Importance: Population studies have recorded an increased, unexplained risk of post-acute cardiovascular and thrombotic events, up to 1 year after acute severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Objectives: To search for clinical variables and biomarkers associated with late post-acute thrombotic and cardiovascular events after SARS-CoV-2 infection. Design: Retrospective cohort study. Setting: Third-level referral hospital in Bergamo (Italy). Participants: Analysis of an existing database of adult patients, who received care for SARS-CoV-2 infection at our institution between 20 February and 30 September 2020, followed up on a single date ("entry date") at 3-6 months. Exposure: Initial infection by SARS-CoV-2. Main outcomes and measures: Primary outcome: occurrence, in the 18 months after entry date, of a composite endpoint, defined by the International Classification of Diseases-9th edition (ICD-9) codes for at least one of: cerebral/cardiac ischemia, venous/arterial thrombosis (any site), pulmonary embolism, cardiac arrhythmia, heart failure. Measures (as recorded on entry date): history of initial infection, symptoms, current medications, pulmonary function test, blood tests results, and semi-quantitative radiographic lung damage (BRIXIA score). Individual clinical data were matched to hospitalizations, voluntary vaccination against SARS-CoV-2 (according to regulations and product availability), and documented reinfections in the following 18 months, as recorded in the provincial Health Authority database. A multivariable Cox proportional hazard model (including vaccine doses as a time-dependent variable) was fitted, adjusting for potential confounders. We report associations as hazard ratios (HR) and 95% confidence intervals (CI). Results: Among 1,515 patients (948 men, 62.6%, median age 59; interquartile range: 50-69), we identified 84 endpoint events, occurring to 75 patients (5%): 30 arterial thromboses, 11 venous thromboses, 28 arrhythmic and 24 heart failure events. From a multivariable Cox model, we found the following significant associations with the outcome: previous occurrence of any outcome event, in the 18 months before infection (HR: 2.38; 95% CI: 1.23-4.62); BRIXIA score ≥ 3 (HR: 2.43; 95% CI: 1.30-4.55); neutrophils-to-lymphocytes ratio ≥ 3.3 (HR: 2.60; 95% CI: 1.43-4.72), and estimated glomerular filtration rate < 45 ml/min/1.73 m2 (HR: 3.84; 95% CI: 1.49-9.91). Conclusions and relevance: We identified four clinical variables, associated with the occurrence of post-acute thrombotic and cardiovascular events, after SARS-CoV-2 infection. Further research is needed, to confirm these results.

RESUMO

The culture of early embryos in the surrogate xeno-oviduct was first developed in the early 1950s to allow transport of embryos at long distances. Later, it was applied to the study of culture requirements of the early embryo especially that of bovine origin. In this article, we review the data available on the culture of in vitro-matured and in vitro-fertilized embryos of Bos taurus, Sus scrofa, Equus caballus and Ovis aries in the surrogate sheep oviduct compared with data on in vitro culture in different media. Short-term and long-term cellular and molecular effects are described mainly for the bovine species where more extensive use of this technique has been made. A comparison with in vitro culture in various conditions and species indicate that embryos cultured in the sheep oviduct have close similarities to totally in vivo-derived embryos. The data provided demonstrate that the technique of in vivo culture in the surrogate sheep oviduct is versatile and allows a high rate of embryonic development in all species examined.

RESUMO

The paper analyses some critical situations in the assessment of the compliance with the human exposure limits to the low-frequency magnetic fields. The study is focused on non-sinusoidal and transient waveforms in the frequency range from 8 Hz to 65 kHz. It is carried out by combining measurements and numerical evaluations, starting from actual magnetic flux density behaviours. The results show that inconsistent indications of compliance with the exposure limits may arise not only because of the evaluation method adopted, but also as a result of the used time or frequency windows in relation to the investigated magnetic field waveform.

Current peaks have been observed and measured in electrolytic ionic current of L-glutamic acid aqueous solutions at room temperature, in static magnetic fields of 20, 40, and 60 muT flux densities, with a superimposed extremely low-frequency, (1/10) Hz, alternating magnetic field flux density of 40 nT. The distributions of the peaks have mean values centered at the cyclotron resonance frequency of the singly charged L-glutamic acid ion molecular mass in the corresponding static field. Amplitudes and widths of the peaks are compared and analyzed to extract their correlation. The results can be considered a contribution to the understanding of the experimental phenomenology in low-frequency electromagnetic fields on ionic currents of L-glutamic amino acid aqueous solutions. The results can be of interest in the studies on the interaction of the electromagnetic fields with some structural neurotransmitters in cellular medium.

