RESUMO

Antibiotic dry cow therapy (DCT) is an important part of most mastitis control programs. Updating DCT recommendations is an ongoing topic due to the global problem of antimicrobial resistance. Finland, along with other Nordic countries, has implemented selective DCT for decades. Our study analyzed Dairy Herd Improvement (DHI) information from 241 Finnish farmers who participated in a survey about their drying-off practices. The aim was to evaluate herd-level associations between milk somatic cell count (SCC), milk production, and various antimicrobial DCT approaches both cross-sectionally in 2016 and longitudinally in 2012-2016. The three DCT approaches in the study were selective, blanket, and no DCT use. An additional aim was to evaluate whether dynamic changes occurred in herd-average SCC and annual milk production over five years, and whether these potential changes differed between different DCT approaches. The method for the longitudinal analyses was growth modeling with random coefficient models. Differences in SCC and milk production between farms with different DCT approaches were minor. Regardless of the farm's DCT approach, annual milk production increased over the years, while average SCC was reasonably constant. The variability in SCC and milk production across all DCT groups was low between years, and most of the variability was between farms. Compared to other milking systems, farms with automatic milking system (AMS) had higher SCC, and in 2016 higher milk production. The results of this study suggest that it is possible to maintain low herd-average SCC and good milk production when using selective DCT and following the guidelines for prudent antimicrobial use. Average SCC and milk production varied across the herds, suggesting that advice on DCT practices should be herd-specific. The methodology of growth modeling using random coefficient models was applicable in analyzing longitudinal data, in which the time frame was relatively short and the number of herds was limited.

Mortality of broilers during transport and lairage before slaughter represents an economic loss to the poultry industry and a welfare issue that needs to be addressed. In Canada, broilers can be transported long distances and be exposed to environmental factors, such as cold temperatures, that can affect the percentage of dead-on-arrivals or DOAs. Slaughter plant records for loads transported over a 19-month period in 2009-2010 were examined to identify factors affecting mortality risk (% DOA) during transportation from the rearing barn to the slaughter plant. Information from 2007 loads was analysed using a multilevel linear model. Most of the variation in the mortality risk occurred at the load level rather than at the producer or barn level. There were significant effects of bird sex, age and weight, catching team, journey duration and holding barn duration on mortality risk. The following environmental risk factors increased mortality risk: cold temperatures during the journey and in the holding barn, low crate stocking density during journeys at cold temperature and increased trailer temperature when in the holding barn. The analyses identified risk factors that can be used to refine management practices to mitigate some of the mortality risk. Increased mortality in some loads was associated with environmental conditions that caused deaths from hypothermia, whereas in other circumstances, some deaths likely occurred from hyperthermia. The climatic conditions in Atlantic Canada were responsible for significant risk factors that affected the mortality risk. Although refinement of the management practices described in this study, such as reduced journey and lairage durations, and increased crate stocking density can mitigate some of the mortality risk, the extreme cold conditions experienced during some parts of the year exceeded the capacity of the systems of transportation used to provide environmental conditions that minimise mortality.

OBJECTIVE: To develop an extension of the STROBE (Strengthening the Reporting of Observational Studies in Epidemiology) statement that addresses unique reporting requirements for observational studies in veterinary medicine related to health, production, welfare, and food safety. DESIGN: Consensus meeting of experts. SETTING: Mississauga, Canada. PARTICIPANTS: Seventeen experts from North America, Europe, and Australia. METHODS: Experts completed a pre-meeting survey about whether items in the STROBE statement should be modified or added to address unique issues related to observational studies in animal species with health, production, welfare, or food safety outcomes. During the meeting, each STROBE item was discussed to determine whether or not rewording was recommended and whether additions were warranted. Anonymous voting was used to determine consensus. RESULTS: Six items required no modifications or additions. Modifications or additions were made to the STROBE items 1 (title and abstract), 3 (objectives), 5 (setting), 6 (participants), 7 (variables), 8 (data sources/measurement), 9 (bias), 10 (study size), 12 (statistical methods), 13 (participants), 14 (descriptive data), 15 (outcome data), 16 (main results), 17 (other analyses), 19 (limitations), and 22 (funding). CONCLUSION: The methods and processes used were similar to those used for other extensions of the STROBE statement. The use of this STROBE statement extension should improve reporting of observational studies in veterinary research by recognizing unique features of observational studies involving food-producing and companion animals, products of animal origin, aquaculture, and wildlife.

