method. Rabbits were randomly divided into a control group (CON), KOA group and KOA + acupotomy group (Acu). Rabbits in the acupotomy group were subjected to acupotomy for 4 weeks after model establishment. The behavior of the rabbits before and after intervention was recorded. Cartilage degeneration was evaluated by optical microscopy and fluorescence microscopy. The level of mitophagy was evaluated by transmission electron microscopy, immunofluorescence and enzyme-linked immunosorbent assay (ELISA). The expression of phosphatase and tensin homolog (PTEN)-induced kinase 1 (Pink1)-Parkin mitophagy pathway components was evaluated by immunofluorescence, Western blotting and real-time polymerase chain reaction. RESULTS: In rabbits with KOA, joint pain, mobility disorders and cartilage degeneration were observed, the Mankin score was increased, collagen type Ⅱ (Col-Ⅱ) expression was significantly decreased, mitophagy was inhibited, mitochondrial function was impaired, and factors associated with the Pink1-Parkin pathway were inhibited. Acupotomy regulated the expression of Pink1-Parkin pathway-related proteins, the mitophagy-related protein microtubule-associated protein-1 light chain-3, the translocase of the outer membrane, and the inner mitochondrial membrane 23; increased the colocalization of mitochondria and autophagosomes; promoted the removal of damaged mitochondria; restored mitochondrial adenosine-triphosphate (ATP) production; and alleviated cartilage degeneration in rabbits with KOA. CONCLUSIONS: Acupotomy played a role in alleviating KOA in rabbits by activating mitophagy in chondrocytes via the regulation of proteins that are related to the Pink1-Parkin pathway.

Knee osteoarthritis (KOA) is one of the most frequently encountered diseases in the orthopedic department, which seriously reduces the quality of life of people with KOA. Among several pathogenic factors, the biomechanical imbalance of the knee joint is one of the main causes of KOA. Acupotomology believes that restoring the mechanical balance of the knee joint is the key to treating KOA. Clinical studies have shown that acupotomy can effectively reduce pain and improve knee mobility by reducing adhesion, contracture of soft tissues, and stress concentration points in muscles and tendons around the knee joint. In this protocol, we used the modified Videman method to establish a KOA model by immobilizing the left hindlimb in a straight position. We have outlined the method of operation and the precautions related to acupotomy in detail and evaluated the efficacy of acupotomy in conjunction with the theory of "Modulating Muscles and Tendons to Treat Bone Disorders" through the detection of the mechanical properties of quadriceps femoris and tendon, as well as cartilage mechanics and morphology. The results show that acupotomy has a protective effect on cartilage by adjusting the mechanical properties of the soft tissues around the knee joint, improving the cartilage stress environment, and delaying cartilage degeneration.

OBJECTIVE: To investigate the effects of acupotomy on the subchondral bone absorption and mechanical properties in rabbits with knee osteoarthritis (KOA). METHODS: The rabbits were divided into blank control, model, acupotomy and electroacupuncture (EA) groups, with 12 rabbits in each. Modified Videman's method was used to prepare KOA model. The acupotomy and EA group were given indicated intervention for 3 weeks. The behavior of rabbits in each group was recorded. Subsequently, cartilage-subchondral bone units were obtained and morphological changes were observed by optical microscope and micro computed tomography. Compression test was used to detect the mechanical properties of subchondral bone, Western blot and real-time polymerase chain reaction (RT-PCR) were applied to detect the expression of bone morphogenetic protein 2-Smad1 (BMP2-Smad1) pathway in subchondral bone. RESULTS: Compared with the control group, rabbits in the KOA group showed lameness, knee pain, and cartilage degradation; the subchondral bone showed active resorption, the mechanical properties decreased significantly and the BMP2-Smad1 pathway downregulated significantly. Both acupotomy and EA intervention could increase the thickness of trabecular bone (Tb. Th), the bone volume fraction (BV/TV) and the thickness of subchondral bone plate, reduce the separation of trabecular bone (Tb. Sp), improve the maximum load and elastic modulus of subchondral bone, and effectively delay cartilage degeneration in KOA rabbits. This process may be achieved through upregulation the related proteins of BMP2-Smad1 pathway. The maximum load and elastic modulus of subchondral bone in the acupotomy group were slightly better than those in the EA group. CONCLUSIONS: Acupotomy could effectively protect cartilage by inhibiting abnormal bone resorption and improving mechanical properties of subchondral bone thorough the related proteins of BMP2-Smad1 pathway in KOA rabbits.

