Purpose: This study aimed to analyze the expression and prognosis of SRY-box transcription factor 11 (SOX11) in neuroblastoma (NB), as well as the biological function and potential regulatory mechanism of SOX11 in NB. Methods: Public RNA sequencing was used to detect the expression level of SOX11. The Kaplan-Meier curve and hazard ratios (HR) were used to determine the prognostic value of SOX11 in NB. Functional analyses were performed using CCK8, wound healing assay, and transwell invasion assay. Finally, the potential target genes of SOX11 were predicted by Harmonizonme (Ma'ayan Laboratory) and Cistrome Data Browser (Cistrome Project) database to explore the potential molecular mechanism of SOX11 in NB. Results: Compared with normal adrenal tissue, the expression of SOX11 in NB tissue was significantly upregulated. The Kaplan-Meier curve showed that high expression of SOX11 was associated with poor prognosis in children with NB (HR, 1.719; P = 0.049). SOX11 knockdown suppressed the migration capacity of SK-N-SH cells but did not affect proliferation and invasion capacity. Enhancer of zeste homolog 2 (EZH2) may be a potential downstream target gene for the transcription factor SOX11 to play a role in NB. Conclusion: The transcription factor SOX11 was significantly upregulated in NB. SOX11 knockdown suppressed the migration capacity of NB cell SK-N-SH. SOX11 may promote the progression of NB by targeting EZH2.

In response to China's policies on pollution control and carbon emission (CE) reductions, more stringent regulations have been implemented to evaluate CE in wastewater treatment facilities. In this study, we have analyzed CE from China's wastewater treatment plants (WWTPs) and influencing factor. Emission factor (EF) and operational data integrated methods (ODIM) were utilized to measure emissions, using data collected from 247 WWTPs over a 1-year period across seven regions in China. The average CE intensity was 0.45 kgCO2-eq/m3, affected by region, season, influent water quality, treatment processes, effluent discharge standards, and facilities. The scale effect was obvious only in the range of 2 × 105 m3/day. Underground WWTPs exhibited significantly higher CE compared to aboveground WWTPs. In summary, the assessment of CE in 247 actual WWTPs not only identifies emission reduction potential but also provides a scientific basis for formulating targeted emission reduction measures.

Ultrasound and ultraviolet light have good inactivation performance against pathogens in sewage. In this study, the inactivation mechanisms of 60 kHz ultrasound and ultraviolet radiation against Staphylococcus aureus (S. aureus) were studied from the perspectives of cell phenotype and transcriptome for the first time. The results showed that both ultrasound and ultraviolet treatments had adverse impacts on the cellular morphology of S. aureus to varying degrees at cellular level. The transcriptomic analysis revealed that there were 225 and 1077 differentially expressed genes (DEGs) in the ultrasound and ultraviolet treatments, respectively. The result revealed that both ultrasound and ultraviolet could interfere with the expression of the genes involved in ABC transporters, amino acid and fatty acid metabolism to influence the membrane permeability. Besides the membrane permeability, ultraviolet also could disturb the ATP synthesis, DNA replication and cell division through restraining the expression of several genes related to carbohydrate metabolism, peptidoglycan synthesis, DNA-binding/repair protein synthesis. Compared with the single inactivation pathway of ultrasound, ultraviolet inactivation of S. aureus is multi-target and multi-pathway. We believe that the bactericidal mechanisms of ultrasound and ultraviolet radiation presented by this study could provide theoretical guidance for the synergistic inactivation of pathogens in sewage by ultrasound and ultraviolet radiation in the future.

