Disturbance can strongly influence ecosystems, yet much remains unknown about the relative importance of key processes (selection, drift, and dispersal) in the recovery of ecological communities following disturbance. We combined field surveys with a field experiment to elucidate mechanisms governing the recovery of aquatic macroinvertebrates in habitats of an alluvial floodplain following flood disturbance. We monitored macroinvertebrates in 24 natural parafluvial habitats over 60 days after a major flood, as well as the colonization of 24 newly-built ponds by macroinvertebrates over 45 days in the same floodplain. We examined the sources of environmental variation and their relative effects on aquatic assemblages using a combination of null models and Mantel tests. We also used a joint species distribution model to investigate the importance of primary metacommunity structuring processes during recovery: selection, dispersal, and drift. Contrary to expectations, we found that beta diversity actually decreased among natural habitats over time after the flood or the creation of the ponds, instead of increasing. This result was despite environmental predictors showing contrasting patterns for explaining community variation over time in the natural habitats compared with the experimental ponds. Flood heterogeneity across the floodplain and spatial scale differences between the experimental ponds and the natural habitats seemingly constrained the balance between deterministic and stochastic processes driving the ecological convergence of assemblages over time. While environmental selection was the dominant structuring process in both groups, biotic interactions also had a prominent influence on community assembly. These findings have profound implications towards understanding metacommunity structuring in riverscapes that includes common linkages between disturbance heterogeneity, spatial scale properties, and community composition. Many inland waters exhibit complete or partial desiccation, or have vanished due to global change, exposing sediments to the atmosphere. Yet, data on carbon dioxide (CO2) emissions from these sediments are too scarce to upscale emissions for global estimates or to understand their fundamental drivers. Here, we present the results of a global survey covering 196 dry inland waters across diverse ecosystem types and climate zones. We show that their CO2 emissions share fundamental drivers and constitute a substantial fraction of the carbon cycled by inland waters. CO2 emissions were consistent across ecosystem types and climate zones, with local characteristics explaining much of the variability. Accounting for such emissions increases global estimates of carbon emissions from inland waters by 6% (~0.12 Pg C y-1). Our results indicate that emissions from dry inland waters represent a significant and likely increasing component of the inland waters carbon cycle.

Streambed colmation by fine sediment, e.g. the deposition, accumulation and storage of fines in the substrate, is known to have severe effects on invertebrate assemblages in both the benthic and hyporheic zones but the changes in biological attributes of invertebrate assemblages related to colmation have never been considered simultaneously for these two zones. We studied the effects of colmation on the invertebrate assemblages of three rivers, testing a priori hypotheses on the biological attributes that should be more selected in communities subjected to different levels of colmation in both zones. Only the proportion of organisms with high fecundity increased and the proportion of small-sized organisms decreased along the colmation gradient in both zones simultaneously. As expected, a higher number of traits were significantly modified with colmation in the benthic vs. hyporheic assemblages. Most of the biological attributes impaired were different in the two zones. In the benthic zone, colmation mainly selected particular physiological or trophic characteristics of species and features related to their resistance or resilience capacities. In contrast, the morphological attributes of species were much more impaired by colmation in the hyporheic zone than in the benthic zone. In clogged benthic habitats, traits seemed to be more impaired by an increase in physico-chemical constraints (e.g. the reduction of oxygen availability) and a reduction of potential exchanges (including exchanges of food resources) due to a decline in stream bed conductivity. The morphological attributes of the hyporheic species were probably more influenced by changes in interstitial space characteristics. A potential indicator of the effects of colmation on river health may be based on the functional traits of benthic communities because they (i) satisfy the WFD recommendations, (ii) respond consistently along a colmation gradient and (iii) are comparable among assemblages even across ecoregions that differ in their taxonomic composition.

