In order to gain knowledge about the potential effects of acidification in aquatic ecosystems, global change research based on microalgae as sentinel species has been often developed. However, these studies are limited to single species tests and there is still a research gap about the behaviour of microalgal communities under this environmental stressor. Thus, the aim of this study was to assess the negative effects of CO2 under an ecologically realistic scenario. To achieve this objective, two types of toxicity tests were developed; i) single toxicity tests and ii) multispecies toxicity tests, in order to evaluate the effects on each species as well as the interspecific competition. For this purpose, three microalgae species (Tetraselmis chuii, Phaeodactylum tricornutum and Nannochloropsis gaditana) were exposed to two selected pH levels (7.4, 6.0) and a control (pH 8.0). The pH values were choosen for testing different scenarios of CO2 enrichment including the exchange atmosphere-ocean (pH 7.4) and natural or anthropogenic sources of CO2 (pH 6.0). The effects on growth, cell viability, oxidative stress, plus inherent cell properties (size, complexity and autofluorescence) were studied using flow cytometry (FCM). Results showed that T. chuii was the most resistant species to CO2 enrichment with less abrupt changes in terms of cell density, inherent cell properties, oxidative stress and cell viability. Although P. tricornutum was the dominant species in both single and multispecies tests, this species showed the highest decrease in cell density under pH 6.0. Effects of competence were recorded in the multispecies control (pH 8) but this competence was eclipsed by the effects of low pH. The knowledge of biological interactions made by different microalgae species is a useful tool to extrapolate research data from laboratory to the field.

This study aims to ascertain the effects of CO2 induced water acidification and leaks from Carbon Capture and Storage activities on the South American amphipod Hyale youngi. A 10-day acute toxicity test was performed using sediments from two sites located inside the Santos Estuarine System. They were subjected to five pH treatments (8.1, 7.6, 7.0, 6.5, and 6.0). Metals (Cd, Cu, Cr, Pb, Ni and Zn) and the metalloid As were analyzed to determine the influence of their acidification-related mobility on the amphipods mortality. The results showed that mortality becomes significant when compared to control in pH 6.5 in the Canal de Piaçaguera sediment (contaminated) and at pH 6.0 in Ilha das Palmas sediment (reference).

The study assesses the effects of carbon dioxide capture and storage (CCS) leaks and ocean acidification (OA) on the metal bioavailability and reproduction of the mytilid Perna perna. In laboratory-scale experiments, CCS leakage scenarios (pH 7.0, 6.5, 6.0) and one OA (pH 7.6) scenario were tested using metal-contaminated sediment elutriates and seawater from Santos Bay. The OA treatment did not have an effect on fertilisation, while significant effects were observed in larval-development bioassays where only 16 to 27 % of larva developed normally. In treatments that simulated CO2 leaks, when compared with control, fertilisation success gradually decreased and no larva developed to the D-shaped stage. A fall in pH increased the bioavailability of metals to marine mussels. Larva shell size was significantly affected by both elutriates when compared with seawater; moreover, a significant difference occurred at pH 6.5 between elutriates in the fertilisation bioassay.

This work offers an environmental assessment of a dredged sediment disposal area in Santos bay, situated on the central coast of the São Paulo State, Brazil. Sediment quality was evaluated through physicochemical analysis and toxicity tests of sediments collected in the disposal site and adjacent area. The physicochemical characterization of the sediments involved grain size distribution, concentrations of polycyclic aromatic hydrocarbons (PAHs), chlorinated and aromatic hydrocarbons, polychlorinated biphenyls (PCBs), pesticides, phthalates, metals and nutrients. Acute and chronic toxicity tests were employed, using amphipods (Tiburonella viscana) and sea urchins (Lythechinus variegatus), respectively. Results revealed toxicity by all the methods applied here, suggesting that the area of disposal of dredged material is significantly altered with respect to sediment quality and probably capable of generating deleterious effects on the local biota. Aiming to elucidate the association between the distinct environmental variables and the biological effects measured in laboratory, Factor Analysis was performed. Results revealed that despite most contaminant concentrations were found below the limits established by Brazilian legislation, biological effects were related to metals (chronic toxicity) and organic compounds (acute toxicity). The application of multivariate analysis proved to be particularly useful to assess and interpret the results in an integrated way, particularly due to the large number of parameters analyzed in environmental assessments, and should be applied in future studies.