Objective:To investigate the characteristics, process, and prognosis of esophageal stricture after circumferential endoscopic submucosal dissection (ESD), and to preliminarily analyze the prevention and treatment effects of simple dilation, stent placement, mucosal transplantation, and glucocorticoid (hereinafter referred to as hormone) application in esophageal stricture.Methods:From August 2017 to March 2022, at the First Affiliated Hospital of Zhengzhou University, the clinical and follow-up data of 55 patients who underwent circumferential ESD for early esophageal cancer and precancerous lesions were retrospectively analyzed. According to the prevention and treatment methods for esophageal stricture, the patients were divided into two groups: simple dilation group (23 cases) and combined dilation group (32 cases). The combined dilation group was divided into mucosal transplantation subgroup (9 cases), stent placement subgroup (14 cases), hormone application subgroup (7 cases), and bleomycin subgroup (2 cases, excluded from comparative analysis due to limited cases). Overall prognosis of patients was observed. Treatment efficacy, prognosis, and adverse events were compared among the simple dilation group, mucosal transplantation subgroup, stent placement subgroup, and hormone application subgroup. Independent samples t-test, chi-square test, and Fisher′s exact test were used for statistical analysis. Results:Among the 55 patients, the follow-up time was (894.1±417.7) days. Refractory esophageal stricture (total dilation times ≥ 5) occurred in 33 patients (60.0%). Fifty-two patients (94.5%) achieved clinical remission of the stricture. The total number of dilations was 5.8±4.0, and the average dysphagia-free period was (52.3±37.1) days. The dysphagia-free period of mucosal transplantation subgroup was longer than that of the simple dilation group, stent placement subgroup, and hormone application subgroup ((114.5±50.0) days vs. (40.9±20.0), (39.7±10.0), and (40.9±25.5) days, respectively), and the differences were statistically significant ( t=4.82, 3.77 and 3.14, P<0.001, =0.011, =0.009). There were no statistically significant differences between the simple dilation group and the mucosal transplantation subgroup, stent placement subgroup, and hormone application subgroup in the total number of dilations (6.8±4.8 vs. 3.0±2.5, 5.8±2.2, and 5.7±5.0), stricture remission rate (95.7%, 22/23 vs. 8/9, 13/14, and 7/7), and incidence of adverse events (17.4%, 4/23 vs. 5/9, 5/14, and 2/7; all P>0.05). Conclusions:Esophageal stricture formed after circumferential ESD shows the characteristics of recurrence and intractability. The over all number of dilations is high, and the average dysphagia-free period is short. Most patients can achieve clinical remission of the stricture after multiple times of endoscopic dilation treatment. However mucosal transplantation, stent placement, and hormone application cannot well intervene the natural process of esophageal stricture.

OBJECTIVE: To investigate the effects of acupotomy on inhibiting abnormal formation of subchondral bone in rabbits with knee osteoarthritis (KOA). METHODS: A total of 24 New Zealand rabbits were randomly divided into four groups of 6 rabbits each [control, model, electroacupuncture (EA) and acupotomy]. Eighteen KOA model rabbits were established using a modified Videman method. Rabbits in EA and acupotomy groups received the intervention for 3 weeks. Then, the cartilage and subchondral bone unit were obtained and the histomorphological changes were recorded. Osteo-protegerin (OPG) and receptor activator of nuclear factor-κB ligand (RANKL) in subchondral bone were evaluated by Western blotting, real-time polymerase chain reaction and immunohistochemistry. RESULTS: Compared with the model group, both the acupotomy and EA groups showed a significant decrease in the Lequesne index (both 0.01) and Mankin score ( 0.01, < 0.05). In addition, both EA and acupotomy groups had a higher expression of total articular cartilage (TAC) ( 0.05, < 0.01) and lower expression of articular calcified cartilage (ACC)/TAC ( 0.05, < 0.05) compared with the model group. The thickness of the subchondral bone plate in EA and acupotomy groups were decreased (both 0.01) compared to the model group. Moreover, trabecular bone volume (BV/TV), protein and relative expression of OPG and the ratio of OPG/RANKL in the subchondral bone of acupotomy group were decreased statistically significant, while these parameters were not significantly changed in the EA group compared with the model group. CONCLUSIONS: In the rabbit model of KOA, acupotomy inhibits aberrant formation of subchondral bone by suppressing OPG/RANKL ratio as a potential therapy for KOA.

