RESUMO

The small-spotted catshark (Scyliorhinus canicula) is a bottom-dwelling elasmobranch that represents the most discarded catch in terms of biomass in the Catalan coast (NW Mediterranean). Potential impacts affecting its population and food safety implications have been assessed in three localities along the Catalan coast. Distinct indicators were integrated, such as biological data, ingested anthropogenic items (plastic and cellulose-like items), parasitological indices, trace metal concentrations and histopathology using liver as target organ. Although high ingestion rates of fibres and levels of some heavy metals, they do not seem negatively affected by any major pathology nor by the current levels of pollutants. Small-scale differences among localities and depths were found and discussed. No zoonotic parasites were found. Encysted larvae of Grillotia adenoplusia and, above all, the levels of Hg found in the musculature, that are well over the European Commission limits, rise concerns regarding human consumption of S. canicula in this region.

Endolimax is a genus of intestinal amoebae which stands among the least known human protists. Previous studies on amoebic systemic granulomatosis of a marine fish (Solea senegalensis) resulted in the unexpected characterization of a new organism which was related to Endolimax and named E. piscium. The existence of multiple reports of systemic granulomatosis caused presumptively by unidentified amoebae in goldfish lead us to investigate the organism involved in goldfish disease. Analysed goldfish presented small whitish nodules in the kidney, which correspond to chronic granulomatous inflammatory reactions with a ring-layer of amoebae in the periphery. Amoebae were amitochondriate and were located in a parasitophorous vacuole within macrophages, as previous studies on this condition in goldfish and other freshwater fish pointed out. SSU rDNA characterization confirmed a new Endolimax lineage which appears closely related to E. piscium, but the molecular evidence, distinct pathological features and lack of ecological overlapping between the hosts support their assignment to a new species, E. carassius. The results support the existence of a considerable unexplored diversity of Endolimax spp. among fish, and their proper characterization can contribute to an understanding of Archamoebae evolution and pathogenic potential.

Prevalence, abundance, concentration, size and composition of anthropogenic items (AIs) (synthetic and non-synthetic) ingested by Merluccius merluccius juvenile specimens and from near-bottom water samples from different localities off the Catalan coast (NW Mediterranean), were characterized. The potential effect of AIs on fish condition was assessed through different health indicators. Virtually all AIs found in fish and near-bottom water samples were fibres. A mean of 0.85 fibres/m3 from the surrounding water was observed. Fish ingested a mean of 1.39 (SD = 1.39) items/individual. Cellulosic fibres were predominant (77.8% of samples), except for Barcelona. No differences in ingested AIs abundance and composition off Barcelona between 2007 and 2019 were found. Small AIs from the environment matched ingested AIs composition. Hakes did not ingest large fibres despite being present in the environment, probably due to their feeding behaviour. No adverse health effects or parasites aggregations were detected to be potentially related to AIs ingestion.

The European anchovy (Engraulis encrasicolus) is a small pelagic fish with an outstanding commercial value supporting important fisheries and is a key component of pelagic ecosystems in the Mediterranean Sea. Progressive reductions in the population size of this species has been observed in the Mediterranean Sea during recent decades, accompanied by a decline in the body condition, as well as the size/age of maturation. Nonetheless, the health status has not been yet assessed using a holistic approach. Herein, we analyse the health status of the European anchovy, integrating distinct indicators from fish condition, enzymatic biomarkers, presence of tissue alterations, and parasite descriptors. In addition, we analyse the presence of anthropogenic items (AIs) in the digestive tract of fish and their potential impact on health status. Additionally, we assess the differences between current AIs values and those recorded over 12 years ago. None of the health indicators studied provided evidence of relevant pathologic conditions affecting this fish species in the studied area. However, changes in the pattern of liver parenchyma were found. Compared with anchovy populations from other distribution areas, no zoonotic parasites were recorded in this study, demonstrating a reduced risks associated with foodborne transmission to humans. AIs, such as fibres and plastic particles, were found in the digestive tract of half of the fish analysed. A significant increase was detected in AIs prevalence between 2007 (40 %) and 2019 (70 %), alongside differences in the abundance and typology of the AIs, though this does not seem to have impacted fish health yet. Therefore, our work underscores the importance of implementing a regular program to monitor the health status of this key species to better understand population dynamics and their drivers.

