RESUMO

With the increasing production and use of polyurethanes (PUs), it is necessary to develop sustainable techniques for the remediation of plastic pollution. The use of microorganisms capable of biodegrading PUs may be an environmentally desirable solution for controlling these plastic contaminants. To contribute to the discovery of alternatives for the mitigation of plastics in the environment, this study aimed to explore the potential of StaphylococcuswarneriUFV_01.21, isolated from the gut of Galleria mellonellalarvae, for biodegradation of PU in pure culture and microbial co-culture with Serratia liquefaciensL135. S. warneri grew using Impranil® PU as the sole carbon source in pure culture and co-culture. With six days of incubation, the biodegradation of Impranil® in Luria Bertani broth was 96, 88 and 76%, while in minimal medium, it was 58, 54 and 42% for S. warneri, S. liquefaciens, and co-culture, respectively. In addition, S. warneri in pure culture or co-culture was able to biodegrade, adhere and form biofilms on the surfaces of Impranil® disks and poly[4,4'-methylenebis (phenyl isocyanate)-alt-1,4-butanediol/di(propylene glycol)/polycaprolactone] (PCLMDI) films. Scanning electron microscopy also revealed biodegradation by detecting the formation of cracks, furrows, pores, and roughness on the surfaces of inoculated PU, both with pure culture and microbial co-culture. This study is the first to demonstrate the potential of S. warneriin PU biodegradation.

Salmonella is a food and waterborne pathogen responsible for outbreaks worldwide, and it can survive during passage through the stomach and inside host phagocytic cells. Virulence genes are required for infection and survival in macrophages, and some are under the regulation of the quorum sensing (QS) system. This study investigated the influence of the autoinducer 1 (AI-1), N-dodecanoyl-homoserine lactone (C12-HSL), on the virulence of Salmonella PT4 using Galleria mellonella as an infection model. Salmonella PT4 was grown in the presence and absence of C12-HSL under anaerobic conditions for 7 h, and the expression of rpoS, arcA, arcB, and invA genes was evaluated. After the inoculation of G. mellonella with the median lethal dose (LD50) of Salmonella PT4, the survival of bacteria inside the larvae and their health status (health index scoring) were monitored, as well as the pigment, nitric oxide (NO), superoxide dismutase (SOD), and catalase (CAT) production. Also, the hemocyte viability, the induction of caspase-3, and microtubule-associated light chain 3 (LC3) protein in hemocytes were evaluated. Salmonella PT4 growing in the presence of C12-HSL showed increased rpoS, arcA, arcB, and invA expression and promoted higher larvae mortality and worse state of health after 24 h of infection. The C12-HSL also increased the persistence of Salmonella PT4 in the hemolymph and in the hemocytes. The highest pigmentation, NO production, and antioxidant enzymes were verified in the larva hemolymph infected with Salmonella PT4 grown with C12-HSL. Hemocytes from larvae infected with Salmonella PT4 grown with C12-HSL showed lower viability and higher production of caspase-3 and LC3. Taken together, these findings suggest that C12-HSL could be involved in the virulence of Salmonella PT4.

Salmonella is a foodborne pathogen that can develop resistance to different stresses, which is essential for successful infection of the host. Some genes directly related to acid resistance are also involved in cationic peptide resistance in Gram-negative bacteria and could be under the control of quorum sensing (QS) mediated by autoinducer 1, known as acyl-homoserine lactone. Here, we investigated the influence of autoinducer 1, N-dodecanoyl-homoserine lactone (C12-HSL) on the resistance of Salmonella enterica subspecies enterica serovar Enteritidis to nisin and acid stress. Salmonella cells growing in anaerobic tryptic soy agar (TSB) at a pH of 7.0 for 7 h were submitted to acid stress at a pH of 4.5 in the presence and absence of nisin and were either supplemented or not with C12-HSL. Viable cell counts, gene expression, membrane charge alterations, fatty acid composition, and intracellular content leakage were observed. The autoinducer C12-HSL increased nisin resistance and survival at a pH of 4.5 in Salmonella. Also, C12-HSL increased the expression of the genes, phoP, phoQ, pmrA, and pmrB, which are involved with antimicrobial and acid resistance. The positive charge on the cell surface and concentration of cyclopropane fatty acid of the cellular membrane were increased in the presence of C12-HSL under acidic conditions, whereas membrane fluidity decreased. The loss of K+ and NADPH, promoted by nisin, was reduced in the presence of C12-HSL at a pH of 4.5. Taken together, these findings suggest that quorum sensing plays an important role in enhanced nisin and acid resistance in Salmonella.

This study aimed to evaluate the physical-chemical, microbiological and sensory characteristics of four formulations of yacon jams. The following characteristics were analyzed: chemical composition, soluble solids, pH, microbiological presence of total coliforms and salmonella at 45 °C, and presence of fungi and yeasts. The yacon jams exhibited high moisture content (> 62%) as well as high concentrations of carbohydrates (> 28%) and total dietary fiber (2.5%), mainly fructooligosaccharides (> 0.9%). The average energy value was 124.6 kcal/100 g. The average pH was 3.8 and the soluble solids measured 27.6 °Brix. All yacon jams complied with standard microbiological requirements and received high acceptance scores, especially the yacon jam and yacon jam with cinnamon. Yacon jam products may offer an excellent alternative to more common jams due to their lower concentrations of simple sugar, lower calorie value, increased presence of dietary fibers, and desirable sensory characteristics.


Este estudio tuvo como objetivo evaluar las características físico-químicas, microbiológicas y sensoriales de cuatro formulaciones de mermeladas de yacón. Las características analizadas fueron: composición química, contenido de sólidos solubles, pH, presencia microbiológica de coliformes totales, coliformes y salmonella a 45 °C, y la presencia de hongos y levaduras. Los atascos de yacón mostraron alto contenido de humedad (> 62%), así como altas concentraciones de hidratos de carbono (> 28%) y fibra dietética total (2, 5%), principalmente fructooligosacáridos (> 0, 9%). El valor energético promedio fue de 124, 6 kcal/100g. El pH promedio fue de 3, 8 y los sólidos solubles mide 27, 6 ° Brix. Todas las mermeladas de yacón cumplido con los requisitos microbiológicos estándar y recibió altos puntajes de aceptación, en especial la mermelada de yacón y mermelada de yacón con canela. Productos de mermelada de yacón pueden ofrecer una excelente alternativa a los atascos más comunes debido a sus concentraciones más bajas de azúcar simple, un valor más bajo de calorías, el aumento de la presencia de fibras dietéticas y las características sensoriales deseadas.

Antibacterial activity of Lentinula edodes against Bacillus subtilis was evaluated in cell-free filtrates obtained after growth in 14 different culture media. The highest B. subtilis growth inhibition was promoted by filtrates of growth media supplemented with rice bran, vermiculite or molasses. L. edodes dry mycelial biomass in liquid culture with 0.5% added rice bran was 3.2 mg/ml, after growth for 30 days at 25ºC without shaking, and 4.3 mg/ml under orbital shaking (150 rpm). However, antibacterial activity, detected between 20 and 24 days of incubation of stationary cultures, was absent in filtrates of aerated cultures. Temperatures of 20-25ºC enhanced both growth and antibacterial activity. Optimum pH for L. edodes mycelial growth was 3.0-3.5, while for production of antibacterial substance(s) it was 4.5. Our results indicated that incubation conditions that enhance mycelial growth are quite different from those necessary for production of antibacterial substance(s) by L. edodes.