In order to facilitate the observation in the process of secondary equipment operation and maintenance supervision and the detection and tracking of operation and maintenance personnel, a secondary operation and maintenance supervision system based on AR modeling and indoor positioning is designed. The whole system is divided into seven levels and a unified information base, in which the basic level contains all kinds of secondary equipment; AR modeling layer uses augmented reality technology to create models for each secondary equipment in the basic layer, and determines the equipment position information based on ranging positioning technology; The data acquisition layer collects all kinds of original management data based on the constructed secondary equipment model; The data analysis layer reads and analyzes the information of the data acquisition layer through the data bus; The process support layer provides task scheduling support for the integrated management application based on the data analysis results; The integrated application layer uniformly monitors the secondary equipment based on the task scheduling results; The presentation layer is responsible for the interface presentation of all operation and maintenance and security management information of the system, and the unified information base provides data support for the whole system. The experimental results show that the secondary equipment model in the designed system has high definition, can obtain more image details, can realize the 3D display and real-time interaction of the secondary equipment operation and maintenance supervision results, and accurately mark the target and track for the staff.

BACKGROUND: Laparoscopic splenectomy (LS), a treatment for both benign and malignant splenic diseases, can prove technically challenging in patients with massive splenomegaly. In particular, the optimal surgical modality for treating massive splenomegaly in children remains controversial. METHODS: The clinicopathologic data of 289 pediatric patients undergoing splenectomy for massive splenomegaly were studied in a retrospective analysis. Accordingly, the patients were classified into the LS surgery group and open splenectomy (OS) surgery group. In the laparoscopy cohort, they were separated into two subgroups according to the method of surgery: the multi-incision laparoscopic splenectomy (MILS) and the single-incision laparoscopic splenectomy (SILS) surgery groups, respectively. Patient demographics, clinical data, surgery, complications, and postoperative recovery underwent analysis. Concurrently, we compared the risk of adverse laparoscopic splenectomy outcomes utilizing univariable and multivariable logistic regression. RESULTS: The total operation time proved remarkably shorter in the OS group in contrast to the LS group (149.87 ± 61.44 versus 188.20 ± 52.51 min, P < 0.001). Relative to the OS group, the LS group exhibited lowered postoperative pain scores, bowel recovery time, and postoperative hospitalization time (P < 0.001). No remarkable difference existed in post-operation complications or mortality (P > 0.05). Nevertheless, the operation duration was remarkably longer in the SILS surgery group than in the MILS surgery group (200 ± 46.11 versus 171.39 ± 40.30 min, P = 0.02). Meanwhile, the operative duration of MILS and SILS displayed a remarkable positive association with splenic length. Moreover, the operative duration of SILS displayed a remarkable positive association with the age, weight, and height of the sick children. Splenic length proved an independent risk factor of adverse outcomes (P < 0.001, OR 1.378). CONCLUSIONS: For pediatric patients with massive splenomegaly who can tolerate prolonged anesthesia and operative procedures, LS surgery proves the optimal treatment regimen. SILS remains a novel surgery therapy which may be deemed a substitutional surgery approach for treating massive splenomegaly.

Poly(vinyl alcohol) (PVA)/modified porous starch (MPS) gel beads were prepared through in situ chemical cross-linking by incorporating with MPS, which was obtained by modifying porous starch (PS) with polyethyleneimine (PEI) and glutaraldehyde (GA). Addition of MPS could improve the storage modulus and the effective crosslinking density (ve) of the gel beads, and the mechanical properties were enhanced. The PVA-MPS gel beads were preserved as immobilized microbial carriers for 40 d and reactivated in wastewater. Scanning electron microscope (SEM) observations showed that the beads were highly porous and conducive for microorganism adhesion. The PVA-MPS gel beads were able to remove 97% of ammonia nitrogen and 80% of chemical oxygen demand (COD) after reactivation under all four preservation conditions. The abundance of Hydrogenophaga as denitrifying bacteria on PVA-MPS gel beads increased, with abundance of 8.44%, 5.55%, 8.90% and 9.48%, respectively. It proved that the carrier provided a partial hypoxic environment for microorganisms.