The STROBE (Strengthening the Reporting of Observational Studies in Epidemiology) statement was first published in 2007 and again in 2014. The purpose of the original STROBE was to provide guidance for authors, reviewers, and editors to improve the comprehensiveness of reporting; however, STROBE has a unique focus on observational studies. Although much of the guidance provided by the original STROBE document is directly applicable, it was deemed useful to map those statements to veterinary concepts, provide veterinary examples, and highlight unique aspects of reporting in veterinary observational studies. The reporting of observational studies in veterinary research presents many challenges that often are not adequately addressed in published reporting guidelines. A consensus meeting of experts was organized to develop an extension of the STROBE statement to address observational studies in veterinary medicine with respect to animal health, animal production, animal welfare and food safety outcomes. The consensus meeting was held 11-13 May 2014 in Mississauga, Ontario, Canada. Seventeen experts from North America, Europe and Australia attended the meeting. The experts were epidemiologists and biostatisticians, many of whom hold or have held editorial positions with relevant journals. Prior to the meeting, 19 experts completed a survey about whether they felt any of the 22 items of the STROBE statement should be modified and whether items should be added to address unique issues related to observational studies in animal species with health, production, welfare or food safety outcomes. At the meeting, the participants were provided with the survey responses and relevant literature concerning the reporting of veterinary observational studies. During the meeting, each STROBE item was discussed to determine whether or not re-wording was recommended, and whether additions were warranted. Anonymous voting was used to determine whether there was consensus for each item change or addition. The consensus was that six items needed no modifications or additions. Modifications or additions were made to the STROBE items numbered as follows: 1 (title and abstract), 3 (objectives), 5 (setting), 6 (participants), 7 (variables), 8 (data sources/measurement), 9 (bias), 10 (study size), 12 (statistical methods), 13 (participants), 14 (descriptive data), 15 (outcome data), 16 (main results), 17 (other analyses), 19 (limitations) and 22 (funding). Badgers are a wildlife host of Mycobacterium bovis, the causative agent of bovine tuberculosis (bTB), and an important contributor to the epidemiology of bTB in cattle in Ireland and Britain. Repeated culling of badgers in high prevalence cattle bTB areas has been used in the Republic of Ireland as one tool to reduce intra- and interspecific transmission of M. bovis. We assessed factors that influenced infection prevalence of culled badgers from 2009 to 2012 (n=4948) where spatial, temporal and intrinsic factor data were available using multivariable modelling. Prevalence appeared higher in western areas than eastern areas of Ireland and badgers were more likely to be test-positive if caught at a sett (burrow system) which was close to other infected setts (spatial clustering of infection). There was a significant positive association between badger test-status and cattle prevalence of M. bovis infection at a spatial scale of 1km around setts. Badgers were more likely to be deemed test positive if they were male (OR: 1.9) or a parous female (OR: 1.7), compared to a female who had never conceived. Our results are consistent with different groups within badger populations having differential exposures and therefore infection risk (for example, parous vs. non-parous females). Furthermore, bTB clusters within the badger population, with greater risk to badgers in setts that are closest to other infected setts. The effective scale of the association of bTB risk between badger and cattle populations may be relatively large in Ireland. Our data indicate that the overall trend in prevalence of M. bovis infection in badgers has decreased in Ireland (P<0.001) while controlling for significant confounders over the study period, and follows a longer temporal trend from 2007 to 2013, where unadjusted apparent prevalence declined from 26% to 11% during 2007 to mid-2011, followed by a stable trend between 9 and 11% thereafter (n=10,267).

