RESUMO

Although studies on non-tuberculous mycobacteria have increased in recent years because they cause a considerable proportion of infections, their cellulolytic system is still poorly studied. This study presents a characterization of the cellulolytic activities of environmental mycobacterial isolates derived from soil and water samples from the central region of Argentina, aimed to evaluate the conservation of the mechanism for the degradation of cellulose in this group of bacteria. The molecular and genomic identification revealed identity with Mycolicibacterium septicum. The endoglucanase and total cellulase activities were assessed both qualitatively and quantitatively and the optimal enzymatic conditions were characterized. A specific protein of around 56 kDa with cellulolytic activity was detected in a zymogram. Protein sequences possibly arising from a cellulase were identified by mass spectrometry-based shotgun proteomics. Results showed that M. septicum encodes for cellulose- and hemicellulose-related degrading enzymes, including at least an active ß-1,4 endoglucanase enzyme that could be useful to improve its survival in the environment. Given the important health issues related to mycobacteria, the results of the present study may contribute to the knowledge of their cellulolytic system, which could be important for their ability to survive in many different types of environments.

Understanding the relationships between structure and properties of aluminosilicate glasses is of interest in magmatic studies as well as for glass applications as mechanical or optical components. Glass properties may be tailored by the incorporation of additional elements, and here we studied the effect of phosphate incorporation on refractive index and the degree of ionic bonding in aluminosilicate glasses. The studied glasses in the system SiO2-Al2O3-Na2O-P2O5 had a metaluminous composition (Al:Na = 1) with the content of SiO2 ranging from 50 to 70 mol% and of P2O5 from 0 to 7.5 mol%. Refractive index was measured at four wavelengths from visible to near-infrared and found to decrease both with increasing P2O5 content (at the expense of NaAlO2) and with increasing SiO2 content (by substitution of SiO4 for AlO4 groups). This trend correlated with a decrease in density while, additionally, the formation of Al-O-P bonds with an SiO2-like structure may account for this change. The degree of ionic bonding, assessed via optical basicity and oxygen polarisability, decreased with increasing P2O5 and SiO2 content. Despite the complexity of the studied glasses, oxygen polarisability and optical basicity were found to follow Duffy's empirical equation for simple oxide glasses. In the high frequency infrared and Raman spectra, band shifts were observed with increasing P2O5 and SiO2 content. They indicated changing average bond strength of the glass network and showed a linear correlation with optical basicity.

Certain individuals have a disproportionate effect on group responses. Characteristics may include susceptibility to pollutants, such as cadmium (Cd), a potent trace metal. Here, we show how a pair of Cd-exposed individuals can impact the behavior of unexposed groups. We used behavioral assessments to characterize the extent of the effects of the Cd-exposed individuals on group boldness, cohesion, foraging, activity, and responses to plants. We found that groups with a pair of Cd-exposed fish remained closer to novel stimuli and plants than did groups with untreated (control) fish. The presence of plants reduced Cd-induced differences in shoal cohesion and delays feeding in male shoals. Shoals with Cd- and water-treated fish were equally active. The results suggest that fish acutely exposed to environmentally relevant Cd concentrations can have profound effects on the un-exposed majority. However, the presence of plants may mitigate the effects of contaminants on some aspects of social behavior.

Crystallisation of bioactive glasses has been claimed to negatively affect the ion release from bioactive glasses. Here, we compare ion release and mineralisation in Tris-HCl buffer solution for a series of glass-ceramics and their parent glasses in the system SiO2-CaO-P2O5-CaF2. Time-resolved X-ray diffraction analysis of glass-ceramic degradation, including quantification of crystal fractions by full pattern refinement, show that the glass-ceramics precipitated apatite faster than the corresponding glasses, in agreement with faster ion release from the glass-ceramics. Imaging by transmission electron microscopy and X-ray nano-computed tomography suggest that this accelerated degradation may be caused by the presence of nano-sized channels along the internal crystal/glassy matrix interfaces. In addition, the presence of crystalline fluorapatite in the glass-ceramics facilitated apatite nucleation and crystallisation during immersion. These results suggest that the popular view of bioactive glass crystallisation being a disadvantage for degradation, apatite formation and, subsequently, bioactivity may depend on the actual system study and, thus, has to be reconsidered.

