Reference intervals for physiologic parameters, crucial for assessing the health status of animals, have been documented for various crocodilian species across the globe. Nonetheless, the establishment of plasma biochemical reference intervals specific to Amazonian crocodilians remains incomplete. In an effort to address this gap, we procured blood samples from 65 black caimans (Melanosuchus niger) and 58 spectacled caimans (Caiman crocodilus) during the period of September-December 2019 within the Anavilhanas National Park in the Brazilian Amazon region We aimed to define reference intervals for 11 key plasma variables measured, namely glucose, triglycerides, total cholesterol, calcium, magnesium, sodium, potassium, albumin, total protein, uric acid, and urea. In general, the determined blood reference intervals aligned closely with those established for other crocodilian species. Some specific measurements, such as total cholesterol, sodium, and magnesium, exhibited distinct variations based on the species. Furthermore, female black caimans showcased elevated cholesterol levels compared with their male counterparts. Within the spectacled caimans, disparities related to sex were evident solely in the case of electrolytes sodium and potassium, with males demonstrating higher levels compared with females. These reference intervals not only provide data for assessing potential fluctuations in the health of wild or captive Amazonian crocodilians but also hold value for veterinary management.

When captured, free-living crocodilians respond by hyperstimulation of the hypothalamic-pituitary-adrenal (HPA) axis, which triggers a cascade of downstream events of physiological stress. We examined these responses in two unstressed, and stressed Amazonian caimans, Caiman crocodilus and Melanosuchus niger. Plasma corticosterone levels increased in both stressed caiman species. In M. niger, the levels of this hormone increased 5.2-fold compared with the basal range values, while in C. crocodilus this was only 1.7-fold. After stress, M. niger needed more than 6 h to return its corticosterone levels to basal range values, whereas in C. crocodilus just 0.5 h was enough. Downstream events were characterized by an increase in glucose levels, which is associated with corticosterone increments. Excessive muscle activity resulted in increased plasma lactate content in both species. Lactate levels were also related to plasma calcium concentration, possibly due to the buffering capacity for preventing lactic acidosis. Clearance of excessive lactate load was faster in M. niger (0.5 h) than in C. crocodilus (more than 6 h). Although both caiman species respond in the same way to capture, the amplitude and duration of activation of the HPA axis are different. M. niger may be potentially more sensitive to acute stress than C. crocodilus. On the other hand, C. crocodilus needs more time to recover from the lactic acid load. Our experiment provides a useful diagnostic tool for management and conservation programs, as well as evaluating the impacts of tourism and recreational capture on caimans in the Amazon.

The Amazon aquatic ecosystems have been modified by the human population growth, going through changes in their water bodies and aquatic biota. The spectacled alligator (Caiman crocodilus crocodilus) has a wide distribution and adaptability to several environments, even those polluted ones. This study aimed to investigate if a Caiman species living in urban streams of Manaus city (Amazonas State, Brazil) is affected by environmental pollution. For that, it was used classical and molecular cytogenetic procedures, in addition to micronucleus and comet assays. Although the karyotype macrostructure remains unaltered (2 n = 42 chromosomes; 24 t + 18 m/sm; NF = 60), the genotoxic analysis and the cytogenetic mapping of repetitive DNA sequences demonstrated that polluted environments alter the genome of the specimens, affecting both the chromosomal organization and the genetic material.

Schneider's dwarf caiman Paleosuchus trigonatus is one of the smallest living crocodilians. Due to its broad distribution, cryptic behavior, and small home range, the species is well suited for the study of phylogeographic patterns on a continental scale. Additionally, this species is under threat due to habitat loss, trade and harvest, but is considered at low conservation risk by the IUCN. In the present study we test the hypothesis that P. trigonatus is comprised of geographically structured lineages. Phylogenetic reconstructions of the mitochondrial cytochrome b gene and single locus species discovery methods revealed the existence of two well-supported lineages within P. trigonatus-an Amazonian and Guianan lineage. Fossil calibrated divergence of these lineages was estimated to have occurred in the Late Miocene (7.5 Ma). The hypothesis that the Atlantic coast drainages might have been colonized from the southeast or central Amazon is supported by demographic metrics and relatively low genetic diversity of the Coastal and upper Branco populations when compared to the Amazon basin populations. The Amazon basin lineage is structured along an east-west gradient, with a sharp transition in haplotype frequencies to the east and west of the Negro and Madeira rivers. These lineages are already under anthropogenic threat and, therefore, are conservation dependent. Recognition of these lineages will foster discussion of conservation future of P. trigonatus and these lineages.

Although nesting ecology is well studied in several crocodilian species, it is not known how nest attendance influences physiology and body condition of nesting females. In this study, we describe body condition and serum biochemical values of nesting female, non-nesting female and male spectacled caiman (Caiman crocodilus) and black caiman (Melanosuchus niger) in two areas of Central Amazonia. We also evaluated the effect of nest age and nest distance to water on body condition and blood parameters of nesting females. Body condition and plasmatic concentrations of glucose, triglycerides, lactate and uric acid of nesting females were significantly different from those of non-nesting females and males in C. crocodilus, but not in M. niger. Our study also demonstrated that nest age and distance to water had a negative effect on female body condition in C. crocodilus, but not in M. niger. Female C. crocodilus attending older nests or nests built further away from permanent water bodies tended to have lower body condition. Our results demonstrate that the nesting strategy of C. crocodilus has a metabolic cost associated with nest attendance for nesting females, which appear to depend on accumulated energetic reserves during nest attendance. In contrast, nest attendance had little effect on the physiology of female M. niger.