Objective:To investigate the correlation between serum anti phospholipase A2 receptor (PLA 2R) antibody level and the condition and prognosis of patients with primary membranous nephropathy (PMN).Methods:61 patients who were diagnosed as PMN in the laboratory department of Jiangsu Province Hospital of Chinese Medicine from January 2017 to August 2019 were selected and divided into PLA 2R positive group (41 cases) and PLA 2R negative group (20 cases) according to the level of PLA 2R antibody. The gender, age and other general data of the two groups were collected, and the clinical pathological characteristics and prognosis of the two groups were analyzed. Spearman correlation was used to analyze the correlation between the level of PLA 2R antibody and the disease condition and treatment prognosis.Results:The levels of estimate glomerular filtration rate (eGFR), total protein (TP) and albumin (ALB) in PLA 2R positive group were lower than those in PLA 2R negative group, while the levels of 24-h urinary protein and IgG4 were higher in PLA 2R positive group than that in PLA 2R negative group ( P<0.05); Multivariate logistic regression analysis showed that 24-hour urinary protein was an independent risk factor for PLA 2R antibody level ( OR=2.242, P<0.05), while TP and ALB levels were protective factors ( OR=0.840, 0.674, P<0.05); The patients in both groups were followed up for one year by rechecking serological indexes. The complete remission rate and total remission rate of PLA 2R positive group were lower than those of PLA 2R negative group ( P<0.05); Spearman correlation analysis showed that the level of PLA 2R antibody was negatively correlated with the levels of TP, ALB, and prognosis of patients ( r=-0.642, -0.547, -0.357, P<0.05), and positively correlated with the level of 24-hour urinary protein ( r=0.347, P<0.05). PLA 2R in renal tissue of 61 patients was found to be positive in 63.93% (39/61). Conclusions:The level of PLA 2R antibody is closely related to serum albumin, TP, 24 h urinary protein and prognosis of patients with PMN.

With Jianyu (LI 15), Jianliao (TE 14) and Jianzhen (SI 9) for instance, the three acupoints used to treat shoulder bi syndrome by needle knife, and through traceability and researching constant structure, the acupoint position, insertion trace, manipulation and clinical significance of needle knife medicine were discussed. Accurate position is one of the characteristics of acupoints selection of needle knife medicine. As for the acupoints selection method, the mean of body surface localization is always used. The phanerous or palpable bone processes, muscles and tendons are taken as positioning marks; pressing areas where appear sour, numb, or distensible and other sensations is considered as the principle of press positioning. So acupoints position method is the combination of observation and palpation. Different insertion methods can effectively relieve the accretive bursae synovialis, tendon, joint capsule and the compressed nerve, so that shoulder bi syndrome is relieved.

RESUMO

The impact factors were explored to determine the horizontal positional relationship between the umbilicus and the 2nd lumbar spinal process in adults and to verify the accuracy of the localization of Shenshu (BL 23) via the umbilicus. The position of the umbilicus and the 2nd lumbar spinal process was measured in 100 participants and the data were analyzed through SPSS 20.0 software. It was found that the umbilicus and the 2nd lumbar process were not positioned horizontally. The positional relationship of these two sites was not apparently correlated with gender, age, body weight, body height, BMI, waistline and discomfort of lumbar region. The umbilicus was commonly and posteriorly projected on the site between the 4th and 5th lumbar vertebra. It is explained that the localization of Shenshu (BL23) via the umbilicus is not accurate.

Objective To explore the mechanism of therapeutic action of needle knife “regulating sinews and treating bones” on knee osteoarthritis (KOA) by observing the effect of needle knife intervention on KOA rabbit behaviors, mechanical characteristics of patellar ligament (PL), and expressions of interleukin-4 (IL-4), matrix metalloproteinase-3 (MMP-3) and aggrecan in knee cartilages.Methods Forty New Zealand rabbits were randomly allocated to blank, model, needle knife and electroacupuncture groups, 10 rabbits each. A rabbit model of KOA was made by the modified Videman method of immobilization in extension position. After model making, the needle knife and electroacupuncture groups received needle knife and electroacupuncture treatments, respectively. A behavioral assessment was made using the modified Lequesne MG knee grade evaluation method in every group after model making and treatment. The samples were taken after treatment. PL tension, stress relaxation and creep state were tested using a Bose Electro Force 3300 protracted test machine. Cartilage cell IL-4 expression was examined by ELISA. MMP-3 mRNA and aggrecan mRNA expressions were detected by real-time PCR.Results After model making, there was a statistically significant difference in the Lequesne MG score between the model and blank groups (P0.05). There was a statistically significant post-treatment difference in the Lequesne MG score between the needle knife or electroacupuncture group and the model group (P0.05).Conclusion The mechanism of action of needle knife treatment on KOA may be that it improves ligament mechanical characteristics, regulates intra-articular stress environment, and modulates aggrecan mRNA and MMP-3 mRNA expressions and inhibits cartilage degeneration through IL-4 mechanical signal pathway, to produce the therapeutic effect of “regulating sinews and treating bones”.