Periodontal disease is a significant burden for oral health, causing progressive and irreversible damage to the support structure of the tooth. This complex structure, the periodontium, is composed of interconnected soft and mineralised tissues, posing a challenge for regenerative approaches. Materials combining silicon and lithium are widely studied in periodontal regeneration, as they stimulate bone repair via silicic acid release while providing regenerative stimuli through lithium activation of the Wnt/ß-catenin pathway. Yet, existing materials for combined lithium and silicon release have limited control over ion release amounts and kinetics. Porous silicon can provide controlled silicic acid release, inducing osteogenesis to support bone regeneration. Prelithiation, a strategy developed for battery technology, can introduce large, controllable amounts of lithium within porous silicon, but yields a highly reactive material, unsuitable for biomedicine. This work debuts a strategy to lithiate porous silicon nanowires (LipSiNs) which generates a biocompatible and bioresorbable material. LipSiNs incorporate lithium to between 1% and 40% of silicon content, releasing lithium and silicic acid in a tailorable fashion from days to weeks. LipSiNs combine osteogenic, cementogenic and Wnt/ß-catenin stimuli to regenerate bone, cementum and periodontal ligament fibres in a murine periodontal defect.

Some individuals have a disproportionate effect on group responses. These individuals may possess distinct attributes that differentiate them from others. These characteristics may include susceptibility to contaminant exposure such as cadmium, a potent trace metal present in water and food. Here, we tested whether a pair of cadmium-exposed individuals could exert an impact on the behavior of the unexposed majority. We used behavioral assessments to characterize the extent of the effects of the cadmium-exposed pair on group boldness, cohesion, activity and responses to landmarks. We found that groups with a pair of cadmium-exposed fish approached and remained closer to novel stimuli and landmarks than did groups with pairs of fish treated with uncontaminated water (control). Shoals with cadmium and water treated fish exhibited similar levels of cohesion and activity. The results suggest that fish acutely exposed to environmentally-relevant cadmium concentrations can have profound effects on the un-exposed majority.

To account for global contamination events, we must identify direct and indirect pollutant effects. Although pollutants can have direct effects on individuals, it is unknown how a few contaminated individuals affect groups, a widespread social organization. We show environmentally relevant levels of cadmium (Cd) can have indirect social effects revealed in the social context of a larger group. Cd-contaminated individuals had poor vision and more aggressive responses, but no other behavioral effects. The presence of experienced Cd-exposed pairs in the groups had an indirect effect on the un-exposed individual's social interactions leading to the shoal becoming bolder and moving closer to a novel object than control groups. Because a few directly affected individuals could indirectly affect social behavior of the un-exposed majority, we believe that such acute but potentially important heavy metal toxicity could inform reliable predictions about the consequences of their use in a changing world.

Cobalt-containing materials are of interest for a wide range of applications, from biomaterials to solid-state lasers in optics. For instance, Co2+ is known to trigger the formation of new blood vessels, i.e. angiogenesis. Here, the use of phosphate glasses as a vehicle for local release of Co2+ ions is an attractive strategy to overcome the vascularisation limitation in tissue engineering. This study aimed to establish structure-property correlations as a function of the coordination environment of cobalt in 45P2O5-(30 - x)CaO-25Na2O-xCoO (x: 0.01 to 10 mol%) glasses. Constant polymerization and O/P ratio, resulting ultimately in constant basicity, were shown by ICP-OES and Raman spectroscopy. The latter, combined with EPR analysis, indicated that Co2+ was the predominant oxidation state and the presence of Co3+ can be excluded. UV-vis-NIR absorption spectra showed that the ratio between Co2+ in four- and six-fold coordination remained constant throughout the glass series. Their thermal properties measured by DSC and heating microscopy did not change much in the substitution range studied here. The steady trend in Tg values suggests a compensation between two opposite effects caused by the presence of four and six-fold coordinated Co2+, both being present at a constant ratio throughout the glasses. Accordingly, the higher field strength of Co2+ compared to that of Ca2+ is expected to strengthen the glass network. In contrast, four-fold coordinated cobalt is expected to weaken the network by connecting fewer fragments of the phosphate glass network than six-fold coordinated cobalt. These results indicate that the structural properties of the glasses with constant basicity are influenced by the coordination number of Co2+.

