RESUMO

Burning incenses and scented candles may provide harmful chemicals. Although many studies have evaluated volatile organic chemicals emitted by their use and related health risks, extension of our understanding for guiding appropriate use under various use conditions is necessary. In this study, emission characteristics of commercial incenses and scented candles were evaluated in a laboratory chamber using real-time measurement and the time-weighted average exposure concentrations of monoaromatic compounds and monoterpenes were assessed using passive samplers while volunteers living in a studio apartment use them. After burning incense, the average levels of benzene increased from 1.4 to 100 µg m-3. The presence of a wood core in commercial incense products was the main cause of high benzene emission by burning them although the increase in benzene was also influenced by factors such as the brand of the products, the number of incense sticks burned, the duration of each burning session, and ventilation period. Electrical warming of scented candles increased the levels of monoterpenes by factors of 16-30 on average. Considering the emission characteristics found in this study, exposure to benzene and monoterpenes could be mitigated by cautious use of those products in residential areas.

The exposure levels of selected terpenes (limonene, α- and ß-pinenes, and γ-terpinene) emitted by essential oil diffusers in vehicles and studio apartments were assessed using a passive sampling method. A previously developed passive sampler composed of an expanded polytetrafluoroethylene membrane and adsorbent (ePTFE PS) was enlarged and made wearable. Before field deployment, the sampling performance of the modified ePTFE PS for selected terpenes was compared with that of active sampling in a lab-scale 5 m3 test chamber under constant exposure conditions, supporting that passive sampling provides reasonable estimates of the time-weighted exposure concentration. Fifty volunteers were recruited and asked to wear the ePTFE PS while using an essential oil diffuser inside their own vehicle while commuting and in their studio apartment while sleeping. Terpene levels without an essential oil diffuser were very low in vehicles and 47, 3.6, 1.6, and 0.62 µg m-3 for average concentrations of limonene, α- and ß-pinenes, and γ-terpinene in studio apartments, respectively, close to those reported in previous studies. The indoor concentrations of all selected terpenes in vehicles and studio apartments were elevated by the use of essential oil diffusers, especially in vehicles. The average concentration of limonene in vehicles after the use of essential oil diffusers was 11 µg m-3, which was greater than that before use by a factor of 30. Therefore, cautious usage of essential oil diffusers indoors where the volume is limited, such as a vehicle, is needed to reduce exposure to terpenes.

Plants sense and integrate diverse stimuli to determine the timing for germination. A smoke compound, 3,4,5-trimethylfuran-2(5H)-one (trimethylbutenolide, TMB), has been identified to inhibit the seed germination of higher plants. To understand the mode of action, we examined various physiological and molecular aspects of the TMB-dependent inhibition of seed germination in Arabidopsis thaliana The results indicated that the effect of TMB is due to the enhanced physiological dormancy, which is modulated by other dormancy regulatory cues such as after-ripening, stratification, and ABA/GA signaling. In addition, gene expression profiling showed that TMB caused genome-wide transcriptional changes, altering the expression of a series of dormancy-related genes. Based on the TMB-responsive physiological contexts in Arabidopsis, we performed mutant screening to isolate genetic components that underpin the TMB-induced seed dormancy. As a result, the TMB-RESISTANT1 (TES1) gene in Arabidopsis, encoding a B2 group Raf-like kinase, was identified. Phenotypic analysis of the tes1 mutant implicated that TES1 has a critical role in the TMB-responsive gene expression and the inhibition of seed germination. Taken together, we propose that plants have been equipped with a TMB sensory pathway through which the TMB induces the seed dormancy in a TES1-dependent way.