Viral nervous necrosis (VNN) is the most important viral disease affecting farmed fish in the Mediterranean. VNN can affect multiple fish species in all production phases (broodstock, hatchery, nursery and ongrowing) and sizes, but it is especially severe in larvae and juvenile stages, where can it cause up to 100% mortalities. European sea bass has been and is still the most affected species, and VNN in gilthead sea bream has become an emerging problem in recent years affecting larvae and juveniles and associated to the presence of new nervous necrosis virus (NNV) reassortants. The relevance of this disease as one of the main biological hazards for Mediterranean finfish farming has been particularly addressed in two recent H2020 projects: PerformFISH and MedAID. The presence of the virus in the environment and in the farming systems poses a serious menace for the development of the Mediterranean finfish aquaculture. Several risks associated to the VNN development in farms have been identified in the different phases of the farming system. The main risks concerning VNN affecting gilthead seabream and European seabass have been identified as restocking from wild fish in broodstock facilities, the origin of eggs and juveniles, quality water supply and live food in hatcheries and nurseries, and infected juveniles and location of farms in endemic areas for on-growing sites. Due to the potential severe impact, a holistic integrated management approach is the best strategy to control VNN in marine fish farms. This approach should include continuous surveillance and early and accurate diagnosis, essential for an early intervention when an outbreak occurs, the implementation of biosecurity and disinfection procedures in the production sites and systematic vaccination with effective vaccines. Outbreak management practices, clinical aspects, diagnostic techniques, and disinfections methods are reviewed in detail in this paper. Additionally, new strategies are becoming more relevant, such as the use of genetic resistant lines and boosting the fish immune system though nutrition.

The present study provides a baseline assessment of the prevalence and densities of splenic melanomacrophage centres (MMCs) in 18 fish species from the NW Mediterranean Sea related to spatiotemporal and environmental factors and fish traits. Their correlation with other established health indicators, such as body condition indices (condition factor, hepatosomatic and gonadosomatic indices), parasite community descriptors and histological assessment of target organs (gills, liver and spleen) is also assessed. Despite MMCs variability is mainly attributed to the species identity and fish size, their potential use as generic biomarkers of health condition is pointed out for certain species (e.g. Spicara maena and Micromesistius poutassou) in which an increased response was identified, and whose potential drivers are discussed. Most importantly, present results provide a comprehensive assessment of MMCs in the fish community for future studies in the area.

Many studies have been carried out along mighty rivers with heavily industrialized watersheds to evaluate pollutants and their effects on freshwater organisms. However, their impact on marine organisms is virtually unknown. In order to cover this gap, Solea solea, one of the most important commercial fish species, together with sediments, were sampled during 2013-2015 offshore from the Ebro Delta river mouth. Fish health indicators (condition indices, histological tissue alterations, and parasite descriptors) were used to assess the potential effect of pollutants, an issue of particular interest in the area following the dredging activities taking place in the river upstream in 2013. No major histopathological alterations were detected, but perivascular inflammatory foci (PIF) were frequently observed, especially in 2014. The most prevalent and abundant parasites were acanthocephalans and digeneans within the digestive tract and copepods on the gills. Levels of trace metals from sediments and fish muscle were below the effects range median and reference levels accepted for human consumption, respectively. However, the lower levels of the hepatosomatic index, higher numbers of PIF, and variations in the abundance of parasites in 2014 and 2015 could suggest a pollutant exposure during these years. These results warn signs of toxicity, which could be associated with sediment leaks during the dredging activities.