Compared with blanket dry cow therapy (DCT), the selective antimicrobial treatment of cows based upon on-farm culture results has the potential to reduce the amount of antimicrobials used in dairy production. The objective of the current study was to determine the effect of a Petrifilm (3M Canada, London, Ontario) on-farm culture-based selective DCT program on milk yield and somatic cell count (SCC) in the following lactation. A total of 729 low-SCC (<200,000 cells/mL) cows from 16 commercial dairy herds with a low bulk tank SCC (<250,000 cells/mL) were randomly assigned to receive either blanket DCT or Petrifilm-based selective DCT. Cows belonging to the blanket DCT group were infused with a commercial DCT product and an internal teat sealant (ITS) at drying off. Using composite milk samples collected on the day before drying off, cows in the selective DCT group were treated at drying off based on the results obtained by the Petrifilm on-farm culture system with DCT and ITS (Petrifilm culture positive) or ITS alone (Petrifilm culture negative). Milk test-day records for the following lactation were obtained from Dairy Herd Improvement for all cows enrolled in the trial. Repeated measures linear mixed models were used to assess the effect of study group (blanket or selective DCT) on test-day milk production and natural logarithm of SCC over the first 180 d of the subsequent lactation. According to the final multivariable models, when low-SCC cows were selectively treated with DCT at drying off based on results obtained using the Petrifilm on-farm culture system, no effect on milk production (least squares means for blanket DCT = 39.3 kg vs. selective DCT = 39.0 kg) or natural logarithm of SCC (least squares means for blanket DCT = 3.95 vs. selective DCT = 3.97) was observed in the subsequent lactation when compared with cows receiving blanket DCT. The results of this study indicate that selective DCT based on results obtained by the Petrifilm on-farm culture system enabled a reduction in the use of DCT without negatively affecting milk production and milk quality.

Dairy cow mortality (unassisted death and euthanasia) has increased, worldwide and in Sweden. On-farm mortality indicates suboptimal herd health or welfare and causes financial loss for the dairy producer. The objective of this study was to identify cow-level risk factors associated with on-farm cow mortality. Cows with at least one calving between 1 July 2008 and 30 June 2009 from herds enrolled in the Swedish official milk recording scheme with >40 cow-years were included. Each cow was followed from the day of calving until she calved again or left the herd (died, slaughtered or sold). The effects of potential risk factors on on-farm cow mortality were analysed using a Weibull proportional hazard model with a gamma distributed frailty effect common to cows within herd. The event of interest (failure) was euthanasia or unassisted death. An observation was right censored if the cow was slaughtered, sold, calved again or had an on-going lactation at 500 days after calving. The lactations were split into seasons (January to April, May to August and September to December) and at 30 and 100 days in milk in order to evaluate seasonal effects and the effect of disease in different lactation stages. Primiparous and multiparous cows were analysed separately. The highest hazards for both primiparous and multiparous cows were found for traumatic events and diseases, both in the lactation stage in which the cow died and in the preceding stage. The hazard was higher in early lactation and lower in 2nd parity compared to higher parities. Increased age at first calving (for primiparous cows), calving between January and April, dystocia and stillbirth also increased the mortality hazard. Differences were also found between breeds, between milk production parameters at first test milking and between management types. The results from this study show the importance of good management and preventive health actions, especially around calving, to avoid mortality in dairy cows.

The objective of the study was to evaluate the utility of a Petrifilm-based on-farm culture system when used to make selective antimicrobial treatment decisions on low somatic cell count cows (<200,000 cells/mL) at drying off. A total of 729 cows from 16 commercial dairy herds with a low bulk tank somatic cell count (<250,000 cells/mL) were randomly assigned to receive either blanket dry cow therapy (DCT) or Petrifilm-based selective DCT. Cows belonging to the blanket DCT group were infused with a commercial dry cow antimicrobial product and an internal teat sealant (ITS) at drying off. Using composite milk samples collected on the day before drying off, cows in the selective DCT group were treated at drying off based on the results obtained by the Petrifilm on-farm culture system with DCT + ITS (Petrifilm culture positive), or ITS alone (Petrifilm culture negative). Quarters of all cows were sampled for standard laboratory bacteriology on the day before drying off, at 3 to 4d in milk (DIM), at 5 to 18 DIM, and from the first case of clinical mastitis occurring within 120 DIM. Multilevel logistic regression was used to assess the effect of study group (blanket or selective DCT) and resulting dry cow treatment (DCT + ITS, or ITS alone) on the risk of intramammary infection (IMI) at calving and the risk of a first case of clinical mastitis between calving and 120 DIM. According to univariable analysis, no difference was observed between study groups with respect to quarter-level cure risk and new IMI risk over the dry period. Likewise, the risk of IMI at calving and the risk of clinical mastitis in the first 120 DIM was not different between quarters belonging to cows in the blanket DCT group and quarters belonging to cows in the selective DCT group. The results of this study indicate that selective DCT based on results obtained by the Petrifilm on-farm culture system achieved the same level of success with respect to treatment and prevention of IMI over the dry period as blanket DCT and did not affect the risk of clinical mastitis in the first 120 d of the subsequent lactation.