The mental health and related quality of life of runaway adolescents are global public health issues. As few intervention studies have considered the family contexts of runaway adolescents, we aimed to develop an intervention tailored specifically to the needs of this population using an Intervention Mapping protocol. First, a literature review and interviews with runaway adolescents and youth shelter workers were conducted to create a logic model of the problem. Second, the behavioral and environmental outcomes were set to adapt to stressful situations and enable families to become more resourceful in dealing with family adversity, based on the results of needs assessment. Performance objectives and changeable determinants were also created by reviewing the pertinent theories and studies. Third, theory- and evidence-based methods to influence changes in the determinants were identified. Fourth, we designed an eight-session family-based mental health program incorporating individual and family approaches for runaway adolescents. Fifth, we determined that mental health nurses at community mental health centers linked to youth shelters would serve as the program implementers. Finally, we planned a randomized controlled trial to evaluate the effects of our program on improving runaway adolescents' mental health status and perceived family functioning.

We introduce UnaG as a green-to-dark photoswitching fluorescent protein capable of high-quality super-resolution imaging with photon numbers equivalent to the brightest photoswitchable red protein. UnaG only fluoresces upon binding of a fluorogenic metabolite, bilirubin, enabling UV-free reversible photoswitching with easily controllable kinetics and low background under Epi illumination. The on- and off-switching rates are controlled by the concentration of the ligand and the excitation light intensity, respectively, where the dissolved oxygen also promotes the off-switching. The photo-oxidation reaction mechanism of bilirubin in UnaG suggests that the lack of ligand-protein covalent bond allows the oxidized ligand to detach from the protein, emptying the binding cavity for rebinding to a fresh ligand molecule. We demonstrate super-resolution single-molecule localization imaging of various subcellular structures genetically encoded with UnaG, which enables facile labeling and simultaneous multicolor imaging of live cells. UnaG has the promise of becoming a default protein for high-performance super-resolution imaging.

1-Aminocyclopropane-1-carboxylic acid (ACC), a biosynthetic precursor of ethylene, has long been proposed to act as a mobile messenger in higher plants. However, little is known about the transport system of ACC. Recently, our genetic characterization of an ACC-resistant mutant with normal ethylene sensitivity revealed that lysine histidine transporter 1 (LHT1) functions as a transporter of ACC. As amino acid transporters might have broad substrate specificity, we hypothesized that other amino acid transporters including LHT1 paralogs might have the ACC-transporter activity. Here, we took a gain-of-function approach by transgenic complementation of lht1 mutant with a selected set of amino acid transporters. When we introduced transgene into the lht1 mutant, the transgenic expression of LHT2, but not of LHT3 or amino acid permease 5 (AAP5), restored the ACC resistance phenotype of the lht1 mutant. The result provides genetic evidence that some, if not all, amino acid transporters in Arabidopsis can function as ACC transporters. In support, when expressed in Xenopus laevis oocytes, both LHT1 and LHT2 exhibited ACC-transporting activity, inducing inward current upon addition of ACC. Interestingly, the transgenic expression of LHT2, but not of LHT3 or AAP5, could also suppress the early senescence phenotypes of the lht1 mutant. Taking together, we propose that plants have evolved a multitude of ACC transporters based on amino acid transporters, which would contribute to the differential distribution of ACC under various spatiotemporal contexts.

Light is an important environmental cue, causing a high degree of developmental plasticity in higher plants. The outcome of light-regulated developmental response is determined by not only photo-sensory systems but also endogenous physiological contexts in plants. KARRIKIN-INSENSITIVE2 (KAI2) functions as a receptor of karrikin and endogenous, as yet to be identified, KAI2 ligand (KL). The loss-of-function of KAI2 caused light-hyposensitive photomorphogenesis, affecting the expression light-responsive genes under the light conditions. However, it remains still unclear how KAI2-KL signaling interacts with light-signaling. Here, we show that the ply2 mutation, a severe loss-of-function allele of KAI2 affected the expression of a subset of light-responsive genes, irrespectively of light condition. The results implied that the overlapping set of light- and KAI2-responsive genes may serve as an integrating node between light- and KAI2-KL signaling. Further, the results of double mutant analyses between the ply2 mutant and mutants of CONSTITUTIVELY PHOTOMORPHOGENIC1 (COP1) or LONG HYPOCOTYL IN FAR-RED (HFR1) implicated that KAI2-KL signaling acts at downstream of COP1, largely independently of HFR1. Together, these results suggest that KAI2-KL signaling intersects with a subset of the light-regulatory network, by which plants adjust their photomorphogenic development.