Melt-derived metaluminous (Al/Na = 1) aluminosilicate glasses in the system SiO2-Al2O3-Na2O-P2O5 were prepared with P2O5 and SiO2 contents varying from 0 to 7.5 and 50 to 70 mol %, respectively. The glass structure was investigated by X-ray absorption near edge structure, far- and medium-infrared, and polarized Raman spectroscopic techniques. The results indicate the incorporation of phosphate into the aluminosilicate network not only as partially depolymerized groups but also as fully polymerized groups charge-balanced by aluminate units in Al-O-P bonds. A new analysis method based on polarized Raman spectra in the bending frequency range indicates a preference of phosphate to reorganize the smallest ring structures. Changes in the glass transition temperature with the increase in phosphate content were found to be consistent with the depolymerization of the network structure shown by spectroscopy. By contrast, increasing the silica content by substituting SiO4 for AlO4 tetrahedra, while keeping the phosphate content constant, was found to have a negligible effect on network polymerization. Still, the glass transition temperature decreased and correlated with a far-infrared sodium band shift to higher frequency. This was interpreted as local changes in bond strength caused by complex interactions between the different network formers and sodium ions.

Background: Behavioral weight loss programs often prescribe physical activity (PA) goals in terms of minutes/week of moderate-to-vigorous PA (MVPA) and steps/day. However, the impact of meeting each type of goal prescription on weight loss is unclear, particularly in digitally-based (eHealth/mHealth) programs. This secondary analysis of a randomized trial examined the effects of meeting steps-based and minutes-based goals on weight loss in an eHealth behavioral weight control program. Methods: Adults in the control arm received a 6-month online behavioral weight loss intervention with prescribed weekly goals for daily steps and minutes of MVPA. The number of weeks steps-based and minutes-based goals were met (≥100% and ≥75% thresholds) based on self-reported PA were examined as predictors of 6-month weight loss among those providing weight outcomes (n = 172; 81% of control arm) using a systems regression approach. Results: Participants (BMI 35.6 kg/m2; 90.1% female; 48.7 years of age) met weekly goals for MVPA (7.1 ± 6.4 weeks) more often than steps (3.5 ± 5.5 weeks, P < .001). Meeting the steps goals (ß = .24, P < .001) and MVPA goals (ß = .20, P < .001) were each statistically significant predictors of weight loss at the 100% threshold; their total effects were not statistically different from one another (χ 2 = 1.12, P = .29). Similarly, at the 75% threshold for steps goals (ß = .19, P < .001) and MVPA goals (ß = .19, P < .001), each independently predicted weight loss; no differences were detected in their total effects (χ2 = .01, P = .92). The probability of reaching ≥5% weight loss was comparable between meeting the steps goals and MVPA goals at both adherence thresholds. Conclusions: Greater attainment of PA goals prescribed as steps and minutes of MVPA independently contribute to similar weight loss outcomes in a 6-month online behavioral weight loss intervention. Future research should determine whether promoting adherence to combined steps-based and minutes-based goals produces better weight loss than utilizing either goal alone and identify strategies that improve adherence.

Background: Concurrent administration of vancomycin and piperacillin/tazobactam (VAN+PTZ) may increase the risk of acute kidney injury (AKI) in hospitalized patients. Comprehensive characterization of VAN+PTZ associated AKI and recovery patterns remains lacking in previous reports. Objective: To compare the incidence of AKI associated with VAN+PTZ compared to either cefepime (CEF) or meropenem (MER) with VAN in adult general ward patients. Methods: A multicenter, retrospective, propensity score cohort study was conducted in non-critically ill adult patients. Included patients were concurrently administered VAN+PTZ or VAN+CEF/MER. Patients developing AKI ≤48 hours following combination therapy were excluded. The primary endpoint was to compare the incidence of AKI between study groups. Multivariable Cox regression modeling in predicting AKI was also conducted. Results: A total of 3199 patients met inclusion criteria and were evaluated. The incidence of AKI in VAN+PTZ and VAN+CEF/MER groups were 16.4% and 8.7%, respectively (P < .001). The onset to AKI was 1.8 days earlier with VAN+PTZ compared to VAN+CEF/MER (P < .001). Multivariable prediction model showed concomitant VAN+PTZ was identified as an independent risk factor of developing AKI (HR 2.34, 1.82-3.01, P < .001). The VAN+PTZ group experienced significantly higher rates of severe AKI (stage II or III) compared to the VAN+CEF/MER group (P = .002). No differences in the AKI recovery patterns were found between study groups. Conclusions: Concomitant VAN+PTZ in adult general ward patients was independently associated with an increased risk of AKI overall. More severe AKI was also associated with VAN+PTZ.