Although the ingestion of plastics has been reported in a wide variety of organisms, there remains a lack of knowledge regarding the extent of spatial and temporal gradients and no consensus concerning the definition of monitor species for benthic marine environments. The present study aims at demonstrating the correlation between the presence of tangled balls of fibres and high levels of total plastic fibre ingestion in Nephrops norvegicus to assess the potential use of the prevalence of tangled balls as indicators of fibre pollution. To do so, the presence of plastics in stomach contents from several European populations of N. norvegicus is characterized in detail, including size distribution and polymer composition, and then its correlation with the prevalence of balls tested. Our results demonstrate that the prevalence of balls (>20%) is significantly correlated to higher levels of plastic ingestion, regardless of polymer composition and size distribution of ingested fibres. Plastic fibre ingestion levels across wide temporal and geographical scales are assessed using only the prevalence of balls, highlighting areas of increased fibre ingestion (e.g., the Gulf of Cadiz, N Barcelona) and areas of potentially lower fibre pollution (e.g., the Ebro Delta, the NW Iberian margin). Moreover, the relationship between the prevalence of balls and diet composition is analysed to discard a potential relationship with geographical differences in diet habits. Finally, we discuss the benefits and drawbacks of the use of the prevalence of tangled balls of fibres as an affordable, cost-effective and easy to implement indicator of fibre pollution for monitoring purposes in this species.

We make new morphological observations not previously reported for the old acanthocephalan Macracanthorhynchus hirudinaceus (Pallas, 1781) Travassos, 1917, described for the first time about 240 years ago. Our specimens were collected from the wild boar, Sus scrofa Linn., in Ukraine in 2005. We provide comparative morphometrics with other populations from Ukraine, Union of Soviet Socialist Republics, Europe, Brazil and the United States of America. Our specimens from the Ukraine had the smallest trunk (110-120 mm long), longest and thickest hooks, with the third hook being the largest, largest eggs, and a proboscis wider than long. We document the morphology of the proboscis, apical organ, hooks, hook roots, sensory pores, micropores, and eggs with scanning electron microscopy for the first time. We also provide chemical analysis of hooks and eggs using energy-dispersive X-ray analysis, discuss its micropores, and provide a new molecular profile based on 18S rDNA from a European population for the first time. Edge of hook tips feature high levels of calcium and phosphorous but the high level of sulphur is mostly found in the cortical layer of eggs. One new partial 18S rDNA sequence (482 nt length) was generated from an adult specimen of M. hirudinaceus. We present the first 18S rDNA published sequence for this cosmopolitan acanthocephalan obtained from Europe. The amplified region corresponded to the approximate middle region of the small subunit ribosomal ribonucleic acid gene, which is ~1800 nt in length. This molecular contribution is especially valuable in light of the extreme scarcity of genetic information about species of Macracanthorhynchus and of the family Oligacanthorhynchidae as a whole.

Parasitic and amphizoic amoebae are ubiquitous and can affect a huge variety of hosts, from invertebrates to humans, and fish are not an exception. Most of the relationships between amoebae and fish are based on four different types: ectocommensals, ectoparasites, endocommensals and endoparasites, although the lines between them are not always clear. As ectocommensals, they are located specially on the gills and particularly the amphizoic Neoparamoeba perurans is the most relevant species, being a real pathogenic parasite in farmed salmon. It causes amoebic gill disease, which causes a progressive hyperplasia of epithelial cells in the gill filaments and lamellae. Nodular gill disease is its analogue in freshwater fish but the causative agent is still not clear, although several amoebae have been identified associated to the lesions. Other species have been described in different fish species, affecting not only gills but also other organs, even internal ones. In some cases, species of the genera Naegleria or Acanthamoeba, which also contain pathogenic species affecting humans, are usually described affecting freshwater fish species. As endocommensals, Entamoebae species have been described in the digestive tract of freshwater and marine fish species, but Endolimax nana can reach other organs and cause systemic infections in farmed Solea senegalensis. Other systemic infections caused by amoebae are usually described in wild fish, although in most cases these are isolated cases without clinical signs or significance.