MAL gene expresses in the mediate and late stage of T-lymphocytes.Many studies have shown that the expression of MAL gene has down regulation or loss in many kinds of tumors including esophageal carcinoma,gastric cancer,colon cancer,head and neck squamous cell carcinoma,and so on.MAL gene is related with the process of generation and development of the tumors.The application of MAL gene for clinical diagnosis,prognostic and instructing therapy needs further studies.

Objective To observe the regulatory effect of acupotomy lysis on SP level in spinal cord and tissues above spinal cord of rats with knee osteoarthritis.Methods 60 healthy SD rats were randomly divided into normal control group,model group,electro-acupuncture(EA)group,and acupotomy lysis(AL)group.Mix 4%papain solution with 0.3 mol/L cysteine solution in the ratio of 1∶1.After pausing for 0.5h,inject the mixture,20 μl each time,into the left knee joint cavities of rats in model,AL,and EA groups on the day of 1,4,7.After 4 weeks AL group was treated with acupotomy lysis and EA group with electro-acupuncture.Three weeks after treatment,take samples of spinal cord,midbrain,pituitary gland,thalamus,and hypothalamus from the swellings of rats'waists.Measure the content of SP therein separately.Results Compared with normal control group,there was a significant rise in the content of SP in spinal cord and the tissues above spinal cord of model group rats (P<0.05,P<0.01),and there was a significant rise in spinal cord,pituitary gland,thalamus and hypothalamus of EA group rats (P<0.05,P<0.01);in AL group,there was a significant rise in spinal cord,pituitary gland,thalamus,and there was no statistically difference in hypothalamus and midbrain.Compared with normal control group,there was a significant rise in spinal cord (P<0.05)and a significant decrease in the SP contents in hypothalamus(P<0.05,P<0.01)in EA group.There was no statistically difference between EA group and AL group except in hypothalamus(P<0.05).Conclusion Acupotomy lysis has positive functions in regulating SP content in centrum of rats with knee osteoarthritis,which helps easing pain.

@#: Acupotome therapy exhibits good efficacy on knee osteoarthritis in clinic. The mechanism based on its dynamic equilibrium theory is that acupotome therapy can restore normal joint stress state by changing the imbalance of mechanical equilibrium around the joint. Furthermore, it can ease inflammation, relieve pain and repair tissue structure so as to achieve its therapeutic effect. This review proposes new thoughts for the etiopathogenisis, pathology and treatment of KOA.

