RESUMO

Contamination of aquatic ecosystems can have cascading effects on terrestrial consumers by altering the availability and quality of aquatic insect prey. Comprehensive assessment of these indirect food-web effects of contaminants on natural resources and their associated services necessitates using both ecological and economic tools. In the present study we present an aquatic-terrestrial assessment tool (AT2), including ecological and economic decision trees, to aid practitioners and researchers in designing contaminant effect studies for linked aquatic-terrestrial insect-based food webs. The tool is tailored to address the development of legal claims by the US Department of the Interior's Natural Resource Damage Assessment and Restoration Program, which aims to restore natural resources injured by oil spills and hazardous substance releases into the environment. Such cases require establishing, through scientific inquiry, the existence of natural resource injury as well as the determination of the monetary or in-kind project-based damages required to restore this injury. However, this tool is also useful to researchers interested in questions involving the effects of contaminants on linked aquatic-terrestrial food webs. Stylized cases exemplify how application of AT2 can help practitioners and researchers design studies when the contaminants present at a site are likely to lead to injury of terrestrial aerial insectivores through loss of aquatic insect prey and/or dietary contaminant exposure. Designing such studies with ecological endpoints and economic modeling inputs in mind will increase the relevance and cost-effectiveness of studies, which can in turn improve the outcomes of cases and studies involving the ecological effects of contaminants on food webs. The US Environmental Protection Agency's short-term freshwater effluent test methods include a fish (Pimephales promelas), a cladoceran (Ceriodaphnia dubia), and a green alga (Raphidocelis subcapitata). There is a recognized need for additional taxa to accompany the three standard species for effluent testing. An appropriate additional taxon is unionid mussels because mussels are widely distributed, live burrowed in sediment and filter particles from the water column for food, and exhibit high sensitivity to a variety of contaminants. Multiple studies were conducted to develop a relevant and robust short-term test method for mussels. We first evaluated the comparative sensitivity of two mussel species (Villosa constricta and Lampsilis siliquoidea) and two standard species (P. promelas and C. dubia) using two mock effluents prepared by mixing ammonia and five metals (cadmium, copper, nickel, lead, and zinc) or a field-collected effluent in 7-day exposures. Both mussel species were equally or more sensitive (more than two-fold) to effluents compared with the standard species. Next, we refined the mussel test method by first determining the best feeding rate of a commercial algal mixture for three age groups (1, 2, and 3 weeks old) of L. siliquoidea in a 7-day feeding experiment, and then used the derived optimal feeding rates to assess the sensitivity of the three ages of juveniles in a 7-day reference toxicant (sodium chloride [NaCl]) test. Juvenile mussels grew substantially (30%-52% length increase) when the 1- or 2-week-old mussels were fed 2 ml twice daily and the 3-week-old mussels were fed 3 ml twice daily. The 25% inhibition concentrations (IC25s) for NaCl were similar (314-520 mg Cl/L) among the three age groups, indicating that an age range of 1- to 3-week-old mussels can be used for a 7-day test. Finally, using the refined test method, we conducted an interlaboratory study among 13 laboratories to evaluate the performance of a 7-day NaCl test with L. siliquoidea. Eleven laboratories successfully completed the test, with more than 80% control survival and reliable growth data. The IC25s ranged from 296 to 1076 mg Cl/L, with a low (34%) coefficient of variation, indicating that the proposed method for L. siliquoidea has acceptable precision. The Clinch and Powell Rivers (Virginia, USA) support diverse mussel assemblages. Extensive coal mining occurs in both watersheds. In large reaches of both rivers, major ion concentrations are elevated and mussels have been extirpated or are declining. We conducted a laboratory study to assess major ion effects on growth and survival of juvenile Villosa iris. Mussels were exposed to pond water and diluted pond water with environmentally relevant major ion mixtures for 55 days. Two treatments were tested to mimic low-flow concentrations of Ca(2+), Mg(2+), [Formula: see text] , [Formula: see text] , K(+) and Cl(-) in the Clinch and Powell Rivers, total ion concentrations of 419 mg/L and 942 mg/L, respectively. Mussel survival (>90%) and growth in the two treatments showed little variation, and were not significantly different than in diluted pond water (control). Results suggest that major ion chronic toxicity is not the primary cause for mussel declines in the Clinch and Powell Rivers.

This project examined the impact of long-term poultry litter application on the chemical signatures of As, Cu, Zn, and P in stream sediments of the Broadkill River watershed within the Delmarva Peninsula, a region of intense poultry production. Thirty-seven sediment samples were collected from Broadkill River drainage systems and analyzed for litter-derived elements (As, Cu, Zn, P) and basic soil parameters such as particle size distribution, organic matter, and soluble salts. Results showed that concentrations of elements in stream sediments are approximately log-normally distributed. Spatial variability in concentrations of elements was evident, with most elements increasing in concentration and enrichment from upgradient headwaters to downgradient reaches draining predominantly agricultural areas. Results of correlation analyses showed positive significant correlation among elements; elements were also positively correlated with percent clay and silt in the sediment. Using GIS maps with overlays of hydrology and land use activities, statistical correlations between As, Cu, Zn, and P enrichment factors and land use were examined. Results showed statistically significant relationships between As, Mn, and Zn enrichment factors and residential areas within the watershed, but did not show a statistically significant relationship between element enrichment factors and agricultural land use. Factors that complicate this type of landscape-scale study include the presence of poultry processing plants, impoundments, changes in land use over time, and the influence of tides, all of which can have direct and indirect influences on element mobility.