PURPOSE: Echinorhynchus gadi is one of the most widely distributed and commonly described acanthocephalans in marine fishes throughout the world. We provide a detailed morphometric and molecular description of a distinct Alaska population collected from the Pacific halibut Hippoglossus stenolepis Schmidt (Pleuronectidae) compared to those from other hosts and regions, illustrating new features never previously reported. METHODS: We described new specimens by microscopical studies, augmented by SEM, Energy Dispersive x-ray and molecular analyses, and histopathology. RESULTS: Specimens from Alaska were distinguished from those collected from the other geographical areas in proboscis size and its armature, especially number of hook rows and hooks per row, and length of hooks. The size of the receptacle, lemnisci, and reproductive structures in some other collections also varied from the Alaska material. X-ray scans of the gallium cut hooks depict prominent layering with high Sulfur content for tip cuts and increased calcium and phosphorus content in the base area of the hook. Sections of E. gadi specimens in the host tissue show prominent hook entanglement with subsequent connective tissue invasion also depicting the internal anatomy of certain worm structures not readily seen by other means. Molecular analyses clearly confirmed the identity of our E. gadi sequences. CONCLUSION: Our Alaska population of the E. gadi complex appears to represent a novel population distinguishable by its distinct morphometrics, geography and host species. We further establish new information on the Energy Dispersive X-ray analysis in our Alaska material for future comparisons with the other siblings and explore genetic relationships among echinorhynchid genera and species.

Microfibres are among the most prevalent type of microplastics in marine environments. Man-made fibres derived from cellulose are distributed worldwide, but are often confused with synthetic plastic fibres and consequently neglected. All these fibres may adversely affect aquatic organisms, but their levels and potential effects in wild fish remain unknown. We analysed anthropogenic fibre (AF) ingestion in the red mullet (Mullus barbatus), at both temporal and geographical scales, to assess potential effects of these fibres on fish health condition. AFs were present in 50% of fish digestive tracts, with a mean of 1.48 AFs per individual (SD = 1.98). In Barcelona, an increase of 46% in AF ingestion was observed in 2018 compared to 2007. AF ingestion also increases by 20% when Barcelona is compared to a less urban area (the town of Blanes). Visual characterization of fibres by typologies-corroborated by Raman spectroscopy-allowed classification and identification of 88% of AFs as cellulosic (57%), and synthetic polymers (PET) (31%). In all sampling stations, the only histopathological alterations were cysts of unknown etiology, and the most abundant parasites were nematodes. None of these alterations, parasite load, or other fish health indicators (condition indices) indicate an effect of AF ingestion.

Rhadinorhynchus hiansi Soota and Bhattacharya, 1981 , has remained unknown since its original incomplete description from 2 male specimens collected from the flat needlefish Ablennes hians Valenciennes (Belonidae) off Trivandrum, Kerala, India. Recent collections of fishes along the Pacific coast of Vietnam in 2016 and 2017 produced many specimens of the same species from the striped bonito Sarda orientalis Temminck and Schlegel (Scombridae) off the southern Pacific coast of Vietnam at Nha Trang. We describe females for the first time, assign a female allotype status, and provide an expanded description of males from a larger collection completing missing information on hooks and hook roots, receptacle, lemnisci, cement glands, Saefftigen's pouch, and trunk spines. Specimens of R. hiansi characteristically have no dorsal spines in the posterior field of trunk spines and a long proboscis with 36-48 dorso-ventrally differentiated proboscis hooks per row becoming progressively smaller posteriorly then increasing in size near the posterior end to a maximum at the posterior-most ring. Trunk, testes, and lemnisci in our specimens were considerably larger than those reported in the original description, but the proboscis was relatively smaller. The females had long reproductive system and corrugated elliptic eggs without polar prolongation of fertilization membrane. Energy Dispersive X-ray Analysis (EDXA) demonstrates high levels of calcium and phosphorus in large gallium cut hooks and high levels of sulfur in tip cuts of large and small hooks and in spines. This EDXA pattern is a characteristic fingerprint of R. hiansi. The molecular profile of R. hiansi is described from 18S rDNA and COI genes, and phylogenetic relationships with most closely related species are discussed.

