RESUMO

Turkey coryza is a major respiratory disease caused by Bordetella avium (B. avium). It occurs in all ages of turkeys and is characterized by high morbidity and low mortality rates. The present study aimed firstly at determination of the prevalence rates of B. avium in turkeys reared in Egypt at different ages using various diagnostic methods including clinical examination, histopathology, enzyme-linked immunosorbent assay (ELISA), bacterial culture, and polymerase chain reaction (PCR). Using PCR, virulence-associated genes were detected in the confirmed B. avium isolates. Furthermore, the antibiotic resistance profiles of the confirmed B. avium isolates were examined. The achieved results indicated isolation and identification of B. avium infection at different ages of turkeys reared in Egypt. The overall PCR-confirmed prevalence rate of B. avium was 22.95%. The identified B. avium strains harbored virulence-associated genes responsible for colonization in the respiratory tract of turkeys including Bordetella virulence gene (100%), fimbriae (71.14%), and filamentous hemagglutinin (85.68%). The isolated B. avium strains showed multidrug resistance profiles. B. avium isolates were resistant to penicillin (92.82%), ceftiofur (85.68%), nalidixic acid (78.54%), and lincomycin (71.40%). The identified B. avium strains showed clear sensitivities to both gentamicin and neomycin, suggesting these as possible antimicrobial candidates for the control of B. avium infection in turkeys.

The objective of this study was to identify potential mRNA expression changes in chicken livers associated with environmental exposure to dichloro-diphenyl-trichloroethane (DDT) and its metabolites (DDTs). In particular, we focused on genes relating to the immune system and metabolism. We analyzed liver samples from free-ranging chickens in KwaZulu-Natal, South Africa, for contamination by DDTs. This area predominantly uses DDT in its malaria control program, and homes are sprayed annually with the pesticide. Genes relating to the immune system and metabolism were selected as potential genetic biomarkers that could be linked to higher contamination with DDTs. RT-qPCR analysis on 39 samples showed strong correlations between DDTs contamination and mRNA expression for the following genes: AvBD1, AvBD2, AvBD6 and AvBD7 (down-regulated), and CYP17, ELOVL2 and SQLE (up-regulated). This study shows for the first time interesting and significant correlations between genetic material collected from environmentally-exposed chickens and mRNA expression of several genes involved in immunity and metabolism. These findings show the usefulness of analysis on field samples from a region with high levels of environmental contamination in detecting potential biomarkers of exposure. In particular, we observed clear effects from DDT contamination on mRNA expression of genes involved in immune suppression, endocrine-disrupting effects, and lipid dysregulation. These results are of interest in guiding future studies to further elucidate the pathways involved in and clinical importance of toxicity associated with DDT exposure from contaminated environments, to ascertain the health risk to livestock and any subsequent risks to food security for people.

Quail meat is an emerging source of high-quality animal protein. Quails are exposed to a wide range of xenobiotics such as heavy metals. In this study, residual concentrations of four toxic metals, of significant public health importance, including cadmium (Cd), lead (Pb), arsenic (As), and nickel (Ni), were determined in edible tissues of quails. In addition, metal loads were measured in water, feed, and litter samples collected from same quail farms as possible sources for quail exposure to heavy metals. The possible use of metallothionein (MT) and heat shock protein 70 (Hsp70) as molecular biomarkers of exposure to heavy metals was further investigated. Furthermore, the dietary intake and the potential risk assessment of the examined heavy metals among children and adults were calculated. The edible tissues of quails contained high concentrations of four heavy metals (contents (ppm/ww) ranging from 0.02 to 0.32 in Cd, 0.05 to 1.96 in Pb, 0.002 to 0.32 in As, and 1.17 to 3.94 in Ni), which corresponded to the high contents of these metals in the feeds, water, and litter. MT and Hsp70 mRNA expressions showed positive correlations with the concentrations of heavy metals in tissues indicating the possibility to use these proteins as biomarkers for quail's exposure to toxic metals. Dietary intake of quail meat and risk assessment revealed potential risks especially for children after prolonged exposure to the examined metals. Thus, legislations should be established and continuous screening of metal residues should be adopted in order to reduce the toxic metal concentrations in feeds and drinking water for quails. Reduction of exposure to heavy metals subsequently would lead to minimization of exposure of such toxicants through consumption of quail meat.
