Introduction: The oncogenic transformation is driven by genetic and epigenetic alterations influencing cancer cell fate. These alterations also result in metabolic reprogramming by modulating the expression of membrane Solute Carrier (SLC) transporters involved in biomolecules trafficking. SLCs act as tumor suppressors or promoters influencing cancer methylome, tumor growth, immune-escape, and chemoresistance. Methods: This in silico study aimed to identify the deregulated SLCs in various tumor types compared to normal tissues by analyzing the TCGA Target GTEx dataset. Furthermore, the relationship between SLCs expression and the most relevant tumor features was tackled along with their genetic regulation mediated by DNA methylation. Results: We identified 62 differentially expressed SLCs, including the downregulated SLC25A27 and SLC17A7, as well as the upregulated SLC27A2 and SLC12A8. Notably, SLC4A4 and SLC7A11 expression was associated with favorable and unfavorable outcome, respectively. Moreover, SLC6A14, SLC34A2, and SLC1A2 were linked to tumor immune responsiveness. Interestingly, SLC24A5 and SLC45A2 positively correlated with anti-MEK and anti-RAF sensitivity. The expression of relevant SLCs was correlated with hypo- and hyper-methylation of promoter and body region, showing an established DNA methylation pattern. Noteworthy, the positive association of cg06690548 (SLC7A11) methylation with cancer outcome suggests the independent predictive role of DNA methylation at a single nucleotide resolution. Discussion: Although our in silico overview revealed a wide heterogeneity depending on different SLCs functions and tumor types, we identified key SLCs and pointed out the role of DNA methylation as regulatory mechanism of their expression. Overall, these findings deserve further studies to identify novel cancer biomarkers and promising therapeutic targets.

Nowadays the rationalization of electrical energy consumption is a serious concern worldwide. Energy consumption reduction and energy efficiency appear to be the two paths to addressing this target. To achieve this goal, many different techniques are promoted, among them, the integration of (artificial) intelligence in the energy workflow is gaining importance. All these approaches have a common need: data. Data that should be collected and provided in a reliable, accurate, secure, and efficient way. For this purpose, sensing technologies that enable ubiquitous data acquisition and the new communication infrastructure that ensure low latency and high density are the key. This article presents a sensing solution devoted to the precise gathering of energy parameters such as voltage, current, active power, and power factor for server farms and datacenters, computing infrastructures that are growing meaningfully to meet the demand for network applications. The designed system enables disaggregated acquisition of energy data from a large number of devices and characterization of their consumption behavior, both in real time. In this work, the creation of a complete multiport power meter system is detailed. The study reports all the steps needed to create the prototype, from the analysis of electronic components, the selection of sensors, the design of the Printed Circuit Board (PCB), the configuration and calibration of the hardware and embedded system, and the implementation of the software layer. The power meter application is geared toward data centers and server farms and has been tested by connecting it to a laboratory server rack, although its designs can be easily adapted to other scenarios where gathering the energy consumption information was needed. The novelty of the system is based on high scalability built upon two factors. Firstly, the one-on-one approach followed to acquire the data from each power source, even if they belong to the same physical equipment, so the system can correlate extremely well the execution of processes with the energy data. Thus, the potential of data to develop tailored solutions rises. Second, the use of temporal multiplexing to keep the real-time data delivery even for a very high number of sources. All these ensure compatibility with standard IoT networks and applications, as the data markup language is used (enabling database storage and computing system processing) and the interconnection is done by well-known protocols.

RESUMO

This paper presents a study on the optimization of the tracking system designed for patients with Parkinson's disease tested at a day hospital center. The work performed significantly improves the efficiency of the computer vision based system in terms of energy consumption and hardware requirements. More specifically, it optimizes the performances of the background subtraction by segmenting every frame previously characterized by a Gaussian mixture model (GMM). This module is the most demanding part in terms of computation resources, and therefore, this paper proposes a method for its implementation by means of a low-cost development board based on Zynq XC7Z020 SoC (system on chip). The platform used is the ZedBoard, which combines an ARM Processor unit and a FPGA. It achieves real-time performance and low power consumption while performing the target request accurately. The results and achievements of this study, validated in real medical settings, are discussed and analyzed within.