Marine litter is one of the most concerning threats for marine wildlife especially regarding plastics and their micro-sized forms, widely known as microplastics. The present study evaluates mesoscale spatial (230 km, Catalan coast) and temporal (2007 vs 2017-2018, Barcelona area) differences on the ingestion of anthropogenic fibers in the deep-sea shrimp Aristeus antennatus in the NW Mediterranean Sea and its relation with shrimp's health condition. Synthetic fibers with lengths ranging between 0.16 and 37.9 mm were found in both stomach (where sometimes they were tangled up in balls) and intestine contents. The percentage of fiber occurrence was >65% at each sampling point. Tangled balls of fibers observed in stomach contents exhibited a wide range of sizes (up to a diameter of 1 cm) and were usually composed of fibers of different polymers, sizes and colours. Differences between locations (2018) were found, with greater fiber loads towards the south during spring and a great variability in summer, as shrimps caught off Barcelona showed a nearly thirty-times higher fiber load compared to shrimps from other localities. Highest concentrations were more likely to be related to major sources of fibers and currents in the area. Fiber load in shrimps from 2007 was comparable to that of shrimps captured in 2017 and 2018 (spring) yet a shift in the proportion of acrylic and polyester polymers was detected. No consistent effect on shrimp's health condition was found, with only a significant negative correlation found between gonadosomatic index and fibers for those shrimps with the highest values of fiber load (caught off Barcelona, summer 2018). Our findings contribute to the knowledge on plastic pollution for the NW Mediterranean Sea and highlight the potential use of this species as a sentinel species for plastic fiber contamination.

We explored the relationships between features of host species and their environment, and the diversity, composition and structure of parasite faunas and communities using a large taxonomically consistent dataset of host-parasite associations and host-prey associations, and original environmental and host trait data (diet, trophic level, population density and habitat depth vagility) for the most abundant demersal fish species off the Catalonian coast of the Western Mediterranean. Altogether 98 species/taxa belonging to seven major parasite groups were recovered in 683 fish belonging to 10 species from seven families and four orders. Our analyses revealed that (i) the parasite fauna of the region is rich and dominated by digeneans; (ii) the host parasite faunas and communities exhibited wide variations in richness, abundance and similarity due to a strong phylogenetic component; (iii) the levels of host sharing were low and involved host generalists and larval parasites; (iv) the multivariate similarity pattern of prey samples showed significant associations with hosts and host trophic guilds; (v) prey compositional similarity was not associated with the similarity of trophically transmitted parasite assemblages; and (vi) phylogeny and fish autecological traits were the best predictors of parasite community metrics in the host-parasite system studied.

Rhadinorhynchus laterospinosus Amin, Heckmann & Ha, 2011 (Rhadinorhynchidae) was described from a single female collected from a trigger fish, Balistes sp. (Balistidae) from the northern Pacific coast of Vietnam in Halong Bay, Gulf of Tonkin. More recent collections of fishes in 2016 and 2017 revealed wider host and geographical distributions. We report this Acanthocephala from nine species of fish representing six families (including the original record from Balistes sp.) along the whole Pacific coast of Vietnam. The fish species are Alectis ciliaris (Carangidae), Auxis rochei (Scombridae), Auxis thazard (Scombridae), Leiognathus equulus (Leiognathidae), Lutjanus bitaeniatus (Lutjanidae), Megalaspis cordyla (Carangidae), Nuchequula flavaxilla (Leiognathidae), and Tylosurus sp. (Belonidae). We provide a complete description of males and females of R. laterospinosus, discuss its hook metal microanalysis using EDAX, and its micropores. Specimens of this species characteristically have lateral trunk spines bridging the anterior ring of spines with posterior field of ventral spines and a proboscis with 15-19 longitudinal alternating rows of 21-26 hooks each varying with host species. We demonstrate the effect of host species on the distribution and size of the trunk, proboscis, proboscis hooks, trunk spines, and reproductive structures. The molecular profile of this acanthocephalan, based on 18S rDNA and cox1 genes, groups with other Rhadinorhynchus species and further seems to confirm the paraphyly of the genus, which is discussed.