In wastewater treatment plants (WWTPs), aeration is vital for microbial oxygen needs. To achieve carbon neutrality, optimizing aeration for energy and emissions reduction is imperative. Machine learning (ML) is used in wastewater treatment to reveal complex rules in large data sets has become a trend. In this vein, the present paper proposes an aeration optimization approach based on the extreme gradient boosting-bidirectional long short-term memory (XGB-Bi-LSTM) model via the online monitoring of oxygen transfer efficiency (OTE) and oxygen uptake rate (OUR), thus allowing WWTPs to conserve energy and reduce indirect carbon emissions. The approach uses gain algorithm of XGB to calculate the importance of features and identify important parameters, and then uses Bi-LSTM to predict the target with important parameters as features. Operational data from a WWTP in Suzhou, China, is employed to train and test the approach, the performance of which is compared with ML models suitable for regression prediction tasks (XGB, random forest, light gradient boosting machine, gradient boosting and LSTM). Experimental results show the approach requires only a small number of input parameters to achieve good performance and outperforms other machine-learning models. When OTE and dissolved oxygen (DO) are used as features to predict the alpha factor (αF; since diffusers were used, multiply by the pollution factor F), the R-squared (R2) is 0.9977, the root mean square error (RMSE) is 0.0043, the mean absolute percentage error (MAPE) is 0.0069 and the median absolute error (MedAE) is 0.0032. When the predicted αF and the OUR are used as features to predict the air flow rate of an aeration unit, the R2 is 0.9901, the RMSE is 3.6150, the MAPE is 0.0209 and the MedAE is 1.5472. Using our optimized aeration approach, the energy consumption can be reduced by 23%.

Cibotium barometz (Linn.) J. Sm., a tree fern in the Dicksoniaceae family, is an economically important industrial exported plant in China and widely used in Traditional Chinese Medicine. C. barometz produces a range of bioactive triterpenes and their metabolites. However, the biosynthetic pathway of triterpenes in C. barometz remains unknown. To clarify the origin of diverse triterpenes in C. barometz, we conducted de novo transcriptome sequencing and analysis of C. barometz rhizomes and leaves to identify the candidate genes involved in C. barometz triterpene biosynthesis. Three C. barometz triterpene synthases (CbTSs) candidate genes were obtained. All of them were highly expressed in C. barometz rhizomes, consisting of the accumulation pattern of triterpenes in C. barometz. To characterize the function of these CbTSs, we constructed a squalene- and oxidosqualene-overproducing yeast chassis by overexpressing all the enzymes in the MVA pathway under the control of GAL-regulated promoter and disrupted the GAL80 gene in Saccharomyces cerevisiae simultaneously. Heterologous expressing CbTS1, CbTS2, and CbTS3 in engineering yeast strain produced cycloartenol, dammaradiene, and diploptene, respectively. Phylogenetic analysis revealed that CbTS1 belongs to oxidosqualene cyclase, while CbTS2 and CbTS3 belong to squalene cyclase. These results decipher enzymatic mechanisms underlying the origin of diverse triterpene in C. barometz.

Lanthanide-doped metal-organic frameworks (Ln-MOFs) have versatile luminescence properties, however it is challenging to achieve lanthanide-based upconversion luminescence in these materials. Here, 1,3,5-benzenetricarboxylic acid (BTC) and trivalent Yb3+ ions were used to generate crystalline Yb-BTC MOF 1D-microrods with upconversion luminescence under near infrared excitation via cooperative luminescence. Subsequently, the Yb-BTC MOFs were doped with a variety of different lanthanides to evaluate the potential for Yb3+ -based upconversion and energy transfer. Yb-BTC MOFs doped with Er3+ , Ho3+ , Tb3+ , and Eu3+ ions exhibit both the cooperative luminescence from Yb3+ and the characteristic emission bands of these ions under 980 nm irradiation. In contrast, only the 497 nm upconversion emission band from Yb3+ is observed in the MOFs doped with Tm3+ , Pr3+ , Sm3+ , and Dy3+ . The effects of different dopants on the efficiency of cooperative luminescence were established and will provide guidance for the exploitation of Ln-MOFs exhibiting upconversion.