Healthy democratic polities feature competing visions of a good society but also require some level of cooperation and institutional trust. Democracy is at risk when citizens become so polarized that an 'us versus them' mentality dominates. Despite a vast multidisciplinary literature, no coherent conceptual framework of the microlevel dynamics that increase or decrease polarization has been presented. In this Review, we provide a conceptual framework to integrate scientific knowledge about cognitive-motivational mechanisms that influence political polarization and the social-communicative contexts in which they are enacted. Ego-justifying and group-justifying motives lead individuals to defend their own pre-existing beliefs and those of their in-group, respectively. However, a distinct class of system-justifying motives contributes to asymmetric forms of polarization. Whereas conservative-rightist ideology is associated with valuing tradition, social order and maintenance of the status quo, liberal-leftist ideology is associated with a push for egalitarian social change. These cognitive-motivational mechanisms interact with social influence processes linked to communication source, message and channel factors, all of which might contribute to increased or decreased polarization. We conclude with a discussion of unanswered questions and ways in which our framework can be extended to the study of culture and institutions.

Objective: Weight control programs that incorporate group sessions produce greater weight losses, but this has not been explored in the context of online programs. Further, counselor-crafted self-monitoring feedback is a core element of lifestyle interventions, although pre-scripted, modular feedback which does not require detailed counselor review may adequately promote weight loss. The current study explored the weight losses achieved in an online program that included facilitated group sessions, as well as outcomes when counselor-crafted self-monitoring feedback was provided. Methods: A 2 × 2 pilot factorial randomized participants (90% women) with overweight/obesity (N = 73) to facilitated group sessions (yes/no) and type of feedback (counselor-crafted/pre-scripted, modular) within a 16-week online behavioral weight control program. Weight change outcomes were collected digitally. Treatment engagement and intervention delivery time were also tracked. Results: Individuals offered weekly facilitated online group sessions lost more weight (-5.3% ± 4.9%) than those receiving the same digital program without group sessions (-3.1% ± 4.0%; p = 0.04). Those receiving group sessions also demonstrated significantly greater treatment engagement. Individuals receiving pre-scripted, modular feedback lost significantly more weight (-5.3% ± 4.8%) than those receiving the more traditional counselor-crafted feedback (-3.1% ± 4.1%; p = 0.04), but treatment engagement did not differ between conditions. However, interventionist time required to provide feedback was markedly lower for pre-scripted than counselor-crafted feedback (1.4 vs. 3.5 h per participant over 16 weeks, respectively, p = 0.01). Conclusions: Incorporating weekly facilitated online group sessions significantly increased weight losses achieved in a digital lifestyle program. Further, pre-scripted, modular feedback required significantly less staff time than counselor-crafted feedback without diminishing weight losses. Thus, group sessions and pre-scripted feedback warrant consideration when designing digital lifestyle programs.

Social context may influence the perception of sensory cues and the ability to display refined behavioral responses. Previous work suggests that effective responses to environmental cues can be contingent on having a sufficient number of individuals in a group. Thus, the changes in group size may have profound impacts, particularly on the behavior of small social groups. Using zebrafish (Danio rerio), here we examined how changes in group size influence the ability to respond to changes in water flow. We found that fish in relatively larger groups displayed stronger rheotaxis even when comparing pairs of fish with groups of four fish, indicating that a small increase in group size can enhance the responsiveness to environmental change. Individual fish in relatively larger groups also spent less time in the energetically costly leading position compared to individuals in pairs, indicating that even a small increase in group size may provide energetic benefits. We also found that the shoal cohesion was dependent on the size of the group but within a given group size, shoal cohesion did not vary with flow rate. Our study highlights that even a small change in group size could significantly affect the way social fish respond to the changes in water flow, which could be an important attribute that shapes the resilience of social animals in changing environments.