RESUMO

Our work exhibits how previous projects on the Active and Healthy Ageing field have advanced to the conception of CAPTAIN, a radically new approach towards increased end-user acceptance. The goal is to create intuitive technology that does not require specific skills for interaction and blends in with real life. CAPTAIN will be co-designed by all types of stakeholders, including older adults, involved in all stages, from the initial design to delivery of the final system.

Trauma care in cave rescue is a unique situation that requires an advanced and organized approach with medical and technical assistance because of the extreme environmental conditions and logistical factors. In caving accidents, the most common injuries involve lower limbs. We describe an advanced medical rescue performed by the Italian Corpo Nazionale del Soccorso Alpino e Speleologico, in which extended focused assessment with sonography for trauma and an ultrasound-guided adductor canal block were performed on a patient with a knee distortion directly in the cave. The rescue team inside the cave shared data on patient monitoring and the ultrasound scanning in real time with rescuers at the entrance, using a video conference powered by the new Ermes system. The use of handheld, battery-powered, low-weight, multiparametric monitors, ultrasound machines, and digital data transmission systems could ensure complete medical assistance in harsh environmental conditions such as those found in a cave.

Repeated (but not acute) exposure to brief, non-injurious seizures evoked by minimal electroconvulsive shock (ECS) decreases neuronal death in limbic system and increases mRNA levels for nerve growth factor (NGF). Thus, the induction of NGF is a potential mechanism for the neuroprotection evoked by repeated ECS. The neuroprotective action of NGF is mediated by the TrkA receptor. This study determined whether repeated ECS exposure increased TrkA and NGF protein levels. To determine the functional significance of changes in these proteins, we compared the effects of ECS given daily either for 7 days (chronic ECS) or for 1 day (acute ECS). After chronic ECS, upregulation of both NGF and TrkA was found in perirhinal cortex, thalamus, and amygdala. In hippocampus, TrkA was upregulated in CA2, CA3 and CA4. NGF increase in hippocampus was found in CA1 and dentate gyrus. In frontal cortex and substantia innominata, an increase in NGF (but not in TrkA) was found. In most brain regions, TrkA and NGF remained unchanged after acute ECS. Our results demonstrate that repeated exposure to ECS causes an upregulation of TrkA and NGF proteins in several limbic areas in which neuroprotective effects are observed suggesting that NGF contributes to ECS-evoked neuroprotection.

It has been suggested that activation of striatal neurons expressing D1 or D2 dopamine receptors elicits opposite changes in the net output of the basal ganglia circuitry and, consequently, in the functional interactions of the circuit with the cerebral cortex. In particular, it has been recently reported that striatal D1 receptors may regulate cortex function. To further address this issue, we mapped cerebral expression of Fos protein following intrastriatal stimulation of D1- or D2-class receptors in freely moving animals. Using permanent cannulas implanted in the right striatum, Sprague-Dawley rats received intrastriatal microinfusions of SKF 38393 (D1 agonist) or quinpirole (D2 agonist) or saline (controls), combined with systemic administration of D1 antagonist SCH 23390 or D2 antagonist eticlopride or saline. Animals treated with SKF 38393 showed dose-dependent, massive Fos increases in the motor, somatosensory, auditory, visual and limbic regions of the cerebral cortex, ipsilaterally to the injected striatum. Consistent Fos expression was also found in the injected striatum and, bilaterally, in the nucleus accumbens shell. These increases were effectively counteracted by systemic SCH 23390. Conversely, quinpirole did not induce significant cortical or striatal expression of Fos, which was instead observed after the systemic administration of eticlopride. Fos was not detected in any of the other basal ganglia nuclei, regardless of the dopamine agonists or antagonists used. Our results confirm that striatal D1 dopamine receptors play a central role in the modulation of cortical activity, thus providing additional information on the functional interaction between basal ganglia circuitry and cerebral cortex.
