Nowadays, different systems for reducing pesticides in table grapes are being tested at different production stages either in the field or in postharvest. The present study tested ozonated water treatments at the beginning of the cold storage of the Princess® seedless table grape variety to reduce the residue contents of some pesticides and to evaluate their effect on gray mold and the berry microbiome. An ozone generator capable of producing an ozone concentration ranging from 18 to 65 Nm3 was utilized for obtaining three ozone concentration levels in water: 3, 5 and 10 mg/L. Ozonated water was placed in a 70 L plastic box where 500 g grape samples closed in perforated plastic clamshell containers were immersed utilizing two washing times (5 and 10 min). Overall, six ozonated water treatments were tested. After the ozonated water treatments, all samples were stored for 30 days at 2 °C and 95% relative humidity to simulate commercial practices. The pesticide residue contents were determined before the ozonated water treatments (T0) and 30 days after the cold storage (T1). The treatments with ozonated water washing reduced the pesticide residues up to 100%, while the SO2 control treatment reduced the pesticide residues ranging from 20.7 to 60.7%. Using 3 mg/L ozonated water to wash grapes for 5 min represented the optimal degradation conditions for all of the analyzed pesticides, except for fludioxonil, which degraded better with a washing time of 10 min. The ozone treatments did not significantly reduce the gray mold and the fungal and bacterial microbiome, while a relevant reduction was observed in the yeast population.

BACKGROUND: 'Sangiovese' (Vitis vinifera L.) is a well-known Italian wine grape utilized for producing the prestigious Chianti Classico brand. Here, 'Sangiovese' is generally trained to a vertical training system, with an imposed crop level of around 3-5 kg per vine to preserve grape quality. Tendone comprises a horizontal trellis system, largely diffused in Mediterranean Countries for table grape production, and is adopted to achieve a high yield and to preserve grapes, by means of its canopy architecture, from excessive solar radiation, which in turn affects the organoleptic aspects of grapes and wines. The present study aimed to evaluate, in terms of phenolic and volatile compounds of grape and wines, 'Sangiovese' trained to Tendone, as affected by two crop levels, namely very high and reduced yields. RESULTS: Reduced yield (18.39 kg per vine) by cluster thinning still appeared to provide a considerable yield and quality of grapes and wines. Thinning increased pH, tartaric acid content, flavonoids, anthocyanins, total polyphenols and proanthocyanins, and total free and bound aroma compounds, without affecting soluble solids and titratable acidity of grapes. As regards wines, reduced yield increased pH, tartaric and malic acids and dry reduced extract, flavonoids, anthocyanins, total polyphenols and proanthocyanins, and, finally, the color intensity of wines. CONCLUSION: The present study highlights that Tendone training systems, combined with 40% of cluster thinning, allows a high yield to be reached, allowing, at the same time, a very interesting quality of grapes, making it possible to produce qualitative wines. © 2022 Society of Chemical Industry.

This study aimed to assess the impact of two soil managements and training systems on yield and wine aroma compounds of Negroamaro variety grown in a warm climate region (southern Italy). Cover crop (CC) and soil tillage (ST) as soil management, whilst bilateral Guyot (BG) and monolateral Guyot (MG) as training systems were compared. Free and bound volatile fractions were evaluated by GC-MS. ST and CC as well as BG and MG significantly affected yield parameters. In particular, yield was higher in ST and BG than in CC and MG, respectively; moreover, it was found to be positively influenced by interaction between BG and ST. Regarding aroma compounds, significant interactions between soil management and training system factors were observed. In case of free volatiles, the most positive interaction was found between BG and ST, whereas, for bound volatiles, the best interaction was represented by MG with both soil tillage and cover crop. Vine leaf area and development over vine growth stages along with water stress levels played an important role in determining the aroma profile as well as yield parameters. In conclusion, the training system significantly interacted with soil management and affected most of important aroma compounds in Negroamaro wine.

The present geochemical study concerns the impact of viticultural practices in the chemical composition of the grape cultivar "Negroamaro" in Apulia, a southern Italian region renowned for its quality wine. Three types of soil management (SM), two cover cropping with different mixtures, and a soil tillage were considered. For each SM, the vines were irrigated according to two irrigation levels. Chemical composition of soil and of berries of Vitis vinifera cultivar "Negroamaro" were analyzed by X-ray fluorescence, inductively coupled plasma-mass spectrometry and multivariate statistics (linear discrimination analysis). In detail, we investigated major and trace elements behavior in the soil according to irrigation levels, the related index of bioaccumulation (BA) and the relationship between trace element concentration and soil management in "Negroamaro" grapes. The results indicate that soil management affects the mobility of major and trace elements. A specific assimilation of these elements in grapes from vines grown under different soil management was confirmed by BA. Multivariate statistics allowed to associate the vines to the type of soil management. This geochemical characterization of elements could be useful to develop fingerprints of vines of the cultivar "Negroamaro" according to soil management and geographical origin.