In-depth knowledge of the deterioration and reactivation of the anaerobic ammonium oxidation (anammox) induced by carbon-to-nitrogen (C/N) is still lacking. Herein, the anammox performance was investigated in an anaerobic sequence biofilm batch reactor fed with low-strength partial nitration effluent in the range of C/N ratio from 0.5 to 3. The anammox was hardly deteriorated at C/N lower than 1.5, while became worsen if C/N was above 2.0. The specific anammox activity (SAA) experiments showed an 85% decrease of SAA at C/N of 3.0 compared with the maximum value (C/N:0). However, anammox capacity was rapidly recovered once influent C/N was adjusted back to zero. Moreover, C/N also highly affected the composition, structure and function of extracellular polymeric substance of the anammox biofilm. High-throughput sequencing revealed a close correlation between C/N change and microbial structure shift. Finally, the potential inhibition and restoration mechanism of the C/N-dependent anammox were proposed based on metagenomic analysis. This research provides some insights into the reinstatement of a mainstream anammox biofilm process after it is interrupted by high C/N influent.

Genistein (Gen), a kind of natural isoflavone drug monomer with poor water solubility and low oral absorption, was incorporated into oral nanoparticles with a new mesoporous carrier material, NH2-MCM-41, which was synthesized by copolycondensation. When the ratio of Gen to NH2-MCM-41 was 1:0.5, the maximum adsorption capacity of Gen was 13.15%, the maximum drug loading was 12.65%, and the particle size of the whole core-shell structure was in the range of 370 nm-390 nm. The particles were characterized by a Malvern particle size scanning machine, XRD, Fourier transform infrared spectroscopy, scanning electron microscopy, and nitrogen adsorption and desorption. Finally, Gen-NH2-MCM-41 was encapsulated by sodium alginate (SA), and the chimerism of this material, denoted as GEN-NH2-MCM-41@SA, was investigated. In vitro release experiments showed that, after 5 h in artificial colon fluid (pH = 8.0), the cumulative release reached 99.56%. In addition, its anti-rotavirus (RV) effect showed that the maximum inhibition rate was 62.24% at a concentration of 30 µM in RV-infected Caco-2 cells, and it significantly reduced the diarrhea rate and diarrhea index in an RV-infected-neonatal mice model at a dose of 0.3 mg/g, which was better than the results of Gen. Ultimately, Gen-NH2-MCM-41@SA was successfully prepared, which solves the problems of low solubility and poor absorption and provides an experimental basis for the application of Gen in the clinical treatment of RV infection.

Background: Hirschsprung's disease (HD) is a commonly digestive malformation in children that usually requires surgery. This study aims to evaluate the short-term efficacy of conventional laparoscopic surgery (CLS), transumbilical single-hole laparoscopic surgery (TU-LESS), and robotic surgery (RS) in the treatment of Hirschsprung's disease. Methods: 90 patients with Hirschsprung's disease undergone laparoscopic surgery at our center between 2015 and 2019, divided into three groups (group CLS, TU-LESS and RS), were retrospectively analysed. Results: CLS and TU-LESS group showed no significant difference in operation duration (P > 0.05) but shorter operation duration than the RS group (P < 0.05). RS group had highest overall SCAR scores, while TU-LESS group had the lowest one (P < 0.05). Other parameters such as operative blood loss, hospital stays, recovery time of digestive function, postoperative complications had no significant difference among the three groups (P > 0.05). Conclusion: The three surgical methods for HD revealed similar efficacy, where TU-LESS and CLS spent less time than RS; TU-LESS led to the most aesthetic effect, followed by CLS and RS.

Camellia sasanqua is an important economic plant that is rich in lipophilic triterpenols with pharmacological activities including antiallergic, anti-inflammatory, and anticancer activities. However, the key enzymes related to triterpene biosynthesis have seldom been studied in C. sasanqua. Oxidosqualene cyclases (OSCs) are the rate-limiting enzymes related to triterpene biosynthesis. In this study, seven putative OSC genes (CsOSC1-7) were mined from the C. sasanqua transcriptome. Six CsOSCs were characterized for the biosynthesis of diverse triterpene skeletons, including α-amyrin, ß-amyrin, δ-amyrin, dammarenediol-II, ψ-taraxasterol, taraxasterol, and cycloartenol by the heterologous expression system. CsOSC3 was a multiple functional α-amyrin synthase. Three key residues, Trp260, Tyr262, and Phe415, are critical to the catalytic performance of CsOSC3 judging from the results of molecular docking and site-directed mutagenesis. These findings provide important insights into the biosynthesis pathway of triterpenes in C. sasanqua.