Alternative brominated flame-retardants (BFRs), 2-ethylhexyl 2,3,4,5-tetrabromobenzoate (TBB), 2-ethylhexyl 2,3,4,5-tetrabromophthalate (TBPH), 1,2-bis(2,4,6-tribromophenoxy) ethane (BTBPE) and decabromodiphenyl ethane (DBDPE), are now being detected in the environment. However, contaminant bioavailability is influenced by the organisms' ecology (i.e., route of uptake) and in situ environmental factors. We observed that the filter-feeding bivalve (Corbicula fluminea) and grazing gastropod (Elimia proxima), collected downstream from a textile manufacturing outfall, exhibited TBB, TBPH, and BTBPE concentrations from 152 to 2230 ng g(-1) lipid weight (lw). These species also contained additional BFRs. Maximum levels of total hexabromocyclododecane diastereomers (∑HBCDs) in these species were 363,000 and 151,000 ng g(-1) lw, and those of polybrominated diphenyl ethers (∑PBDEs) were 64,900 and 47,200 ng g(-1) lw, respectively. These concentrations are among the highest reported to date worldwide. While BDE-209 was once thought to be nonbioavailable and resistant to degradation, it was the dominant BFR present and likely debromination products were detected. Contributions of α- and ß-HBCD were higher in tissues than sediments, consistent with γ-HBCD bioisomerization. Mollusk bioaccumulation factors were similar between HBCD and PBDEs with 4 to 6 bromines, but factors for TBB, TBPH, and BTBPE were lower. Despite different feeding strategies, the bivalves and gastropods exhibited similar BFR water and sediment accumulation factors.

Trace elements used in animal feed additives can be introduced to aquatic environments through application of manures from animal feeding operations to agricultural land as fertilizer. The use of poultry feed additives containing arsenic (As) is of particular concern in the Shenandoah River watershed (Virginia, USA), an agricultural landscape with a high density of poultry operations. This study investigated the relationship between watershed characteristics of Shenandoah River tributaries and trace element concentrations in streambed sediment and tissue of resident mollusks, including: Asian clams (Corbicula fluminea), which are commonly used biomonitors, and pleurocerid snails (Leptoxis carinata), which are generally understudied. Results failed to support the primary hypothesis of a predictive relationship between watershed densities of poultry operations and As concentrations in sediment and mollusk tissue. However, there were statistical relationships between land use in tributary watersheds and other trace elements in sediment (Cu, Mn, Pb, Zn) and tissue (Cd, Hg, Pb). Principal components analysis of the sediment data suggested a possible geologic source of As at some sites. Tissue concentrations of As were significantly higher in snails than in clams, but clams accumulated higher concentrations of other trace elements (Cd, Cr, Hg, Pb, Se). Snails may be useful biomonitors of environmental As, but appear to be less suitable than clams for studies of landscape sources of other trace elements.

Application of manures from animal feeding operations (AFOs) as fertilizer on agricultural land can introduce nutrients and hormones (e.g. estrogens) to streams. A landscape-scale study was conducted in the Shenandoah River watershed (Virginia, USA) in order to assess the relationship between densities of AFOs in watersheds of agricultural streams and in-stream nutrient concentrations and estrogenic activity. The effect of wastewater treatment plants (WWTPs) on nutrients and estrogenic activity was also evaluated. During periods of high and low flow, dissolved inorganic nitrogen (DIN) and orthophosphate (PO(4)-P) concentrations were analyzed and estrogens/estrogenic compounds were extracted and quantified as17ß-estradiol equivalents (E2Eq) using a bioluminescent yeast estrogen screen. Estrogenic activity was measurable in the majority of collected samples, and 20% had E2Eq concentrations >1 ng/L. Relatively high concentrations of DIN (>1000 µg/L) were also frequently detected. During all sampling periods, there were strong relationships between watershed densities of AFOs and in-stream concentrations of DIN (R(2) = 0.56-0.81) and E2Eq (R(2) = 0.39-0.75). Relationships between watershed densities of AFOs and PO(4)-P were weaker, but were also significant (R(2) = 0.27-0.57). When combined with the effect of watershed AFO density, streams receiving WWTP effluent had higher concentrations of PO(4)-P than streams without WWTP discharges, and PO(4)-P was the only analyte with a consistent relationship to WWTPs. The results of this study suggest that as the watershed density of AFOs increases, there is a proportional increase in the potential for nonpoint source pollution of agricultural streams and their receiving waters by nutrients, particularly DIN, and compounds that can cause endocrine disruption in aquatic organisms.

Polybrominated diphenyl ether (PBDE) flame retardants have become distributed ubiquitously in the environment. High concentrations have been reported in U.S. sewage sludge (biosolids). The burgeoning practice of land-applying biosolids as fertilizer creates an avenue for reintroduction of PBDEs to surface waters and aquatic sediments. Bioavailability of biosolids- and sediment-associated PBDEs was assessed using the freshwater oligochaete, Lumbriculus variegatus. Oligochaetes were exposed to composted biosolids (1,600 ng/g total PBDEs) and artificial sediment spiked with penta- and deca-brominated diphenyl ether (BDE) formulations (1,300 ng/g total PBDEs). Uptake (28-d exposure) and depuration (21 d) of eight congeners were studied. Polybrominated diphenyl ethers in both substrates were bioavailable, but bioaccumulation was 5 to 10 times greater from spiked artificial sediment. The congeners BDE 47 and BDE 99 were the most prevalent congeners in oligochaetes after exposure. Congener BDE 47 was more bioaccumulative, possibly due to the threefold greater depuration rate of BDE 99. Bioaccumulation of penta- and hexa-brominated congeners appeared to be affected more strongly by substitution pattern than degree of bromination. Uptake of BDE 209, the dominant congener in deca-BDE, was minimal. Accumulation of certain PBDE congeners from biosolids and sediments by benthos provides a pathway for transfer to higher trophic levels, and congener discrimination may increase with each trophic transfer.