Metabolic profiles of 32 Negroamaro red wines were analysed using (1)H NMR spectroscopy and multivariate statistical analyses (Principal Component Analysis, PCA, and Orthogonal Partial Least Square Discriminant Analysis, OPLS-DA). Among winemaking technologies three were compared: ultrasounds (U; 12 samples), cryomaceration using dry ice (C; 12 samples) and traditional (T; 8 samples). Moreover, each vinification technology was used for grapes grown by two different soil management practices, soil tillage (ST; 16 samples) and cover crop (CC; 16 samples), and by two different training systems, monolateral (M; 16 samples) and bilateral Guyot (B; 16 samples). All statistical models applied on NMR data revealed a good separation between ST (soil tillage) and CC (cover crop), showing a higher influence of the soil management practices compared to the winemaking technologies (ultrasound, cryomaceration and traditional). The differentiation among samples, due to soil management practices, was mainly caused by metabolites such as glycerol, 2,3-butanediol, malic acid, α/ß-glucose and phenolic compounds, such as tyrosine and caffeic acid.

This study aims to assess the effect of three wine grape varieties, three training systems and two bud loads on the Total Antioxidant Capacity (TAC) and polyphenolic composition of Southern Italy red wines produced, during two vintages. Overall, Primitivo, Malvasia nera of Brindisi-Lecce and Montepulciano as grape varieties, single Guyot (SG), single spur pruned low cordon (SLC) and single spur pruned high wire cordon (HSLC) as training systems, 8 and 12 buds/plant as bud loads were compared. Significant differences in the polyphenolic families were shown by the grape varieties and by modifying the vine growing practices. Moreover, the results demonstrated that varieties influenced the TAC (indicating the Malvasia as the more effective one), that SLC led to the lowest level of TAC and that 8 buds/plant increased it. The relationship between antioxidant indexes and the concentration of single polyphenolic families was evaluated and the highest correlation was found between the total polyphenols and the proanthocyanidins family.

RATIONALE: Due to their health benefits, there is growing interest in the production and use of carotenoids from natural sources, e.g. microalgae. To date, only Haematococcus pluvialis and Dunaliella, that accumulate, respectively, astaxanthin and ß-carotene in large quantities, are grown commercially. However, interest is also being focused on other xanthophylls, such as (all-E)-fucoxanthin characterized by anti-obesity and anti-carcinogenic effects. In this regard, rigorous chemical and analytical techniques following preparative isolation of components are needed to unequivocally identify individual carotenoids in microalgae. METHODS: The carotenoid profile of Isochrysis sp. biomass, produced in closed photobioreactors, was determined by reversed-phase C30 (RP-30) high-performance liquid chromatography coupled with diode-array detector mass spectrometry using positive electrospray ionization (HPLC/DAD-MS/ESI(+) ) analysis. Additionally, multistage mass spectrometry (MS(n) ) analyses, together with fine structures of the UV-vis spectra, were used to differentiate structural and geometrical isomers. RESULTS: This technique allowed the simultaneous determination of geometrical, isomers of fucoxanthin (all-E-fucoxanthin, 13Z-, 13'Z- and 9'Z-fucoxanthin), diatoxanthin and 5,8-epoxydiadinoxanthin diasteroisomers (R/S). The analyzed extracts contained fucoxanthin isomers as the major carotenoids and, in particular, (all-E)-fucoxanthin was the main geometrical isomer (~85%) found at a concentration of 17 mg/g of the lyophilized biomass. CONCLUSIONS: Considering the high content of fucoxanthin in Isochrysis sp. biomass, the microalga could be proposed as a source of this compound for nutraceutical and pharmaceutical applications.

Negroamaro is an autochthonous wine grape variety of Southern Italy, which is becoming very important for the Italian wine market. The wine aroma is primary affected by the chemical composition of grapes, which can be influenced also by agronomic practices such as soil management. In this study, the free and bound aroma characterization was performed by gas chromatography-mass spectrometry analyses, and the influence of two soil managements (cover cropping and soil tillage) was evaluated. A total of 40 volatile compounds were observed in the wine samples. Alcohols (55.7 mg/L), fatty acids (7.0 mg/L) and esters (6.6 mg/L) were found as the main classes in Negroamaro wine. The results showed that the aroma composition of Negroamaro wine was positively affected by soil tillage probably because of the higher water stress (ψ(s)) recorded in the vines from this treatment. Indeed, among the free volatile compounds, higher contents of esters, carboxylic acids, alcohols, phenolics and acetamides together with lower contents of sulfurs compounds were found in soil tillage wine. Conversely, no difference was observed in glycoside volatile compounds.