RESUMO

The species diversity of Lepidapedon Stafford, 1904 (Lepidapediae) in the Western Mediterranean was assessed based on samples from five deep-sea gadiform fishes collected between the Balearic Islands and the Catalonian coast of Spain: Lepidion lepidion (Risso) and Mora moro (Risso) (both Moridae); Coelorinchus mediterraneus Iwamoto & Ungaro and Trachyrincus scabrus (Rafinesque) (both Macrouridae); and Phycis blennoides (Brünnich) (Phycidae). Integration of morphological and molecular evidence (28S rRNA gene and the mitochondrial nad1 gene) revealed that the deep-sea fishes in the Western Mediterranean share two species of Lepidapedon. Lepidapedon desclersae Bray & Gibson, 1995 was recovered in all five fish species of three families [Moridae (L. lepidion and M. moro); Macrouridae (C. mediterraneus and T. scabrus); and Phycidae (P. blennoides)] and L. guevarai Lopez-Roman & Maillard, 1973 was recovered in three fish species of three families [Phycidae (P. blennoides); Macrouridae (T. scabrus); and Moridae (L. lepidion)]. Therefore, both species are considered to be euryxenic but restricted to gadiforms. The voucher material for the two species from different fish hosts is described in detail and the host-related variability based on the morphometric data is assessed.

Hamaticolax juanji n. sp. is described from specimens collected from the blackbelly rosefish Helicolenus dactylopterus Delaroche 1809 (Scorpaeniformes: Sebastidae). It is the second Hamaticolax species described and reported from the Mediterranean Sea, after Ha. resupinus Pérez-i-García, Carrassón and Boxshall, 2017. It is distinguished from Ha. resupinus by the presence of only one dorsal naked seta on the third segment of the antennule (vs. four), two unequal short naked setae in distal part of the antenna (vs. four), and the absence of a minute spine on the third endopodal segment of leg 1, among others. It is differentiated from Ha. prolixus Cressey 1969 by a comparatively reduced fourth pedigerous somite, the presence of two naked setae on the third segment of the antennule (vs. three), two naked setae and three curved claws in the distal part of the apical segment of the antenna (vs. three and four), an outer naked seta on the basis of leg 2, and by larger length/width ratio of the third endopodal segment, among others. Hamaticolax juanji n. sp. also has relatively longer inner setae on the first and second endopodal segments of leg 4 than the two former species. Patterns of host-use and host-specificity of the genus Hamaticolax are also discussed. The frequently observed low host-specificity of its members may be better explained by host ecological similarity and host availability phenomena, rather than by host phylogenetic distance.

Microplastic (MP) ingestion has been reported in a wide variety of organisms, however, its spatial occurrence and effects on wild populations remain quite unknown. The present study targets an economically and ecologically key species in the Mediterranean Sea, the shrimp Aristeus antennatus. 39.2% of the individuals sampled had MP in their stomachs, albeit in areas close to Barcelona city the percentage reached values of 100%. Overall, MP ingestion was confirmed in a wide spatial and depth (630-1870 m) range, pointing out the great dispersion of this pollutant. The benthophagous diet and close relationship with the sea bottom of A. antennatus might enhance MP exposure and ultimately lead to accidental ingestion. Detailed analysis of shrimps' diet revealed that individuals with MP had a higher presence of endobenthic prey. Microplastic fibers are probably retained for long periods due to stomach's morphology, but no negative effects on shrimp's biological condition were observed.

A new nematode species, Raphidascaris (Raphidascaris) macrouri n. sp. (Anisakidae), is described from male and female specimens found in the intestine, and occasionally in stomach and pyloric caeca, of two deep-water macrourid fishes (Gadiformes) off Barcelona, Mediterranean Sea: Nezumia aequalis (Günther) (type-host) and Trachyrincus scabrus (Rafinesque). Based on light and scanning electron microscopy examination, the new species shows similar morphological features as the other four valid species of the subgenus Raphidascaris Railliet & Henry, 1915, but it differs from Raphidascaris (Raphidascaris) acus (Bloch, 1779), Raphidascaris (Raphidascaris) lutjani Olsen, 1952 and Raphidascaris (Raphidascaris) mediterraneus Lèbre & Petter, 1983 in the high number of precloacal papillae (23-32) and from Raphidascaris (Raphidascaris) gigi Fujita, 1928 in the length of the spicules. Moreover, Raphidascaris (Raphidascaris) macrouri n. sp. exhibits a high variability on the number and distribution of caudal papillae, which was not recorded in the other four mentioned species. This is the first species of this subgenus reported from the family Macrouridae. Sequences of ITS1-5.8S-ITS2 region are analysed and compared with closely related nematode species. Molecular analysis confirmed the uniformity of the R. (R.) macrouri n. sp. between hosts.