Objective To investigate the relationship between lower limb artherosclerosis and cardiac function in elderly patients with diabetes mellitus(DM)complicated with coronary heart disease (CHD). Methods 441 cases of elderly patients were divided into four groups: the group with CHD (n=117), the group with DM(n=42), the group with DM compllicated with CHD( DM±CHD, n=132),the group without DM and CHD(NC,n=150). Systolic blood pressure (SBP), diastolic blood pressure(DBP), pulse pressure (PP), braehial-ankle pulse wave velocity (baPWV), ankle-brachial index(ABl) and toe-brachial index(TBI) were measured by the auto-survey artherosclerosis apparatus.Left ventricular posterior wall (LVPW), left atrium (LA), fractional shortening (FS), ejection fraction(EF) and EF slope were measured by the two-dimensional echocardiography. Results The baPWV was significantly higher in DM+CHD group than in DM group and NC group[(20125±354)cm/s vs (1849±467)cm/s, (1863±347) cm/s. P<0.05]. The ABI was significantly lower in DM±CHD group compared with NC, CHD and DM groups (0.91±0.23 vs 1.15±0.11,1.07±0.16 and 1.06±0.14. all P<0.05). There were significant differences in TBI among four groups(all P<0.05) :NC group(0.905±0.15)>CHD group(0.805±0.16)>DM group(0.71±0.17)>DM+CHD group(0.625±0.17). The LA and the LVPW were significantly higher in DM+CHD group than in NC group(P<0.05). The FS and EF slopes were significantly lower in the DM+CHD group than in DM group (P<0.05) and NC group(P<0.05). The EF in DM+CHD group was significantly lower than that in other three groups (P<0.05). BaPWV showed a strong negative association with FS,EF and EF slope(r:-0.112,-0.151,-0.275. all P<0.05). ABI showed a strong positive association with FS,EF and EF slope(r:0.150,0.186,0.260. all P<0.01) ,TBI was found to have strong negative association with LA and LVPW ( r:-0.158,-0.171;all P<0.01) and have strong positive association with FS,EF and EF slope(r:0.163,0.197,0.243.all P<0.01). Conclusions The lower limb artherosclerosis is serious and cardiac function is significantly decreased in elderly patients with diabetes mellitus complicated with coronary heart disease. PWV,ABI and TBI are closely related with cardiac function and can reflect the state of cardiac function in some extent.

BACKGROUND: At present, there is no ideal way to treat auditory injury, and acupuncture would become an effective method to prevent and cure drug auditory injury.OBJECTIVE: To observe the effect of acupuncture at Yifeng, Ermen and Zhongzhu acupoints on improving histopathologic form of basilar membrane of guinea pig with drug poisoning auditory injury.DESIGN: Completely randomized design on the basis of experiment al animals.SETTING: Department of Neurology, Xuanwu Hospital, Capital University of Medical Sciences.MATERIALS: A total of 45 white male guinea pigs with normal Preyer's reflex were selected in this study. Guinea pigs were 3-4 months old, of general grade and weighted from 250 g to 300 g. All guinea pigs were randomly divided into 5 groups: normal group, model group, Yifeng group, Ermen group and Zhongzhu group with 9 in each group.METHODS: The experiment was completed at the Laboratory of College of Acupuncture and Moxibustion Affiliated to Beijing University of Traditional Chinese Medicine from April to July 2002. ① Normal group: Guinea pigs were fed with normal feed. Other four groups: Guinea pigs were injected into muscle with 80 mg/kg gentamycin for 20 days, and guinea pigs in Yifeng group, Ermen group and Zhongzhu group were acupunctured at corresponding acupoints at the same time. Before acupuncture, guinea pigs were fixed and rapidly acupunctured into subcutaneous tissue with stainless steel needle sized 0.35 mm × 25 mm. The needle was inserted about 10 mm. According to uniform reinforcing-reducing method, the needle was thrusted once every 10 minutes with each for 30 seconds and retained for 30 minutes. The course was 20 days. ② Basilar membrane of guinea pigs was established. Amount of hair cell was calculated under optic microscope (×200) to record deletion amount of hair cell and observe activity of succinic dehydrogenase; meanwhile, ultrastructure of hair cells was observed with transmission electron microscope (×5 000). Measurement data were compared with t test among groups.MAIN OUTCOME MEASURES: Activities of succinic dehydrogenase of hair cell in stretched preparation and changes of ultrastructure of hair cell in cochlea of guinea pigs.RESULTS: All 45 guinea pigs were revolved in the final analysis. ① Comparing activities of succinic dehydrogenase: Activity in normal group was great, and structures of inter and outer hair cells were normal. Activity in mode group was reduced gradually till disappeared. Outer hair cell at base was damaged severely, and there was significant difference from that in normal group (P < 0.01). Activity of succinic dehydrogenase in Yifeng group was changed a little as compared with that in normal group, and difference was not obviously (P > 0.05). Activity of succinic dehydrogenase in Ermen group was decreased, and hair cell showed a little swelling and deletion. Activity of succinic dehydrogenase in Zhongzhu group was decreased remarkably, and outer hair cell at base was damaged severely.There was significant difference between Ermen group and Zhongzhu group as compared with that in normal group (P < 0.01). ② Deletion amount of outer hair cells in basilar membrane of guinea pigs: Amounts in Yifeng group, Ermen group and normal group were less than those in model group (P < 0.05-0.01), but those in Zhongzhu group were similar to those in model group. ③ Ultrastructure: Structure of hair cell of cochlea was normal in normal group, pathological changes were observed on hair cells in model group, and pathological changes in acupuncture groups were milder than those in model group. Pathological changes of outer hair cells were mild in Yifeng group, structure of hair cells was changed a little in Ermen group,pathological changes in Zhongzhu group were severe, but its degree was milder than that in model group.CONCLUSION: Acupuncture can improve and maintain the activity of succinic dehydrogenase, relieve or resist toxic effect of gentamycin, especially at Yifeng, then at Ermen and Zhongzhu.