UNLABELLED: Flavonoids present in skin extracts of red seedless table grape varieties Summer Royal, Autumn Royal, and Crimson, and white seedless varieties Carati and Thompson were analyzed by HPLC-DAD-MS, in 3 y of study (2006 to 2008). The anthocyanins, delphinidin-3-O-glucoside, cyanidin-3-O-glucoside, petunidin-3-O-glucoside (with their corresponding p-coumaroyl derivatives), peonidin-3-O-glucoside, and malvidin-3-O-glucoside (with their corresponding acetyl, caffeoyl, and p-coumaroyl derivatives) were found. In addition the flavonols quercetin-3-O-glucuronide, quercetin-3-O-rutinoside, quercetin-3-O-glucoside, kaempferol-3-O-glucoside, kaempferol-3-O-galactoside, and the flavan-3-ols procyanidin B1, procyanidin B2, and catechin were also detected. Anthocyanins were the main flavonoids in red grapes ranging from 24 (Crimson) to 500 (Summer Royal) mg/kg fresh weight of grapes; consistent levels of flavonols and flavan-3-ols were also quantified in all varieties. To determine the effective climatic influence on flavonoids content in field conditions, viticultural practices have been developed, that could exclude the effects of direct solar radiation from confounding the assessment of those related to thermal conditions alone. A strong positive correlation was determined between flavonoids and temperature data that seem to be responsible for the difference of these metabolites along the years; furthermore, it has been possible to define a linear relationship (R(2) = 0.6871, P = 0.0057) between thermal amplitude and total flavonoids values in the red grapes. PRACTICAL APPLICATION: Grapes are economically the most important fruit species in the world and approximately 30% of its production is used as fresh fruit. Because of the very important role of flavonoids in food quality as well as their health-promoting properties, and considering that our experiments were performed along 3 consecutive years, gathered results in this research are quite promising to give a useful information on the flavonoid contents and their evolution in 5 seedless table grapes that are widespread in Mediterranean regions but also in California and South America, and are grown in a viticultural climate (Apulia, South Italy) very close to some regions of Spain, Turkey, Tunisia, and Israel.

To determine a correlation between carotenoid precursors in grapes and norisoprenoid varietal aroma of wine, carotenoids were identified and quantified by HPLC-DAD-MS (ESI+) from four representative wine grape varieties of the Apulian region (Chardonnay, Merlot, Negroamaro, Primitivo) in two years of study (2006-2007), and C13-norisoprenoid aroma potential, DeltaC (microg/kg), was calculated from the difference of total carotenoid concentration between veraison and maturity. C13-norisoprenoids were analyzed by GC-MS in the obtained wines from 2006 and 2007 vintages. Higher DeltaC values, found in Chardonnay and Merlot grapes, corresponded to higher norisoprenoid contents in the respective wines, particularly characterized by highly flavorant compounds such as beta-damascenone and 3-oxo-alpha-ionol. A linear regression was determined that was significant at the 0.01% level (F=36.12, p=0.00096) with R=0.9261, between grape DeltaC values and total norisoprenoid contents in wine. These findings support the hypothesis that DeltaC could be a useful technological tool to predict norisoprenoid aroma of wine and, consequently, to identify grapes with higher aroma potential.

Carotenoid compounds in wine grapes (Chardonnay, Merlot, Primitivo, Negroamaro) grown in Apulian region were investigated by chromatographic and spectrometric analyses. Cis-isomers of lutein and beta-carotene (9Z, 9'Z-lutein and 9Z-beta-carotene) and 5,6-epoxyxanthophylls were detected: 9'Z-neoxanthin, violaxanthin, and 5,6-epoxylutein. Moreover, zeaxanthin was efficiently resolved from lutein by a selective factor > 1 (alpha= 1.06) and was found in high amounts (50 to 300 microg/kg) in the grape extracts analyzed in 3 y of study (2006 to 2008). At grape maturity, beta-carotene had concentration approximately 2-to 4-fold higher than (all-E)-lutein in all varieties. Because carotenoids are potential precursors of aroma compounds, it was determined carotenoids change DeltaC (microg/kg), from the difference of total carotenoids concentration between veraison and maturity. Chardonnay and Merlot had the highest DeltaC values and principal component analysis showed that they were characterized by 5,6-epoxyxanthophylls derivatives and zeaxanthin, lutein, and beta-carotene derivatives, respectively. An important effect of vintage on DeltaC values in the analyzed grapes was also observed. A strong positive correlation was determined between DeltaC and temperature data that seem to be responsible for the difference of DeltaC in the Chardonnay and Merlot compared to the Primitivo and Negroamaro varieties.