One-stage anammox-based autotrophic nitrogen removal technology has attracted increasing interest to sustainable biological nitrogen removal for future wastewater treatment. However, its application in mainstream municipal wastewater treatment is still challenging due to low nitrogen and high organics of raw wastewater. Herein, a novel Simultaneous Carbon Oxidation, partial Comammox, Denitratation and Anammox (SCOCDA) was firstly developed in a single sequencing batch biofilm reactor operated at a dissolved oxygen concentration of ∼0.5 mg/L for treating synthetic municipal wastewater (50 mg/L NH4+-N and 100-250 mg/L COD). The long-term operation showed that almost complete COD and nitrogen removal performance could be achieved at a carbon/nitrogen ratio (COD/NH4+-N) of 3-5 with the corresponding effluent total nitrogen (TN)<5 mg/L. Microbial community and amoA-targeting amplicon sequencing analysis further verified that comammox Nitrospira spp., denitrifier Thauera and other aerobic/facultative heterotrophs could work synergistically with anammox bacteria, Candidatus Kuenenia. Moreover, nitrogen metabolic and inorganic carbon fixation pathways through the interaction between comammox and anammox were also revealed with the aid of Kyoto Encyclopedia of Genes and Genomes (KEGG). Lastly, potential application of proposed SCOCDA process was illustrated. This research sheds new light on advanced nitrogen removal towards limit of technology via the synergy of comammox and anammox.

Background: There are many reports on the application of minimally invasive technology in correction of children's vesicoureteral junction obstruction (VUJO), but there is no report on the treatment of children's VUJO with the transumbilical laparoendoscopic single-site surgery (TU-LESS) Lich-Gregoir method. We aimed to comparatively analyze the therapeutic outcomes of transvesicoscopic ureteral reimplantation Cohen (TUR-C) procedure and TU-LESS Lich-Gregoir (TU-LESS-LG) procedure in pediatric VUJO. Materials and Methods: The data of 49 children with VUJO, admitted from January 2016 to January 2020, were retrospectively analyzed. Based on different surgical methods, they were divided into the TUR-C group (23 cases) and the TU-LESS-LG group (26 cases). Demographic characteristics, perioperative characteristics, postoperative complications, recovery of renal function, and improvement of hydronephrosis were compared between the two groups. Results: There were no statistical differences in demographic characteristics and preoperative data between the two groups. The TU-LESS-LG group was superior to the TUR-C group in terms of average operation time and postoperative hospital stay. There was no statistical difference between the two groups in terms of postoperative complications, postoperative recovery of renal function, and improvement of hydronephrosis. Conclusions: The two surgical methods can achieve a similar curative effect in the treatment of VUJO. The TU-LESS-LG procedure has more advantages of operation time, postoperative hospital stay, wider age range for selection of cases, megaureter tapering, and cosmetic incision, but the operation is more difficult. Anemarrhena asphodeloides is an immensely popular medicinal herb in China, which contains an abundant of mangiferin. As an important bioactive xanthone C-glycoside, mangiferin possesses a variety of pharmacological activities and is derived from the cyclization reaction of a benzophenone C-glycoside (maclurin). Biosynthetically, C-glycosyltransferases are critical for the formation of benzophenone C-glycosides. However, the benzophenone C-glycosyltransferases from Anemarrhena asphodeloides have not been discovered. Herein, a promiscuous C-glycosyltransferase (AaCGT) was identified from Anemarrhena asphodeloides. It was able to catalyze efficiently mono-C-glycosylation of benzophenone, together with di-C-glycosylation of dihydrochalcone. It also exhibited the weak O-glycosylation or potent S-glycosylation capacities toward 12 other types of flavonoid scaffolds and a simple aromatic compound with -SH group. Homology modeling and mutagenesis experiments revealed that the glycosylation reaction of AaCGT was initiated by the conserved residue H23 as the catalytic base. Three critical residues H356, W359 and D380 were involved in the recognition of sugar donor through hydrogen-bonding interactions. In particular, the double mutant of F94W/L378M led to an unexpected enzymatic conversion of mono-C- to di-C-glycosylation. This study highlights the important value of AaCGT as a potential biocatalyst for efficiently synthesizing high-value C-glycosides.