A current peak has been observed and measured in ionic electrolytic current of a glutamic acid aqueous solution, placed in a static magnetic field of flux density of 40 microT, with a superimposed low-frequency alternating magnetic field of flux density of 40 nT. The peak occurs at the frequency of the cyclotronic resonance of the molecular mass of a single charged glutamic acid ion, placed in a magnetic flux density equal to that of the static field. The amplitude of the current peak is about 30% of the background electrolytic current. Qualitative considerations and a listing of unsolved problems related to the phenomenology are given. The result is the first contribution to the study we have undertaken on the effects of low-frequency alternating electromagnetic fields on the ionic current of amino acid aqueous solutions which are the basic structural units of the proteins.

RESUMO

Many factors influence the efficiency of the in vitro embryo production technology in cattle but the most important are the physiological conditions of the donor and the culture protocols for oocyte maturation and fertilization and for embryo culture from zygote to blastocyst. Therefore, general factors such as age, body conditions and herd management play a pivotal role together with more specific factors such as reproductive soundness and ovarian cyclicity. Given that good quality and competent oocytes are available a complex series of processes, including oocyte maturation, fertilization and culture of the derived zygotes, must be completed to generate viable embryos.

A survey network for congenital toxoplasmosis (TOXO-NET) was set up in December 1996 in Piedmont (Italy). Participants were asked to classify the infections in pregnant mothers and newborns by the criteria of the European Network on Congenital Toxoplasmosis published by Lebech in 1996. Because the IgG Avidity test is largely employed as a 2nd level test in toxoplasmosis diagnosis and it could be helpful to date infection, the co-ordinators of TOXO-NET suggested including it in the "case definition" of "probable" infection and "unlikely" infection. 117 cases of toxoplasmosis in pregnancy divided into the risk categories under Lebech's criteria were re-examined using the "new" case definitions. 77 out of 117 (65.8%) Toxoplasma gondii infections during pregnancy could be defined with only one serum sample using the IgG Avidity test. The IgG Avidity test proved a useful method to classify the Toxoplasma gondii infections in pregnancy, especially when we had only one serum sample.

Embryo technologies are a combination of assisted reproduction, cellular and molecular biology and genomic techniques. Their classical use in animal breeding has been to increase the number of superior genotypes but with advancement in biotechnology and genomics they have become a tool for transgenesis and genotyping. Multiple ovulation and embryo transfer (MOET) has been well established for many years and still accounts for the majority of the embryos produced worldwide. However, no progress has been made in the last 20 years to increase the number of transferable embryos and to reduce the side effects on the reproductive performance of the donors. In vitro embryo production (IVP) is a newer and more flexible approach, although it is technically more demanding and requires specific laboratory expertise and equipment that are most important for the quality of the embryos produced. Somatic cell cloning is a rapidly developing area and a very valuable technique to copy superior genotypes and to produce or copy transgenic animals. More knowledge in oocyte and embryo biology is expected to shed new light on the early developmental events, including epigenetic changes and their long lasting effect on the newborn.Embryo technologies are here to stay and their use will increase as advances in the understanding of the mechanisms governing basic biological processes are made.

Butyrolactone I (BL-I) and Roscovitine (ROS), two specific and potent inhibitors of M-phase promoting factor (MPF) kinase activity, were used to block germinal vesicle breakdown (GVBD) of cattle oocytes. A concentration 6.25 microM BL-I and 12.5 microM ROS blocked over 93.3 +/- 2.5% of oocytes in germinal vesicle (GV) stage during a 24-hr culture period. Following a second 24-hr culture step in maturation medium (IVM) almost all (91.5 +/- 3.0%) inhibited oocytes resumed meiosis and reached the metaphase II (MII) stage. The MII kinetics was different for inhibited and control oocytes. Fifty percent MII was reached at 13-14 hr in BL-I + ROS treated oocytes, compared to 18 hr in control oocytes. Therefore, control oocytes were fertilised (IVF) after 22 hr IVM and inhibited oocytes after 16 or 22 hr IVM. After IVF, percentage of grade 1 freezable embryos on day 7 (D + 7) as well as percentage of blastocyst formation on D + 8 in the group of BL-I + ROS treated oocytes fertilised after 16 hr IVM were higher (P < 0.05) compared with the other experimental group fertilised after 22 hr IVM but not different in comparison with the control. Survival to freezing and thawing of grade 1 embryos frozen on D + 7 was employed as viability criteria and was similar in all groups. Thus, the presence of BL-I + ROS in the prematuration medium of bovine oocytes determines a reversible meiotic block, without compromising their subsequent developmental competence.