Cow-level milk ELISA results can be used to determine herd Mycobacterium avium ssp. paratuberculosis (MAP) status. Milk sample collection is minimally invasive and ELISA results can be obtained quickly and economically. The objectives were to evaluate the herd-level test characteristics of 3 commercial milk ELISA, and to determine the impact of within-herd MAP prevalence on the performance of the milk ELISA herd test. A total of 32 purposively selected herds with a median herd size of 66 milking cows were used in this 2-yr project. Fecal and milk samples were collected from all milking cows at 6-mo intervals. Fecal samples were pooled by cow age, with 5 cow samples per pool; individual fecal culture was completed on cow samples from positive pools. Herd MAP status was defined as MAP positive if, at any point during the longitudinal study, a pooled fecal culture from the herd was positive. Milk samples were analyzed using each of 3 commercial milk ELISA kits; a cow-level result from each ELISA was classified as positive following the respective manufacturer's recommended threshold for a positive result. Herd-level milk ELISA test characteristics were estimated using generalized estimating equations logistic models, which accounted for repeated measurements. Using a cutoff of 2% milk ELISA-positive cows, milk ELISA herd sensitivity relative to a herd MAP status based on all pooled fecal culture results collected during the study was as follows: ELISA A: 59% [95% confidence interval (CI): 36-78%), ELISA B: 56% (95% CI: 32-77%), and ELISA C: 63% (95% CI: 41-81%). Herd specificity for ELISA A, B, and C was 80% (95% CI: 71-88%), 96% (95% CI: 89-98%), and 92% (95% CI: 86-96%), respectively. The remainder of the analyses focused on results from ELISA B. Herd sensitivity of ELISA B increased as MAP prevalence increased. In herds with a mean MAP prevalence ≤5%, the herd sensitivity of the milk ELISA was low, ranging from 11% when MAP prevalence was 1%, to 62% when MAP prevalence was 5%. Categorical likelihood ratios based on milk ELISA within-herd prevalence predicted that herds with milk ELISA prevalence above 0 but <2% had a similar likelihood to be MAP positive or MAP negative, whereas herds with a milk ELISA prevalence between 2 and 4% were 3.7 times more likely to be MAP positive than MAP negative. All herds with a milk ELISA prevalence >4% were MAP positive. Although milk ELISA B worked well to establish herd MAP status in high-prevalence herds, interpretation was unreliable in MAP-negative and low-prevalence herds.

The purpose of this study was to evaluate a 3M Petrifilm-based on-farm culture system for the detection of intramammary infection (IMI) in low somatic cell count (SCC) cows (<200,000 cells/mL) at drying off. The main objectives were to determine the test characteristics and the predictive values of the Petrifilm on-farm culture system. The ability of dairy producers to correctly classify cows as infected or uninfected based on Petrifilm culture and a set colony count threshold was also assessed. A total of 360 cows originating from 16 low bulk tank SCC (<250,000 cells/mL) dairy herds were enrolled at drying off. Enrolled cows had an expected dry period of 30-90 days, a SCC<200,000 cells/mL on the last 3 tests prior to drying off, no clinical mastitis in the same time period, and no antibiotic treatment in the last 14 days. Quarter milk samples were collected on the day prior to drying off, and a composite milk sample was created by combining 5 mL of milk from each quarter sample. Composite milk samples were cultured on-farm using the Petrifilm culture system, which provided results within 24h. Quarter milk samples were cultured in a reference laboratory, and the results were aggregated to the cow level. On the day of drying off, the Petrifilm was read by the producer and cows were classified as positive if ≥5 colonies (equivalent to 50 colony forming units/mL) were present. When read by the producer, 47.8% of the cows cultured negative on Petrifilm and were infused with only an internal teat sealant at drying off. The test characteristics of the Petrifilm on-farm culture system were calculated by comparing the producer-derived Petrifilm results to those obtained by standard laboratory culture. The sensitivity and specificity of the Petrifilm on-farm culture system were 85.2% (78.5-90.5) and 73.2% (66.4-79.3), respectively. The negative predictive value of the Petrifilm test system was high (86.6%) when estimated using the prevalence of IMI in this data set, and the positive predictive value was moderate (70.9%). An automated 3M Petrifilm reader was used to obtain accurate colony counts. The agreement between Petrifilm results obtained by the producer and those obtained by the automated Petrifilm reader was high, with a kappa value of 0.82 (0.75-0.89).