PURPOSE: Financial incentives are a promising approach to enhance weight loss outcomes; however, little guidance exists on the optimal incentive structure. DESIGN: Mixed methods. SETTING: An online weight management trial, combining outcome (i.e., weight loss) and behavioral (i.e., self-weighing, dietary self-monitoring, and steps) incentives over 12 months (up to $665). SUBJECTS: 116 participants who completed the incentive preference assessment at the 18-month follow-up visit. METHOD: Response distributions on the form, magnitude, certainty, and target of the incentives and content analysis of the qualitative responses. RESULTS: Nearly all (96.6%) participants indicated they liked receiving electronic Amazon gift cards, more so than the alternatives presented. Most participants (81.0%) thought they would have lost a similar amount of weight if the incentives were smaller. Few (18.1%) indicated they would have preferred a lottery structure, but 50.8% indicated the variable incentive schedule was beneficial during the maintenance period. Most (77.6%) felt incentives were most helpful when starting to lose weight. In both phases, most participants (85.3% and 72.4%, respectively) indicated appropriate behaviors were incentivized. Participants had mixed views on whether outcome or behavioral incentives were most motivating. CONCLUSION: There was notable variation in preferences for the magnitude, duration, and timing of incentives; it will be important to examine in future research whether incentive design should be tailored to individual preferences.

PURPOSE: The risk of acute kidney injury (AKI) associated with concomitant vancomycin and piperacillin/tazobactam in the intensive care unit (ICU) remains controversial. The aim of this study was to compare the AKI incidence associated with concomitant vancomycin and piperacillin/tazobactam compared to either cefepime or meropenem with vancomycin in the ICU. MATERIALS AND METHODS: A multicenter, retrospective, propensity score-matched cohort study was conducted in adult ICU patients administered vancomycin in combination with either piperacillin/tazobactam, cefepime, or meropenem were included. Patients developing AKI ≤48 h following combination therapy initiation were excluded. The primary endpoint was to compare the incidence of AKI associated with concomitant antimicrobial therapy. Multivariable Cox regression modeling in predicting AKI was also conducted. RESULTS: A total of 1044 patients were matched. The AKI incidence in vancomycin- piperacillin/tazobactam and vancomycin-cefepime/meropenem groups were 21.9% and 16.8%, respectively (p = 0.068). Multivariable prediction models showed concomitant vancomycin-piperacillin/tazobactam was an independent risk factor of AKI using serum creatinine only (HR 1.52, 1.10-2.10, p = 0.011) and serum creatinine with urine output-based KDIGO criteria (HR 1.77, 1.18-2.67, p = 0.006). No significant differences between groups were observed for AKI recovery patterns or mortality. CONCLUSION: Concomitant vancomycin and piperacillin/tazobactam administration in adult ICU patients was independently associated with an increased risk of AKI.

OBJECTIVE: This study examined the impact of a financial incentive scheme integrating process and outcome incentives across weight-loss induction and weight maintenance on 18-month weight outcomes. METHODS: This was a randomized controlled trial. Participants with overweight or obesity (n = 418; 91% female; 28% racial/ethnic minority) were randomized to an 18-month, online, group-based behavioral weight-control program (Internet-Only) or the same program with financial incentives provided for 12 months, contingent on self-regulatory weight-control behaviors (self-weighing, dietary self-monitoring, and physical activity) and weight-outcome benchmarks (Internet+Incentives). No financial incentives were provided from Months 13 to 18 to examine the durability of weight-control behaviors and outcomes without incentives. RESULTS: Weight-loss induction at Month 6 was significantly greater for Internet+Incentives than Internet-Only (6.8% vs. 4.9%, respectively, p = 0.01). Individuals receiving incentives were significantly more likely to maintain weight loss ≥ 5% at Month 12 (45% in Internet+Incentives vs. 32% in Internet-Only, p < 0.02) and remain weight stable (39% vs. 27%, respectively, p < 0.01). Internet+Incentives participants also reported significantly greater behavioral engagement through Month 12. However, once incentives ceased, there were no differences in sustained weight outcomes (Month 18), and engagement declined dramatically. CONCLUSIONS: Despite promoting greater treatment engagement and initial weight loss, financial incentives as offered in this study did not promote better extended weight control.