In urban area, management of stormwater leads to the accumulation of polluted sediments at the water-sediment interface of various aquatic ecosystems. In many cases, these sediments are colonised by dense populations of tubificid worms. However, the influence of tubificid worms on the fate of stormwater sediments has never been tackled. The aim of this study was to measure in sediment columns the influence of tubificid worms on sediment reworking, organic matter processing (O(2) uptake and release of NH(4)(+), NO(3)(-), PO(4)(3-), and dissolved organic carbon), release of hydrocarbons and heavy metals, and microbial characteristics. Results showed that tubificid worms increased the release of NH(4)(+), PO(4)(3-), and dissolved organic carbon by 2-, 4-, and 3-fold, respectively. O(2) uptake also increased by more than 35% due to tubificid activity. The increase in the percentages of active bacteria and hydrolytic activity in the presence of worms indicated that the higher sediment respiration was caused by the stimulation of microbial communities. A reduction of the number of sulphate-reducing bacteria in the uppermost layers of the sediment was attributed to the penetration of O(2) due to worm activity. These significant effects of tubificid worms were probably linked to the dense network of burrows, which enhanced the exchange surface between the water column and the sediment. No release of heavy metals and hydrocarbons to the water phase was detected in the sediment columns. Understanding the fate and effect of organic stormwater sediments in the natural environment requires the integration of the role of bioturbation in urban pollution studies.

Artificial recharge of urban aquifers with stormwater has been used extensively in urban areas to dispose of stormwater and compensate for reduced groundwater recharge. However, stormwater-derived sediments accumulating in infiltration beds may act as a source of dissolved contaminants for groundwater. Concentrations of hydrocarbons, heavy metals, nutrients and dissolved oxygen (DO) were monitored at multiple depths in shallow groundwater below a stormwater infiltration basin retaining large amounts of contaminated organic sediments. Multilevel wells and multiparameter loggers were used to examine changes in groundwater chemistry occurring over small spatial and temporal scales. Rainfall events produced a plume of low-salinity stormwater in the first 2 m below the groundwater table, thereby generating steep vertical physico-chemical gradients that resorbed during dry weather. Heavy metals and hydrocarbons were below reference concentrations in groundwater and aquifer sediments, indicating that they remained adsorbed onto the bed sediments. However, mineralization of organic sediments was the most probable cause of elevated concentrations of phosphate and DOC in groundwater. DO supply in groundwater was severely limited by bed respiration which increased with temperature. Cold winter stormwater slightly re-oxygenated groundwater, whereas warm summer stormwater lowered DO concentrations in groundwater. Among several results provided by this study, it is recommended for management purposes that infiltration practices should minimize the contact between inflow stormwater and organic sediments retained in infiltration basins.

BACKGROUND: Image cytometry has proved to provide a good alternative to flow cytometry for DNA ploidy measurement of archival tumors. However, when interactively done this technique is unable to give statistically valuable results within an acceptable time for clinical oncology. METHODS: An image cytometer was developed for fully automatic DNA ploidy quantitation, focusing efforts on speed and accuracy. Software functionalities include systematic acquisition of fields on a microscopic slide, detection, localization and sorting of nuclei, computation of the DNA content together with post-processing tools, for a deeper analysis of the DNA ploidy diagram. RESULTS: DNA ploidy analysis of archival breast carcinoma samples illustrates the accuracy of DNA ploidy measurements and the sensitivity in the detection of DNA ploidy abnormalities as a result of cell sorting. CONCLUSIONS: Fully automatic image cytometry is able to combine qualities of flow cytometry (automatic analysis of a statistically significant collection of cell nuclei) with additional advantages: sorting of unwanted events (debris, stromal and inflammatory cell nuclei) and facilities for an a posteriori control of the quality of cell selection. This method is well suited to DNA ploidy analysis of archival cancer samples.

RESUMO

An automatic machine, dedicated to solid tumor DNA ploidy quantitation has been built in order to provide pathologists with a tool usable in clinical practice. Main efforts were focused on an automation of each step of the analysis and on an elimination of any subjective choice, while preserving the quality of measurement. As the software is independent of the machine architecture, it offers performances which increase in parallel with the rapid evolution of the computers. An illustration of the various functionalities of the automaton is proposed through the study of deparaffined breast cancer samples.