The objective of the study was to explore the relationship between herd dry period (DP) intramammary infection (IMI) incidence and elimination rates. A cohort of 91 Canadian dairy herds was recruited and followed in 2007 and 2008. Universal dry cow therapy was widely adopted among the participating herds. At the beginning of years 2007 and 2008, a sample of 15 cows was selected in each herd. A series of quarter-milk samples consisting of 2 predry and 2 postcalving samples were collected on each quarter of these 30 cows. Milk samples were analyzed using routine bacteriological milk culture, and predry and postcalving IMI statuses of these quarters were established using parallel interpretation of the 2 predry and 2 postcalving tests, respectively. Intramammary infection status was defined as IMI by Staphylococcus aureus, streptococci other than Streptococcus agalactiae, coagulase-negative staphylococci (CNS), or Corynebacterium spp. Incidence and elimination rates of DP IMI were computed for each herd. Lowess curves and linear regression were used to investigate the association between DP IMI incidence and elimination rates. Significant negative associations were found between incidence and elimination rates of Staph. aureus and CNS. The relationship between incidence and elimination rates was nonlinear for CNS, with a relatively strong negative relationship between DP IMI incidence and elimination rates for herds with relatively low DP IMI incidence (<0.10 new IMI/quarter-month). For herds with higher DP IMI incidence (≥ 0.10 new IMI/quarter-month), a weaker negative relationship was observed between rates. No significant associations could be seen between DP incidence and elimination rates of streptococci other than Strep. agalactiae and of Corynebacterium spp. These results suggest that, conversely to the general belief, acquisition of new IMI and elimination of existing IMI during the DP may be driven, at least for staphylococci, by common mechanisms. The use of management strategies that would lead to the selection of pathogens less well adapted to the host could explain the associations observed between acquisition and elimination of IMI over the DP.

The objective of the study was to evaluate the ability of an estimate of the herd intramammary infection (IMI) incidence rate computed using repeated somatic cell count (SCC) measurements (quarter- and composite-SCC; hereafter, the SCC-derived herd IMI incidence, SCCI)to predict the incidence rate computed using repeated quarter-milk bacteriological culture (hereafter, bacteriological culture incidence, BCI) during the lactating period. A cohort of 91 Canadian dairy herds was followed in 2007 and 2008. In each herd and at each of 4 sampling periods, a series of 3 to 7 quarter-milk samples was collected from a sample of 15 cows. Routine milk bacteriological culture was conducted to identify IMI, SCC was measured on the quarter-milk samples, and composite-SCC of the preceding and following dairy herd improvement (DHI) tests were obtained. Mastitis pathogens were grouped in 3 categories: major, minor, and any pathogens. For each herd and for each period, BCI was computed for each group of organisms. Similarly, SCCI were computed using quarter- and DHI composite-SCC and using a threshold of 200,000 cells/mL to define infected quarters or cows. A linear regression model taking into account the structure of the data was used to compare the SCCI to the BCI. A similar model was used to compare fluctuations (i.e., changes from one sampling period to the next) over time of the SCCI and BCI. Measures of correlation between observed and predicted rates were computed and limits of agreement plots sketched to better explore the predictive ability of the SCCI. The quarter-milk SCC measurements that could be obtained-for instance, using on-line milking system measurements-appeared to be particularly valuable. Quarter-SCCI showed a positive and significant association with the BCI. However, limits of agreement plots indicated important disagreement for the small proportion of observations with very high BCI. Quarter-level SCCI and BCI fluctuations were also significantly associated, and a substantial correlation (Spearman rho ranging from 0.54 to 0.58) could be seen between observed and predicted rates. Conversely, the predictive value of composite-DHI SCC for monitoring IMI incidence during the lactation seemed to be quite limited. Composite SCCI was strictly associated with major IMI BCI, showed a relatively low correlation with the observed rate (Spearman rho: 0.14), and was of little help for longitudinal monitoring of the IMI incidence.

