RESUMO

Traditional "one for one channel" long-wavelength probes in hematology analyzers limit their resolution and detection efficiency. In this study, we developed a "one for two channels" probe named NATO, which shows a short wavelength (λabs = 460 nm), good nucleus and nucleolus location, and a high signal-to-noise ratio to nucleic acids. When NATO was made into a hematology analysis kit and applied in an automated hematology analyzer, short-wavelength absorbance endows NATO with higher resolution, which in turn leads to better separation of red blood cells and platelets in the blood shadow of the differentiating (DIFF) channel. In addition, this kit showed terrific performance in both DIFF and reticulocytes channels. Our study sheds light on the development of hematology analysis in an automated hematology analyzer by proposing a nucleic acid probe with difunction and higher resolution.

RESUMO

The introduction of Lawesson's reagent into a bis-rhodamine spirolactam system afforded a new fluorescent sensor for Cu(2+) ions, SRR, which contained a new tridentate sulfur ligand. SRR showed excellent specificity for Cu(2+) ions over other cations (including Cu(+), Hg(2+), and Fe(3+)), very high sensitivity (10 nM), and a rapid response time (3 min). The detection mechanism was investigated by (1)H NMR, (13)C NMR, (31)P NMR, and ESR spectroscopy, MS, and Gaussian calculations. Coordination of a Cu(2+) ion to the tridentate sulfur ligand, which promotes ring-opening of the rhodamine groups, followed by a spontaneous reduction reaction (Cu(2+) into Cu(+)), has been proposed as the sensing mechanism.

RESUMO

Born to dye: Five fluorescein analogues were synthesised (see scheme). One analogue was found to emit in the NIR region with a high quantum yield, excellent photostability and good permeability. Three derivatives were found to specifically stain mitochondria and one dye responds to thiols with a strong turn-on NIR fluorescence signal and colorimetric change, in vitro and in vivo.

RESUMO

OBJECTIVE: To provide scientific basis data for revising the national hygiene criteria of "Classification of hazard conditions of productive dust" (GB5817-86). METHODS: The data of the retrospective study and the field survey data were analyzed with correlation and regression analysis. The product of total dust concentration of respiratory exposure (mg/m(3)), total ventilation during exposure (m(3)/d per psrson), and level of free SiO(2) in dust (%) was the respiratory exposure dose of free SiO(2) (mg per day per person) which was used as dose criteria value of classification of hazard degree of dust. RESULTS: Using free SiO(2) exposure dose and the dose-effect relationship, the hazard degrees of the dust were divided into 5 grades: 0, I, II, III, IV (0 - 8.0, 8.1 - 12.0, 12.1 - 16.0, > 24.1 mg per day per person). CONCLUSION: The exposure dose of free SiO(2) is closely related to the pathogenesis of silicosis. Using the exposure dose of free SiO(2) as the classification indicator of hazard degree of dust is reliable, simple and easy to execute.