Antibiotics in environment can be of concern as they can enter the food chain posing risks to ecosystems and human health. Photodegradation has been considered as a promising way of naturally degrading antibiotics in environment. Antibiotics are usually present in mixtures in environment; however, previous studies focused on individual compounds. Therefore, this study investigated the effect of UV irradiation on the degradation of tetracycline (TC) and sulfathiazole (STH) in individual solutions and mixtures. Under dark conditions, the initial masses of TC and STH were reduced by about 35% and 26%, respectively, over a 35 d-reaction period. With UV irradiation TC and STH were completely removed within 14 d and 35 d, respectively, regardless of the initial concentrations. Both the TC and STH removals were faster (i.e., 2-4 times) when they were in mixtures. This may be partly attributed to the byproducts such as sulfate that can promote indirect photolysis and partly to the enhanced hydrolysis due to changes in the solution pH. Overall, this study suggests that when photodegradation is used to remove antibiotics in water, the removal kinetics of antibiotics individually and in mixtures can be considered to develop more efficient treatment technologies.

We investigated the relation of sputtering powers with structural and morphological properties of nickel oxide (NiO) thin films. NiO thin films were fabricated by using an rf-reactive sputtering method on Si(100) substrates with a Ni target in a partial pressure of oxygen and argon. The films were deposited by various rf-sputtering powers from 100 to 200 W at room temperature. The phases and crystalline structures of the deposited films were investigated by using grazing incident X-ray diffraction (XRD). The thickness and surface morphology of the films were investigated by using a field emission-scanning electron microscopy (FE-SEM). The different sputtering conditions drastically affected the crystallinity and the surface morphology of NiO thin films. A combined analysis of the data obtained from X-ray diffraction and SEM images demonstrates that the preferred orientation of NiO films tends to grow from (111) to (200) direction as increasing the sputtering power, which can be explained by in terms of the surface energy along the indexing plane in an fcc structure. As increasing the rf power, lattice constants decreased from 4.26 to 4.20 angstroms and samples became high-quality crystals. Under our experimental condition, NiO films prepared at 150 W with 20% partial pressure of oxygen and 7 cm distance from the sample to the target show the best quality of the crystal.

When hydrogen atoms slowly diffuse into a metastable structure, a coherence length of thin TiZrNi quasicrystal metallic ribbons is increased. Improved atomic order with addition of hydrogen is an uncommon phenomenon in metals, and may reflect the unordinary structural property of quasicrystals. Metastable TiZrNi quasicrystals prepared by rapid quenching of molten ingots were hydrogenated in a low pressure of hydrogen (lower than 1000 Torr) at high temperature. After completion of the pressure-composition-temperature measurements between 200 to 300 degrees C, X-ray diffraction data revealed that the full width at half maximum values of the diffraction peaks were decreased suggesting that the quasicrystal phase may stabilized by hydrogen inclusion. Annealing of the same sample at 200 degrees C without hydrogen yields no change of the relative intensity of diffraction peaks.

Distinctive thin layers of TiZr and Ni were deposited by using a magnetron sputtering method and a thermal annealing was applied to discover metallic films of quasicrystals. After a heat treatment in vacuum, 70 nm thick deposited layers were well mixed with nominal compositions of 49.7, 29.3 and 21.0 for Ti, Zr and Ni, respectively, which is very close with the one forming a quasicrystalline phase. The magnetization values were significantly decreased from 0.286 to 0.142 emu/mm3 at 2000 Oe, after annealing, while a shape of magnetic hysteresis was maintained. It is believed that a different magnetic behavior after thermal annealing is due to the homogeneous mixing of atomic elements and possible existence of a metastable phase.