RESUMO

A theoretical model for the magnetic forces on ferromagnetic drug particles in a guiding magnetic field is established with the consideration of the electromagnetic field theory. Our model shows that the ferromagnetic drug particles can concentrate on the targeted area with the guidance of the outside magnetic field. Both the distribution of the intensity of the guiding magnetic field and the forces on ferromagnetic drug particles are studied numerically. The theoretical results are in good agreement with the experimental results reported recently.

Objective: To deduce the physiological function of Yangming Stomach Meridian of Foot,enrich the content of channel theory.Methods: Unscramble the related classic literature of Yangming Stomach Meridian of Foot,and analyze its channel courses and links,amount of qi and blood and the relationship between the manifestations of disease,main therapy rule of acupoints and the physiological function.Results: Stomach meridian is the head of the twelve meridians;abounds in qi and blood;smoothes the convergent tendon.Conclusion: Yangming Stomach Meridian of Foot is one of the most important meridians in the fourteen meridians.It is distributed on the anterior aspect of the body and communicates with the spleen and stomach,connects with many other organs,and keeps the functions and activities of all parts of the body in harmony and balance relatively through creating and transporting qi and blood.

Objective To observe the changes of serum endotoxin and interleukin-6 level before and after treatment at intestine get rid of filth and adjustment intestine microenviroment in patients with advanced stage liver cirrhosis and to oberserve the incidence of spontaneous bacterial peritonitis(SBP) and to research a method that can more efficient reduce the incidence of SBP.Methods Group A was given conventional treatment such as protection liver,supplement human albumin and uragogue;goup B was given FPA 0 2g tid po,continuous three days on the basis of two weeks conventional treatment,group C was given Tab comfort get 0 5g tid po,Changtai orally taken liquid,10ml tid po,lactulose 10ml tid po, for all 8 weeks,on the basis of group B treatment after pausing use FPA.Results Serum endotoxin and interleukin-6 levels in group A was not significantly changes after treatment,,SBP incidence was higher than both group B and group C;at group B,serum endotoxin and interleukin-6 levels decline,SBP incidence significantly drop afer treatment,at group C,serum endotoxin,interleukin-6 level and SBP incidence significantly drop,the difference was significant at statistics as compared with group A and B.Conclusions The treatment at intestine get rid of filth and adjustment intestine microenviroment can effectively reduce serum endotoxin and interleukin-6 levels and can significantly reduce the incidence of SBP.

Objective To study the status of cystatin B gene expression in human esophageal cancer. Methods Expression of cystatin B gene was detected by RT PCR in 41 pairs of human esophageal cancer tissues, matched adjacent normal mucosa, human esophageal cancer cell line EC9706 and human lung cancer cell line GLC82. Results cystatin B gene expression was down regulated in 82.9% (34/41) of human esophageal cancer tissues while expressed at high level in all matched adjacent normal esophageal mucosa, and was significantly corelated to lymph node metastasis ( P

Objective To investigate the mechanisms of Mal gene down regulated in human esophageal cancer. Methods We amplified the 5′regulatory region of Mal gene in normal placenta tissue and human esophageal cancer cell lines EC9706、EC8712、EC109 by optimization the PCR methods, cloned and sequenced the PCR products and compare the sequence. Results Human esophageal cancer cell lines EC9706、EC8712、EC109 all have the 654 th of 5′regulatory region of Mal gene T to C changes, corresponding codon changed from TCC to CCC, and the amino acid which it codes changed from serine to proline; EC9706 also has the 657 th of 5′regulatory region of Mal gene T to C changes, corresponding codon changed from TGC to CGC, the amino acid which it codes changed from cysteine to arginine; EC8712 has the 139 th of 5′regulatory region of Mal gene A to G changed, corresponding codon changed from GGA to GGG, the amino acid which it codes remains glycine. Conclusion Our results suggested there are may point mutation in 5′ region of Mal gene in human esophageal cancer.