Rotavirus (RV) is one of the main pathogens that induce infantile diarrhea and by now no effective drugs are available for RV-induced infantile diarrhea. Thus the development of novel models is of vital importance for the pathological research of RV-induced infantile diarrhea, as well as the progress of the associated treatment strategy. Here we introduced for the first time that RV-Wa strain and RV-SA-11 strain could infect 5 dpf(day post fertilization) and 28 dpf larvae, to induce infantile diarrhea model that was highly consistent with the clinical infection of infants. RV infection significantly changed the signs, survival rate and inflammation of larvae. Some important indicators, including the levels of RV antigen VP4 and VP6, the in vivo RV tracking, and the RV particles were also analyzed, which collectively demonstrated that the model was successfully established. More importantly, we also determined the potentials of the proposed RV-infected zebrafish model for anti-viral drug assessment. In conclusion, we established a RV-infected zebrafish model with formulated relevant indicators both larvae and adult fish, which might be served as a high throughput platform for antiviral drug screening.

PURPOSE: The molecular mechanisms and signal pathways of ferroptosis in hepatoblastoma (HB) have not yet been clarified. In previous studies, activating transcription factor 3 (ATF3) was reported to be correlated with several tumors, but the clinical significance of ATF3 has never been determined. Herein, we investigated the clinicopathological value and mechanisms of ATF3 in regulating ferroptosis in HB. METHODS: The mRNA microarray and RNA-sequencing data of 402 samples from our hospital and public databases were used to estimate ATF3 expression and assess its clinical role in HB. The standard mean difference (SMD) and summary receiver operating characteristic curves were utilized to judge the discrimination ability of ATF3 between HB and non-HB liver tissues. We examined the expression variation of ATF3 in HB cells after the treatment with erastin. We also predicted the target genes of ATF3 as a transcriptional factor from public Chromatin Immunoprecipitation-sequencing data and selected the ferroptosis-related genes for a signaling pathway analysis. RESULTS: In ten series, the pooled SMD for ATF3 was -0.91, demonstrating that ATF3 expression was predominantly lower in HB than in non-HB liver tissues. ATF3 down-regulation showed moderate potential to distinguish HB from non-HB liver tissues (area under curves = 0.83, 95% confidence interval = 0.79-0.86). Altogether, 4855 putative targets of ATF3 as a transcriptional factor were collected, among which, 60 genes were ferroptosis-related. CONCLUSION: The down-regulated ATF3 expression may play a vital role in the occurrence of HB possible partially by regulating ferroptosis.

Photothermal therapy has been considered a powerful means of cancer therapy due to its minimal invasiveness, effectiveness, and convenience. Although promising, the therapeutic effects are greatly limited as they rely on the photothermal agent (PTA). It is urgent to develop new PTAs with high photothermal conversion performance, especially under irradiation in the long-wavelength biowindows. Herein, a dual-biowindow-responsive PTA made of NbS2-PVP nanosheets was fabricated to be used both in the first near-infrared (NIR-I) and the second near-infrared (NIR-II) biowindows. With excellent hydrophilicity and biocompatibility, the nanosheets could effectively convert the near-infrared (NIR) light into heat, showing prominent photothermal stability. The calculated photothermal conversion efficiencies reached 59.2% (under NIR-I excitation) and 69.1% (under NIR-II excitation), respectively, which are comparable to those of metallic PTAs. The NbS2-PVP nanosheets had low cytotoxicity and could trigger strong photothermal treatment and cause cancer cell death upon irradiation by NIR-I or NIR-II light in vitro. Moreover, we have also demonstrated the highly efficient tissue ablation and tumor inhibition capability of NbS2-PVP nanosheets in vivo. This work explores an effective PTA of two-dimensional nanomaterials in NIR-I and NIR-II biowindows and offers a reference for the design of new kinds of PTAs.