Total mercury (Hg) concentrations of muscle, liver, blood, and epidermal keratin were measured in typically consumed, economically and culturally important species of turtle (Podocnemis unifilis and Podocnemis expansa) and caiman (Melanosuchus niger and Caiman crocodilus) from the Rio Purus in the Amazon basin, Brazil. Methylmercury (MeHg) concentrations were also measured in muscle tissue, representing the first analysis of MeHg concentrations in Amazonian reptile species. In muscle tissues Hg was mostly MeHg (79-96%) for all species. No correlations existed between animal size and total Hg or MeHg concentrations for any species other than M. niger, possibly as a result of growth dilution or the evolution of efficient Hg elimination mechanisms. Significant linear correlations were found between total Hg concentrations in all pairs of nonlethally sampled tissues (keratin and blood) and internal tissues (muscle and liver) for M. niger and between keratin and internal tissues for P. expansa, indicating that nonlethally sampled tissues can be analyzed to achieve more widespread and representative monitoring of Hg bioaccumulation in Amazonian reptiles. Although mean Hg concentrations in muscle for all species were below the World Health Organization guideline for safe consumption (500 µg kg(-1)), mean concentrations in caiman liver were above the safe limit for pregnant women and children (200 µg kg(-1)). No significant differences were found between total Hg and MeHg concentrations in tissues from wild-caught and farm-raised P. expansa, suggesting that farming may not reduce Hg exposure to humans.

Reptiles are ideal organisms for the non-invasive monitoring of mercury (Hg) contamination. We have investigated Hg bioaccumulation in tissue layers of reptile dermis as a basis for establishing a standardized collection method for Hg analysis. Tissue samples from freshwater turtle species Podocnemis unifilis and Podocnemis expansa and caiman species Melanosuchus niger and Caiman crocodilus, all from the Amazonian region, were analysed in this study. We first tested the relationships between Hg concentrations in keratin and bone to Hg concentrations in muscle to determine the best predictor of Hg concentration in muscle tissue. We then investigated the potential for measuring Hg concentrations across turtle carapace growth rings as an indicator of longer term changes in Hg concentration in the environment. Hg concentrations were significantly lower in bone (120 ng g(-1) caimans and 1 ng g(-1) turtles) than keratin (3600 ng g(-1) caimans and 2200 ng g(-1) turtles). Keratin was found to be a better predictor of exposure to Hg than muscle and bone tissues for both turtles and caimans and also to be a reliable non-invasive tissue for Hg analysis in turtles. Measurement of Hg in carapace growth rings has significant potential for estimating Hg bioaccumulation by turtles over time, but full quantification awaits development and use of a matrix-matched reference material for laser ablation ICPMS analysis of Hg concentrations in keratin. Realising this potential would make a valuable advance to the study of the history of contamination in mining and industrial sites, which have until now relied on the analysis of Hg concentrations in sediments.

Mercury (Hg) concentrations in the Amazon are generally high, but no studies have been published on Hg concentrations in caimans (Alligatoridae) from the region. Aiming for sizes representative of caimans traded for food in the Amazon, we measured Hg concentration in tail muscle of spectacled caiman (Caiman crocodilus crocodilus) and black caiman (Melanosuchus niger) from the Purus River basin. The information on Hg concentration in caimans from this area is important because of the potential health risk to humans and other animals that eat them as well as the potential use of these top-level predators as bioindicators. There were no significant interspecific or sex differences in Hg concentrations. The mean Hg concentration was 291.2 µg/kg in C. c. crocodilus and 193.9 µg/kg in M. niger. A significant positive correlation between Hg concentration and size was found for M. niger (p = 0.005) but not for C. c. crocodilus. Our Hg sample from M. niger corresponded to the size of M. niger collected for commercial trade, but our Hg sample from C. c. crocodilus turned out to be significantly smaller than the trade samples (p = 0.004), but this difference is not pertinent in the absence of a correlation between size and Hg concentration for this species. Although there are no standards for reptile meat, both species had mean Hg concentrations lower than the maximum allowable level of 500 µg/kg Hg recommended by the World Health Organization and by the Brazilian Health Ministry for fish. However, by calculating daily consumptions limits and number of meals per month that can be safely consumed, we found that consumers who eat caimans frequently may be at risk for Hg-related health problems.

We assessed the spatial distribution of the genetic variability of Melanosuchus niger from 11 localities in South America using 1,027 base pairs of the mitochondrial cytochrome b gene. Screening 132 animals, we found 41 haplotypes, high values of genetic diversity, low values of nucleotide diversity and significant deviations from neutral expectation of allelic frequencies in some localities. Mantel test and nested-clade analysis indicated that isolation-by-distance was an important population dynamic for the species as a whole. Wright's fixation indexes analyses showed that hydrogeographically separated populations from French Guiana together with Amapá state population in Brazil are genetically differentiated from all other populations that are found in the Amazon drainage basin. These indexes also disclosed that the population from Ecuador is genetically differentiated in relation to the populations from Brazil, Peru and French Guiana. Within the Amazon Basin little differentiation exists, and genetic and geographic distances are not correlated. Demographic data as well as population genetic data suggest that M. niger is recovering in some protected regions. However, part of this apparent recovery may be owing to the movement of animals into protected regions.