RESUMO

Weaving the future of the field of comparative psychology is dependent on the career advancement of early-career scientists. Despite concerted efforts to increase diversity in science, technology, engineering, and mathematics, scholars from marginalized groups are disproportionately underrepresented in the field-especially at advanced career stages. New approaches to sponsorship, mentoring, and community building are necessary to retain talent from marginalized communities and to create a culture and a system where all individuals can thrive. We describe the unique and supportive role of senior women scientists united through a professional society in initiating peer coaching circles to facilitate the success of a diverse cohort of early-career women scientists. We offer our experiences with the Weaving the Future of Animal Behavior program as a case study that illustrates the cascading impacts of professional societies investing in the success and career development of marginalized scholars. We focus on our peer coaching circle experience and share the products and outcomes after 2 years of meeting. Peer coaching transformed us from a group of loosely organized, anxious individuals into a collective of empowered agents of change with an enhanced sense of belonging. We end by presenting recommendations to institutions seeking to expand the landscape of opportunities to other marginalized scholars. (PsycInfo Database Record (c) 2021 APA, all rights reserved).

Physical activity (PA) goal adherence is consistently associated with greater weight loss during behavioral obesity treatment, and early weight loss response predicts future weight loss success. However, it remains unclear which behaviors during the initial weeks of treatment distinguish responders from nonresponders and might be effective targets for improving treatment outcomes. To characterize subgroups with distinct patterns of PA goal adherence during the initial 2 months of an online, group-based weight control program and determine associations between these patterns and 6-month weight loss. Participants received an online behavioral obesity intervention with PA goals and daily self-monitoring. Weekly adherence to step goals and moderate-to-vigorous PA (MVPA) minute goals based on self-monitoring records were examined using latent class analysis. Body weight was objectively measured at 0, 2, and 6 months. Participants (N = 212; 91.5% female, 31.6% race/ethnic minority, mean body mass index: 35.8 ± 5.9 kg/m2) clustered into three subgroups based on early goal attainment: "Both PA Goals," "MVPA Goals Only," and "Neither PA Goal." The "Both PA Goals" class had significantly greater 6-month weight loss (estimated mean weight loss [95% CI]: -9.4% [7.4 to 11.5]) compared to the "MVPA Goals Only" (-4.8% [3.4 to 6.1]) and "Neither PA Goal" classes (-2.5% [1.4 to 3.6]). Individuals meeting both PA goals early in treatment achieve greater weight losses than those meeting MVPA but not step goals, pointing to the need to explore factors associated with nonadherence to each of the PA goals to better understand these potential targets for treatment refinement and adaptive interventions.


This study is the first to characterize subgroups of individuals engaged in a behavioral weight control program with distinct patterns of early physical activity (PA) goal attainment. These early PA patterns emerged as a novel factor associated with subsequent weight loss and provide an important lens to view early treatment engagement. The greatest weight losses were seen in the subgroup likely to meet program goals for both weekly minutes of moderate-to-vigorous PA and daily steps. Understanding the factors associated with PA goal attainment during the initial 2 months of a behavioral weight control program may provide insights that will allow early identification of likely treatment success and detect individuals at risk for reduced weight losses, which could signal individuals for whom additional or different support may then be directed to increase weight loss success.

Bioactive glasses convert to a biomimetic apatite when in contact with physiological solutions; however, the number and type of phases precipitating depends on glass composition and reactivity. This process is typically followed by X-ray diffraction and infrared spectroscopy. Here, we visualise surface mineralisation in a series of sodium-free bioactive glasses, using transmission electron microscopy (TEM) with energy-dispersive X-ray spectroscopy (EDXS) and X-ray nano-computed tomography (nano-CT). In the glasses, the phosphate content was increased while adding stoichiometric amounts of calcium to maintain phosphate in an orthophosphate environment in the glass. Calcium fluoride was added to keep the melting temperature low. TEM brought to light the presence of phosphate clustering and nearly crystalline calcium fluoride environments in the glasses. A combination of analytical methods, including solid-state NMR, shows how with increasing phosphate content in the glass, precipitation of calcium fluoride during immersion is superseded by fluorapatite precipitation. Nano-CT gives insight into bioactive glass particle morphology after immersion, while TEM illustrates how compositional changes in the glass affect microstructure at a sub-micron to nanometre-level.