Embryo production by in vitro techniques has increased steadily over the years. For cattle where this technology is more advanced and is applied more, the number of in vitro produced embryos transferred to final recipients was over 30,000 in 1998. An increasing proportion of in vitro produced embryos are coming from oocytes collected from live donors by ultrasound-guided follicular aspiration (ovum pick up, OPU). This procedure allows the repeated production of embryos from live donors of particular value and is a serious alternative to superovulation. Ovum pick up is a very flexible technique. It can be performed twice a week for many weeks without side effects on the donor's reproductive career. The donor can be in almost any physiological status and still be suitable for oocyte recovery. A scanner with a sectorial or convex probe and a vacuum pump are required. Collection is performed with minimal stress to the donor. An average of 8 to 10 oocytes are collected per OPU with an average production of 2 transferable embryos. The laboratory production of embryos from such oocytes does not differ from that of oocytes harvested at slaughter as the results after transfer to final recipients. For other species such as buffalo and horses OPU has been attempted similarly to cattle and data will be presented and reviewed. For small ruminants, laparotomy or laparoscopy seems the only reliable route so far to collect oocytes from live donors.

After a brief description of the reference systems for the calibration of the electromagnetic field meters set up at IEN and the Ivrea Department of ARPA, the procedures for the evaluation of the measurement uncertainty are discussed, with reference to the problem of identifying and quantifying the different uncertainty components. which are due both to the field meter characteristics and to the measurement procedure and conditions. The problem of how to compare a measurement result, which consists of the measurement value and its associated uncertainty, with a limit fixed by the standards is also faced.

RESUMO

Fifteen patients affected by chronic uremia and treated with hemodialysis were studied. The main hemorheological parameters were evaluated in basal conditions and after hemodialytic therapy and were correlated with some hematochemical indices. Although some limitations to the purpose of this study derive from the heparinization of the blood during hemodialysis, the results of this study demonstrate that erythrocyte filtration does not significantly change during the hemodialytic therapy. The only hemorheological parameter altered in the uremic patient is represented by plasma viscosity; this is probably due to the increase of fibrinogenemia in these patients.

RESUMO

We describe the case of a man, previously affected by B-CLL, who developed Sweet's syndrome four months later the previous diagnosis. Sweet's syndrome was diagnosed by cutaneous biopsy. Neither infections nor drugs administration were demonstrated, in order to explain beginning of Sweet's syndrome. Altered immune responsiveness, as seen in B-CLL, could cause Sweet's syndrome.

Only few patients (3,8%) affected by ANLL survive for more than 3 years from the time of diagnosis. In the present study the Authors try to determine which clinical and haematological factors can determine long-terme survival. The Authors describe the case of a male patient (25 years old) affected by ANLL (M2), who became and is still now a long-survivor (80 months from 1th complete remission). He has been treated with DNB, ARA-C and TG (TRA schedule) for two cycles. After CR he refused consolidation and reinduction therapy and was treated only with 6-MP and MTX for 3 years, as maintenance chemotherapy. The Authors suggest that important factors for a favourable prognosis are early diagnosis and young age. A relatively high number of platelets can be correlated with a good prognosis.

RESUMO

Free radicals and peroxides of our organism, that are very unstable, are now recognized as a product of biological oxido-reductive processes. The evaluation of free radicals and peroxides in blood was done by counting Heinz-bodies (H.B.) (Beutler method). The Authors examined 40 healthy subjects, at rest, and 37 neoplastic patients (19 carcinomas, 10 leukemias, 8 lymphomas) at the admission to hospital. The results show that the rate per cent of H.B. in neoplastic patients is significantly higher than in healthy subjects. According to us, neoplastic patients have free radicals hematic levels higher than healthy volunteers.
