RESUMO

Photoinduced changes of human ceruloplasmin (hCp) under laser and ultraviolet (UV) radiation have been studied. A complex character of hCp spectral modifications points to a different degree of the protein molecule folding. After the influence of UV radiation (240-390 nm) in a dose range 906-4530 J/m2 the decrease of oxidase hCp activity was registered. These changes are thought to be connected with modification of the conformation of hCp active centre or copper reduction that was confirmed by the decrease of optical density at 610 nm. An inactivation constant (kf = 3.8 x 10(-4) 1/s) and a photoinactivation quantum yield (1.4 x 10(-4)) were calculated. Kinetic dependencies of o-phenilen-diamine oxidation by intact and UV-modified glycoprotein do not correspond to Michaelis-Menten equation. UV radiation in a dose range 151-4530 J/m2 induced increasing of ceruloplasmin superoxiddismutase activity. The influence of He-Ne laser radiation (1.3 mW; lambda = 632.8 nm, exposure time from 1 up to 30 min) leads to the raise of the hCp functional activity; superoxiddismutase activity changes more significantly. Possible photophysical and photochemical processes in hCp molecule, leading to photomodification and changes of protein functional activity have been represented on the scheme.

RESUMO

The content of medium molecular weight (MMW) substances in the blood and urine was studied in patients with chronic dermatoses (psoriasis and atopic dermatitis). Intoxication was detected in all these patients, which was confirmed by clinical symptoms and a high level of plasma MMW substances, indicating pronounced catabolic processes in this condition. The distribution of MMW substances in biological media and the absence of positive changes after treatment in patients with atopic dermatitis in comparison with psoriasis patients may be indicative of different mechanisms of biochemical homeostasis and different adaptation potential, which should be taken into consideration when prescribing correction.

RESUMO

A new enzyme immunoassay EIA-HCV-Spectr test system constructed on the base of recombinant proteins and synthetic peptides allows separate detection of antibodies to E1/E2, core, HS3, NS4, and NS5 antigens of hepatitis C virus (HCV). The system is highly specific and more sensitive than the test systems used in screening studies, which allows its use as a final test for antiHCV antibodies. Antibodies to various HCV antigens were analyzed using this test system in patients with acute and chronic hepatitis C and asymptomatic donors with antiHCV. In acute hepatitis C during the first-second week after clinical manifestation, antibodies to nonstructural virus proteins are detected 3-4 times less often than in chronic hepatitis C. Acute hepatitis C is characterized by the presence of antibodies only to core antigen (66%). In chronic condition combinations of antibodies to structural and nonstructural HCV antigens predominate: core + NS4, core + NS3 + NS4, core + NS3 + NS5, core + NS4 + NS5, and core + NS3 + NS4 + NS5. In asymptomatic donors with antiHCV and in patients with chronic hepatitis C the spectra of antibodies were similar in 45.7% cases.

RESUMO

A number of enzyme preparations of microbial origin such as immunomodulators and immunostimulators have been studied in vitro according to reactions of rosette technique of cell (ROC) active and general, adhesion, activation of a complement and also the spontaneous activity and phagocytosis of neutrophils have been determined. The enzyme preparations of microbial origin have been shown to be capable of causing immunomodulation and of influencing the specific and nonspecific immune response. Unlike the enzymes of animal origin the enzymes of microbial origin are effective in smaller concentrations. The studied preparations modulate the immune response differently according to ROC, adhesion. Each of preparations has specific features of biological activity--bacterio-stationary in reaction to a certain group of microorganisms and some specific features of reaction with cell and levels of immune reactions.

RESUMO

Estimation and analysis of correlations permit disclosing principally new mechanisms in assessment of the homeostasis. The authors have derived curvilinear correlations between medium-molecular peptides, immune complexes, and lipids in coronary patients with the acute forms of the condition. Close relationships (high correlations) between homeostasis parameters were revealed, that were found more manifest in the patients with unstable angina pectoris. The immune complexes and lipids are regarded as the principal signs, and the medium-molecular peptides are functionally related to these. A lower correlation between the parameters and change of the argument/functional relationships in the patients with myocardial infarction are characteristic of essential imbalance of the homeostasis system.

RESUMO

Dynamic alterations and interrelationships of the patterns of humoral immunity (immunoglobulins and immune complexes) were studied in 77 patients with myocardium infarction and in 23 healthy donors. Stimulation of immunity homeostasis in the patients with acute form of myocardium infarction was manifested as an increase in content of immunoglobulins and immune complexes. Content of immunoglobulin G was distinctly decreased in epistenocardic pericarditis. High rate of correlations was found between the patterns studied in healthy persons (r = 0.7 = 0.85). During the acute period of myocardium infarction disbalance of homeostasis was observed, which was expressed as a decrease in the correlation coefficient down to r = 0.41 = 0.54. An increase in the interrelation and redistribution of the argument-functional parameters were detected in dynamics of the disease.

RESUMO

Effect of lysozyme on some immunochemical reactions was studied, involving rosette-developing properties of T lymphocytes and lymphocyte adhesion. The rosette-forming activity of the cells was shown to increase after addition of lysozyme in vitro as well as after treatment with lysozyme in vivo. Participation of lysozyme in activation of complement via various pathways is considered.

Content of sialic acids was increased 2.9-3.4-fold in soluble fraction of pathologically altered skin (parakeratotoic scales in psoriasis and allergo dermatoses) as compared with a skin of healthy persons. In parakeratotic scales 7-11 zones of PAS (positive glycoproteins) were found by polyacrylamide gel disc electrophoresis; they occurred in skin of healthy persons only in several cases. Hemoglobin-binding activity (haptoglobin) was observed in pathologically altered skin of 64-93% patients with psoriasis; it was not found in healthy persons. The spectrum of haptoglobin fractions in skin correlated with its phenotype in blood.