Evidence in the literature exists to support the theory that mastitis and intramammary infection (IMI) tend to cluster within herds, within cows, and within quarters, facts which may have overarching ramifications on mastitis management in modern dairy herds. Most previous studies, however, have been carried out on prevalent IMI instead of new IMI (NIMI), although reducing incidence of NIMI is a major step toward controlling mastitis. The Canadian Bovine Mastitis Research Network (Saint-Hyacinthe, QC, Canada) has a large mastitis database derived from a 2-yr data collection on a national cohort of dairy farms, and data from this initiative were used to investigate the effect of clustering on the acquisition of NIMI. Longitudinal milk samplings of clinically normal udders taken over several 6-wk periods as well as samples from cows pre-dry-off and postcalving were used (n=73,772 quarter milk samples). Multilevel logistic models were used to evaluate the effect of location of IMI in quarters of the bovine udder previous to occurrence of an NIMI with Staphylococcus aureus, coagulase-negative staphylococci, and Corynebacterium spp. Several factors were investigated, including the number and location of quarters infected with the pathogen of interest before occurrence of an NIMI, the number of quarters infected with any other pathogen before occurrence of an NIMI (a measure of susceptibility), somatic cell count of the quarter before occurrence of an NIMI, somatic cell count of the other 3 quarters before occurrence of an NIMI, prevalence of the specific pathogen in the herd, and the average somatic cell count of the herd. The amount of variation occurring at different levels (herd, cow, and quarter) for the various pathogens was also calculated. The presence of an IMI in the ipsilateral quarter was associated with an elevated risk of an NIMI occurring for all pathogens investigated. Risk of an NIMI increased considerably as herd prevalence of the pathogen rose. Substantial clustering was found at all levels, with roughly equal amounts of variation found in all 3 levels for coagulase-negative staphylococci, most variation at the cow-level for Corynebacterium spp., and most variation found at the quarter-level for Staph. aureus. Simulation was used to calculate exact values of intraclass correlation coefficients to estimate clustering within cows and within quarters--these exact values were, for the most part, lower than estimates calculated using the latent variable approach, but also increased as pathogen prevalence and number of infections in a cow at the previous sampling increased. These results of these analyses can be used to inform approaches to preventing NIMI in modern dairy operations.

The objective of the study was to evaluate the predictive ability of the herd dry period (DP) intramammary infection (IMI) incidence and elimination rates derived from predry and postcalving somatic cell count (SCC) measurements [quarter-level SCC and dairy herd improvement (DHI) composite-level SCC] for monitoring the herd DP IMI incidence and elimination rates. Routine milk bacteriological culture was conducted to identify IMI, SCC was measured on the quarter milk samples, and composite SCC of the last predry and first postcalving DHI tests were obtained. Mastitis pathogens were grouped into 3 categories: major pathogens, minor pathogens, and any pathogens. For each herd, DP bacteriological culture-derived IMI incidence and elimination rates were computed using quarter milk culture data. Similarly, SCC-derived herd incidence and elimination rates were computed using quarter and DHI composite-level SCC measurements and using various SCC thresholds to define new and eliminated IMI. Linear regression was used to compare herd quarter-level and composite-level SCC-derived herd incidence and elimination with DP bacteriological culture-derived IMI incidence and elimination. Herd DP incidences computed by using quarter-level SCC, and with most of the SCC thresholds tested, were significant predictors of the DP major, minor, and any IMI incidences (F-test; P≤0.05). The highest coefficients of determination (R(2)) were obtained with thresholds of 200,000 (R(2): 12%) and 50,000 cells/mL (R(2): 25%) for predicting major and minor IMI, respectively. When using composite DHI SCC measurements, however, substantial losses of predictive power were seen for minor and any IMI incidences compared with quarter-level SCC. For DP major IMI incidence, composite SCC yielded similar, but modest, predictive power when a cutoff value of 150,000 cells/mL was chosen to define new IMI. To predict DP elimination rates, the value of quarter-level SCC seemed limited to predicting the DP major IMI elimination rate. Composite SCC, on the other hand, showed modest predictive power for major and minor IMI elimination rates, with thresholds of 200,000 and 50,000 cells/mL, respectively. Results from the current study suggest that quarter and composite SCC-derived rates could be used as substitutes for bacteriological culture-derived rates for some groups of mastitis pathogens.