The injection and storage of CO2 into marine geological formations has been suggested as a mitigation measure to prevent global warming. However, storage leaks are possible resulting in several effects in the ecosystem. Laboratory-scale experiments were performed to evaluate the effects of CO2 leakage on the fate of metals and on the growth of the microalgae Phaeodactylum tricornutum. Metal contaminated sediments were collected and submitted to acidification by means of CO2 injection or by adding HCl. Sediments elutriate were prepared to perform toxicity tests. The results showed that sediment acidification enhanced the release of metals to elutriates. Iron and zinc were the metals most influenced by this process and their concentration increased greatly with pH decreases. Diatom growth was inhibited by both processes: acidification and the presence of metals. Data obtained is this study is useful to calculate the potential risk of CCS activities to the marine environment.

In aquaculture, disinfection of facilities, prevention of fish diseases, and stimulation of fish growth are priority goals and the most important sources of toxic substances to the environment, together with excretory products from fish. In the present study, embryos of two species of sea urchin (Paracentrotus lividus and Arbacia lixula) were exposed to serial dilutions of six antibiotics (amoxicillin (AMOX), ampicillin, flumequine (FLU), oxytetracycline (OTC), streptomycin (ST), and sulfadiazine [SFD]) and two disinfectants (sodium hypochlorite (NaClO) and formaldehyde [CH(2)O]). Alterations in larval development were studied, and the effective concentrations (ECs) were calculated to evaluate the toxicity of the substances. Both species showed similar sensitivities to all substances tested. Disinfectants (EC(50) = 1.78 and 1.79 mg/l for CH(2)O; EC(50) = 10.15 and 11.1 mg/l for NaClO) were found to be more toxic than antibiotics. AMOX, OTC, and ST caused <20 % of alterations, even at the highest concentrations tested. FLU was the most toxic to P. lividus (EC(50) = 31.0 mg/l) and SFD to A. lixula (EC(50) = 12.7 mg/l). The sea urchin bioassay should be considered within toxicity assessment-monitoring plans because of the sensitivity of larvae to disinfectants.

Environmental monitoring plans (EMP) that include chemical analysis of water, a battery of bioassays and the study of local hydrodynamic conditions are required for land-based marine aquaculture. In this study, the following standardized toxicity tests were performed to assess the toxicity of effluents from eight land-base marine fish farms (LBMFFs) located on the northwest coast of Spain: bacterial bioluminescence (with Vibrio fischeri at 15 and 30 min), microalgal growth (with Phaeodactyllum tricornutum and Isochrysis galbana) and sea urchin larval development (with Paracentrotus lividus and Arbacia lixula). These bioassays were evaluated for inclusion in routine fish farm monitoring. Effective concentrations (EC(5), EC(10), EC(20), EC(50)) for each bioassay were calculated from dose-response curves, obtained by fitting the bioassay results to the best parametric model. Moreover, a graphical method of integrating the results from the battery of bioassays and classifying the toxicity was proposed, and the potential ecotoxic effects probe (PEEP) index was calculated. The bacterial bioluminiscence test at 30min, growth of I. galbana and larval development of A. lixula were found to be the most sensitive and useful tests. Graphical integration of these test results enabled definition of the ecotoxicological profiles of the different farms. The PEEP index, considering EC(20), efficiently reflected the toxic loading potential of LBMFF effluents. In conclusion, a battery of bioassays with species from different low trophic levels is recommended as a rapid and cost-effective methodology for assessing LBMFF discharges. The graphical integration method and the PEEP index are proposed for consideration in EMPs for such farms.