Major mastitis pathogens such as Staphylococcus aureus, Streptococcus agalactiae, Streptococcus uberis, Streptococcus dysgalactiae, and the coliforms are usually considered more virulent and damaging to the udder than minor mastitis pathogens such as Corynebacterium bovis and coagulase-negative staphylococci (CNS). The current literature contains several studies detailing analyses with conflicting results as to whether intramammary infection (IMI) with the minor pathogens decreases, increases, or has no effect on the risk of a quarter acquiring a new intramammary infection (NIMI) with a major pathogen. To investigate the available scientific evidence regarding the effect of IMI with minor pathogens on the acquisition of NIMI with major pathogens, a systematic review and meta-analysis were conducted. The total extant English- and French-language literature in electronic databases was searched and all publications cited by relevant papers were investigated. Results from 68 studies were extracted from 38 relevant papers. Random-effects models were used to investigate the effects of CNS and C. bovis on acquisition of new IMI with any of the major pathogens, as well as individually for the minor pathogens and Staph. aureus. Significant heterogeneity among studies exists, some of which could be accounted for by using meta-regression. Overall, observational studies showed no effect, whereas challenge studies showed strong and significant protective effects, specifically when major pathogens were introduced into the mammary gland via methods bypassing the teat end. Underlying risk can account for several unmeasured factors, and studies with higher underlying risk found more protective effects of minor pathogens. Larger doses of challenge organisms reduced the protective effect of minor pathogens, and studies with more stringent diagnostic criteria for pathogen IMI identified less protection. Smaller studies (those utilizing fewer than 40 cows) also showed a greater protective effect than larger studies.

The current literature comprises several studies (n=38) detailing analyses with conflicting results as to whether intramammary infections (IMI) with the minor pathogens decrease, increase, or have no effect on the risk of a quarter acquiring a new IMI (NIMI) with a major pathogen. The Canadian Bovine Mastitis Research Network has a large mastitis database derived from a 2-yr data collection on a national cohort of dairy farms, and data from this initiative were used to further investigate the effect of IMI with minor pathogens on the acquisition of new major pathogen infections (defined as a culture-positive quarter sample in a quarter that had been free of that major pathogen in previous samples in the sampling period). Longitudinal milk samplings of clinically normal udders taken over several 6-wk periods as well as samples from cows pre-dry-off and postcalving were used to this end (n=80,397 quarter milk samples). The effects of CNS and Corynebacterium spp. on the major mastitis pathogens Staph. aureus, Strep. uberis, Strep. dysgalactiae, and coliform bacteria (Escherichia coli and Klebsiella spp.) were investigated using risk ratio analyses and multilevel logistic regression models. Quarter-, cow- and herd-level susceptibility parameters were also evaluated and were able to account for the increased susceptibility that exists within herds, cows and quarters, removing it from estimates for the effects of the minor pathogens. Increased quarter-level susceptibility was associated with increased risk of major pathogen NIMI for all pathogens except the coliforms. Increased somatic cell count was consistently associated with elevated risk of new major pathogen infections, but this was assumed to be a result of low sensitivity of bacteriology to diagnose major pathogen NIMI expediently and accurately. The presence of CNS in the sample 2 samplings before the occurrence of a NIMI increased the odds of experiencing a Staph. aureus NIMI 2.0 times, making the presence of CNS a risk factor for acquiring a Staph. aureus NIMI. Even with this extensive data set, power was insufficient to make a definitive statement about the effect of minor pathogen IMI on the acquisition of major pathogen NIMI. Definitively answering questions of this nature are likely to require an extremely large data set dedicated particularly to minor pathogen presence and NIMI with major pathogens.
